摘要

目的

研究黄芩汤对溃疡性结肠炎（UC）小鼠肠道辅助性T细胞（Th）17/调节性T细胞（Treg）、Th1/Th2细胞平衡的影响。

方法

60只雄性C57BL/6J小鼠，按体质量随机分为正常组、模型组、柳氮磺胺吡啶组（0.2 g·kg

-1

），黄芩汤低、中、高剂量组（4.55、9.1、18.2 g·kg

-1

），除正常组外均予以2.5%右旋葡聚糖硫酸钠（DSS）自由饮水5 d诱导UC，连续灌胃给药7 d后处死小鼠，取肠系膜淋巴结（MLNs）和结肠组织，流式细胞术检测小鼠肠系膜淋巴结Th17/Treg、Th1/Th2，流式细胞微球芯片捕获技术检测结肠组织白细胞介素（IL）-2、IL-17A、IL-6、肿瘤坏死因子-

（TNF-

）、

干扰素（IFN-

）、IL-4、IL-10等细胞因子含量，对结肠组织样本进行病理切片观察，并采用免疫组化法检测结肠组织中T细胞表达的T-box（T-bet）、GATA结合蛋白-3（GATA-3）、IL-17A、叉头蛋白P3（FoxP3）的表达水平，酶联免疫吸附测定法（ELISA）检测小鼠结肠组织中转化生长因子-

（TGF-

）的含量。

结果

与正常组比较，模型组Th1、Th17细胞数量显著增高，而Th2、Treg细胞数量显著减少，Th1/Th2和Th17/Treg明显升高（

0.05，

0.01），小鼠结肠组织中T-bet、GATA-3、IL-17A、FoxP3表达明显升高（

0.05，

0.01），细胞因子IL-2、IL-17A、IL-6、TNF-

、IFN-

的含量明显升高，IL-4、TGF-

、IL-10的含量明显降低（

0.05，

0.01）。与模型组比较，黄芩汤各剂量组均能逆转Th1/Th2和Th17/Treg细胞的比例失衡（

0.01）；黄芩汤低、中、高剂量组均可明显降低小鼠结肠T-bet、GATA-3的表达（

0.05，

0.01），黄芩汤高低剂量组可显著降低IL-17A的表达（

0.01）；黄芩汤高、中剂量组可显著降低细胞因子IL-2、IL-17A、IFN-

、IL-6、TNF-

的含量，黄芩汤高剂量组还可明显增加TGF-

、IL-10的含量（

0.05，

0.01）。

结论

黄芩汤可减轻UC小鼠结肠炎症，改善UC小鼠结肠组织损伤，这可能是通过调节Th17/Treg、Th1/Th2细胞平衡及相关细胞因子水平，调节促炎细胞因子的释放水平来实现的。

Abstract

Objective

To study the effect of Huangqintang on intestinal T helper cells（Th）17/regulatory T cells（Treg） and Th1/Th2 balance in experimental ulcerative colitis （UC） mice.

Method

Sixty male C57BL/6J mice were randomly divided into normal group， model group， sulfasalazine group （0.2 g·kg

-1

）， and Huangqintang low-dose， medium-dose and high-dose groups （4.55，9.1，18.2 g·kg

-1

， respectively） according to body weight. Except for the normal group， all mice were given 2.5% dextran sodium sulfate （DSS， added to free drinking water） for 5 days to induce UC. After 7 days of continuous intragastric administration， the mice were sacrificed and the mesenteric lymph nodes （MLNs） and colon tissues were collected. The Th17/Treg and Th1/Th2 cell ratios in MLNs of mice were measured by flow cytometry. Interleukin-2 （IL-2）， interleukin-17A （IL-17A）， interleukin-6 （IL-6）， tumor necrosis factor-

（TNF-

）， interferon-

（IFN-

）， interleukin-4 （IL-4）， interleukin-10 （IL-10） and other cytokines in colon tissues were detected by flow cytometric microsphere-based assay. The colon tissues samples were sectioned， and immunohistochemical method was used to detect the expression levels of T-box transcription factor （T-bet）， GATA binding protein 3 （GATA-3）， IL-17A and forkhead box P3 （FoxP3） in colon tissues. The content of transforming growth factor-

（TGF-

） in colon tissue of mice was determined by enzyme-linked immunosorbent assay （ELISA）.

Result

Compared with the normal group， the number of Th1 and Th17 cells in the model group was significantly increased， while the number of Th2 and Treg cells was significantly decreased， and Th1/Th2 and Th17/Treg were significantly increased (

0.05，

0.01). The expressions of T-bet， GATA-3， IL-17A and FoxP3 in colon tissues of mice were significantly increased (

0.05，

0.01)， and the contents of cytokines IL-2， IL-17A， IL-6， TNF-

and IFN-

were significantly increased. The contents of IL-4， TGF-

and IL-10 were significantly decreased (

0.05，

0.01). Compared with the model group， the imbalance of Th1/Th2 and Th17/Treg cells was reversed in each dose group of Huangqin decoction (

0.01). The expressions of T-bet and GATA-3 in colon of mice were significantly decreased in low-dose， medium-dose and high-dose groups of Huangqintang (

0.05，

0.01)， and the expression of IL-17A was significantly decreased in high-dose and low-dose groups of Huangqintang (

0.01). The contents of cytokines IL-2， IL-17A， IFN-

， IL-6 and TNF-

were significantly decreased in the high-dose and medium-dose groups of Huangqintang， and the contents of TGF-

and IL-10 were significantly increased in the high-dose group of Huangqintang (

0.05，

0.01).

Conclusion

Huangqintang could reduce colonic inflammation and improve colonic tissue damage in UC mice， possibly by regulating Th17/Treg and Th1/Th2 balance and related cytokines levels， and the release level of pro-inflammatory cytokines.
