研究黄连解毒汤对高脂饮食诱导的载脂蛋白E基因敲除(ApoE
-/-
)小鼠动脉粥样硬化的干预作用。
方法

高脂饮食建立ApoE
-/-
小鼠动脉粥样硬化模型,给予黄连解毒汤进行干预。通过苏木素-伊红(HE)染色观察动脉粥样硬化病变后主动脉病理形态变化;全自动生化分析仪检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)水平变化;蛋白免疫印迹法(Western blot)检测主动脉沉默信息调节因子2相关酶1(SIRT1)与核转录因子-
κ
B(NF-
κ
B)蛋白表达水平;实时荧光定量聚合酶链式反应(Real-time PCR)检测肝脏腺苷酸活化蛋白激酶(AMPK)与过氧化物酶体增殖物激活受体
α
(PPAR
α
) mRNA表达水平,检测主动脉组织肿瘤坏死因子-
α
(TNF-
α
)、白细胞介素-6(IL-6)、白细胞介素-1
β
(IL-1
β
)与NOD样受体热蛋白结构域3(NLRP3) mRNA表达水平。
结果

与正常组比较,模型组血管中有大量脂质积累,血清中TG、TC、LDL-C水平均显著升高(

小鼠动脉粥样硬化主动脉斑块形成有一定的干预作用,其机制可能与降低血清TC、TG、LDL-C水平、升高HDL-C水平从而发挥降血脂作用,上调SIRT1蛋白、降低NF-
κ
B蛋白、降低TNF-
α
、IL-6等炎症因子等保护血管免受炎症损伤及提高AMPK与PPAR
α
水平参与自噬与凋亡反应有关。
Objective

To study the intervention of Huanglian Jiedutang on atherosclerosis (AS) in apolipoprotein E knockout (ApoE
-/-
) mice induced by the high-fat diet.
Method

The ApoE
-/-
mouse model of AS was induced by the high-fat diet, and Huanglian Jiedutang was used to intervene in the AS in the ApoE
-/-
mice. The pathological changes of aorta were observed by hematoxylin-eosin (HE) staining. The levels of serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were detected by an automatic biochemical analyzer. The protein expression levels of sirtuin-1 (SIRT1) and nuclear factor-kappa B (NF-
κ
B) were determined by Western blot assay, and the mRNA expression levels of adenosine 5'-monophosphate-activated protein kinase (AMPK), peroxisome proliferators-activated receptors
α
(PPAR
α
), tumor necrosis factor-
α
(TNF-
α
), interleukin-6 (IL-6), interleukin-1
β
(IL-1
β
), and NOD-like receptor pyrin domain-containing 3 (NLRP3) were determined by real-time quantitative polymerase chain reaction (Real-time PCR).
Result

As compared with the normal group, there was a large amount of lipid accumulation in the blood vessels of the model group. In the model group, the levels of serum TG, TC, and LDL-C were increased (
P

0.01), and the level of HDL-C was decreased (
P
<
0.01). The protein expression level of SIRT1 in the aorta was decreased, while that of NF-
κ
B was increased in the model group (
P
<
0.01). The mRNA expression levels of IL-6, TNF-
α
, and IL-1
β
were higher (

<
0.01), while those of AMPK in the liver were lower in the model group (
P
<
0.01). Compared with the model group, the Huanglian Jiedutang group reduced the lipid accumulation and inflammatory reaction in the aorta of mice with AS, reduced the levels of TC, TG, and LDL-C (

0.01), and increased the level of HDL-C (

<
0.01). Huanglian Jiedutang significantly increased the protein expression level of SIRT1 in the aorta of ApoE
-/-
mice (

0.01) and decreased the protein expression levels of NF-

B in the aorta (

0.01). Huanglian Jiedutang down-regulated the mRNA expression levels of TNF-

, IL-6, IL-1

Conclusion

Huanglian Jiedutang has a certain intervention effect on the formation of atherosclerotic aortic plaque in ApoE
-/-
mice. Its mechanism may be related to the decrease of serum TC, TG, and LDL-C levels, the increase of HDL-C levels, thus playing a role in lowering blood lipid, the increase of SIRT1 protein, the decrease of NF-
κ
B protein, the decrease of inflammatory factors such as TNF-
α
and IL-6, which protects blood vessels from inflammatory injury, and the improvement of AMPK and PPAR
α
levels to participate in autophagy and apoptosis.
