Mechanism of Linggui Zhugantang in Promoting Astrocyte Endocytosis and Degradation of Amyloid β
“The research team delved into the potential of traditional Chinese medicine Linggui Zhugan Tang (LGZGT) in the treatment of Alzheimer's disease (AD). The study constructed a rat primary astrocyte (AS) AD model induced by β - amyloid protein 1-42 (A β 1-42) and observed the effect of LGZGT containing serum on the model. The experimental results showed that A β 1-42 has an inhibitory effect on the proliferation of AS, while LGZGT containing serum can significantly improve cell survival rate, reduce lactate dehydrogenase (LDH) activity, and alleviate cell damage. In addition, LGZGT can upregulate the expression of A β - related degradation enzymes such as insulin degrading enzyme (IDE) and tissue protease D (CTSD), enhance the fluorescence intensity of tissue protease B (CTSB), and reduce the intracellular A β 1-42 content. This discovery indicates that LGZGT has a protective effect on A β 1-42-induced AS and can promote the degradation of A β. The mechanism may be related to reducing A β toxicity, enhancing cell viability, promoting the expression of IDE, CTSD, CTSB, and restoring lysosomal function. This provides new candidate drugs and research directions for the treatment of AD.”
