摘要:By reviewing ancient materia medica, medical books and modern literature, the name, origin, quality evaluation, producing area and processing methods of Huoxiang herbs were systematically investigated and researched, so as to provide reference and basis for the development and utilization of famous classical formulas containing Huoxiang herbs. Through the herbal textual research, it can be seen that most of materia medica in past dynasties have taken Huoxiang as the nominal rectification, and the mainstream base used is Pogostemon cablin. In order to distinguish another plant of the same family, Agastache rugosa, which has been widely used in Chinese folk since the Ming dynasty, and respect geo-authentic region, Pogostemonis Herba is also named Guanghuoxiang. Pogostemonis Herba is native to Vietnam and other Southeast Asian countries, and was introduced to China as a spice through Guangdong and other places in the early days, and has been successfully cultivated in the south of China since the Song dynasty. The medicinal parts are mostly dried aboveground parts, and the leaves and stems are also separated for medicine sometimes. The geo-authentic region of Pogostemonis Herba is Guangdong in the past dynasties, and it is currently cultivated in Guangzhou, Zhaoqing, Zhanjiang of Guangdong province and Hainan province, with the most famous one cultivated in Shipai. Pogostemonis Herba is mainly planted by cutting propagation. It usually sprouts in February and is harvested in June. The main processed method in region is stuffy dry, which is placed in the sun and repeatedly suffocated until it has an aromatic smell and the color turns yellow. The processing method is mostly to use the raw product as medicine after being selected. Based on the research results, it is suggested that the leaves of P. cablin are used in Yangweitang, for Huopo Xialingtang, it is recommended to choose the raw product of A. rugosa that is removed the roots and old stems.
摘要:Benzoinum has a long history of medicinal use. In order to standardize its clinical use, the author made systematic textual research on the name, origin, evolution of scientific name, producing area, property, quality evaluation, harvesting and processing of Benzoinum by reviewing the ancient materia medica, medical books and modern literature. According to the research, the mainstream variety of Benzoinum recorded in ancient materia medica should be the dry resin of Styrax tonkinensis of Styracaceae. In addition to S. benzoin, which was also used in many applications. In modern times, S. benzoin was the mainstream, but in practice, it was mostly the resin of various plants of the genus Styrax, and then S. tonkinensis and S. benzoides were recorded as the main sources of Benzoinum. Nowadays, it mainly comes from S. tonkinensis. In ancient times, the producing areas of Benzoinum were Persia, Xirong, Annan and other places, whereas now are distributed in Indonesia, Sumatra, Thailand, Vietnam, and Fujian, Guizhou, Yunnan, Guangxi, Guangdong and other places in China, but most of the medicinal materials in China are imported. Traditionally, the medicinal materials of Benzoinum with properties of oily, waxy luster, brittle and fragile, strong aroma, sand feeling when chewing and no impurities are considered better. In antiquity, the harvesting time of Benzoinum was concentrated in July and August, while in modern times, it is harvested in summer and autumn, and collected in various ways, usually the trees are selected to be harvested if they are more than 10 years old, and the best quality being the first, milky resin collected when the time of cutting the trees is from April to June. In the past dynasties, the powdered resin was mainly ground and used in formulas, in addition, there were also records of wine steaming into paste and processing with honey, etc. The 1953 edition of Chinese Pharmacopoeia added records such as sevum benzoinatum and adeps benzoinatus, and now it is mostly used as powder into pills or powers, or used after processing with wine. Based on the results of textual research, it is suggested that although the dry resin of S. benzoin is not a medicinal variety of Benzoinum included in the 2020 edition of Chinese Pharmacopoeia, it has a long history of application and has some clinical value, so its medicinal feasibility can be explored in depth. In the development of famous classical formulas containing Benzoinum, the dry resin of S. tonkinensis or S. benzoin can be used, which should be used in medicine as powder after dried or processed according to the requirements of prescriptions.

摘要:By reviewing the ancient materia medica, medical books and modern literature, this paper made a systematic textual research on Haliotidis Concha in famous classical formulas, including the name, origin, producing area, quality evaluation, harvesting and processing, in order to provide a basis for the development of famous classical formulas containing Haliotidis Concha. The textual research showed that Shijueming was the official name of Haliotidis Concha in past dynasties, and there were also aliases such as Qianliguang, Jiukongluo and Zhenzhumu named after its efficacy, properties and near-phonetic characters. Before the Tang dynasty, the original description of Haliotidis Concha was too concise, which could only be identified as the animal of genus Haliotis, family Haliotidae. During the Song, Yuan, Ming and Qing dynasties, the main varieties were H. diversicolor and H. discus hannai. At the beginning of the Republic of China, a variety of animals from genus Haliotis were used as Haliotidis Concha, and varieties were numerous and continued to this day. In ancient and modern times, the main producing areas in China are Hainan, Guangdong and Shandong, while the foreign producing areas are mainly Japan and Vietnam. The quality evaluation of Haliotidis Concha in ancient books was roughly determined by the number of openings of the expiratory orifice, and seven-hole and nine-hole abalone shells were preferred. In modern times, characters as big, neat, unbroken, clean inside and outside, lustrous, thick shells are preferred. Based on the textual research results and combined with the record years of the Shijueming San, it is suggested that the shells of H. diversicolor or H. discus hannai should be used in the development of this formula, and the raw products should be used as medicine.
摘要:By reviewing the materia medica, medical books and scripture history, combining modern and contemporary literature and field investigation, this paper systematically reviewed the name, origin, scientific name, producing area, quality, harvesting and processing of Scorpio in famous classical formulas to clarify the relationship between ancient and modern times and provide a reference basis for the development of related famous classical formulas. After the textual research, it can be seen that there are many names of Scorpio, and most of the materia medica use Xie or Quanxie as the official name. The origin of Scorpio used in the past dynasties is Buthus martensii, which is the same in ancient and modern times. B. martensii is mainly distributed in the central and eastern parts of China, as well as Mongolia, Korea and other places, located in East Asia. Therefore, pharmaceutical workers in China mostly continue to use the early Chinese name, that is, Dongya Qianxie, while modern Scorpiones taxonomists set its Chinese name as Mashi Zhengqianxie. In order to maintain the stability and continuity of the origin of Scorpio, the previous editions of Chinese Pharmacopoeia have always used the name of B. martensii. The geo-authentic producing area of Scorpio, which has been respected in the past dynasties, was Qingzhou, Shandong. Until the Republic of China, due to the different processing methods in the production area, the geo-authentic producing area expanded to Yu county, Henan, with Yu county as the distribution center, the best quality of Scorpio is produced by boiling in clear water and drying after boiling. The origin processing of Scorpio is mainly divided into clean water and salt water boiling and then drying, where the method of boiling with salt water was first described in Bencao Yuanshi, the purpose is antiseptic and suitable for storage, and the salt should be washed away when used clinically. There are few processing methods of Scorpio, in ancient times, it was used for roasting or frying after removing the feet, in modern times, it is mostly to remove impurities, wash and dry. Scorpio is not only used for medicinal purposes, but also has the habit of eating in many areas, so the consumption of resources is relatively large, and it is still mainly harvested from the wild, resulting in the decrease of wild resources year by year. Based on the research conclusion, it is recommended that B. martensii, which is produced in Qingzhou, Luyi or Yuzhou, should be used in the the development of famous classical formulas containing Scorpio, and the origin processing is preferable to be dried after boiling with water, and the processing specification should be selected as raw products. And B. martensii in geo-authentic producing areas was used as the seed source to establish a standardized breeding base to ensure the sustainable development of the resources of Scorpio.
摘要:ObjectiveTo study the effect of modified Erchentang on the expression of key molecules in the high mobility group Box 1 protein (HMGB1)/receptor for advanced glycation endproduct (RAGE)/nuclear factor-κB (NF-κB) signaling pathway in bronchioles of rats with chronic obstructive pulmonary disease (COPD), to explore the mechanism of modified Erchentang against bronchiolar inflammation of COPD rats via HMGB1/RAGE/NF-κB signaling pathway.MethodSixty SD rats were randomly divided into normal group, model group, modified Erchentang low-, medium- and high-dose groups (5, 10, 20 g·kg-1·d-1) and ethyl pyruvate (HMGB1 inhibitor) group, with 10 in each group. The COPD rat model was prepared by cigarette smoke combined with tracheal injection of lipopolysaccharide (LPS). After modeling, the modified Erchentang groups were given corresponding drugs (ig) and Ringer's solution (4 mL, ip), while the EP group was treated with equal volume of normal saline (ig) and EP (0.04 g·kg-1·d-1, ip). The normal group and the model group received equal volume of normal saline (ig) and Ringer's solution (ip) for 21 consecutive days. The contents of HMGB1, chemokine (C-X-C motif) ligand 1 (CXCL1), CXCL2 and monocyte chemotactic protein-1 (MCP-1) in bronchoalveolar lavage fluid (BALF) were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA expressions of HMGB1, RAGE and NF-κB p65 were determined by Real-time polymerase chain reaction (Real-time PCR), and the protein expressions of HMGB1, RAGE, p-NF-κB p65, and alpha-smooth muscle actin (α-SMA) in bronchioles tissue of rats were determined by immunohistochemistry (IHC).ResultCompared with the conditions in the normal group, the forced vital capacity (FVC), forced expiratory volume in the first second (FEV1) and FEV1/FVC in the model group were decreased (P<0.01) while the contents of HMGB1, CXCL1, CXCL2 and MCP-1 in BALF were increased (P<0.01). And the model group presented higher mRNA expressions of HMGB1, RAGE and NF-κB p65 (P<0.01) and protein expressions of HMGB1, RAGE, p-NF-κB p65 and α-SMA (P<0.05, P<0.01) than the normal group. Compared with the model group, the modified Erchentang medium- and high-dose groups had increased FEV1/FVC (P<0.05, P<0.01), lowered contents of HMGB1, CXCL1, CXCL2 and MCP-1 in BALF (P<0.05, P<0.05), and reduced mRNA expressions of HMGB1, RAGE and NF-κB p65 (P<0.05, P<0.01) and protein expressions of HMGB1, RAGE, p-NF-κB p65 and α-SMA (P<0.05, P<0.01).ConclusionModified Erchentang can resist bronchiolar inflammation of COPD rats. The mechanism may be related to down-regulating the mRNA expressiona of HMGB1 and RAGE, inhibiting the activity of NF-κB, and reducing the release of HMGB1, CXCL1, CXCL2 and MCP-1, thus suppressing the inflammatory injury and abnormal repair of bronchioles.

摘要:ObjectiveTo observe the effect of Banxia Xiexintang on the autophagy of interstitial cells of Cajal (ICCs) in the gastric antrum of rats with gastric electric dysrhythmia, and explore the protective effect and regulatory mechanism.MethodThirty-two SD rats were randomly assigned into a normal group, a model group, a Banxia Xiexintang (24.68 g·kg-1) group, and a positive drug (2.7 mg·kg-1) group. The rat model of gastric electric dysrhythmia was established by the method of dieting every other day and drinking dilute hydrochloric acid, and Banxia Xiexintang and the positive drug were administrated for intervention. The body weight of each rat was recorded weekly. The gastric electric activity was recorded by the biological function experimental system. The ultrastructural changes of the gastric antrum tissue were observed by a transmission electron microscope. The co-expression of receptor tyrosine kinase (c-kit)/mammalian homolog of yeast Atg6 (Beclin1) in the gastric antrum tissue was detected by double immunofluorescence labeling method. The expression of microtubule-associated protein 1 light chain 3B (LC3B) and p62 protein in the gastric antrum tissue was determined by Western blot, and the LC3BⅡ/Ⅰ ratio was calculated.ResultCompared with the normal group, the modeling reduced the body weight (P<0.01) and decreased the dominant frequency and dominant power of gastric electricity (P<0.01). In addition, the modeling caused ultrastructural damage of ICCs in gastric antrum, degeneration and necrosis of organelles, and appearance of a small number of autophagic vesicles. The results of double immunofluorescence labeling showed that the modeling inhibited the positive expression of c-kit and promoted the positive expression of Beclin1 in gastric antrum tissue. Western blot results showed that the modeling increased the ratio of LC3BⅡ/Ⅰ (P<0.01) and down-regulated the expression of p62 protein (P<0.01) in the gastric antrum tissue. Compared with the model group, Banxia Xiexintang and the positive drug increased the body weight (P<0.01) and the dominant frequency and dominant power of gastric electricity (P<0.01), repaired the ultrastructural damage of ICCs in gastric antrum tissue, promoted the positive expression of c-kit and inhibited the positive expression of Beclin1 in the gastric antrum tissue. Furthermore, Banxia Xiexintang up-regulated the expression of p62 (P<0.05) and inhibited the transformation of LC3BⅠ into LC3BⅡ in gastric antrum tissue (P<0.05).ConclusionBy regulating the expression of autophagy-related proteins, Banxia Xiexintang can reduce the autophagy and regulate the number and structure of ICCs and thus improve the gastric electric rhythm of rats, which preliminarily explains the mechanism of Banxia Xiexintang in the treatment of epigastric stuffiness.

摘要:ObjectiveTo investigate the effect of Danzhi Xiaoyaosan on the phosphorylation of tau protein and different sites of glycogen synthase kinase-3β (GSK-3β) and phosphoseryl/suanyl phosphate protein phosphatase 2A (PP2A) in the hippocampus of rats with Alzheimer's disease (AD) and its mechanism.MethodThe rat model of AD was established by injecting okadaic acid into the bilateral hippocampus of 90 male Wistar rats in SPF grades. The rats with successful modeling were selected and randomly divided into model group, aricept group (0.5 mg·kg-1), and Danzhi Xiaoyaosan high, medium, and low groups (17.55, 8.77, and 4.38 g·kg-1), and then gavaged for 42 d, once a day. Morris water maze was used to detect the learning and memory ability of rats, Nissl's staining was used to observe the morphological structure of neurons in the hippocampus, and Real-time polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression levels of tau protein, GSK-3β, and PP2A. Western blot was used to determine the protein expression levels of tau protein, GSK-3β, and PP2A.ResultAs compared with the control group, the learning and memory abilities of the rats in the model group were significantly decreased (P<0.01), and the hippocampal CA3 region cells had abnormal structure, disorderly arrangement, and decreased number. The expression levels of GSK-3β mRNA, GSK-3β, p-GSK-3β-Tyr216, p-PP2A, and p-tau were increased in the model group as compared with the control group (P<0.01), and those of p-GSK-3β-Ser9 and PP2A decreased significantly (P<0.01). As compared with the model group, the learning and memory ability of the Aricept group and the Danzhi Xiaoyaosan groups were improved (P<0.05, P<0.01), and the cell morphology and the number of hippocampal CA3 regions were better. The mRNA expression levels of PP2A and tau in the Aricept group were significantly up-regulated (P<0.05), the mRNA expression level of GSK-3β was significantly down-regulated (P<0.01), and the protein expression levels of GSK-3β, p-GSK-3β-Tyr216, and p-PP2A were down-regulated (P<0.05, P<0.01), and the protein expression level of PP2A was significantly up-regulated (P<0.01). As compared with the model group, the mRNA expression level of PP2A in the high-dose Danzhi Xiaoyaosan group was significantly up-regulated (P<0.01), and that of GSK-3β was significantly down-regulated (P<0.01), whereas the protein expression levels of p-PP2A, p-GSK-3β-Tyr216, and p-tau were down-regulated (P<0.05, P<0.01), and the protein expression level of PP2A was significantly up-regulated (P<0.01). As compared with the model group, the mRNA expression level of GSK-3β was significantly down-regulated in the medium-dose Danzhi Xiaoyaosan group (P<0.01), the protein expression levels of GSK-3β, p-GSK-3β-Tyr216, and p-tau were down-regulated (P<0.05, P<0.01), and the protein expression level of PP2A was significantly up-regulated (P<0.01). As compared with the model group, the mRNA expression level of PP2A was significantly up-regulated in the low-dose Danzhi Xiaoyaosan group (P<0.01), and that of GSK-3β was significantly down-regulated (P<0.01), whereas the protein expression levels of GSK-3β and p-GSK-3β-Tyr216 were down-regulated (P<0.05, P<0.01), and those of p-GSK-3β-Ser9 and PP2A were significantly up-regulated (P<0.01).ConclusionDanzhi Xiaoyaosan can improve the learning and memory ability of rats with AD, and its mechanism may be related to the regulation of the activities of GSK-3β and PP2A protein-related sites and the phosphorylation of tau protein.

摘要:ObjectiveTo explore the inhibitory effect of different concentration of baicalin (0, 100, 200, 400 μmol·L-1) on the proliferation of human gastric cancer SGC-7901 cells and the underlying mechanism.MethodSGC-7901 cells were treated with baicalin. Then methyl thiazolyl tetrazolium (MTT) assay was employed to examine the inhibitory effect of baicalin on the cells. At the same time, ferrostatin-1 (Fer-1) was added to observe the viability of cells after baicalin treatment. The expression of ferroptosis-related genes was detected by Real-time polymerase chain reaction (Real-time PCR) and Western blot. The content of malondialdehyde (MDA) and the level of glutathione (GSH) were detected respectively by MTT assay and enzyme-linked immunosorbent assay. The role of tumor protein 53 (p53)/solute carrier family 7 member 11 (SLC7A11) pathway in the regulation of ferroptosis was investigated respectively via overexpression and small interfering RNA (siRNA) methods.ResultCompared with the blank group, baicalin decreased the viability of SGC-7901 (P<0.05, P<0.01) in a dose- and time-dependent manner. The intervention of Fer-1 significantly alleviated the decrease of SGC-7901 cell viability caused by baicalin (P<0.01). In addition, compared with the baicalin group, Fer-1+baicalin group showed decrease in MDA content and the mRNA and protein levels of prostaglandin-endoperoxide synthase 2 (PTGS2) in the cells (P<0.01), and increase in GSH activity and mRNA and protein levels of glutathione peroxidase 4 (GPX4) (P<0.01). The protein level of SLC7A11 in the baicalin group was decreased compared with that in the blank group (P<0.05, P<0.01) in a dose-dependent manner. Compared with the baicalin group, the reactive oxygen species (ROS) level and MDA content in SLC7A11-overexpressing cells were significantly decreased after baicalin treatment (P<0.01), and the GSH activity was significantly increased (P<0.01). The fluorescence intensity of p53 in the cells of the baicalin group was increased compared with that of the blank group (P<0.01). Compared with the baicalin group, the expression level of p53 protein in the cells transfected with p53 siRNA was significantly decreased after baicalin treatment (P<0.01), and the expression level of SLC7A11 was significantly increased (P<0.01).ConclusionBaicalin can effectively inhibit the proliferation of SGC-7901 cells by regulating p53/SLC7A11-mediated ferroptosis.

摘要:ObjectiveTo investigate the protective effect and regulatory mechanism of berberine (BBR) against the senescence of ovarian granulosa cells.MethodA cell senescence model in the human ovarian granulosa-like tumor (KGN) cell line was induced by H2O2. A control group, a model group, and high-dose (1 μmol·L-1) and low-dose (0.5 μmol·L-1) BBR groups were set up. The cells in the model group and the BBR groups were incubated with 10 μmol·L-1 H2O2 for 40 min. The effect of BBR on KGN cell proliferation was detected by cell counting kit-8 (CCK-8) assay. The effect of BBR on the senescence of KGN cells was detected by β-galactosidase staining. The effects of BBR on the apoptosis and ROS content of KGN cells were detected by flow cytometry. The effects of BBR on the mRNA expression of B-cell lymphoma-2 (Bcl-2)/Bcl-2-associated X protein (Bax), cysteinyl aspartate-specific protease-3 (Caspase-3), forkhead transcription factor O1 (FoxO1), and catalase (CAT) was detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Western blot was used to detect the effects of BBR on protein expression of silent information regulator1 (SIRT1), superoxide dismutase 2 (SOD2), c-Jun N-terminal kinase (JNK), FoxO1, autophagy-associated protein microtubule-associated protein light chain 3Ⅱ (LC3BⅡ), mammalian ortholog of yeast Atg6 (Beclin-1), and ubiquitin-binding protein p62.ResultAfter H2O2 induction for 40 min, the cell proliferation rate of the model group decreased compared with that of the control group (P<0.01), and the cell proliferation rates of the BBR groups increased compared with that of the model group (P<0.05). The results of β-galactosidase staining showed that the cells of the model group showed significant senescence compared with those of the control group (P<0.01), and the cellular senescence in the BBR groups was reduced compared with that of the model group (P<0.01). As revealed by flow cytometry, compared with the control group, the model group showed increased apoptosis rate (P<0.01), and compared with the model group, BBR groups showed decreased apoptosis rates (P<0.05). Meanwhile, the ROS content in the model group increased compared with that in the control group (P<0.01), and compared with the model group, the BBR groups showed reduced cellular ROS content (P<0.01). The Real-time PCR results showed that compared with the control group, the model group showed decreased mRNA expression of CAT and Bcl-2/Bax in KGN cells and increased mRNA expression of Caspase-3 and FoxO1 (P<0.05), and compared with the model group, the BBR groups showed increased mRNA expression of CAT and Bcl-2/Bax (P<0.05) and reduced mRNA expression of Caspase-3 and FoxO1 in KGN cells (P<0.05). As revealed by Western blot results, SIRT1, SOD2, and p62 protein levels decreased in the model group compared with those in the control group (P<0.01), and JNK FoxO1, LC3BⅡ, and Beclin-1 protein levels increased (P<0.05). After BBR intervention, SIRT1, SOD2, and p62 protein levels increased (P<0.01), and JNK, FoxO1, LC3BⅡ, and Beclin-1 protein levels decreased compared with those in the model group (P<0.05).ConclusionBBR has an inhibitory effect on ovarian granulosa cell senescence, and the mechanism is related to the inhibition of apoptosis and autophagy mediated by the SIRT1/FoxO1 pathway.
摘要:ObjectiveTo investigate the effect of Ganoderma lucidum polysaccharides (GLP) on the proliferation, migration, cycle, and apoptosis of hepatocellular carcinoma SKHEP1 and Huh7 cells and to explore the underlying mechanism.MethodSK-HEP-1 and Huh-7 cells were classified into the blank group and low-, medium-, and high-dose GLP groups (3.5, 7, 14 g·L-1). The proliferation of the cells was examined by cell counting kit-8 (CCK8) assay, and the migration by scratch assay. Cell cycle was measured by flow cytometry and apoptosis was detected based on Hoechst33258 staining. In addition, the expression of phosphatidylinositol 3-kinase (PI3K), protein kinase B (Akt), phosphorylated PI3K (pPI3K), and phosphorylated Akt (pAkt) in the cells was determined by Western blot.ResultCompared with the blank group, the three doses of GLP reduced the proliferation and migration of SKHEP1 and Huh7 cells (P<0.05), increased the percentage of cells in G1 phase (P<0.05), and decreased percentage of cells in S and G2 phase (P<0.05). In addition, the three doses can induce apoptosis of both SK-HEP-1 and Huh-7 cells, particularly the high dose. Moreover, the three doses of GLP lowered the levels of pPI3K and pAkt (P<0.05).ConclusionGLP significantly inhibited the malignant phenotype of SK-HEP-1 and Huh-7 cells through the PI3K/Akt signaling pathway.
摘要:ObjectiveTo evaluate the lipid-lowering activity of Quansanqi tablets(QSQ), an innovative new drug of Panax notoginseng.MethodMice and golden hamsters were used to establish a hyperlipidemia model by injecting egg yolk milk and feeding high-fat diets. The levels of total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were detected, and liver function indicators [alanine aminotransferase (ALT), aspartate amino-transferase (AST), and alkaline phosphatase (ALP)] of golden hamsters were detected. Hematoxylin-eosin (HE) staining was used to observe the degree of liver injury. In the experiments, a normal group, a model group, an atorvastatin calcium group, and low-, medium-, and high-dose QSQ groups (0.32, 0.64, 1.28 g·kg-1 for mice, and 0.16, 0.32, 0.64 g·kg-1 for golden hamsters) were set up.ResultCompared with the normal group, the acute hyperlipidemia model mice showed increased TC, TG, and LDL-C levels (P<0.01), and the hyperlipidemia model mice showed increased TC and LDL-C levels (P<0.01). Additionally, the hyperlipidemia model golden hamsters showed increased serum TC, TG, LDL-C, ALT, AST, and ALP levels (P<0.05, P<0.01). HE staining indicated the presence of fat accumulation in the liver, accompanied by inflammatory reactions. Compared with the model group, QSQ of various doses could reduce TC, TG, and LDL-C levels in acute hyperlipidemia model mice (P<0.05, P<0.01), and the high-dose QSQ could reduce TC and LDL-C levels (P<0.01) and increase HDL-C level (P<0.05) in hyperlipidemia model mice, as well as reduce TC, TG, and LDL-C levels in hyperlipidemia model golden hamsters (P<0.05, P<0.01), especially in the first two weeks. In addition, atorvastatin calcium could further increase ALT, AST, and ALP levels (P<0.05, P<0.01) and aggravate liver function damage, while low-dose QSQ could reduce ALT, AST, and ALP (P<0.05), and medium- and high-dose QSQ did not cause further liver function damage.ConclusionQSQ have a significant lipid-lowering effect on different hyperlipidemia model animals and can improve liver function and liver injury.
摘要:ObjectiveTo observe the clinical efficacy of compound Huangbai liquid fomentation on diabetic foot ulcer of Wagner grade 1-2.MethodPatients were classified into the observation group and control group, with 41 cases in either group. The observation group received routine therapy and compound Huangbai liquid fomentation, while the control group was treated by routine therapy and medical silver nanoparticles-containing dressing. Ulcer area, ulcer depth, traditional Chinese medicine (TCM) syndrome score, ankle-brachial index, transcutaneous oxygen pressure, vascular endothelial growth factor, epidermal growth factor, advanced glycation end product, high-sensitivity C-reactive protein, interleukin-6, erythrocyte sedimentation rate, and adverse events were observed before and after treatment in two groups.ResultAfter treatment, the ulcer area, ulcer depth, TCM syndrome score, vascular endothelial growth factor, epidermal growth factor, advanced glycation end product, high-sensitivity C-reactive protein, interleukin-6(IL-6), and erythrocyte sedimentation rate were lower than those before treatment in two groups (P<0.05), and ankle-brachial index, transcutaneous oxygen pressure, vascular endothelial growth factor, and epidermal growth factor were higher than those before treatment in two groups (P<0.05). After treatment, the ulcer area, ulcer depth, TCM syndrome score, advanced glycation end product, high-sensitivity C-reactive protein, IL-6, and erythrocyte sedimentation rate were lower in the observation group than in the control group (P<0.05), and ankle-brachial index, transcutaneous oxygen pressure, vascular endothelial growth factor, and epidermal growth factor were higher in the observation group than in the control group (P<0.05). The clinical efficacy of observation group was better than that of the control group (P<0.05). There was no significant difference in the incidence of adverse events and endpoint events between two groups.ConclusionCompound Huangbai liquid fomentation is effective for diabetic foot ulcer of Wagner grade 1-2, as it is beneficial to the healing of the ulcer. This study provides clinical evidence for the further promotion of Chinese medicine fomentation.

摘要:ObjectiveTo investigate the distribution of vascular cognitive impairment (VCI) with kidney Yang deficiency syndrome and explore the biological nature of VCI with kidney Yang deficiency syndrome from the perspective of DNA methylation under the combination of disease and syndrome, so as to provide an epigenetic target for traditional Chinese medicine (TCM) treatment of this disease with this syndrome in the future.MethodCommunity residents in Beijing were screened out for cognitive impairment from September 2020 to November 2022 through the scale, and VCI patients were analyzed for the syndrome. VCI patients with kidney Yang deficiency syndrome and healthy people were enrolled in this study. Peripheral venous blood was collected and subjected to genome-wide DNA methylation detection by Illumina Human Methylation 850K BeadChip. Then, differentially methylated genes (DMGs) were screened out for bioinformatics analysis.ResultA total of 1 902 people were investigated in this study, and 201 of them had VCI, accounting for 10.57%, including 72.14% with kidney Yang deficiency syndrome. The methylation results showed that compared with the normal group, the VCI group had 386 differential methylation sites, and 136 DMGs were annotated. The Kyoto Encyclopedia of Gene and Genomes(KEGG) signaling pathway enrichment analysis showed that the DMGs between the two groups were mainly involved in mammalian target of rapamycin(mTOR) signaling pathway, Estrogen signaling pathway, cyclic adenosine monophosphate(cAMP) signaling pathway, etc. Protein-protein interaction (PPI) analysis showed that DMGs, such as epidermal growth factor receptor(EGFR), epidermal growth factor (EGF), and signal transducer and activator of transcription 3(STAT3), played important roles in the network.ConclusionKidney Yang deficiency is the main syndrome in VCI patients. DMGs including EGFR, EGF, and STAT3 and the related pathways such as mTOR signaling pathway, Estrogen signaling pathway, and cAMP signaling pathway may play a vital role in the occurrence and development of VCI with kidney Yang deficiency syndrome.

摘要:ObjectiveTo explore the medication characteristics and clinical efficacy of the Tenghuang Jiangu tablets in the treatment of knee osteoarthritis (KOA) in the remission stage in the real world,providing references for rational clinical use of this prescription.MethodBased on the "registration system of KOA treated with Tenghuang Jiangu tablets",2 439 KOA cases in the remission stage were analyzed by SPSS 25.0,IBM SPSS Modeler18.0,and Apriori algorithm. To be specific,the age,body mass index (BMI),and course of treatment were described in the form of x̄±s. The information on gender,K-L grade,daily dose,and frequency of drug use was described by frequency analysis. The number of cases,course of treatment,daily dose,and drug use frequency of the single-use group and the combined-use group were described by frequency analysis,and the combination of drugs was described by frequency analysis and Apriori algorithm. Mann-Whitney U test was employed to compare the scores of Visual Analogue Scale (VAS),Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC),pain,stiffness,and joint function between the single-use group and the combined-use group.ResultThe results of clinical treatment showed that 2 439 patients with KOA in the remission stage were treated with Tenghuang Jiangu tablets,with 1 432 (58.71%) in the single-use group and 1 007 (41.29%) in the combined-use group. The average daily dose of Tenghuang Jiangu tablets was (3.90±1.44) g,and the majority of the patients were at grade Ⅱ (54.47%). The daily average daily dose of Tenghuang Jiangu tablets in the single-use group was (3.64±1.35) g,which was lower than that in the combined-use group [(4.26±1.48) g,P<0.05]. In the combined use,the top three western medicines were glucosamine (270 times,14.68%),sodium hyaluronate (126 times,6.85%),and imrecoxib (116 times,6.31%),and the top three Chinese medicines were Huoxuezhitong capsules/tablets/ointments (31 times,1.69%),Biqi capsules (25 times,1.36%),and Maizhiling (23 times,1.25%). As for the overall clinical efficacy,the VAS score was (5.13±0.93) score before treatment and (2.22±1.18) score after treatment (P<0.05),with an overall average decrease of (2.91±1.14) score, and the average decrease in the single-use group was (2.76±1.43) score, which was lower than that in the combined-use group [(3.12±1.36) score,(P<0.01)]. The WOMAC score was (31.05±11.84) score before treatment and (13.55±9.91) score after treatment (P<0.05). The overall average decrease was (17.50±11.79) score, and the average decrease in the single-use group and combined-use group was (16.39±11.14) score and (19.08±12.50) score,respectively (P<0.01). The patients with KOA>grade Ⅱ accounted for 91.34%(1 308/1 432) and 93.55%(942/1 007) in the single-use group and combined-use group,respectively (χ2=80.026,P<0.05). A total of 43.37%(621/1 432) of the patients in the single-use group had other complications,lower than that in the combined-use group [54.92%(553/1 432),(χ2=20.087,P<0.01)].ConclusionMore than half of the patients with KOA in the remission stage are treated with Tenghuang Jiangu tablets alone,and the combination therapy is mainly applied in patients with severe conditions or other complications. In relieving knee joint pain and improving joint stiffness and joint function,both the Tenghuang Jiangu tablets alone and the combination therapy are effective.
摘要:ObjectiveTo explore the distribution and characteristics of traditional Chinese medicine (TCM) syndromes in patients with endometriosis (EMS).MethodA cross-sectional survey was conducted to analyze the characteristics of TCM syndromes in 1 895 cases of EMS in hospitals of 19 provinces, autonomous regions, and municipalities.ResultAmong the 1 895 patients, Qistagnation and blood stasis syndrome accounted for the highest proportion, followed by Qi deficiency and blood stasis syndrome, cold coagulation and blood stasis syndrome, and kidney deficiency and blood stasis syndrome. The distribution of TCM syndrome types of patients with EMS in different regions, different treatment stages, and different disease types and with different therapeutic goals was different, and the differences were statistically significant. However, under different conditions, the Qi stagnation and blood stasis syndrome accounted for the highest proportion. Under the stratification of different regions, the cold coagulation and blood stasis syndrome in north China was relatively high, the kidney deficiency and blood stasis syndrome in south China was relatively high, and the combined phlegm and stasis syndrome was relatively high in southwest China. Different diagnosis and treatment goals corresponded to different clinical syndromes. With pelvic pain as the main manifestation, the proportion of cold coagulation and blood stasis syndrome was higher. The proportion of kidney deficiency and blood stasis syndrome was higher in married patients with infertility. Patients with the main diagnosis and treatment goals of controlling mass and inhibiting recurrence had a higher proportion of Qi deficiency and blood stasis syndrome. In different treatment stages, the proportion of Qi deficiency and blood stasis syndrome in postoperative patients and those with recurrent EMS was higher. Among different disease types, the Qi deficiency and blood stasis syndrome accounted for a higher proportion in patients with ovarian endometriosis (OEM). The cold coagulation and blood stasis syndrome accounted for a higher proportion in patients with deep invasive endometriosis (DIE). The kidney deficiency and blood stasis syndrome accounted for a higher proportion in patients with peritoneal EMS. There were significant differences in age, body mass index (BMI), and course of disease among patients with different syndromes. Patients with Qi deficiency and blood stasis syndrome was relatively older, and their course of the disease was relatively long. Patients with combined phlegm and stasis syndrome had relatively high BMI. There was no significant difference in CA125 and CA199 levels among all syndrome types.ConclusionThe distribution of TCM syndromes of EMS has a certain regularity, and there are differences in regional distribution, therapeutic goals, treatment stages, and disease types(P<0.05). However, the Qi stagnation and blood stasis syndrome accounts for a large proportion under different conditions, suggesting that Qi stagnation is the key link of EMS. In the early stage, the team took relieving depression and activating blood as the primary treatment, and created Huoxue Xiaoyi prescription, which was the core prescription for the treatment of EMS with Qi stagnation and blood stasis syndrome, achieving good clinical effect. At the same time, it is emphasized that EMS treatment should be integrated into the concept of chronic disease management and combined with health management. Through psychological counseling, cognitive behavior intervention, popular science lectures, and other methods, it is advised to adjust the emotion of patients with EMS, thereby increasing the curative effect. This study is expected to provide references for the clinical treatment of EMS.

摘要:ObjectiveTo re-evaluate the systematic reviews/Meta-analyses (SRs/MAs) of tonic traditional Chinese medicine (TCM) injections against cerebral ischemic stroke (CIS) and provide evidence support for clinical practice and decision-making.MethodTCM injections of different varieties were obtained after searching the three major drug catalogues. Seven Chinese and English databases were searched from database inception to March 13,2022,for the relevant SRs/MAs. The methodological quality,risk of bias,reporting quality,and quality of evidence were assessed by Assessment of Multiple Systematic Reviews-2 (AMSTAR-2),the Risk of Bias in Systematic Review (ROBIS),the Preferred Reporting Items for Systematic Reviews and Meta-analyses 2020 (PRISMA 2020),and the Grading of Recommendations Assessment,Development,and Evaluation (GRADE). In addition,the literature overlap matrix was established to calculate the corrected covered area (CCA) and evaluate the rate of overlaps of the original literature.ResultFive types of TCM injections and 18 SRs/MAs were included. AMSTAR 2 evaluation showed that the methodological quality of 18 SRs/MAs was extremely low,and 14 SRs/MAs had a high risk of bias assessed by ROBIS. The quality evaluation results reported by the PRISMA 2020 showed that the scores of the studies included ranged from 19.5 to 28.5,with 10 being of medium quality and eight of low quality. The evaluation with the GRADE system demonstrated that one outcome was moderate-quality evidence,15 outcomes were low-quality evidence,and 41 outcomes were very low-quality evidence. The CCA of the included SRs/MAs was 0.263,indicating a low rate of overlaps of the original literature.ConclusionTonic TCM injections are effective and safe in the treatment of CIS,but this conclusion should be treated with caution because of the low quality of methodology,reports,and evidence in published SRs/MAs. It is recommended to improve the study design,obtain clinical evidence of higher quality,and conduct systematic evaluations in strict accordance with procedures to standardize the reporting of research results.

摘要:ObjectiveThis study performed a scoping review to comprehensively analyze and report the information on the instructions of Chinese patent medicines and clinical research evidence for the treatment of respiratory diseases in children.MethodChinese patent medicines against respiratory diseases in children were obtained by searching the three major drug catalogues. The Chinese and English databases were searched for relevant literature,followed by data statistics and visualized analysis.ResultAfter screening and analysis,52 Chinese patent medicines were included,involving nine dosage forms. The main drugs were Scutellariae Radix,Armeniacae Semen Amarum,Forsythiae Fructus,etc. The main functions included clearing heat and releasing exterior syndrome,and relieving cough and dissipating phlegm. The indications mainly included common cold with wind-heat syndrome and cough in children. Adverse drug reactions and contraindications were only specified in 19.23% (10/52) of Chinese patent medicines,and the rest only displayed "unclear". A total of 279 articles were included,including 277 articles from Chinese Core Periodicals and two articles from SCIE. In terms of research type,those articles included 253 randomized controlled trials (RCTs,with six dosage form/dose comparisons involved),11 retrospective analyses based on Hospital Information System (HIS) data,one case series,13 systematic reviews/Meta-analyses (with two network Meta-analyses involved),and one economic evaluation article. Among them,72.76% (203/279) of the articles were published in the Core Journals of Chinese Science and Technology. Only 33 Chinese patent medicines were involved,and Xiaoer Feire Kechuan Oral Liquid was the top 1 under investigation,accounting for 15.71% (44/280). The indicated diseases were mainly infantile pneumonia,bronchitis,respiratory tract infection,cough,asthma,and other western medicine diseases. Xiaoer Chiqiao Qingre Granules and Xiaoer Dingchuan Oral Liquid were used off-label. The sample size was concentrated in 51-150 cases,accounting for 67.17% (178/265). The interventions in the experimental group were mainly Chinese patent medicine + western medicine + basic treatment or Chinese patent medicine + western medicine. The main outcomes were the effective rate and the improvement of clinical symptoms. The adverse reactions were mainly gastrointestinal reactions,drug-induced skin symptoms,etc.,and two studies have shown that drug doses were associated with adverse reactions.ConclusionIn research years,the research on Chinese patent medicines in the treatment of respiratory diseases in children has advanced rapidly. However,there are still some problems that need to be resolved in the future,such as incomplete information on drug content in the instruction,concentrated drugs to be studied,limited indications,failure to highlight the characteristics of traditional Chinese medicine (TCM) syndromes,unstandardized research design,and an incomplete reflection of Chinese patent medicine.

摘要:ObjectiveTo review the drug information and research progress on oral Chinese patent medicines in the treatment of cardiac arrhythmia to identify existing problems and provide references for follow-up research.MethodChinese patent medicines against cardiac arrhythmia were retrieved from the three major drug catalogues,Yaozh.com,and relevant guidelines with arrhythmia as the retrieval term. The instructions for included Chinese patent medicines were retrieved through Yaozh.com and specific information was extracted. The research articles on Chinese patent medicines included were retrieved from the five databases,and the information meeting the inclusion and exclusion criteria was extracted and displayed in the form of text description and graphs after statistical analysis.ResultSixty-five oral Chinese patent medicines were included in this study,with the main functions of activating the blood and resolving stasis. The average daily cost of medicines was 8.17 yuan,and there were 42 medicines with an average daily cost of less than 10 yuan,showing a moderate medicine cost. A total of 351 research articles on Chinese patent medicines were screened out,including 259 randomized controlled trials (RCTs),16 non-RCTs,eight non-controlled trials,62 systematic reviews,two guidelines,and two expert consensuses. Eighteen types of Chinese patent medicines were involved,whose clinical trials had been conducted in 28 provinces,cities,autonomous regions,and municipalities in China. Wenxin granules and Shensong Yangxin capsules were the top medicines under investigation,accounting for 75.21% of all research articles. Among the included studies,the most common comparison design was Chinese patent medicine combined with western medicine vs western medicine (64.25%). The outcome evaluation was mainly based on clinical efficacy,symptom efficacy,arrhythmia efficacy,adverse reactions,and heart rate changes.ConclusionThe number of clinical studies of oral Chinese patent medicines against cardiac arrhythmia varies greatly,but traditional Chinese medicine (TCM) syndrome differentiation thinking is less considered in practical application. Due to unstandardized clinical research and low-quality literature,further advancement is required in the future.

摘要:Health research priority setting, based on the existing disease burden or healthcare needs, screens out specific areas or topics with relatively high research priority by scientific and systematic methods, and optimizes the allocation of health resources by influencing healthcare decision-making, so as to alleviate the imbalance between regional or global health and development. Many developed countries have carried out related research and practical work on different scales, and the World Health Organization (WHO) attaches great importance to the transformation and application of relevant achievements in developing countries. As the largest developing country in the world, China's research in this field started relatively late, and only a small number of scholars have carried out part of the localization methodology research and practice according to the specific national conditions. However, health research priority setting has not yet attracted the attention of large-scale research institutions or government organizations in China. Although the priority setting is rarely mentioned in the research on traditional Chinese medicine (TCM), the research and decision-making on the diseases responding specifically to TCM can also be regarded as the practical work of exploring the priority of TCM. Policymakers have a sense of priority support in the "priority of TCM research", but the decisions from the top design are mainly based on the consensus reached by high-level think tanks. There is a lack of extensive research, and moreover, the data of multiple stakeholders are not included. Therefore, it is urgent to introduce appropriate priority setting methods to solve the problem of transparency and scientificity in the decision-making process. Given the perspective of the specific implementation, the present study introduced three international priority setting methods, i.e., the James Lind Alliance and Priority Setting Partnerships(JLAPSP,)the Child Health and Nutrition Research Initiative(CHNRI), and the Council on Health Research and Development (COHRED), and presented relevant recommendations on how to apply them in the research of TCM, which is expected to provide references for the local research.

摘要:ObjectiveTo review the drug information and clinical researches on Chinese patent drug in Pharmacopoeia of the People's Republic of China,National Essential Drugs List and Medicine List for National Basic Medical Insurance,Employment Injury Insurance and Maternity Insurance.MethodSearch Chinese patent medicine,which can reduce blood sugar in the three major catalogues above. CNKI,VIP,Wanfang,Embase and PubMed were searched from their inception dates to August 14th,2021 for the clinical researches on Chinese patent drug. A database was established based on the collected Chinese patent drug for the treatment of diabetes. And then,descriptive analysis was performed to analyze the general condition of clinical researches.ResultFrom the three catalogues above,28 kinds of Chinese patent drugs were retrieved, and Supplementing Qi and Nourishing Yin was the basic effect of 22 kinds of Chinese patent drugs. A total of 1 069 clinical researches published and peaked in 2017 before August 14th,2021 were included. Clinical studies have been carried out and published in all 30 provinces and autonomous regions,and the province with the largest number of published literature was Henan.What's more,16.65% of the projects were supported by government funding. The number of research to Shenqi Jiangtang tablets/granules/capsules was the largest,among the 28 kinds of Chinese patent drugs.Besides,the most frequent type of interventions in the 958 two-arm trials was the load test,accounting for 78.91%.Most types of diabetes,including type 1 diabetes,type 2 diabetes and its complications,gestational diabetes,other types of diabetes and pre-diabetes,were covered in in this study. And the results showed that different drugs with different suitable crowd.ConclusionA summary of the current status of clinical research on Chinese patent drug by means of scoping review can provide direction for the next research.

摘要:ObjectiveTo establish a method for seahorse identification based on graphene oxide fluorescence sensing technology, and to provide a new research idea for identification of traditional Chinese medicine.MethodThe fluorophore FAM was labeled at the 5' end of the specificity upstream primer Ja-F of Hippocampus japonicus as the nucleic acid probe FAM-ssDNA (single strand DNA). The recognition site of RNA polymerase Ⅱ was added to its specific downstream primer Ja-R as Ja-R1. The seahorse samples were amplified with Ja-F/Ja-R1 primers, and the ssDNA of H. japonicus was obtained by reverse transcription of the amplification products using vitro transcription method. The 20 μL nucleic acid probe FAM-ssDNA (500 nmol·L-1) was incubated at 90 ℃ for 5 min, and was gradually cooled to room temperature. Different volume of graphene oxide solution (100 mg·L-1) and Tris hydroxymethyl amino methane HCl (Tris-HCl) buffer (50 mmol·L-1) were added into each probe solution to make a final reaction volume of 1 mL. The fluorescence intensity of each sample was measured after mixing and placing different times at room temperature away from the light. So that the most appropriate graphene oxide concentration and reaction time were screened for constructing the best nucleic acid probe-graphene oxide biosensor. Adding probe complementary sequence FAM-ssDNA-match solution into the nucleic acid probe-graphene oxide solution, the fluorescence intensity of the reaction mixture was measured after being placed different times at room temperature. Therefore, the optimal reaction time of fluorescence recovery was screened and the feasibility of the sensor was tested. The sensitivity was detected via adding ddH2O as the blank control and different concentration H. japonicus ssDNA into each nucleic acid probe-graphene oxide solution, respectively. Finally, the commercial hippocampal were identified using the optimal experimental condition, and the feasibility of this method for the identification of Chinese medicinal materials was verified.ResultThe fluorescence of 1 mL reaction mixture including 10 nmol·L-1 nucleic acid probe FAM-ssDNA and 12 mg·L-1 go solution for 20 min at room temperature away from the light could be completely quenched. Feasibility test of the biosensor showed that when probe complementary sequence FAM-ssDNA-match solution (final concentration 90 nmol·L-1) was added to the biosensor solution and reacted 1 h reaction at room temperature, the fluorescence signal was significantly enhanced. Sensitivity test showed that the minimum concentration of ssDNA detected by this method was about 10 mg·L-1. This method was used to detect commercial seahorses, and only H. japonicus samples had obvious fluorescence signal.ConclusionThe graphene oxide-based fluorescent sensing technology could be used for zoological origin survey of commercial hippocampus.

摘要:ObjectiveIn recent years, with the sharp decline of wild resources in Arisaematis Rhizoma and Pinelliae Rhizoma and the immaturity of medicinal cultivation technology, their adulterants have appeared frequently in the market, and the main identifying characteristics have mostly disappeared in the circulation of medicinal materials. Therefore, there is an urgent need to establish a molecular identification method that can quickly and effectively identify the specificity of Arisaematis Rhizoma and Pinelliae Rhizoma.MethodAfter comparison of the rbcL sequences of Arisaematis Rhizoma,Pinelliae Rhizoma, and their adulterants, the specific enzyme cleavage sites Hae Ⅲ and Dra Ⅰ of Arisaematis Rhizoma and Pinelliae Rhizoma, respectively, were selected and identified by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The main system conditions of PCR-RFLP reaction were established and optimized, and their durability and the ability to detect genuine, adulterants, and mixed counterfeits were investigated.ResultThe PCR-RFLP identification method of Arisaematis Rhizoma and Pinelliae Rhizoma was established. After specific primer amplification, Arisaematis Rhizoma and Pinelliae Rhizoma could be digested by Hae Ⅲ and Dra Ⅰ-restricted endonucleases respectively, at annealing temperature of 54 ℃, the number of cycles of 35, and the amount of DNA template of 3-30 ng, producing two fragments or small cut fragments with a single band between 100-250 bp, whereas the mixed counterfeits were not cleaved and both showed a band at 250 bp. The method is highly accurate in identifying adulterants and mixed counterfeits of Arisaematis Rhizoma or Pinelliae Rhizoma.ConclusionThe PCR-RFLP method developed in this study allows for the rapid identification of Arisaematis Rhizoma and Pinelliae Rhizoma.

摘要:ObjectiveTo investigate the effect of exogenous H2O2 on secondary metabolism in Atractylodes chinensis and its mechanism.MethodFresh rhizomes of A. chinensis were treated with 5.0, 1.0, 0.2, 0.04 mmol·L-1 H2O2 solution and clean water, and the relationships between the contents of reactive oxygen species, activities of antioxidant enzymes, activities of key enzymes of secondary metabolites, and contents of secondary metabolites in A. chinensis were compared.ResultUnder treatment with exogenous H2O2, the content of reactive oxygen species and malondialdehyde (MDA) in the fresh rhizomes of A. chinensis were significantly elevated on the 4th day, and returned to normal level on the 6th-8th day. The activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) were all increased first and then decreased, and reached the peak on the 4th, 4th-6th and 2th-4th day, respectively. The activities of 3-hydroxy-3-methyl glutaryl coenzyme A reductase (HMGR) and acetyl CoA carboxylase (ACC), key enzymes of the secondary metabolites, were remarkably enhanced, and under treatments with different concentrations of H2O2, the activities of key synthetic enzymes of the secondary metabolites in 0.2 mmol·L-1 H2O2 groupwere increased most, with the highest biosynthesis of secondary metabolites. The contents of atractylodin, β-eudesmol, atractylone, atractylenolide Ⅱ, and atractylenolide Ⅲ on the 6thday of 0.2 mmol·L-1 H2O2 treatment were 89.5%, 108.7%, 308.8%, 64.7% and 9.3%, respectively higher than those in the control.ConclusionThe antioxidant enzymes and secondary metabolites in A. chinensis synergistically maintain the balance of reactive oxygen species, and exogenous H2O2 can improve the medicinal quality of A. chinensis remarkably.

摘要:ObjectiveTo compare the regulatory effects of five plant polysaccharides on immune function of cyclophosphamide (CTX)-induced immunosuppressed mice by network Meta-analysis, to provide evidence for the clinical application of polysaccharides and the development of effective polysaccharides and oligosaccharides.MethodSeven databases including PubMed, Embase and Web of Science were searched, and studies that met the inclusion criteria were selected. The methodological quality of the included studies was evaluated using the risk of bias tool of Systematic Review Centre for Laboratory Animal Experimentation (SYRCLE), and the data were analyzed using RStudio and StataSE 17.ResultA total of 62 randomized controlled trials (RCTs) were included, involving 1 512 mice and five plant polysaccharides: Astragalus polysaccharide (APS), lentinan (LNT), Ganoderma lucidum polysaccharide (GLP), Poria cocos polysaccharide (PCC), and Codonopsis pilosula polysaccharide (CPP). The network Meta-analysis showed that APS ranked first in increasing spleen index (mean deviation (MD)=3.54, 95% confidence interval (CI) [2.10, 5.96]), thymus index (MD=1.98, 95%CI [1.55, 2.54]) and T helper cells (CD4+)/T suppressor cells (CD8+) (MD=1.63, 95%CI [1.13, 2.37]), while CPP ranked first in up-regulating the number of peripheral blood leukocytes (MD=24.16, 95%CI [8.21, 71.12]), macrophage phagocytosis index (MD=2.52, 95%CI [1.32, 4.82]) and immunoglobulin M (IgM) content (MD=1.79, 95%CI [1.12, 2.85]).ConclusionAll the five plant polysaccharides can regulate the immune function of immunosuppressed mice. Among them, APS has advantages in elevating spleen index, thymus index and CD4+/CD8+, while CPP focuses on increasing the number of peripheral blood leukocytes, macrophage phagocytosis index and IgM content. Due to limited number and quality of included studies, the conclusions needs to be further verified with large samples and high-quality studies.
摘要:ObjectiveTo investigate the pharmacodynamic characteristics and explore the molecular mechanism of Honghua oral liquid (HOL) in relieving neuropathic pain (NP).MethodHealthy male SD rats were randomly assigned into sham group, model group, low-, medium-, high-dose (0.5, 1.0, 2.0 mL·kg-1·d-1, respectively) HOL groups, and a positive drug (pregabalin, 25 mg·kg-1·d-1) group, with 6 rats in each group. Spinal nerve ligation (SNL) of L5 was conducted in other groups except the sham group. Drug administration was performed 3 days after the SNL surgery for 2 consecutive weeks, and samples were collected after the end of the administration. During the treatment period, the mechanical pain threshold and cold pain threshold were determined to measure the pain-relieving effect of HOL. Transcriptome sequencing was performed on hippocampal tissue samples from the sham, model, and high-dose HOL groups, and differentially expressed genes between the sham group and the model group as well as the model group and HOL high-dose group were obtained. After pathway enrichment analysis, we selected the targets which were closely related to neuroinflammation for validation, and predicted the specific binding sites of the major active components in HOL with the targets through molecular docking. In addition, the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) were determined by enzyme-linked immunosorbent assay (ELISA) to evaluate the effect of HOL on neuroinflammation in NP rats.ResultCompared with the sham group, SNL decreased the mechanical pain threshold and cold pain threshold (P<0.05). Compared with the model group, HOL recovered the mechanical pain threshold and cold pain threshold (P<0.05). The transcriptome data showed that 376 differentially expressed genes (DEGs) were identified between the model group and the sham group, including 124 upregulated genes and 252 downregulated genes, and 194 DEGs between the model group and the high-dose HOL group, including 33 upregulated genes and 161 downregulated genes. Among them, insulin-like growth factor 1(IGF1), matrix metallopeptidase-2 (MMP-2), matrix metallopeptidase-14 (MMP-14), erb-B2 receptor tyrosine kinase 2 (ERBB2), and integrin subunit alpha 5 (ITGA5) associated with NP were selected for further validation. The Real-time fluorescence quantitative polymerase chain reaction(Real-time PCR) results showed that compared with the sham group, the modeling up-gurelated the mRNA levels of the above five molecules in the hippocampus (P<0.01). Compared with model group, HOL down-regulated the mRNA levels of these molecules (P<0.01). The molecular docking results showed that the main active components of safflower, hydroxysafflor yellow A, kaempferol, and quercetin, formed stable hydrogen bonds with the amino acid residues of IGF1, MMP-2, MMP-14, ERBB2, and ITGA5. The enzyme-linked immunosorbent assay(ELISA) results showed that compared with those in the sham group, the serum levels of TNF-α and IL-10 were out of balance in the model rats (P<0.01). Compared with the model group, HOL lowered the level of the pro-inflammatory cytokine TNF-α (P<0.01) and elevated that of the anti-inflammatory cytokine IL-10 (P<0.05).ConclusionHOL exerts analgesic effect on SNL rats by inhibiting neuroinflammation.
摘要:Under the national policy background of traditional Chinese medicine(TCM)registration review and approval reform, TCM preparations in medical institutions(hereinafter referred to as medical institution preparations) have human use experience, conform to the characteristics of TCM clinical practice, and have advantages in the research and development of innovative Chinese medicines. Therefore, the research and development mode based on clinical experience prescription-medical institution preparations-innovative Chinese medicines has attracted widespread attention from the industry. However, in the process of development and use of medical institution preparations, there are generally clinical problems that restrict their transformation into innovative Chinese medicines, resulting in a relatively weak collection basis of human use experience and insufficient clinical evidence for supporting the research and development of new TCM medicines. In this paper, on the basis of sorting out the supporting regulations and relevant technical requirements of human use experience, and analyzing the clinical problems restricting the transformation of medical institution preparations, it provides suggestions for medical institutions to carry out high-quality research on human use experience of preparations in the process of TCM clinical practice from the aspects of continuously exploring clinical value, improving the construction of information system, focusing on the TCM clinical practice and giving full play to the advantages of discipline cooperation. By realizing the whole life cycle management of medical institution preparations based on three-combination evaluation evidence system, we can promote the transformation of medical institution preparations into innovative Chinese medicines.

摘要:At present, major depressive disorder (MDD) is highly prevalent with advanced neurological disorders as the main pathological manifestations. As the physiological function bearer of higher neural activity, gray matter has become the focus of MDD treatment. However, recent research has shown that white matter and gray matter are independent of each other in the central nervous system (CNS), and their functions are integrated and linked. In addition to gray matter damage, white matter damage is also the core driving event of disease progression and determines the outcome of MDD. At the treatment level, the current drug treatment of MDD mainly focuses on gray matter repair, while ignoring the importance of white matter integrity for the treatment of the disease, which has become the weakness of the current treatment of MDD. Traditional Chinese medicine (TCM) has good application potential in white matter repair. This paper elaborated on the following three aspects. ① The roles of white matter damage in the occurrence and development of MDD were summarized. ② The key link of white matter repair in MDD was elaborated with microglia microenvironment regulation as the entry point. ③ The application value of TCM in white matter repair in MDD was analyzed. This review aims to highlight the importance of white matter integrity in the treatment of MDD and is expected to expand the understanding dimension of the activity of related Chinese medicines in MDD from the perspective of white matter repair and analyze its potential application value.
摘要:In China, malignant tumors have become the main cause of death. In the past half century, the incidence and mortality of malignant tumors have been on the rise, posing a threat to health of patients, and the burden of cancer has been increasing. At the moment, malignant tumors are mainly treated by surgery, radiotherapy, and cytotoxic drugs, which, however, have limitations and induce great adverse reactions. As biological technology and the research on tumor microenvironment, immunology, cell biology, and molecular biology advance, high-efficiency low-toxicity targeted therapy has attracted wide attention in the treatment of tumors. Epidermal growth factor receptor (EGFR) plays an important role in many cellular processes such as cell proliferation, survival, differentiation, migration, inflammation, and stromal homeostasis. EGFR promotes tumor growth, proliferation, and metastasis in a variety of ways. Chinese medicine has unique efficacy in the comprehensive treatment of malignant tumors. Through multiple components, multiple targets, and multiple pathways, it enhances the efficacy, reduces toxicity, prolongs life, and improves life quality in the treatment of tumors. Many Chinese medicines and their active components exert anti-tumor effect by inhibiting the EGFR signal transduction pathway. Through targeted inhibition of EGFR, Chinese medicine can promote the apoptosis and autophagy of tumor cells, suppress the proliferation and metastasis of tumor cells, and delay the progression of tumors. Thus, EGFR is a potential target for suppressing tumor. This paper summarizes the relationship between EGFR signal transduction pathway and tumor occurrence and development and analyzes the anti-tumor action mode and possible mechanisms of Chinese medicine and the active components by regulating EGFR signaling pathway, which is expected to provide a reference for clinical practice.

摘要:Zhishi Xiebai Guizhitang is a classical prescription for the treatment of chest impediment with the method of warming Yang. It is included in the Catalogue of Ancient Classical Prescriptions issued by the National Administration of Traditional Chinese Medicine (First Batch), with the effect of activating Yang, dissipating mass, moving Qi and resolving phlegm. Its main symptoms include chest fullness and pain, or even chest pain radiating to the back, wheezing, coughing, shortness of breath, and Qi reversal from the hypochondrium. In modern traditional Chinese medicine, Zhishi Xiebai Guizhitang is clinically used in the treatment of cardiovascular system, digestive system, respiratory system and other diseases, among which coronary heart disease, unstable angina pectoris, myocardial infarction, sinus bradycardia and other cardiovascular diseases have particularly significant effects. This paper reviewed the pharmacological studies of Zhishi Xiebai Guizhitang in the past 10 years. The results showed that each single medicine and the whole prescription alleviated the above cardiovascular diseases to a certain extent, with the pharmacological effects of improving intravascular environment, myocardial ischemia, myocardial ischemia-reperfusion injury, and myocardial hypoxia, anti-inflammation, plaque stabilisation, etc., and the pharmacological mechanism involved the regulation of relevant active substances in vivo as well as related signaling pathways and ion channels, mainly including thromboxane B2 (TXB2), prostacyclin I2(PGI2) and phosphatidylinositol 3-kinases/protein kinase B/endothelial nitric oxide synthase (PI3K/Akt/eNOS) signaling pathways, and ATP-sensitive potassium channels. In addition, the relationship between the pharmacological effects of some single medicines and transient receptor potential ankyrin 1 (TRPA1) has been reported that TRPA1 is a key to understanding the mechanism of Zhishi Xiebai Guizhitang in treating cardiovascular diseases, which is worth of further study.
摘要:Diabetes and its complications are major public health issues of worldwide concern. Diabetic microangiopathy is a vascular complication of diabetes caused by blood stasis and deficiency, characterized by impaired microcirculation with hyaline deposits. Diabetic nephropathy, diabetic retinopathy, and diabetic neuropathy are the most common types of diabetic microangiopathy, which can be traced back to the pre-diabetes period and is aggravated by the dynamic evolution of diabetes. Therefore, early intervention is required. Anti-oxidative, anti-inflammatory, and microcirculation-improving drugs should be chosen to treat diabetic microangiopathy based on hypoglycemic, lipid-lowering, and hypotensive treatment in clinical practice. Diabetic microangiopathy belongs to the theoretical concept of "collateral disease" in traditional Chinese medicine (TCM). The core of the treatment of diabetic microangiopathy with Chinese medicine is to protect "tertiary collateral vessels-microvascular", and Chinese medicines with Qi-replenishing, Yin-nourishing, heat-clearing, and blood-activating effect are used for compatibility in Chinese medicine prescriptions. Based on the understanding and treatment principles of TCM and western medicine for diabetic microangiopathy, this review briefly summarized the research progress of commonly used prescriptions such as Renshen Baihutang, Yuye Tang, Simiao Yongantang, Gegen Qiliantang, Liuwei Dihuangwan, and modern Chinese medicine preparations for the treatment of diabetic microangiopathy. Moreover, the research progress of Chinese medicines including Ginseng Radix et Rhizoma, Astragali Radix, Rehmannia Radix, Lycii Fructus, Notoginseng Radix et Rhizoma, Salviea Miltiorrhizae Radix et Rhizoma, Lonicera Japonica Flos, and Puerariae Lobatae Radix were outlined. This review is expected to provide the clinical basis and theoretical guidance for the treatment of diabetic microangiopathy with Chinese medicine.
摘要:Osteoporosis (OP) is a systemic metabolic bone disease. Amid population aging, OP has become a major health problem for the middle-aged and the elderly in China. Aging, iron load, and estrogen deficiency break the balance between oxidation and antioxidant systems, and the increase of reactive oxygen species mediates oxidative stress to damage DNA, lipids, proteins and other macromolecules, thus accelerating cell apoptosis and inducing OP, obesity, and neurodegenerative disorders. It has been found that oxidative stress is of great significance in the pathogenesis of OP. Oxidative stress regulates the signaling pathways, cytokines, and proteins related to the mesenchymal stem cells, osteoblasts, and osteoclasts, thereby weakening the osteogenic differentiation of mesenchymal stem cells, inhibiting osteoblast mineralization, and promoting the activation, proliferation, and maturation of osteoclasts. As a result, the dynamic imbalance between bone resorption and bone formation occurs, influencing bone remodeling and promoting the progression of OP. At the moment, anti-bone resorption drugs, bone formation-promoting drugs, and hormones are mainly used in clinical settings in western medicine. However, due to the long treatment cycle and the occurrence of serious gastrointestinal reactions, hypocalcemia, osteonecrosis, and others, patients show poor compliance and thus the effect is not as expected. Traditional Chinese medicine (TCM) demonstrates remarkable effect on OP attributing to the multi-pathway and multi-target characteristics. With low price and few adverse reactions, TCM is widely applied in clinical practice in comparison with western medicine. TCM has unique advantages in the treatment of OP by regulating oxidative stress. It exerts the therapeutic effect on OP by modulating different signaling pathways, providing new mindset for the treatment of this disease. Therefore, through literature research, this study summarized the research on mechanism of oxidative stress in OP and the treatment by TCM, which is expected to lay a foundation for further research.
摘要:Osteosarcoma (OS) is the most common primary malignant bone tumor originating from mesenchymal stem cells, which features high degree of malignancy, strong invasiveness, easy early metastasis, and high recurrence rate. The clinical manifestations of OS are pain, local mass, limited movement, and pathological fracture. OS mainly occurs in children, adolescents, and the elderly, seriously threatening physical and mental health of patients, as well as their quality of life. The currently available therapies for OS are surgery, chemoradiotherapy, and the combination of the two. Although the therapeutic effect has been improved, tumor recurrence and metastasis and multidrug resistance still occur. Thus, the therapeutic effect is not satisfactory, especially in improving the overall survival rate of patients with metastatic OS. As a result, clinicians and researchers have been making efforts to find an effective therapy. In recent years, the mechanism of curcumin (CUR) against OS has attracted wide attention. CUR, a pigment extracted from the rhizomes or tubers of many plants, such as Curcuma longa, C. rcenyujin, and C. phaeocaulis, has a variety of pharmacological effects. Scholars have found that CUR has the effects of inhibiting proliferation, inducing apoptosis, and reversing multidrug resistance (MDR) of tumor cells, but also it has poor water solubility and low bioavailability, which limit the clinical application. This paper mainly discusses the mechanism of CUR against OS, the existing problems, new treatment methods, and future research directions, which is expected to provide new ideas for scientific researchers and provide a reference for the development and utilization of CUR in the future.
