    • Mechanism of Xiezhuo Jiedu Recipe Regulating Ferroptosis in Treatment of Ulcerative Colitis Based on Bioinformatics and Animal Experiments

    • A bioinformatics study on the treatment of ulcerative colitis (UC) has made significant progress. Researchers have successfully screened iron death differentially expressed genes (FRGs) closely related to UC using bioinformatics methods, and verified the mechanism of action and regulation of cell iron death in traditional Chinese medicine Xiezhuo Jiedu Formula for the treatment of UC. The research team first obtained differentially expressed genes in colon mucosal tissue of UC patients from the GEO database, and performed intersection analysis with iron death genes to screen out 21 FRGs. After cluster analysis, LASSO regression, and ROC curve analysis, FBXW7, ZFP36, SLC7A11, and TLR4 were identified as the core FRGs. Animal experiments further confirmed that these core FRGs are significantly correlated with immune cell infiltration in the colon mucosa of UC mice. The experimental results showed that the colon mucosal tissue structure of the model group mice was disordered, and inflammatory cell infiltration was severe. However, after treatment with the formula for relieving turbidity and detoxification and mesalazine, the inflammatory response of the colon mucosal tissue in mice was alleviated to varying degrees. It is worth mentioning that the colon mucosa of mice in the high-dose group of Xiezhuo Jiedu Formula showed almost no inflammatory changes. In addition, compared with the normal group, the expression levels of FBXW7, ZFP36, SLC7A11, TLR4 protein and mRNA in the colon mucosa of the model group mice were significantly increased. But after treatment with the formula for clearing turbidity and detoxifying and mesalazine, the expression of these core FRGs was significantly downregulated. This research achievement provides a new perspective and solution for the treatment of UC, as well as a scientific basis for the clinical application of traditional Chinese medicine formulas for clearing turbidity and detoxifying. In the future, the research team will continue to explore the details of this mechanism of action in order to bring more effective treatment methods for UC patients.
