目的

从调控低氧应激及线粒体功能角度探讨温经汤防治子宫内膜异位症(EMT)的作用机制。
方法

采用消化法原代培养EMT患者异位子宫内膜间质细胞(ESCs),实验设正常(Control)组、5%空白血清(5% KBXQ)组、10%空白血清(10% KBXQ)组、5%温经汤血清(5% WJTXQ)组、10%温经汤血清(10% WJTXQ)组。噻唑蓝(MTT)比色法检测不同组ESCs增殖情况,实时荧光定量聚合酶链式反应(Real-time PCR)及蛋白免疫印迹法(Western blot)检测缺氧诱导因子-1
α
(HIF-1
α
)mRNA及蛋白表达,透射电镜观察ESCs线粒体超微结构,高内涵分析(HCS)法多参数同时检测细胞线粒体功能[线粒体膜电位,线粒体膜电位(MMP)和细胞色素C(Cyt C)表达]及细胞凋亡情况(细胞膜通透性、细胞核荧光强度、细胞核大小及细胞数量),流式细胞术检测细胞凋亡率,Western blot检测B细胞淋巴瘤-2(Bcl-2)相关X蛋白(Bax),Bcl-2和剪切的半胱氨酸天冬氨酸蛋白水解酶-3(cleaved Caspase-3)蛋白表达。
结果

与Control组比较,5% KBXQ和10% KBXQ组细胞活力显著增加(

mRNA及蛋白表达水平无明显变化;透射电镜观察可见线粒体嵴明显,线粒体内外膜清晰;HCS多通道荧光染色显示,MMP及Cyt C表达、细胞膜通透性无显著变化,细胞核呈现均匀淡染;细胞凋亡率、cleaved Caspase-3蛋白表达及Bax/Bcl-2值均无明显变化。与Control组及相应体积分数KBXQ组比较,5% WJTXQ及10% WJTXQ组细胞活力显著下降(

温经汤能够下调异位ESCs中HIF-1
α
表达而改善低氧应激,抑制细胞增殖,降低线粒体生物活性,诱导细胞凋亡,这可能是其防治EMT的作用机制。
Objective

This study investigated the mechanism of Wenjingtang in the prevention and treatment of endometriosis (EMT) from the perspective of regulating hypoxia stress and mitochondrial function.
Method

Primary human endometrial stromal cells (ESCs) form ectopic endometrial tissues were isolated and cultured, the cells were divided into control group (Control), 5% control serum group (5% KBXQ), 10% control serum group (10% KBXQ), 5% Wenjingtang serum group (5% WJTXQ) and 10% Wenjingtang serum group (10% WJTXQ). ESCs in different groups were detected for proliferation by methyl thiazolyl tetrazolium (MTT) assay, mRNA and protein expression of hypoxia inducible factor-1
α
(HIF-1
α
) by real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) and Western blot analysis, mitochondrial ultrastructure by transmission electron microscope, mitochondrial function [mitochondrial membrane potential, mitochondrial membrane potential(MMP) and cytochrome C(Cyt C) content] and apoptosis (cell membrane permeability, nuclear fluorescence intensity, nuclear size and cell counts) by high content screening (HCS) assay, apoptosis rate by flow cytometry, and proteins of B-cell lymphoma-2 (Bcl-2) associated X (Bax), Bcl-2 and cleaved cysteinyl aspartate specific proteinase-3 (cleaved Caspase-3) by Western blot.
Result

Compared with Control group, the 5% KBXQ and 10% KBXQ groups showed increased cell viability (

0.01), there was no significant change in HIF-1
α
mRNA and protein expression, transmission electron microscopy showed that the mitochondrial cristae were obvious and the inner and outer membranes of mitochondria were clear, HCS multichannel fluorescence staining showed that there were no significant changes in the expression of MMP, Cyt C and cell membrane permeability, and the nuclei showed uniform light staining, there were no significant changes in apoptosis rate, cleaved Caspase-3 protein expression and Bax/Bcl-2 ratio. Compared with Control group and corresponding concentration KBXQ group, the 5% WJTXQ and 10% WJTXQ group showed decreased cell viability (

mRNA and protein levels (

0.01), the ultrastructure of mitochondria was destroyed, some mitochondria were swollen and the cristae were blurred, moreover, decreased MMP and up-regulated Cyt C release (

0.01), and apoptosis characteristics included nuclear pyknosis, DNA agglutination in nucleus and decrease of cell numbers were observed (

0.01), which was consistent with the results of HCS analysis, and up-regulated expression levels of cleaved Caspase-3 protein and Bax/Bcl-2 ratio (

Conclusion

In conclusion, the results suggest that Wenjingtang can improve hypoxia stress via down-regulating HIF-1
α
expression in ectopic ESCs, and inhibit cell proliferation, reduce mitochondrial biological activity and induce apoptosis, which might be the internal mechanism of Wenjingtang in preventing and treating EMT.
