    • Improvement of Colonic Mucosa Inflammatory Response in Mice with Ulcerative Colitis by Xiezhuo Jiedu Recipe Through miRNA-155-5p/JAK2/STAT3 Pathway

    • A study on the treatment mechanism of ulcerative colitis (UC) has made significant progress. The research team used bioinformatics methods to conduct in-depth analysis of microRNAs (miRNAs) between the active and remission phases of UC. By screening differentially expressed genes (DEGs) and constructing regulatory relationships, the mechanism of action of Xiezhuo Jiedu Formula in treating UC was speculated and validated through animal experiments. The research team first obtained a miRNA dataset of colon mucosal tissue from UC patients from the Gene Expression Database (GEO). Using tools such as GEO2R and Excel, the most significantly differentially expressed miRNAs were selected as the research subjects. Subsequently, multiple database tools were used to screen key DEGs, predict downstream transcription factors (TFs), and perform gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. These analyses reveal the key signaling pathways involved in the occurrence and development of UC. To verify the speculated mechanism of action, the research team conducted animal experiments. A UC mouse model was prepared using a 2.5% dextran sulfate sodium (DSS) free drink method, and the formula for clearing turbidity and detoxifying and mesalazine were orally administered for 7 days. The experimental results showed that the formula for relieving turbidity and detoxification can effectively downregulate the expression of specific miRNAs in the colon tissue of UC mice and regulate the expression of key signaling pathway related proteins. These changes effectively increased the levels of anti-inflammatory factors and reduced the infiltration of inflammatory cells. This study not only provides new ideas and methods for the treatment of UC, but also opens up new directions for research in related fields. In the future, it is expected to bring better treatment outcomes to UC patients through further optimization of treatment plans.
