摘要:This article has systematically sorted out and verified the name, origin, producing area, quality evaluation, harvesting and processing of Pruni Semen by consulting ancient materia medica, medical books, prescription books and modern literature, in order to provide a basis for the development of famous classical formulas containing Pruni Semen. The results showed that Pruni Semen, as a medicinal material, has been widely used in medical literature of past dynasties since it was collected in Shennong Bencaojing, and also included under the names such as Yuhe, Yuzi and Yuli, and aliases such as Jueli, Queli and Chexiali. The primordial plants mentioned in the past dynasties involve about 12 species of Rosaceae, but with Prunus humilis, P. japonica and P. glandulosa as mainstream varieties used in the past dynasties, while the 2020 edition of Chinese Pharmacopoeia stipulates that the basal plants are P. humilis, P. japonica and P. pedunculata. Most of the ancient records for the origin of Pruni Semen are found everywhere in high mountains, valleys and hills, modern literature records that its origin varies according to its base, for example, P. humilis and P. japonica are mainly produced in Hebei, eastern Inner Mongolia, Liaoning, Shandong and other regions of China, and P. pedunculata is mainly produced in Inner Mongolia. Modern literature summarizes its quality as faint yellow, full and fulfilling, neat and not broken, and non-oiling, and the small Pruni Semen is better than the big Pruni Semen. The ancient processing methods of Pruni Semen mainly include blanching and peeling, blanching and peeling followed by frying, and blanching and peeling followed by pounding, with the common feature of blanching and peeling. The successive editions of Chinese Pharmacopoeia stipulate that it should be pounded when used. Based on the results of the herbal textual research and the writing time of Bianzhenglu, and combined with the market survey of Pruni Semen, it is suggested that P. humilis or P. japonica should be used as the origin of Pruni Semen in Sanpiantang, and it is harvested when the fruits are ripe, the kernels are collected by removing the stones, and processed by blanching, peeling and pounding consulting the decoction method in the current edition of Chinese Pharmacopoeia.

摘要:By reviewing ancient materia medica, medical books, prescription books and modern literature, this paper conducted a systematic research on name, origin, medicinal parts, producing area, quality, harvesting and processing methods, functions and toxicity of Polygoni Multiflori Radix(PMR) and Polygoni Multiflori Caulis(PMC) in famous classical formulas. It was found that the name of Heshouwu was first found in the Biography of Heshouwu and originated from its discoverer, and then still in use today. Since the Song dynasty, Heshouwu has been included in the materia medica with Fallopia multiflora as the mainstream origin. Since the Ming dynasty, in addition to F. multiflora, Cynanchum bungei, C. wilfordii, C. auriculatum have been used as the origin of Heshouwu. Heshouwu is widely distributed, the production areas recorded in ancient times are mainly Luchuan, Guangxi and Ganzhou, Jiangxi, and in modern times, Deqing, Guangdong and other places are respected as the geo-authentic habitats. Its origin processing is mostly dug out, washed, sliced and dried in the sun. Modern literature concludes that the quality of PMR is better in terms of weight, solid quality and powder, while PMC is better in terms of uniformity and purplish-red skin. In the Qing dynasty and before that, it was recorded in the materia medica that PMR was harvested mainly in late spring, mid-summer and early autumn, in modern times, it was harvested mainly in spring and autumn, while PMC was harvested in autumn. In Song dynasty, the processing method of PMR was mainly soaked with rice-washed water, the Ming dynasty for black bean steaming method, the Qing dynasty for steaming followed by black bean juice maceration, while in modern times, it is mostly sliced and steamed with black bean juice and yellow rice wine until the juice is absorbed. PMC is prepared by removing impurities, washing or moistening, and cutting into sections to dry. During the five dynasties, PMR was used to treat infertility as well as gastrointestinal diseases and blood in the stool caused by wind chill, and during the Song dynasty, it was mostly used raw products for the treatment of scrofula and carbuncles, and in the Yuan and Ming dynasties, raw and processed products were used, and Polygoni Multiflori Radix Praeparata was used to treat hemorrhoids, waist and knee soreness. PMC is mainly used for treating insomnia, fatigue and sweating, wind sores and scabies, etc. In the Song dynasty, the ancients considered that PMR was non-toxic, but its toxicity was gradually discovered after the Ming dynasty, and the toxicity mechanism has not been clearly analyzed yet. Based on the results of the textual research, it is recommended that F. multiflora be used as the base for development of famous classical formulas containing PMR, and from the safety perspective, it is recommended to use raw products with caution, oral administration must be used processed products. PMC is recommended to use raw products.
摘要:By consulting ancient materia medica, medical books, prescription books and modern literature, this paper systematically combed and reviewed the name, origin, scientific name evolution, producting area, quality evaluation, medicinal parts, harvesting and processing and traditional efficacy of Lasiosphaera Calvatia. The results show that Mabo was first recorded in Mingyi Bielu. Since then, all dynasties have taken Mabo as a legitimate name. Before the Song dynasty, only Calvatia lilacina was used as the original plant of Lasiosphaera Calvatia, which was expanded after the Song dynasty with the appearance of C. gigantea, Lasiosphaera fenzlii, Bovistella radicata and other varieties. Until modern times, there was an addition of Lycoperdon perlatum, L. pyriforme and other original plants of Lasiosphaera Calvatia. Since 1975, the original plant of Lasiosphaera Calvatia in various regulations and academic monographs has been basically uniform for C. lilacina, Lasiosphaera fenzlii and C. gigantea. Resource of the medicinal fungus was widely distributed in China and was mainly wild. From ancient times to the present, the medicinal parts of Lasiosphaera Calvatia are all fruiting body, which is harvested in summer and autumn, and its processing method was to take powder in ancient times, but to cut blocks in modern times. In recent times, its quality has been summarized as large, thin-skinned, intact, full, loose-bubbled and elastic. The medicinal efficacy has been developed from very good for all scores, and after the Ming and Qing dynasties, it is consistent with the 2020 edition of Chinese Pharmacopoeia, with the efficacy of clearing the lung, promoting pharynx, relieving fever and hemostasis, mainly treating cough aphonia, throat obstruction and pharyngeal pain, vomiting blood, epistaxis, hemoptysis, and external treating sores and bleeding from cuts and wounds. Based on the results of herbal textual research, it is suggested that C. lilacina is the first choice for the origin of Lasiosphaera Calvatia involved in famous classical formulas, and it is processed into block or powder for medicine.

摘要:ObjectiveTo establish the specific chromatogram and thin layer chromatography(TLC) of Qingxin Lianziyin(QXLZY) benchmark samples, in order to clarify the key quality attributes and provide a reference for the quality evaluation of QXLZY.MethodHigh performance liquid chromatography(HPLC) specific chromatogram of QXLZY benchmark samples was developed by using a YMC Hydrosphere C18 column(4.6 mm×250 mm, 5 μm) with the mobile phase of acetonitrile(A)-0.2% formic acid aqueous solution(B) for gradient elution(0-10 min, 5%-20%A; 10-20 min, 20%A; 20-25 min, 20%-24%A; 25-40 min, 24%-30%A; 40-55 min, 30%-50%A; 55-65 min, 50%-100%A; 65-75 min, 100%A; 75-75.1 min, 100%-5%A; 75.1-90 min, 5%A), and the detection wavelength was 360 nm. Ultra-high performance liquid chromatography-linear ion trap/orbitrap mass spectrometry(UHPLC-LTQ-Orbitrap MS) with electrospray ionization(ESI) was used to identify the components of QXLZY benchmark samples by accurate relative molecular weight and multilevel MS fragment ion information, the detection conditions were positive and negative ion modes and data dependency scanning mode. TLC identification methods for Ophiopogonis Radix, Lycii Cortex, Nelumbinis Semen, Poria, Astragali Radix and Ginseng Radix et Rhizoma in QXLZY were established.ResultA total of 15 characteristic peaks were identified from Glycyrrhizae Radix et Rhizoma, Plantaginis Semen and Scutellariae Radix, and the relative standard deviations of the retention times of 15 characteristic peaks in 15 batches of QXLZY benchmark samples were≤3% with peak 8(baicalin) as the reference peak. A total of 100 compounds, including flavonoids, organic acids, saponins, amino acids and others, were identified in the benchmark samples by UHPLC-LTQ-Orbitrap MS. The established TLC had good separation and was suitable for the identification of Ophiopogonis Radix, Lycii Cortex, Nelumbinis Semen, Poria, Astragali Radix and Ginseng Radix et Rhizoma in QXLZY.ConclusionThe material basis of QXLZY benchmark samples is basically determined by MS designation and source attribution. The established specific chromatogram and TLC of QXLZY are simple, stable and reproducible, which can provide a reference for the development and quality control of QXLZY.

摘要:ObjectiveTo explore the anti-inflammatory effect of Duhuo Jishengtang (DHJST) on collagen-induced arthritis (CIA) model rats and its effect on the Toll-like receptor 2 (TLR2)/p38 mitogen-activated protein kinase (MAPK)/nuclear factor-κB (NF-κB) signaling pathway.MethodForty-eight male SD rats were randomly divided into the following six groups (n=8): normal group, model group, methotrexate (MTX) group, low-dose DHJST (DHJST-L) group, medium-dose DHJST (DHJST-M) group, and high-dose DHJST (DHJST-H) group. The CIA model was established by injecting bovine type Ⅱ collagen into the rat tail root with the collagen antibody induction method. After model induction, rats were treated with drugs by gavage. The rats in the MTX group received MTX at 2.0 mg·kg-1, three times a week, and those in the DHJST groups received DHJST at 3.8, 7.6, 15.2 g·kg-1·d-1 for 28 days. The rats in the normal group and the model group were given the same dose of normal saline. The weight of the rats was recorded, and the paw swelling degree was observed. The arthritis index and immune organ index were measured, and the changes in the microcirculation indexes of the rats were detected with a microcirculation detector. Hematoxylin-eosin (HE) staining was used to detect the pathological morphologic changes in rat synovial tissues and the apoptosis rate of synovial cells was detected by flow cytometry to determine the therapeutic effect of DHJST on rheumatoid arthritis. Enzyme-linked immunosorbent assay (ELISA) was used to detect the changes in serum levels of tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-17A, and interferon-γ (IFN-γ). The protein expression of TLR2, NF-κB p65, phosphorylated NF-κB p65 (p-NF-κB p65), p38 MAPK, and p-p38 MAPK was detected by Western blot.ResultCompared with the normal group, the model group showed reduced body weight (P<0.01), increased paw swelling degree, arthritis index, and immune organ index (P<0.01), increased comprehensive microvascular score and vascular resistance (P<0.01), significant hyperplasia of synovial tissues and massive infiltration of inflammatory cells as revealed by pathological sections, and up-regulated expression levels of TNF-α, IL-1β, IL-17A, and IFN-γ in serum, and TLR2, p-NF-κB p65/NF-κB p65 and p-p38 MAPK/p38 MAPK in synovial tissues (P<0.01). Compared with the model group, the DHJST groups showed increased body weight of rats (P<0.01), decreased paw swelling degree, arthritis index, and immune organ index (P<0.05, P<0.01), reduced comprehensive microvascular score and vascular resistance (P<0.05, P<0.01), improved synovial histopathological injury, increased apoptosis rate of synovial cells (P<0.01), and down-regulated levels of TNF-α, IL-1β, IL-17A, and IFN-γ in serum (P<0.05, P<0.01) and TLR2, p-NF-κB p65/NF-κB p65 and p-p38 MAPK/p38 MAPK in synovial tissues (P<0.05, P<0.01).ConclusionDHJST may alleviate the inflammatory reaction in CIA rats by regulating the TLR2/p38 MAPK/NF-κB signaling pathway, thus exerting its anti-rheumatoid arthritis effect.
摘要:ObjectiveTo investigate the anti-inflammatory effect of the component compatibility of Gentianae Macrophyllae Radix and Clematidis Radix et Rhizoma on the rat model of rheumatoid arthritis (RA) and the mechanism.MethodSeventy-two SPF-grade SD rats (male and female) aged 5 to 6 weeks were selected. Except the blank group, the rat model of collagen-induced arthritis (CIA) was replicated by the type Ⅱ collagen induction method. The 64 rats after successfully modeling were randomly divided into model group, methotrexate group (0.375 mg·kg-1), gentianoside with magnoflorine group (150.454 1 mg·kg-1+5.061 8 mg·kg-1), gentianoside with clematichinenoside AR group (150.454 1 mg·kg-1+16.433 1 mg·kg-1), sweroside with magnoflorine group (3.455 8 mg·kg-1+5.061 8 mg·kg-1), sweroside with clematichinenoside AR group (3.455 8 mg·kg-1+16.433 1 mg·kg-1), swertiamarin with magnoflorine group (9.303 2 mg·kg-1+5.061 8 mg·kg-1), and swertiamarin with clematichinenoside AR group (9.303 2 mg·kg-1+16.433 1 mg·kg-1), with 8 rats in each group. Each group was given the corresponding medicinal solution or normal saline by gavage for 15 d. During the experiment, the general status, of rats in each group were observed and recorded. Tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), rheumatoid factor (RF), C reactive protein (CRP), prostaglandin E2 (PGE2), and anti-cyclic peptide containing citrulline antibody (anti-CCP Ab) in the serum of rats were measured by enzyme-linked immunosorbent assay (ELISA). The histopathological changes in rat ankle joints were observed by hematoxylin-eosin (HE) staining. Immunohistochemistry (IHC) and Western blot were used to detect the protein expression of nuclear factor-κB (NF-κB) and vascular endothelial growth factor (VEGF) in rat ankle joints. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression of NF-κB and VEGF in rat ankle joints.ResultCompared with those in the blank group, rats in the model group were in poor general conditions with significant foot-plantar swelling, and the content of CRP, anti-CCP Ab, and IL-1β in the rat serum was significantly increased (P<0.01). In the model group, the tissue structure of the ankle joint was severely damaged, and the protein and mRNA expression of NF-κB and VEGF in the rat ankle joints were significantly up-regulated (P<0.01). As compared with the model group, the general status of rats in each administration group was significantly improved. The levels of serum TNF-α, IL-1β, RF, CRP, PGE2, and anti-CCP Ab were reduced to different degrees in these administration groups, among which the effects of the gentianoside with clematichinenoside AR group on down-regulating serum TNF-α and IL-1β, the gentianoside with magnoflorine group on down-regulating serum RF and CRP, the sweroside with magnoflorine group on down-regulating serum PGE2, and the swertiamarin with clematichinenoside AR group on lowering serum anti-CCP Ab were better than those of administration groups. The histopathological changes in the ankle joint were improved to different degrees. The protein and mRNA expression of NF-κB and VEGF in rat ankle joints in the administration groups was significantly down-regulated (P<0.05, P<0.01), and the swertiamarin paired with clematichinenoside AR group had the most significant effect.ConclusionThe component compatibility of Gentianae Macrophyllae Radix and Clematidis Radix et Rhizoma exerts a good therapeutic effect on the rat model of RA, and the compatibility of components from the two medicines has a multi-channel, multi-target, and synergistic effect. The five component compatibility patterns, namely gentiobioside with magnoflorine, gentiobioside with clematichinenoside AR, sweroside with clematichinenoside AR, swertiamarin with magnoflorine, and swertiamarin with clematichinenoside AR, all have potential advantages. The mechanism may be related to the reduction of inflammatory factor secretion and the inhibition of abnormal protein and mRNA expression of NF-κB and VEGF.
摘要:ObjectiveTo investigate the protective effect of Jianpi Huogu prescription (JPHGP) on the functional injury of vascular endothelial cells caused by alcohol and explore its mechanism based on protein kinase B/c-Jun amino-terminal kinase/p38 MAPK (Akt/JNK/p38 MAPK) signaling pathway.MethodThrough chick embryo allantoic membrane, thoracic aortic ring, and migration, invasion, adhesion, and lumen formation of human umbilical vein endothelial cells (HUVEC), the effect of JPHGP with different concentrations (8, 16 and 32 μg·L-1) on angiogenesis was observed in the presence or absence of alcohol. The expression levels of phosphorylation of Akt, JNK, and p38 MAPK were determined by Western blot.ResultAs compared with the normal group, the number and length of capillaries around the arterial ring in the model group were decreased, and the migration, invasion, and lumen formation capacity of HUVEC were decreased (P<0.05, P<0.01). After treatment with 16 and 32 μg·L-1 JPHGP, the length of neovascularization in chick embryo allantoic membrane was significantly increased (P<0.05, P<0.01). Compared with the model group, the 8, 16, and 32 μg·L-1 JPHGP groups increased the number of capillaries around the thoracic aortic ring in a concentration-dependent manner (P<0.05, P<0.01), and the 32 μg·L-1 JPHGP group increased the length of capillaries around the thoracic aortic ring (P<0.05). The 16 and 32 μg·L-1 JPHGP groups enhanced the migration, invasion, and lumen formation capacity of HUVEC. The results of Western blot showed that, as compared with the normal group, the protein expression levels of p-JNK/JNK, p-p38 MAPK/p38 MAPK, and p-Akt/Akt were significantly decreased in the model group (P<0.01), and as compared with the model group, the protein expression levels of p-p38 MAPK/p38 MAPK and p-Akt/Akt were significantly increased in the 8, 16, and 32 μg·L-1 JPHGP groups (P<0.01) and the protein expression level of p-JNK/JNK was increased significantly in the 16 and 32 μg·L-1 JPHGP groups (P<0.01).ConclusionJPHGP has a protective effect on the functional injury of vascular endothelial cells caused by alcohol, and its mechanism may be related to the activation of Akt/JNK/p38 MAPK signaling pathway. Relevant research results will provide certain scientific basis for clarifying the effect of JPHGP on 'invigorating spleen and promoting blood circulation'.
摘要:ObjectiveTo observe the protective effect and mechanism of Tianhuang formula (THF) against renal injury in hyperuricemia nephropathy (HN) mice through network pharmacology.MethodAll mice were randomly divided into a normal group, a model group, a febuxostat group (5 mg·kg-1), a low-dose THF group (L-THF, 60 mg·kg-1), and a high-dose THF group (H-THF, 120 mg·kg-1). The mice in the normal group were treated with 0.5% sodium carboxymethylcellulose (CMC-Na) by gavage daily. The HN model was induced by oral administration of 500 mg·kg-1 hypoxanthine and intraperitoneal injection of 200 mg·kg-1 oteracil potassium in mice except for those in the blank group. The mice in the groups with drug intervention were treated with corresponding drugs by gavage for three weeks. The levels of serum uric acid, creatinine, urea nitrogen, and 24-h albuminuria were measured. The renal injury was observed by hematoxylin-eosin (HE) staining and PAS staining, and renal fibrosis was observed by Sirius red staining. The effects and molecular mechanism of THF in HN mice were analyzed by Western blot, network pharmacology, and molecular docking.ResultBiochemical results indicated that compared with model group, BUN and 24 h urinary protein levels were significantly decreased in L-THF group (P<0.05), SUA and SCr levels were significantly decreased (P<0.01), and SUA, BUN, SCr and 24 h urinary protein levels in H-THF group were significantly decreased (P<0.01). The results of pathological staining showed that the kidney injury and interstitial fibrosis were improved in different doses of THF groups (P<0.05). Western blot results showed that the Nod-like receptor heat protein domain associated protein 3 (NLRP3) inflammatorome, interleukin-1β (IL-1β), fibronectin (FN), uric acid transporter 1 (URAT1), phosphorylated p65 (p-p65) and phosphorylated nuclear transcription factor (NF) -κB were inhibited in the H-THF group The expression of protein-producing α (p-IκBα) was reduced to the normal level (P<0.01), but the expression of IL-1β, URAT1 and p-IκBα in HN mice was not affected in the L-THF group.ConclusionTHF ameliorates renal inflammation and fibrosis by inhibiting the activation of NF-κB and NLRP3 inflammasomes to alleviate HN
摘要:ObjectiveTo observe the effect of Jianpi Yangzheng Xiaozheng decoction (JYXD) on the proliferation and stemness of the human gastric cancer (GC) cell line HGC-27 by inhibiting aerobic glycolysis, and explore the underlying mechanism.MethodMethyl thiazolyl tetrazolium (MTT) assay was employed to determine the survival rate and chemotherapy sensitivity of HGC-27 cells treated with JYXD (0.25, 0.5, 1, 2, 4, 8, 16, 32 g·L-1). Colony formation assay was employed to detect the effect of JYXD (2, 4, 8 g·L-1) on the colony formation of the cells. The aerobic glycolysis level of HGC-27 cells after treatment with JYXD was measured by glucose assay kit and lactic acid assay kit. The proportion of stem cell subsets in HGC-27 cells was detected by flow cytometry. Western blot was employed to determine the expression of glycolysis-associated proteins such as lactate dehydrogenase (LDH), hexokinase 2 (HK2), glucose transporter 1 (GLUT1), and pyruvate kinase isozyme M2 (PKM2), and the expression of stemness-associated proteins such as octamer-binding transcription factor 4 (OCT4), SRY-box transcription factor 2 (SOX2), and Nanog.ResultJYXD (0.5, 1, 2, 4, 8, 16, 32 g·L-1) inhibited the activity of HGC-27 cells (P<0.05, P<0.01), with the inhibitory concentration 50(IC50) of 4.83 g·L-1, and it improved the sensitivity of HGC-27 cells to cisplatin chemotherapy. Compared with the control group, JYXD (2, 4, 8 g·L-1) reduced the colony formation number of HGC-27 cells (P<0.01) in a concentration-dependent manner. Flow cytometry showed that compared with that in the control group, the proportion of CD44+CD24+ALDH+ population in the cells treated with JYXD (2, 4, 8 g·L-1) decreased (P<0.05). In addition, JYXD (2, 4, 8 g·L-1) inhibited the glucose uptake and lactic acid production of HGC-27 cells. Western blot showed that compared with the control group, JYXD (2, 4, 8 g·L-1) down-regulated the expression levels of SOX2, Nanog, OCT4, PKM2, LDH, GLUT1, and HK2 (P<0.05, P<0.01) in a concentration-dependent manner.ConclusionJYXD may inhibit the proliferation and reduce the stemness of HGC-27 cells by regulating the aerobic glycolysis.

摘要:ObjectiveTransforming growth factor-β1 (TGF-β1) was used to stimulate human fetal lung fibroblast 1 (HFL1) for simulating the pathological process of idiopathic pulmonary fibrosis (IPF) and thereby the effects and mechanism of medicated serum of Bupleuri Radix against IPF were investigated.MethodTGF-β1 (10 μg·L-1) was employed to stimulate HFL1, and cells were treated with medicated serum of Bupleuri Radix (5%, 10%, 15%, 20%) for 24 h. Then cell proliferation rate was determined with cell counting kit-8 (CCK-8). Subsequently, cells were classified into the control group (20% blank serum), TGF-β1 group (20% blank serum and 10 μg·L-1 TGF-β1), TGF-β1 + medicated serum of Bupleuri Radix group (5% blank serum, 15% medicated serum, and 10 μg·L-1 TGF-β1), and TGF-β1 + SIS3 group (3 μmol·L-1 SIS3, 20% blank serum, 10 μg·L-1 TGF-β1). Based on in situ end labeling (TUNEL) staining, the apoptosis rate was examined, and mRNA expression of apoptosis-related proteins B-cell lymphoma 2 (Bcl-2), Bcl-2 associated X protein (Bax) and myofibroblast marker α-smooth muscle actin (α-SMA) was detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). The protein expression of α-SMA, Ras homolog enriched in brain (Rheb), and phosphorylated (p)-Smad3 was determined by immunofluorescence. Expression of Rheb, p-Smad3, and Smad3 was examined by Western blot.ResultThe cell proliferation rate of TGF-β1 group increased compared with that of the control group (P<0.05). The cell proliferation rate of TGF+15% medicated serum of Bupleuri Radix group and TGF+20% medicated serum of Bupleuri Radix group decreased compared with that of the TGF-β1 group (P<0.01). Compared with the control group, TGF-β1 group showed decrease in apoptosis rate, increase in mRNA expression of Bcl-2 and α-SMA, reduction in Bax mRNA expression, and rise of α-SMA and Rheb protein expression and p-Smad3 level (P<0.05). Compared with TGF-β1 group, TGF-β1 + medicated serum of Bupleuri Radix group and TGF-β1 + SIS3 group demonstrated high apoptosis rate, low Bcl-2 and α-SMA mRNA expression, high Bax mRNA expression, and low α-SMA and Rheb protein expression and p-Smad3 level (P<0.05).ConclusionMedicated serum of Bupleuri Radix can inhibit TGF-β1-induced HFL1 proliferation and fibroblast-myofibroblast transition and promote fibroblast apoptosis by regulating the Smad3/Rheb axis.

摘要:ObjectiveTo explore the mechanism of Dendrobium huoshanense polysaccharide (DHP) against inflammatory damage of neurons in Parkinson's disease (PD) model.MethodSH-SY5Y cells were randomized into blank group, model group, and DHP group. The survival rate of cells was measured by thiazole blue(MTT) assay, and the levels of lactate dehydrogenase (LDH), reactive oxygen species (ROS), malondialdehyde (MDA), and superoxide dismutase (SOD) were measured by colorimetric analysis. BV-2 microglia were classified into blank group, model group, DHP group, and MCC950 group (positive control group), and enzyme-linked immunosorbent assay (ELISA) was applied to detect the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-18 (IL-18). The expression of NOD-like receptor protein 3 (NLRP3), adaptor protein apoptosis-associated dot protein (ASC), cysteine aspartic protease-1 (Caspase-1), and IL-1β was measured by Western blot. A total of 50 C57BL/6 mice were randomized into blank group, model group, DHP low-dose (100 mg·kg-1) group, DHP equivalent-dose (350 mg·kg-1) group, and MCC950 group (positive control group), 10 mice in each group. The motor balance and coordination of C57BL/6 mice were observed by beam walking test, tail suspension test and rotarod test. The levels of Iba-1 and tyrosine hydroxylase (TH) were detected by immunofluorescence staining. The damage of dopaminergic neurons in the substantia nigra was detected by FJB staining. The levels of inflammatory factors such as IL-1β, IL-18, and TNF-α in mouse midbrain tissues were detected by ELISA and the protein levels of NLRP3, ASC, Caspase-1, and IL-1β protein were measured by Western blot.ResultCompared with the blank group, the SH-SY5Y model group showed decreased cell survival, increased levels of LDH, ROS, and MDA (P<0.05), and decreased levels of SOD (P<0.05). Compared with the model group, the DHP group demonstrated increased cell survival, decreased levels of LDH, ROS, and MDA (P<0.01), and increased level of SOD (P<0.01). Compared with the blank group, BV-2 model group had high levels of IL-1β, IL-18, and TNF-α (P<0.05) and high protein expression of NLRP3, Caspase-1, IL-1β, and ASC (P<0.05). Compared with the model group, DHP and MCC950 groups demonstrated low levels of IL-1β, IL-18, and TNF-α (P<0.01) and low protein expression of NLRP3, Caspase-1, IL-1β, and ASC (P<0.01). Compared with the blank group, the C57BL/6 model group displayed long time to pass the balance wood (P<0.05), short time spent on the rod in the rotarod test (P<0.05), high levels of IL-1β, IL-18, and TNF-α (P<0.05) and expression of Iba-1 in the midbrain substantia nigra (P<0.05), low TH expression (P<0.05), more positive neurons in the FJB staining (P<0.05), and high expression of NLRP3, Caspase-1, ASC, and IL-1β proteins (P < 0.05). Compared with the model group, the mice in the DHP and MCC950 groups had short time to pass the balance beam (P<0.01), long time spent on the rod (P<0.01), low levels of IL-1β, IL-18, and TNF-α (P<0.01), low Iba-1 expression in midbrain substantia nigra (P<0.01), high TH expression (P<0.01), and small number of positive neurons in the midbrain substantia nigra (P<0.01). The expression of NLRP3, ASC, and IL-1β proteins was lower in the MCC950 group (P<0.01), and the expression of NLRP3, ASC, Caspase-1 and IL-1β proteins was lower in the DHP equivalent-dose group (P<0.01) than in the model group.ConclusionDHP has anti-oxidative stress effect. It regulates the expression of NLRP3 inflammasome and inhibits the overactivation of microglia, thereby alleviating the neuroinflammatory injury in PD and exerting the neuroprotective effect.

摘要:ObjectiveTo investigate the mechanism of Lycium barbarum polysaccharides (LBP) in promoting the activation of RAW264.7 macrophages.MethodRAW264.7 macrophages were stimulated with LBP at different concentrations (50, 100, 200 mg·L-1), and those stimulated with lipopolysaccharide (LPS) at 100 μg·L-1 and galactose (Gal) at 100 mg·L-1 as positive controls. After 24 h of LBP stimulation, the cell counting kit-8 (CCK-8) was used to detect the survival rate of RAW264.7 macrophages treated with LBP (0, 50, 100, 200, 400, 800 mg·L-1). The levels of interleukin-6 (IL-6) and interleukin-12 (IL-12) in cell culture supernatant were detected by enzyme-linked immunosorbent assay (ELISA). The protein and mRNA expression of p38 mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and nuclear factor κB (NF-κB) in Toll-like receptor 4 (TLR4)/Toll-like receptor 2 (TLR2)/macrophage galactose-type lectin (MGL) pathway of RAW264.7 macrophages was detected by Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) and Western blot.ResultCCK-8 results showed that compared with the results in the blank group, the survival rate of RAW264.7 macrophages decreased in the 400, 800 mg·L-1 LBP groups (P<0.05). ELISA results showed that compared with the blank group, 50 mg·L-1 LBP could promote the secretion of IL-12 in RAW264.7 macrophages (P<0.01). Compared with the blank group, 100 mg·L-1 LBP and 200 mg·L-1 LBP could promote the secretion of IL-6 in RAW264.7 macrophages (P<0.05, P<0.01). Western blot results showed that compared with the blank group, the LBP groups (50, 100, 200 mg·L-1) enhanced protein expression levels of MAPK key molecules (p-p38 MAPK, p-ERK, p-NF-κB, and p-JNK) in TLR4, TLR2, and MGL pathways (P<0.05, P<0.01). Compared with the model group, the 200 mg·L-1 LBP group could promote the expression level of p-NF-κB protein in RAW264.7 macrophages (P<0.01). Real-time PCR results showed that compared with the blank group, the LBP groups (50, 100, and 200 mg·L-1) enhanced the mRNA expression levels of MAPK key molecules (p38 MAPK, ERK, NF-κB, and JNK) in TLR4 and TLR2 pathways (P<0.05, P<0.01). Compared with the model group, the 50 and 200 mg·L-1 LBP groups could promote the mRNA expression levels of JNK and ERK2 in RAW264.7 macrophages (P<0.05, P<0.01).ConclusionLBP can regulate the activation of RAW264.7 macrophages and participate in the immune response through the TLR2/TLR4/MGL pathway.
摘要:ObjectiveStudy on the mechanism of Guishao Yunpi decoction in preventing and treating breast hyperplasia based on phosphatase and tensin homolog (PTEN)/ phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) pathway.MethodSeventy SPF-grade SD rats were randomly assigned into blank group (n=15) and modeling group (n=55). The mammary gland hyperplasia model of liver stagnation and spleen deficiency was established by the comprehensive modeling method (hunger and satiety abnormality + tail stimulation + estradiol benzoate + progesterone), and then 5 rats were randomly selected for model verification. The modeled rats were then randomly assigned into five groups: model group, positive control (4 mg·kg-1·d-1 tamoxifen) group, and high-, medium-, and low-dose (17.2, 8.6, 4.3 g·kg-1·d-1, respectively) Guishao Yunpi decoction groups, with 10 rats in each group. The rats in the blank group and model group were given 10 mL·kg-1·d-1 distilled water, and those in other groups were orally administrated with corresponding drugs. After 30 days of treatment, the general living conditions of rats were observed, and the thickness of breast tissue was measured by an ultrasonic diagnostic instrument. The open field test was carried out for behavioral evaluation. The levels of B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X (Bax) in the breast tissue were determined by enzyme-linked immunosorbent assay (ELISA). The mRNA and protein levels of PTEN, PI3K, and Akt in the breast tissue were determined by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively.ResultCompared with those in the blank group, the rats in the model group were irritable, curled up in clusters, and showed positive behavior in the open field test. The modeling led to nipple swelling and increased the breast thickness (P<0.05). Moreover, the modeling elevated the level of Bcl-2 and lowered that of Bax in the breast tissue (P<0.05), down-regulated the mRNA and protein levels of PTEN, and up-regulated the mRNA and protein levels of PI3K and Akt (P<0.05). Compared with the model group, drug administration relieved the general survival state, the degree of nipple swelling, and the positive behavior in the open field test and reduced the breast thickness (P<0.05). In addition, drug administration reduced the level of Bcl-2 and increased that of Bax in the breast tissue (P<0.05), up-regulated the mRNA and protein levels of PTEN, and down-regulated the mRNA and protein levels of PI3K and Akt (P<0.05).ConclusionGuishao Yunpi decoction can improve the general living conditions and alleviate the mammary gland hyperplasia of rats with the syndrome of liver depression and spleen deficiency, which may be realized by the regulation of the PTEN/PI3K/Akt pathway.

摘要:ObjectiveTo observe the clinical effect of modified Yinqiaosan combined with antibiotics in the treatment of acute tonsillitis in children with wind-heat invading lung syndrome.MethodA total of 96 children with acute tonsillitis of wind-heat invading the lung syndrome were randomized into control group and observation group. The control group was treated with routine antibiotics, and the observation group was treated with modified Yinqiaosan and antibiotics for 7 days. The scores of major symptoms (sore throat, erythmatous throat, dysphagia, erythmatous tonsils and suppuration) and minor symptoms (fever, cough, stool, and tongue) and the levels of inflammation- and immune-related indexes [white blood cell (WBC), C-reactive protein (CRP), serum amyloid A (SAA), interleukin-6 (IL-6), and interferon-γ (IFN-γ)] were compared between two groups.ResultThe data of 92 children were statistically analyzed: 45 in the observation group and 47 cases in the control group. The total clinical effective rate of the observation group was 95.56%, as compared with the 93.62% of the control group. After treatment, the scores of major symptoms in the observation group were lower than those in the control group (P<0.05), and the scores of cough, defecation, and tongue in the observation group were lower than those in the control group (P<0.05). The levels of inflammation- and immune-related indexes after treatment in the observation group were lower than those before treatment (P<0.05). Except IFN-γ, the levels of the inflammation- and immune-related indexes in the control group were lower than those before treatment (P<0.05). After treatment, the levels of SAA and IL-6 in the observation group were lower than those in the control group (P<0.05).ConclusionModified Yinqiaosan combined with antibiotics can significantly reduce the expression of SAA and IL-6 in the treatment of children with acute tonsillitis, thereby alleviating inflammation and clinical symptoms and improving prognosis.

摘要:ObjectiveTo observe the clinical effect of modified Jichuanjian on senile patients with slow transit constipation of spleen-kidney yang deficiency syndrome and the influence on brain-gut peptide.MethodA total of 150 senile patients with slow transit constipation were randomized into control group (75 cases) and observation group (75 cases) with the random number table method. The observation group was given modified Jichuanjian (oral, 1 dose/day, 4 weeks), and the control group was treated with Biantong Capsules (oral, 3 capsules/time, twice/day, 4 weeks). Data before and after treatment were recorded, including the score of major constipation symptoms, score of Patient Assessment of Constipation Quality of Life (PAC-QOL), TCM syndrome score, spontaneous complete bowel movements (SCBM), colonic transit test, serum 5-hydroxytryptamine (5-HT), 5-HT 4 receptor (5-HT4R), somatostatin (SS), and vasoactive intestinal peptide (VIP), and recurrence.ResultThe total effective rate of the observation group was 93.06% (67/72), as compared with the 74.65% (53/71) in the control group (χ2=8.974 6, P<0.01). After treatment, the scores of major constipation symptoms, scores of four dimensions of PAC-QOL, total score of PAC-QOL, and TCM syndrome score were lower than those before treatment in the two groups (P<0.01), and lower in the observation group than in the control group (P<0.01). The SCBM in the observation group were more than those in the control group at the 2nd, 3rd, 4th weeks after treatment (P<0.01). The proportions of residual markers at 24, 48, 72 h after treatment were smaller than those before treatment in the two groups (P<0.01), and smaller in the observation group than in the control group (P<0.01). After treatment, the levels of serum 5-HT and 5-HT4R were higher (P<0.01) and the levels of serum SS and VIP were lower (P<0.01) than those before treatment in the two groups. In addition, the levels of serum 5-HT and 5-HT4R in the observation group were higher (P<0.01) and the levels of serum SS and VIP were lower (P<0.01) in the observation group than in the control group. The recurrence in the observation group was 29.85% (20/67) in comparison with the 58.49% (31/53) in the control group (χ2=9.932 4, P<0.01).ConclusionModified Jichuanjian is effective for senile patients with slow transit constipation of spleen-kidney yang deficiency syndrome, which can alleviate clinical symptoms, improve quality of life, regulate the level of serum brain-gut peptide, improve the colonic transit function, increase SCBM, and reduce the recurrence.
摘要:ObjectiveTo study the clinical efficacy of the Qingre Lishi Huazhuo method on patients with chronic gouty arthritis of dampness-heat obstruction syndrome and the effect on nucleotide-binding oligomerization domain-like receptor 3(NLRP3)/interleukin-1β (IL-1β) signaling pathway to preliminarily explore its mechanism.MethodSixty patients with chronic gouty arthritis of dampness-heat obstruction syndrome were enrolled and divided into a treatment group (30 cases) and a control group (30 cases) according to the random number table method. Thirty people were assigned to the healthy group. Patients in the control group were treated with oral Febuxostat, while those in the treatment group were treated with modified Simiaosan combined with Febuxostat. Treatment lasted four weeks. The general clinical data, traditional Chinese medicine (TCM) syndrome scores, serum uric acid (UA), serum creatinine (SCr), blood urea nitrogen (BUN), fasting blood glucose (FPG), low-density lipoprotein (LDL), triglyceride (TG), total cholesterol (TC), erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP) of patients were recorded. Enzyme-linked immunosorbent assay (ELISA) was employed to measure the levels of IL-1β,TNF-α, and IL-6,and the levels of NLRP3,cysteinyl aspartate-specific protease-1 (Caspase-1), and apoptosis-associated speck-like protein containing a CARD (ASC) were detected by Western blot.ResultBefore treatment, the levels of body mass index (BMI), systolic blood pressure (SBP),diastolic blood pressure (DBP),UA,SCr,BUN,FPG,LDL,TG,and TC in both groups significantly increased (P<0.05,P<0.01),and the levels of HDL significantly decreased as compared with those in the healthy group(P<0.05). Additionally, the levels of IL-1β, TNF-α, and IL-6 in both groups significantly increased before treatment (P<0.01). Compared with the results before treatment, patients in the two groups had significant reductions in tube pain, joint tenderness, joint swelling,joint fever, activity disorders, body fatigue, sliminess, bitter mouth, yellow and red urine, and tongue manifestation scores (P<0.05,P<0.01). Compared with patients in the control group after treatment, those in the treatment group had a significant decrease in joint fever, body fatigue, sliminess, bitter mouth,sticky stool,yellow and red urine, tongue manifestation score, and pulse score (P<0.05). The total effective rate in the treatment group was 80.0% (24/30), higher than 56.7% (17/30)in the control group(χ2=11.916,P<0.05). Compared with the results before treatment, BMI, SBP, DBP, UA, SCr, BUN, FPG, LDL, TG, TC, ESR,CRP, IL-1β, TNF-α, IL-6 levels, and VAS score in both groups significantly decreased (P<0.05,P<0.01). Compared with patients in the control group after treatment, those in the treatment group had decreased DBP,ESR, IL-1β levels, and VAS score (P<0.05). Western blot results showed that before treatment, the protein expression of NLRP3, Caspase-1, and ASC in peripheral blood mononuclear cells (PBMCs) of patients in both groups were higher than those in the healthy group (P<0.01). Compared with the results before treatment, the protein expression of NLRP3, Caspase-1, and ASC in PBMCs in patients of both groups after treatment decreased (P<0.05,P<0.01). Compared with the control group after treatment, the treatment group showed decreased expression levels of NLRP3 and Caspase-1(P<0.05).ConclusionThe Qingre Lishi Huazhuo method can effectively improve the clinical symptoms and reduce inflammation of chronic gouty arthritis of dampness-heat obstruction syndrome with good safety. The mechanism may be related to the inhibition of the NLRP3/IL-1β signaling pathway.
摘要:ObjectiveTo analyze the characteristics of kidney Yang deficiency syndrome in different stages and time evolution of chronic kidney disease (CKD) to explore the evolution patterns of kidney Yang deficiency syndrome in CKD.MethodThe evidence information of 256 patients with CKD was collected from October 2020 to September 2022 according to relevant standards, and the "Kidney Yang Deficiency Syndrome Evaluation Scale for Chronic Kidney Disease" was developed. With SPSS Statistics 20.0, SPSS Modeler 18.0, Gephi 0.9.2, and R 4.2.1, the syndrome information of CKD patients at various stages and the syndrome changes after one year were statistically analyzed using complex network analysis, association rule analysis, probability transition matrix analysis, and chi-square test, and the kidney Yang deficiency syndrome of patients at various stages was comprehensively evaluated.ResultIn the CKD population, the proportion of females with kidney Yang deficiency syndrome was higher than that of males (P<0.01), and the proportion of people over 65 years old was higher than in people under 65 years old. The proportion of people with kidney Yang deficiency syndrome increased with the progression of kidney disease, and the proportion of Ⅳ-Ⅴ CKD patients with kidney Yang deficiency syndrome was higher than that of Ⅰ-Ⅱ CKD patients (P<0.01). From Ⅰ CKD to Ⅴ CKD, the frequency of dull tongue continued to increase, and the frequency of enlarged tongue and tooth-marked tongue continued to increase after Ⅲ CKD. The frequency of thick coating and greasy coating ranked in the top 3 of frequency distribution in Ⅴ CKD. After Ⅲ CKD, the top 3 tongue characteristics were weak pulse, deep pulse, and thready pulse, all of which were characteristics of kidney Yang deficiency syndrome. Complex network analysis of the tongue and pulse showed that the core tongue and pulse characteristics of patients with end-stage CKD were tooth-marked tongue with white coating and deep and thready pulse. The results of symptom frequency analysis and complex network analysis showed that aversion to cold and preference for warmth, weakness of the knees, and cold extremities were the top 3 symptoms in Ⅰ-Ⅲ CKD patients with kidney Yang deficiency syndrome, and in Ⅳ-Ⅴ CKD, the manifestations of the syndrome of Yang deficiency and water diffusion, such as drowsiness and fatigue, edema, and frequent urination at night became characteristic symptoms. The scores of edema, pale complexion, soreness and weakness of the waist and knees, loose stools, and mental depression symptoms, as well as the total score of kidney Yang deficiency syndrome gradually increased with disease progression, with statistical differences between different stages of CKD (P<0.05, P<0.01). The frequency analysis of disease-related syndrome elements showed that the frequencies of Yang deficiency syndrome, phlegm-dampness syndrome, blood stasis syndrome, and turbidity-toxin syndrome gradually increased with disease progression, and there were statistically significant differences in the distribution between different stages of CKD (P<0.05, P<0.01). The results of complex network analysis showed that Yang deficiency syndrome was the core syndrome element throughout all stages of CKD and was the main syndrome element type of CKD, while phlegm-dampness syndrome, blood stasis syndrome, and turbidity-toxin syndrome were gradually revealed in the middle and late stages of CKD. In the CKD population with kidney-Yang deficiency syndrome, the distribution of phlegm-dampness syndrome, blood stasis syndrome, and turbidity-toxin syndrome as concurrent syndromes in different CKD stages had statistically significant differences (P<0.05, P<0.01). The association rule analysis showed that as the disease progressed, associations between the concurrent syndromes, such as phlegm-dampness syndrome, blood stasis syndrome, turbidity-toxin syndrome, and fluid retention syndrome, and kidney-Yang deficiency syndrome were gradually enhanced. The comparison of the changes in CKD with kidney Yang deficiency syndrome within one year showed that the disease location was centered on the kidney and transmitted between the spleen, stomach, heart, and liver. There is a 23.81% probability of kidney-Yang deficiency syndrome transforming into Qi deficiency syndromes (Qi deficiency in the spleen and kidney, Qi deficiency in the liver, and Qi deficiency in the heart), 23.79% into Yin deficiency syndromes (Yin deficiency in the liver and kidney, Qi and Yin deficiency, and Yin deficiency in the liver and stomach), and 9.52% into dampness syndromes (phlegm-dampness internal obstruction and wind-dampness obstruction). In contrast, 20% of spleen and kidney Qi deficiency syndrome transformed into kidney Yang deficiency syndrome, and 33.33% of Qi deficiency and blood stasis syndrome transformed into kidney Yang deficiency syndrome.ConclusionAs Ⅰ CKD progresses to Ⅴ CKD, the severity of kidney Yang deficiency syndrome gradually increases, and the syndrome characteristics of kidney Yang deficiency become pronounced. Furthermore, the pathogenic factors, such as phlegm-dampness, blood stasis, and turbidity-toxin, gradually increase. With the change of time, kidney Yang deficiency syndrome in CKD tends to evolve into syndromes related to Qi deficiency, Yin deficiency, and dampness. The discovery of these rules provides a theoretical basis and reference guidance for the treatment of CKD based on syndrome differentiation.

摘要:ObjectiveTo comprehensively evaluate the clinical value of Jintiange Capsules (JCs) in the treatment of osteoporosis (OP) and clarify the intrinsic advantages and clinical treatment characteristics of JCs, providing references for relevant departments of national health and medicine decision-making and the basis and clues for clinical and basic in-depth research.MethodBased on evidence-based medical evidence, this study integrated quantitative and qualitative methods and combined with questionnaires, official website data, human experience, pharmacoeconomic evaluation, and other research methods. From the effectiveness, safety, economy, innovation, suitability, accessibility, and traditional Chinese medicine (TCM) characteristics of the '6+1' dimension, the clinical evidence and value of JCs in the treatment of OP were comprehensively evaluated, forming the 'clinical evidence and value evaluation index'. The comprehensive evaluation of clinical value was based on the multi-criteria decision analysis framework. The expert meeting method was used to empower each dimension and value index. The clinical evidence and value evaluation software of Chinese patent medicine (CSC v2.0) was used to calculate the total value score, and the clinical advantages of JCs were comprehensively evaluated.ResultBased on randomized controlled clinical studies and systematic review, data analysis of spontaneous reporting system (SRS), case reports, non-clinical safety studies, etc., serious adverse drug reactions (ADRs) were reported after the launch of this product monitored by SRS, mainly involving abnormal liver function and adverse reactions of cardiovascular system. Therefore, the safety evidence adequacy of this product should be further improved, and the safety evaluation was Grade B. Meta-analysis showed that JCs were superior to the control group in improving the total clinical effective rate, improving bone mineral density, reducing visual analogue scale (VAS) score, and shortening fracture healing time. Combined with Grading of Recommendations Assessment, Development and Evaluation (GRADE) evidence evaluation, the comprehensive evaluation of effectiveness was Grade A. Pharmacoeconomic evaluation showed that JCs combined with calcium carbonate D3 tablets were more cost-effective than calcium carbonate D3 tablets alone in patients with OP. Compared with Gushukang capsules, JCs had more cost-effectiveness advantages, but the sample size included in the study was small, and the results needed to be verified by further studies. Combined with the results of the Comparative Assessment of Structure Prediction (CASP) evaluation list, the comprehensive economic evaluation was Grade B. JCs were the only bionic medicine of tiger bone in China with all intellectual property rights, with 3 national invention patents. Its process preparation and fingerprint detection had obvious technical advantages. It had innovative advantages in the supply base equipment, medicine resource management, production technology, and other aspects. Thus, its innovative comprehensive evaluation was Grade A. JCs were in capsule dosage form, which was relatively convenient for storage and transportation. The dosage form was suitable for indications, and the usage was easy for patients to grasp and accept. The statutory information and non-statutory information met the national standards. The comprehensive evaluation of suitability was Grade A. JCs did not contain toxic ingredients, had no restrictions on origin and prescription, and had abundant resources of original medicinal materials. The affordability of JCs in the treatment of OP was good in urban areas (14.97%) but not in rural areas (39.76%). The price was higher than that of similar Chinese patent medicines, and the comprehensive evaluation of availability was Grade B. JCs had a reasonable proportion of natural animal bones, and their composition was basically the same as that of natural tiger bones. After marketing, more than 2 000 cases of real-world clinical research evidence was accumulated, and the comprehensive evaluation of TCM characteristics was Grade B. CSC v2.0 software was used for quantitative synthesis of the '6+1' dimension, and the comprehensive clinical value of JCs in the treatment of OP was Grade A.ConclusionJCs have good clinical value in the treatment of OP, and the TCM characteristics are prominent. It is suggested that JCs can be directly transformed into the related policy results of basic clinical drug management according to procedures.
摘要:ObjectiveCYP71 gene family is one of the CYP71 clans belonging to cytochrome P450, which plays an important role in secondary metabolites, especially terpenoid biosynthesis. To understand the characteristics of CYP71 family of diploid Perilla frutescens and predict its function, this study identified and systematically analyzed the family by bioinformatics.MethodOn the basis of the whole genome of diploid P. frutescens PC99, the conserved domains of CYP71 family of diploid P. frutescens were screened, and the sequence characteristics, gene structure, chromosome location, phylogeny and cis-acting elements were analyzed by National Center for Biotechnology Information (NCBI), TBtools, MEME, Molecular Evolutionary Genetics Analysis (MEGA), Cytoscape and other tools.ResultA total of 68 CYP71 genes were identified from diploid P. frutescens, which were unevenly distributed on 34 chromosomes and belonged to two subfamilies. They encoded 481-530 amino acids and contained 10 conserved motifs, with the isoelectric point of 5.70-9.03 and the molecular weight of 54 217.07-60 031.79 Da. The enrichment analysis and functional annotation analysis revealed 11 enriched pathways and 114 categories, and the genes were mainly annotated in biological processes. There were many cis-acting elements in the promoter region of CYP71, mainly light-responsive and methyl jasmonate-responsive elements. Protein-protein interaction analysis indicated that CYP71 protein had multiple functions such as terpene cyclase activity.ConclusionThis study lays a foundation for the functional study of CYP71 family, and provides a reference for the biosynthesis of monoterpenes in P. frutescens and the directional cultivation of excellent varieties.
摘要:ObjectiveBased on the supramolecular "imprinting template" theory, the autonomous action law of the component groups of Shentong Zhuyutang in the preparation process of medicinal materials-decoction pieces-formulas was studied to clarify the quantitative transfer law of its quality attributes.MethodUltra performance liquid chromatography(UPLC) fingerprint of Shentong Zhuyutang was established with mobile phase of 0.4% phosphoric acid aqueous solution(A)-acetonitrile(B) for gradient elution(0-2.5 min, 100%A; 2.5-6 min, 100%-96%A; 6-15 min, 96%-92%A; 15-25 min, 92%-88%A; 25-35 min, 88%-75%A; 35-50 min, 75%-65%A; 50-60 min, 65%-50%A; 60-65 min, 50%-30%A; 65-70 min, 100%A) and detection wavelength of 235 nm, and the total statistical moments, information entropy and primary feeding amount of fingerprint of medicinal materials, decoction pieces and benchmark samples were calculated. Dry extract rate of the benchmark samples, the transfer rates and the addition parameters of medicinal materials-decoction pieces-formulas were calculated.ResultSimilarities of the total statistical moments of UPLC fingerprint of 15 batches of medicinal materials and decoction pieces were>0.89, the relative standard deviations(RSDs) of information entropy of UPLC fingerprint of 12 medicinal materials and decoction pieces were<10%. RSDs of total first-order moment(MCRTT) and information entropy of Shentong Zhuyutang(medicinal materials) were 5.5% and 2.3%, while the RSDs of MCRTT and information entropy of Shentong Zhuyutang(decoction pieces) were 4.8% and 2.6%, respectively. The dry extract rate of 45 batches of Shentong Zhuyutang was 17.2%-20.2%. The transfer rate of medicinal materials to decoction pieces was within the range of data fluctuation, which was 70%-130% of the average value. The overall transfer rates of medicinal materials to decoction pieces and decoction pieces to benchmark samples were 101.8% and 83.0%, respectively.ConclusionThe quality properties of Shentong Zhuyutang benchmark samples can be studied by total statistical moment analysis and primary feeding amount analysis, which can confirm the supramolecular "imprinting template" theory to a certain extent.
摘要:Lancang-Mekong countries refer to the six countries that the Lancang-Mekong River flows through, including China, Laos, Myanmar, Thailand, Cambodia, and Vietnam. These countries are geographically adjacent with similar cultures and have long history of traditional medicine and high plant diversity. Since ancient times, medicinal plants have been introduced into China from the countries along the river, including a variety of medicinal plants with edible and healthcare values, which is an important way for the transnational circulation of medicinal resources. This paper briefly described the history and application of edible medicinal plants in the six Lancang-Mekong countries and summarized more than 150 edible medicinal plants from the other five countries except China. These 150 medicinal plants belong to 66 families such as Labiatae, and 12 species of them are used as edible medicinal plants in all the six countries. Further, we collected the information of these edible medicinal plants, including the origins, efficacy, indications, medicinal edible parts, edible values, and the traditional application of these plants in China. Some valuable edible medicinal plants in the other five countries are considered to have a promising prospect of application in China and may be introduced to China as new medicinal resources. These efforts will be conducive to the cooperation in traditional medicine among Lancang-Mekong countries.

摘要:ObjectiveIn view of the standardization of clinical diagnosis and treatment of the acute abdomen and the inheritance of diagnosis and treatment experience of prestigious veteran traditional Chinese medicine(TCM) doctors, a diagnosis and treatment reasoning algorithm based on association rule mining under incomplete evidence(AMIE)+ random walk was proposed to provide information services and technical support for primary doctors by recommending personalized diagnosis and treatment plans based on medical records.MethodThe experience of diagnosis and treatment of acute abdomen of prestigious veteran TCM doctors and the text data of clinical diagnosis and treatment guidelines of integrated TCM and western medicine were collected to complete the task of knowledge extraction and construct acute abdomen knowledge graph based on Neo4j. On the basis of ontology-supported rule-based reasoning, the rule reasoning based on similar syndromes was used to expand the syndrome combinations whose Jaccard similarity was greater than the threshold in the syndrome recommendation results. The semantic path coverage algorithm was used to calculate the semantic similarity between the symptom nodes. The symptom nodes were divided into 10 categories, and the symptom nodes in the same category were extended. The random walk algorithm was used to search the symptom nodes connected with the syndrome, and the connection rules between the syndrome and symptom nodes were extended to realize the knowledge reasoning of AMIE+ random walk.ResultThe acute abdomen knowledge graph included 1 320 nodes and 2 464 relationships. According to the link prediction evaluation index of knowledge reasoning, the reasoning results of the three algorithms in the auxiliary diagnosis and treatment of acute abdomen were compared. The AMIE+ random walk algorithm complemented the knowledge graph by extending the similar syndrome connection rules and the syndrome-symptom connection rules. Compared with the knowledge reasoning algorithm based on ontology rules, the area under the curve (AUC) was 15.18% higher and the accuracy was 30.36% higher, which achieved more accurate and effective knowledge inference.ConclusionThis study used knowledge graph technology to visualize the diagnosis and treatment of acute abdomen with TCM and western medicine, assisting primary clinicians in intuitively viewing the diagnosis and treatment process and data relationship. The proposed diagnosis and treatment reasoning algorithm can realize the personalized diagnosis and treatment plan recommendation at the level of "disease-syndrome-diagnosis-treatment-prescription", which can assist primary doctors in disease diagnosis and treatment and clinical decision-making, contribute to the knowledge sharing and application of diagnosis and treatment experience and clinical guidelines of prestigious veteran TCM doctors, improve the level of primary clinical diagnosis and treatment, and promote the normalization and standardization of the diagnosis and treatment process of acute abdomen with integrated TCM and western medicine.

摘要:Epilepsy is a chronic brain disease characterized by seizures, and is one of the most common nervous system diseases in clinic practice with the recurrent, transient, and refractory characteristics. Clinically, western medicine therapy is mainly adopted in the treatment of epilepsy, but it is not conducive to long-term use for patients on account of severe side effects, which can result in abnormalities in the digestive system, central nervous system, hematopoietic system, urinary system, and liver function to varying degrees. Syndrome differentiation is usually used for the treatment of epilepsy by traditional Chinese medicine (TCM), which can avoid the side effects of western medicine treatment on the basis of improving patients' syndromes. The literature on TCM in the treatment of epilepsy in China and abroad indicates that the syndrome differentiation in TCM is often based on phlegm, blood stasis, wind, and deficiency, and the treatment methods include acupuncture, acupoint catgut embedding, moxibustion, Chinese medicine monomer, drug pair, and compound decoction. The various treatments of TCM play an important role in the comprehensive treatment of epilepsy through multiple channels and links, such as reducing the degree and number of seizures. This paper comprehensively summarized the clinical experience of TCM in the treatment of epilepsy, systematically expounded various treatment methods and ideas of TCM in the treatment of epilepsy, and deeply discussed the mechanism of TCM in the treatment of epilepsy, aiming to provide a theoretical basis for the clinical formulation of a reasonable individualized treatment plan for epilepsy and diversified ideas for the more effective treatment of epilepsy by TCM.

摘要:Diabetic kidney disease (DKD), a major microvascular complication of diabetes mellitus, serves as the most common cause of end-stage renal disease worldwide. The progression of DKD is closely related to oxidative stress, inflammatory response, apoptosis, and fibrosis in renal tissues activated by high glucose. Numerous studies have shown that the transduction of the p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway is involved in the pathological process of DKD in renal tissues, activating various pathological mechanisms, such as oxidation, inflammation, apoptosis, and fibrosis. Therefore, blocking the transduction of the p38 MAPK signaling pathway is beneficial to alleviating DKD. At present, the main treatment principles of western medicine are glucose lowering, lipid lowering, and blood pressure lowering, as well as medications with new drugs renal sodium-glucose co-transporter 2 (SGLT2), mineralocorticoid receptor, and endothelin receptor, but the progression of DKD still cannot be stopped. The treatment of DKD by traditional Chinese medicine (TCM) has the advantages of simplicity, low cost, and convenience, and the symptoms and root causes can be both treated. In recent years, the basic research on Chinese medicine intervention in DKD has greatly advanced, and p38 MAPK is the key factor of Chinese medicine intervention in DKD. The present study searched and reviewed the literature on the Chinese medicine intervention in the p38 MAPK signaling pathway in DKD treatment in the past decade. The results showed that p38 MAPK interacted with transforming growth factor-β1 (TGF-β1), cysteinyl aspartate-specific protease-3 (Caspase-3), nuclear factor-κB (NF-κB), and other factors to activate fibrosis, inflammation, oxidative stress, and apoptosis. By acting on p38 MAPK and its upstream and downstream factors, Chinese medicine blocked the pathological processes of DKD and inhibited the pathological injury of DKD and the deterioration of renal function. This study is expected to provide new ideas and directions for the prevention and treatment of DKD with Chinese medicine.

摘要:Ulcerative colitis (UC) is a common chronic digestive system disease in clinic. The disease is repeated and difficult to treat, and the pathogenesis is complex, which is related to oxidative stress response. Nuclear factor E2-related factor 2 (Nrf2) is an important factor in antioxidant reaction, which regulates the expression of downstream heme oxygen-1 (HO-1), and plays a role in maintaining redox homeostasis. In the course of UC, the biological activity and content of Nrf2 and HO-1 are decreased, the antioxidant and anti-inflammatory abilities of tissues are weakened, the intestinal epithelial cells are damaged, and the intestinal mucosal barrier is destroyed. At present, western medicine mainly focuses on controlling inflammation and alleviating symptoms in clinical treatment. Although it has a certain effect, there are many problems such as easy recurrence after drug withdrawal and long-term side effects. Studies have shown that Chinese medicine has rich and flexible therapeutic methods and has broad application prospects in the prevention and treatment of UC. In recent years, with Nrf2/HO-1 pathway as the entry point, a large number of basic and clinical experiments on this signal in UC have been carried out in the field of Chinese medicine, and the results show that the intervention of Nrf2/HO-1 pathway is an important potential target for the treatment of UC by Chinese medicines. Based on the etiology and pathogenesis of deficiency-excess in complexity, Chinese medicine regulates Nrf2/HO-1 pathway by clearing heat and detoxifying dampness, activating blood circulation to remove stasis and relieve pain, invigorating Qi, tonifying middle, and warming interior, and treating both symptoms and root causes, to improve the tissue's ability to resist oxidative stress, maintain the balance between pro-inflammatory factors and anti-inflammatory factors, relieve inflammatory response, and play a therapeutic role in UC. This paper summarized and analyzed the effect of Chinese medicine targeting the Nrf2/HO-1 signaling pathway on interfering with UC and its mechanism. The purpose of this study is to provide references for researchers to have a more comprehensive understanding of the mechanism of Chinese medicines in the Nrf2/HO-1 signaling pathway in UC and promote the rational application of Chinese medicines in the prevention and treatment of UC in the future.
摘要:Bone metastasis-caused pain (BMP) is a common complication of cancer, and the incidence has been on the rise with the increase in the overall prevalence of cancer, threatening the survival and quality of life of patients. BMP is a kind of special pain with the characteristics of inflammatory pain and neuropathic pain, but is different from the two. Therefore, its pathogenesis is very complicated, and it is of great significance to understand the pathogenesis. The currently available studies mainly focused on osteoclast activation, changes in the bone microenvironment, glial cell activation, spinal cord neuron activation, and miRNA dysregulation. Modern therapies include the three-step analgesics, bisphosphonates, palliative radiotherapy, and interventional therapy for bone metastases, which show definite efficacy in short term. However, the long-term effect is unsatisfactory due to the adverse reactions, addiction, and drug resistance. Studies have shown that traditional Chinese medicine (TCM) has definite curative effect on BMP, which is safe, enhances efficacy, reduces toxicity, and boosts immunity. Moreover, it exerts the effect through multiple components, multiple targets, and multiple pathways. As a result, it has unique advantages in the prevention and treatment of BMP and has become a research focus. This paper summarizes the research on the pathogenesis of BMP, the intervention of TCM (compound Chinese medicine prescriptions, Chinese medicinals, and monomers from Chinese medicinals), and the mechanisms of TCM, such as inhibiting osteoclast activation, glial cell activation, and spinal cord neuron activation, regulating pain mediators and abnormal expression of microRNA, and anti-tumor, which is expected to further clarify the pathogenesis of BMP and provide ideas and methods for the effective prevention and treatment of BMP with TCM.

摘要:Heart failure is the final stage of heart disease caused by a variety of etiologies and has a high morbidity, mortality, and disability rate, making it a major challenge in the field of medicine. Cardiomyocytes, the most basic unit of the heart, are irreversible in nature and can be damaged or necrotic in various ways in the presence of heart failure. Myocardial cell injury is also an important cause of cardiac dysfunction and affects the prognosis and quality of life of patients. Therefore, reducing the level of myocardial cell damage and delaying the process of cell death can help patients with heart failure lessen the extent of cardiac damage and improve their prognosis, thereby lowering the incidence of death and disability and times of hospitalization. Ferroptosis is a new form of cell death that has been widely concerned in recent years, with studies confirming its occurrence in cardiac myocytes. As a modifiable form of cell death, interfering with ferroptosis can modulate the extent of injury and death in cardiac myocytes. Studies have shown that the inhibition of iron death has a protective effect on cardiomyocytes, thereby alleviating heart failure. Chinese medicine has been widely used in the clinical treatment of heart failure, and has the advantages of multiple approaches and entry points, with significant therapeutic effects, low side effects, and low medical costs. It also reduces the clinical side effects of western medicine, with good clinical results. The use of Chinese medicine to modulate ferroptosis may be a new direction for the future treatment of heart failure. This paper briefly elaborated on the mechanism of ferroptosis, investigated the role of ferroptosis in heart failure, and discussed the current status of research on ferroptosis in Chinese medicine interventions in heart failure, to provide references for further improving the efficacy of Chinese medicine in the treatment of heart failure.
摘要:Hyperlipidemia is a dyslipidemia caused by dyslipidemia of lipid metabolism, which can be divided into primary and secondary types. The current clinical diagnostic criteria are mainly changes in lipid levels, which are the inducers of high-risk cardiovascular diseases such as atherosclerosis, pancreatitis and coronary heart disease. As a key target in lipid metabolism, peroxisome proliferator-activated receptor α (PPARα) is involved in a variety of metabolic activities, including fatty acid degradation, synthesis, transport, storage, lipoprotein metabolism, etc. Activation of PPARα can maintain the balance of lipid metabolism through a variety of ways, which is an important way to treat hyperlipidemia. At present, chemical drugs such as statins and bettes are mainly used in the clinical treatment of hyperlipidemia. Although they can slow down the disease to a certain extent, there are many adverse reactions and drug resistance. By reviewing the literature in recent years, the author found that the activation of PPARα pathway by traditional Chinese medicine in the treatment of hyperlipidemia has significant effect and small adverse reactions. The lipid-lowering active ingredients include flavonoids, alkaloids, phenols, terpenoids and other compounds. These active components mainly affect the expression of downstream effectors through the activation of PPARα pathway, thereby inhibiting the synthesis of total cholesterol and promoting fatty acid oxidation, and play a role in the treatment of hyperlipidemia. In this paper, we systematically reviewed the structure types and mechanism of active components of traditional Chinese medicine that activate PPARα pathway, so as to provide guidance for the rational development and clinical application of lipid-lowering traditional Chinese medicine new drugs.

摘要:Age-related ovarian hypofunction includes a decrease in follicle quantity and quality as well as alterations in the ovarian microenvironment,the mechanisms of which are mainly related to mitochondrial dysfunction,free radical and antioxidant systems,telomere and telomerase alterations,and apoptosis,and is one of the major factors contributing to infertility in advanced maternal age (AMA). Despite the tremendous progress in assisted reproductive technology in recent decades,few breakthroughs have been made in alleviating age-related ovarian hypofunction and improving reproductive outcomes for AMA. In recent years,there has been an increasing number of studies on the multi-level and multi-targeted mechanisms of traditional Chinese medicine (TCM) to improve age-related ovarian hypofunction by modulating mitochondrial homeostasis,alleviating oxidative stress,and inhibiting apoptosis,while more high-quality randomized controlled trials have demonstrated the clinical efficacy of TCM in assisted reproductive technology. Given this,this article presented a systematic review of recent research and randomized controlled trials on the mechanism of Chinese medicine active ingredients,single Chinese medicine, and Chinese medicine compounds in delaying age-related ovarian hypofunction,to clarify the current status and shortcomings of the research. This paper provides medication management of TCM for effectively alleviating age-related ovarian hypofunction and improving reproductive outcomes for AMA.
摘要:Type 2 diabetes mellitus (T2DM) is a chronic metabolic disease characterized by insulin resistance, hyperinsulinemia, and disturbance of glucose and lipid metabolism, with elevated blood glucose as the main clinical manifestation. Due to its complex etiology and pathogenesis, there is no effective treatment, which critically threatens human health and places a heavy burden on society and families. Saponins are a class of glycosides with complex structures that have the advantage of a wide range of sources, elevated safety, and low adverse effects. As an essential active ingredient in Chinese medicine, Chinese medicine saponins have a variety of biological activities such as hypoglycemia, hypoglycaemia, anti-inflammation, antioxidation, anti-tumor, and immune modulation. In recent years, numerous studies have shown that Chinese medicine saponins are effective in preventing and treating T2DM. Although there have been numerous studies on the hypoglycemic effects and mechanisms of Chinese medicine saponins, there has been no systematic review of the mechanisms of Chinese medicine saponins in the treatment of T2DM. Therefore, to provide a theoretical basis for an in-depth study of the hypoglycemic effects of Chinese medicine saponins and a scientific basis for the development and clinical application of drugs, this paper systematically summarized the hypoglycemic mechanisms of Chinese medicine saponins, such as improving islet β-cell function, improving insulin resistance, inhibiting glycosidase activity, reducing the inflammatory response, anti-oxidative stress, and regulating intestinal flora, and analyzed the current research problems and development trends.

摘要:Type 2 diabetes mellitus (T2DM) is a heterogeneous disease with insulin deficiency and insulin resistance (IR) as the main etiology and is often accompanied by complications. Volatile oil is a volatile oily liquid extracted from natural plants, which has many pharmacological effects such as regulating Qi, relieving pain, inhibiting bacteria, and reducing inflammation. In recent years, there have been numerous reports on the treatment of T2DM by natural plant volatile oil and its effective components, which has become one of the new directions in the treatment of T2DM. With natural plant essential oil and its active components as the starting point, this paper comprehensively analyzed and summarized the material basis, mechanism, and signaling pathways of essential oil in the treatment of T2DM and its complications in China and abroad in recent years, and focused on the inhibitory effect of essential oil and its active components, such as carvacrol, paeonol, and β-caryophylene, on IR to improve T2DM by protecting pancreatic β-cells, inhibiting α-glucosidase activity, regulating the abundance and diversity of intestinal microbiota, and regulating glucose transporter protein type4 (GLUT4), adenylate 5′-monophosphate-activated protein kinase (AMPK), phosphatidylinositol-3 kinase (PI3K)/protein kinase B (Akt) signaling pathways to provide some references for the volatile oil intervention in T2DM and the development of new green antidiabetic drugs.
