摘要:Astrocytes are important nerve cells in the central nervous system (CNS), which mainly play a key role in nutrition and support. Astrocytes and neurons undergo close energy coupling and substance coupling, which are closely related and interact with each other. In recent years, many studies have shown that the astrocyte-neuron coupling imbalance plays a central role in the occurrence and progression of Alzheimer's disease (AD) and serves as an important therapeutic target receiving increasing attention. According to traditional Chinese medicine (TCM) theory, the main pathogenesis of AD is kidney deficiency and marrow inadequacy, and in clinical medication, kidney-tonifying and marrow-filling TCM prescriptions are often employed with satisfactory results achieved. As reported, many kidney-tonifying and marrow-filling prescriptions exhibit regulatory and protective effects on the imbalance of astrocyte-neuron coupling, suggesting that the effect of kidney-tonifying and marrow-filling prescriptions in treating AD may have some internal relationship with its regulation of the imbalance of astrocyte-neuron coupling. This article reviewed the underlying internal relationship between the imbalance of astrocyte-neuron coupling and the pathogenesis of kidney deficiency and marrow inadequacy in AD and the research progress in the intervention mechanism of TCM for tonifying the kidney and filling the marrow.

摘要:ObjectiveTo explore the mechanism of Dihuang Yinzi in improving astrocyte injury and glycolysis in Alzheimer's disease (AD) mice via regulating the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway, thereby improving the cognitive function of AD mice.MethodForty male APP/PS1 transgenic mice aged four months were randomly divided into a model group and a model + Dihuang Yinzi (0.25 g·kg-1) group, with 20 mice in each group. Forty C57BL/6J mice with the same background and same age were randomly divided into a control group and a control + Dihuang Yinzi (0.25 g·kg-1) group, with 20 mice in each group. The mice in the control + Dihuang Yinzi group and the model + Dihuang Yinzi group were administered with Dihuang Yinzi by gavage, and those in the control group and the model group received an equal volume of sterilized normal saline, once a day for 150 days. Morris water maze test was performed to test the ability of navigation and space exploration of mice. The protein expression of p-PI3K, PI3K, p-Akt, Akt, phosphofructokinase-1 (PFK-1), and aldehyde dehydrogenase 3 family member B2 (ALDH3B2) in mouse brain tissues was measured by Western blot. An immunofluorescence assay was performed to detect astrocyte morphology and the expression level of ALDH3B2.ResultAs compared with the control group, the model group showed prolonged escape latency during the 2nd to 5th days of the location-based navigation (P<0.05, P<0.01), reduced number of times crossing the target area of the platform, shortened residence time in the target quadrant (P<0.05, P<0.01), prolonged residence time in the opposite quadrant (P<0.05), increased surface area of the cell body and total length of cell protrusions of astrocytes (P<0.05, P<0.01), and down-regulated protein expression of p-PI3K, p-Akt, ALDH3B2, and PFK-1 (P<0.01), while the above experimental indexes were not significantly different in the control + Dihuang Yinzi group. Compared with the model group, the model + Dihuang Yinzi group showed shortened escape latency of APP/PS1 mice during the 2nd to 5th days of the location-based navigation (P<0.05, P<0.01), increased number of times crossing the platform, prolonged target quadrant residence time (P<0.05, P<0.01), shortened residence time in the opposite quadrant (P<0.05), reduced surface area of the cell body and total length of cell protrusions of astrocytes (P<0.05), and up-regulated protein expression of p-PI3K, p-Akt, ALDH3B2, and PFK-1 (P<0.01).ConclusionDihuang Yinzi can improve the learning and memory ability of AD mice by activating the PI3K/Akt signaling pathway and up-regulating the protein expression of PFK-1 and ALDH3B2 to protect against astrocyte injury in brain tissues and improve glycolysis.

摘要:ObjectiveTo explore the mechanism of Dihuang Yinzi (DHYZ)in improving astrocyte injury in the brain and regulating energy metabolism and autophagy disorder in Alzheimer's disease (AD) model mice.MethodForty male APP/PS1 transgenic mice aged four months were randomly divided into a model group and a model + DHYZ group (2.5 g·kg-1), with 20 mice in each group. Forty C57BL/6J mice with the same background and same age were randomly divided into a control group and a control + DHYZ group (2.5 g·kg-1), with 20 mice in each group. The mice in the control group and the model group were administered with an equal volume of sterilized normal saline by gavage, once a day for 150 days. Novel object recognition test and step-down test were performed to evaluate the learning and memory ability of mice. The expression of glial fibrillary acidic protein (GFAP) in astrocytes was detected by immunofluorescence and Western blot. High-performance liquid chromatography (HPLC) was used to detect adenosine triphosphate (ATP), adenosine diphosphate (ADP), and adenosine monophosphate (AMP) in brain tissues of mice, and the data obtained were used to calculate energy charge (EC) levels. The phosphorylation levels of liver kinase B1 (LKB1), adenosine 5′-monophosphate (AMP)-activated protein kinase (AMPK), UNC-51-like kinase 1 (ULK1), and mammalian target of rapamycin (mTOR) and the expression levels of autophagy-related proteins Beclin-1, microtuble-associated protein 1 light chain 3 (LC3)-Ⅱ/LC3-Ⅰ, and p62 in mouse brain were measured by Western blot.ResultCompared with the control group, the model group showed decreased novel object recognition index, shortened retention latency, increased error times in the step-down test, up-regulated protein expression of GFAP, decreased content of ATP, ADP, and EC in brain tissues, elevated AMP , increased levels of p-AMPK, p-LKB1, and p-mTOR, and protein expression of p62 , and down-regulated p-ULK1 level and protein expression of Beclin-1 and LC3-Ⅱ/LC3-Ⅰ(P<0.01), while the above experimental indexes were not significantly different in the control + DHYZ group. Compared with the model group, the model + DHYZ group showed increased novel object recognition index(P<0.05), prolonged retention latency(P<0.01), decreased error times(P<0.01) in the step-down test, reduced protein expression of GFAP(P<0.05), increased content of ATP, ADP, and EC in brain tissues (P<0.05, P<0.01), decreased AMP content(P<0.05), reduced p-AMPK, p-LKB1, and p-mTOR levels and protein expression of p62, and up-regulated p-ULK1 level and protein expression of Beclin-1 and LC3-Ⅱ/LC3-Ⅰ(P<0.01).ConclusionBy protecting astrocytes, DHYZ can improve energy metabolism and autophagy disorder in AD mice to improve the learning and memory ability of model mice.

Forty C57BL/6J mice with the same background and same age were randomly divided into a control group and a control + Dihuang Yinzi (0.25 g·kg-1) group, with 20 mice in each group. The learning and memory ability of mice was tested by the light-dark box test and Y-maze spontaneous alternation test. The content of glutamate (Glu) and glutamine (Gln) was measured by liquid chromatography-tandem mass spectrometry (LC-MS). Long-term potentiation (LTP) assay was used to detect synaptic plasticity in brain tissues. The protein expression levels of excitatory amino acid transporter 2 (EAAT2), postsynaptic density protein95 (PSD95), and synaptophysin (SYN) in brain tissues were measured by Western blot. Immunofluorescence was used to assess the localization and expression of EAAT2. Colorimetry was performed to detect Na+-K+ ATPase activity in mouse brain tissues.ResultAs compared with the control group, the model group showed shortened residence latency (P<0.01), increased number of errors (P<0.01) in the light-dark box test, reduced spontaneous alternation behaviors (P<0.01), no significant difference in the total number of arm entries in the Y-maze spontaneous alternation test, down-regulated expression of EAAT2, PSD95, and SYN (P<0.01), blunted activity of Na+-K+ ATPase (P<0.01), up-regulated Glu level (P<0.01), down-regulated Gln level (P<0.01), and reduced relative population spike (PS) amplitude and the slope of excitatory postsynaptic potential (EPSP) (P<0.05, P<0.01), while the above experimental indexes were not significantly different in the control + Dihuang Yinzi group. Compared with the model group, the model + Dihuang Yinzi group displayed prolonged residence latency (P<0.05), decreased number of errors (P<0.01) in the light-dark box test, increased spontaneous alternation behaviors (P<0.01), no significant difference in the total number of arm entries in the Y-maze spontaneous alternation test, up-regulated expression of EAAT2, PSD95, and SYN (P<0.01), potentiated activity of Na+-K+ ATPase (P<0.01), reduced Glu level (P<0.01), up-regulated Gln level (P<0.01), and increased PS amplitude and EPSP slope (P<0.01).ConclusionDihuang Yinzi can improve cognitive dysfunction in AD mice by protecting astrocytes, increasing Glu uptake to reduce its abnormal accumulation, and protecting synaptic structure and function.
摘要:ObjectiveTo investigate the effects of Huashi Runzao prescription (HRP) on the histopathological injury and function of submandibular gland in naive non-obese diabetic (NOD/Ltj) mouse model of Sjögren's syndrome (SS) and its regulatory effect on aquaporin 5 (AQP5) expression in submandibular gland cells.MethodThe SS model was induced in NOD/Ltj mice. The NOD/Ltj female mice aged nine weeks were selected and randomly assigned into model group,HRP group (7.15 g·kg-1·d-1),and hydroxychloroquine (HCQ) group (1.30 g·kg-1·d-1), and female BALB/c mice in the same age were selected and assigned into the normal group, with six mice in each group. Drug intervention lasted eight weeks. The water consumption and salivary flow rate (SFR) of each group were recorded. The pathological staining results of the submandibular gland of mice in each group were observed and scored. AQP5 expression was determined by immunohistochemistry (IHC) and Western blot.ResultCompared with the normal group, the model group showed increased water consumption (P<0.05) and reduced SFR (P<0.05). Compared with the model group, the HRP group showed decreased water consumption (P<0.05) and increased SFR (P<0.05), and the HCQ group showed increased SFR (P<0.05). In terms of histopathological results of the submandibular gland,compared with the normal group,the model group showed increased pathological score, number of lymphocyte infiltration foci,and percentage of lymphatic infiltration area (P<0.05). Compared with the model group, the HRP group showed reduced pathological scores and number of lymphocyte infiltration foci (P<0.05), and the HRP group and the HCQ group showed reduced percentage of lymphatic infiltration area(P<0.05). The results of IHC and Western blot showed that compared with the normal group,the model group showed down-regulated expression level of AQP5 protein (P<0.05), and compared with the model group and the HCQ group,the HRP group showed up-regulated expression level of AQP5 protein (P<0.05).ConclusionHRP can improve the secretion function of submandibular gland acinous cells and glandular structure injury in SS model mice, and its mechanism may be related to the up-regulation of AQP5 protein expression level in submandibular gland cells.
摘要:ObjectiveTo observe the efficacy and safety of Huashi Runzao prescription for patients with primary Sjögren's syndrome (pSS) of combined dryness and dampness pattern.MethodA total of 105 eligible patients were randomized into the experimental group (65 cases) and control group (40 cases), and they were respectively treated with Huashi Runzao prescription and hydroxychloroquine for 12 weeks. Visual Analogue Scale (VAS) was employed to assess the symptoms. The symptoms of dryness, fatigue, and pain, European League Against Rheumatism (EULAR) Sjögren's Syndrome Patient Reported Index (ESSPRI), EULAR Sjögren's syndrome disease activity index (ESSDAI), and immune inflammatory indicators before and after treatment were compared between the two groups, and adverse reactions were observed.ResultAfter treatment, the ESSPRI score was lower than that before treatment in the experimental groups (P<0.01) and was lower in the experimental group than in the control group (P<0.05). The VAS scores of dry mouth, dry eyes, overall dryness, fatigue, and pain in the experimental group decreased compared with those before treatment (P<0.01), and the experimental group had lower VAS scores of dry mouth and overall dryness than the control group (P<0.01). After treatment, the ESSDAI score of both groups decreased compared with that before treatment (P<0.05, P<0.01), but there was no significant difference between the groups. After treatment, the level of immunoglobulin M (IgM) decreased (P<0.01) and the level of complement C3 increased (P<0.01) in the experimental group, while the level of complement C3 decreased in the control group (P<0.05). There was no significant difference in the laboratory indexes between groups. During the treatment, stomachache occurred to one case in the experimental group, which was alleviated after the treatment, and no adverse reaction was observed in the control group. According to the chi-square test, the occurrence of adverse reactions was insignificantly different between the two groups.ConclusionHuashi Runzao prescription can alleviate the symptoms of dryness, fatigue, and pain, and reduce disease activity without associated side effects in pSS patients with combined dampness and dryness pattern.

摘要:ObjectiveTo explore the characteristics of tongue manifestations and traditional Chinese medicine (TCM) syndromes in patients with primary Sjogren's syndrome (pSS) of different time.MethodpSS patients who visited TCM Department of Rheumatism in China-Japan Friendship Hospital from December 2018 to October 2021 were included and grouped according to the disease courses (short<5 years, medium 5-10 years, long > 10 years). Chi-square test was used for comparison between three groups and Bonferroni method for multiple comparisons. In the pairwise comparison, P<0.017 indicated significant difference. Chi-square test was performed on the syndrome and specific tongue manifestations with inter-group differences to analyze the trend of tongue manifestations and TCM syndromes over time (interval: two years).ResultA total of 193 pSS patients were enrolled, with 85 (44.0%) of short disease course, 69 (35.8%) of medium disease course, and 39 (20.2%) of long disease course. The common tongue manifestations were crimson tongue, fissured tongue, thin tongue, lack of fluid, and dry coating, which showed no significant difference among the three groups. Higher proportion of patients with light red tongue was observed in the group with short disease course than in group with medium disease course (χ2=6.407, P<0.017). Higher proportions of patients with thick coating (χ2=6.784, P<0.017) and phlegm-dampness syndrome (χ2=11.545, P<0.017) and lower proportion of patients with Qi deficiency syndrome (χ2=12.706, P<0.017) were found in the group with short disease course than in the group with long disease course. Patients with medium (χ2=6.358, P<0.017) and long (χ2=8.279, P<0.017) disease course tended to have exfoliated coating compared with those with short disease course, and the proportion of patients with exfoliated coating rose and the proportion of patients with thick greasy coating decreased over time (Ptrend<0.05). In addition, the proportion of patients with phlegm-dampness syndrome decreased and that with Qi deficiency syndrome increased over time (Ptrend<0.05).ConclusionIn the early stage, patients with pSS often show both dryness and dampness, as manifested by the thick greasy coating and phlegm-dampness syndrome. In the medium and late stage, patients often have Qi-Yin deficiency, as evidenced by exfoliated coating and Qi deficiency. In the clinical practice, medicines should be prescribed based on tongue manifestations and TCM syndrome of patients.

摘要:ObjectiveTo analyze the clinical characteristics of geographic tongue in patients with primary Sjögren's syndrome (pSS).MethodPatients with pSS treated in the China-Japan Friendship Hospital from December 2018 to October 2021 were enrolled and divided into different groups according to the presence of geographic tongue or the severity. Phi correlation analysis,Chi-square test, and Mann-Whitney test were used to analyze the clinical characteristics of patients with geographic tongue.ResultA total of 182 pSS patients were included in this study,including 75 (41.2%) patients with geographic tongue and 107 (58.8%) without geographic tongue. Partial exfoliation was more common than total exfoliation, and the anterior part of the tongue was the most common exfoliation site. The number of patients with severe geographic tongue was more than those with mild-to-moderate geographic tongue. Compared with pSS patients without geographic tongue,those with geographic tongue were more common in fissured tongue (92.0%/41.1%,χ2=48.491,P<0.05),red or crimson tongue (48.0%/23.3%,χ2=12.009,P<0.05),Yin deficiency syndrome (100.0%/43.9%,χ2=62.739,P<0.05),and Qi deficiency syndrome (94.7%∶50.5%,χ2=40.046,P<0.05),less common in phlegm-dampness syndrome (33.3%/72.0%,χ2=26.709,P<0.05),and showed higher proportions in hyperglobulinemia (89.3%/65.4%,χ2=13.547,P<0.01),ANA ≥1∶160 (78.1%/57.3%,χ2=8.227,P<0.01),and positive RF (51.4%/36.5%,χ2=3.877,P<0.05). Compared with pSS patients with mild geographic tongue,pSS patients with moderate-to-severe geographic tongue had higher proportions in hyperglobulinemia (98.1%/68.2%,χ2=14.617,P<0.01),positive anti-CENP-B (26.0%/4.8%,χ2=4.214,P<0.05),and reduced complement 3 (26.4%/4.5%,χ2=4.647,P<0.05). The geographic tongue was positively associated with fissured tongue (φ=0.531),Yin deficiency syndrome (φ=0.587),and Qi deficiency syndrome (φ=0.469),negatively associated with phlegm-dampness syndrome (φ=-0.447),and weakly associated with tongue color (φ<0.4). There was no statistical difference in the disease activity index between patients with or without geographic tongue and severity.ConclusionMore than 40% of pSS patients had geographic tongue accompanied by fissured tongue. Geographic tongue is positively associated with Yin deficiency syndrome and Qi deficiency syndrome,and negatively associated with phlegm-dampness syndrome, indicating that treatment should be based on tonifying Qi and nourishing Yin. Compared with pSS patients without geographic tongue, those with geographic tongue may have higher positive rate of some immune indicators,which deserves further exploration.
摘要:ObjectiveTo summarize the characteristics of traditional Chinese medicine (TCM) syndrome in primary Sjögren's syndrome (pSS) patients with interstitial lung disease (ILD) and to explore associated factors.MethodA survey was conducted and pSS patients who were treated in TCM department of rheumatism at China-Japan Friendship Hospital from December 2018 to April 2022 were included. Tongue manifestations and syndromes of patients were recorded. pSS patients with ILD were classified into the pSS-ILD group and those without the ILD were included in the pSS-non-ILD group. The tongue manifestations, syndromes, and laboratory indexes were compared between the two groups, and logistic regression was used to explore the factors associated with pSS-ILD.ResultA total of 200 pSS patients were included, with 186 (93.0%) females, median age of 57 years, and median disease course of 60 months, of which 44 (22%) had pSS-ILD. In terms of tongue manifestations, pSS-ILD patients generally had dark/purple/stasis tongue, fissured tongue, and tongue with little fluid, thick coating, yellow coating, and greasy coating. The proportion patients with yellow coating was higher in pSS-ILD group than in the pSS-non-ILD group (χ2=4.799,P<0.05). In terms of syndrome, more than 40% of pSS-ILD patients had Qi deficiency, Yin deficiency, phlegm-dampness, Qi stagnation, and/or blood stasis syndrome. As for Yin deficiency, liver-kidney Yin deficiency syndrome ranked the first. For Qi deficiency, lung Qi deficiency syndrome was most commonly seen. The proportion of patients with lung Qi deficiency was higher in the pSS-ILD group than in the pSS-non-ILD group (χ2=18.667,P<0.01). As to laboratory indexes, compared with the pSS-non-ILD group, pSS-ILD group had high proportion of anti-SSA-positive patients (P<0.05) and high levels of C-reactive protein (CRP) (P<0.01), complement C3 (χ2=4.332,P<0.05), and complement C4 (P<0.05). Logistic regression analysis showed that pSS with ILD was positively associated with lung Qi deficiency [odds ratio (OR)=6.079, 95% confidence interval (CI) 2.585-14.298, P<0.01)] and yellow coating (OR=5.260, 95% CI 1.337-20.692, P<0.05) and negatively associated with low C4 (OR=0.199, 95% CI 0.070-0.564, P<0.01).ConclusionAbout 22% of pSS patients had ILD, and patients with pSS-ILD generally have Qi deficiency, Yin deficiency, phlegm-dampness, Qi stagnation, and/or blood stasis syndrome. Yellow coating, lung Qi deficiency and C4 level are factors associated with pSS combined with ILD.
摘要:Sjögren's syndrome (SS), a disorder of immune system, is one of the dominant diseases treated by traditional Chinese medicine (TCM). China Association of Chinese Medicine organized experts in the field of TCM and western medicine rheumatology and pharmacology to discuss the advantages and optimal regimens of TCM for the treatment of SS. The experts generally agreed on the low early diagnosis rate of SS and the lack of targeted therapeutic drugs. In addition, autoimmune abnormality is the key factor in the occurrence of SS and deficiency of both Qi and Yin is the core pathogenesis. SS has unique tongue manifestations, which is expected to allow for the early diagnosis and treatment with integrated traditional Chinese and western medicine. TCM has advantages in treating SS in terms of alleviating clinical symptoms and systemic involvement, individualized treatment, relieving sleep and mood disorders, preventing the occurrence in the early stage, and enhancing the effectiveness and reducing toxicity in the treatment by integrated TCM and western medicine. In general, TCM has advantages in different stages of SS. Internal and external use of TCM, acupuncture, and acupotome are all available options. The optimal regimens should be determined on the basis of pattern identification, stage of disease, and the advantages of TCM. Clinical characteristics and biomarkers of SS should be studied to classify patients, so as to design precision evidence-based TCM regimens for SS. On the basis of unique tongue manifestations of SS, models for early diagnosis and poor prognosis identification of SS should also be established to achieve early prevention and treatment and to improve the prognosis. In the future, we should vigorously carry out high-quality evidence-based medical research on the treatment of SS by TCM and integrated traditional Chinese and western medicine and develop relevant guidelines to optimize and standardize current diagnosis and treatment, thereby laying a basis for clarifying and explaining the advantages of TCM in treating SS.

摘要:By reviewing ancient materia medica and combining with modern literature, the textual research of Magnoliae Flos has been conducted to verify the name, origin, producing area, harvesting and processing methods, in order to provide basis for the selection and use of this herb in the development of famous classical formulas. After the textual research, it could be seen that the correct name of Magnoliae Flos was Xinyi in the past dynasties, meaning spicy flower buds. The main original plants used in past dynasties are Magnolia denudata and M. biondii. The history of the research on its scientific name in recent times is complicated, many foreign scholars have given several different scientific names, but most of them are inconsistent with the actual situation of Magnoliae Flos used in ancient China, because foreign scholars failed to collect the original plants of Magnoliae Flos for accurate identification. Before the Ming dynasty, Magnoliae Flos was mainly produced in Shaanxi, and then the recorded production areas gradually increased. After the founding of the People's Republic of China, the products produced in Henan named M. biondii were highly respected, and Henan was regarded as authentic producing area, and because of the collection and distribution through Yuzhou, it was customarily called Huichunhua. In ancient times, the harvesting period of Magnoliae Flos mostly concentrated in the first and second months of the lunar calendar, and the flower buds of M. biondii were also recommended to be used as medicine, but nowadays the flower buds are mostly collected in winter and spring, and those with dry buds, large size, yellow-green color, tight inner petals, fragrant smell, and no impurities are preferred. In the past dynasties, raw products were the mainstream, and there were also frying, soaking and so on. Based on the results, it is suggested that the flower buds of M. biondii should be used in the development of famous classical formulas. If the original formula does not specify the processing requirements, the raw products can be used as medicine.

摘要:Through consulting the ancient materia medica and medical books, combined with modern literature, this paper made a textual research on the name, origin, producing area, harvesting time and processing method of Piperis Kadsurae Caulis, in order to provide basis for the development of the famous classical formulas containing this herb. According to textual research, it is shown that the earliest name for Piperis Kadsurae Caulis as medicine was Nanteng in Bencao Shiyi, and there were other names such as Dinggongteng and Shinanteng in the ancient materia medica. The name of Haifengteng should appear in the Ming dynasty. Before the Song dynasty, the origin of Piperis Kadsurae Caulis was probably derived from caulis of Piper wallichii. After the Song dynasty, the main origins should be some species in Piper, such as P. kadsura and P. hancei, and its origin in the successive editions of Chinese Pharmacopoeia was only P. kadsura. Combining the original plant research, market survey and distribution of wild resources, it is suggested that the Haifengteng used in the famous classical formulas apart form the P. kadsura, the P. hancei should be add as original plant. Due to climate change and the heat-loving habit of Piper, the producing area of Haifengteng gradually moved from the Qinling Mountains to the southern areas rich in Piper, and Quanzhou area of Fujian province has been recommended since the Ming dynasty. The harvesting period of Piperis Kadsurae Caulis is from July to August in the lunar calendar, the above-ground parts are dried by removing fibrous roots, thin stems and leaves. In the past dynasties, there are few records on the processing of this herb, so it is suggested that Piperis Kadsurae Caulis in famous classical formulas without special processing requirements should be used as raw products.

摘要:By reviewing ancient materia medica, medical and prescription books, combined with modern literature, the textual research of Stephaniae Tetrandrae Radix has been conducted to verify the name, origin, producing area, harvesting and processing methods. Through textual research, the results show that the mainstream name of this herb recorded in the past dynasties is Fangji, which is also called Hanzhong Fangji because it is produced in Hanzhong city, and after the Tang dynasty, it was gradually divided into Hanfangji and Mufangji, and there is the saying that Han Zhushuiqi, Mu Zhufengqi. The names of Fenfangji and Guangfangji were first seen in the republic of China. In addition, Fenfangji was once distributed in Hankou, so it was also known as "Hanfangji", which is easily confused with the traditional Hanzhong Fangji for short. Based on the original research, it is concluded that Aristolochia heterophylla(Hanzhong Fangji)is the mainstream of Stephaniae Tetrandrae Radix used in the Qing dynasty and before, and the application history of Cocculus orbiculatus can be traced back to before the Tang dynasty. After the Ming dynasty, Stephania tetrandra gradually became another main origin, and in the Republic of China, A. fangchi was used as a medicine for Stephaniae Tetrandrae Radix, but in modern times it was banned because it contained aristolochic acid as a toxic ingredient, and S. tetrandra has become the mainstream legal origin. The traditional production area of Hanzhong Fangji is Hanzhong, Shaanxi province, while today the mainstream of S. tetrandra is manly produced in Jiangxi and other places. Based on the quality evaluation research, the quality of Hanzhong Fangji is better with the radial texture of section used as radial solution, yellow solid and fragrant. Fenfangji with solid quality, white inside, powdered enough, less fiber and radiating texture is better. From the harvesting and processing research, the root of Fangji is mostly harvested in spring and autumn, and the outer bark should be removed in some literature. Before the Ming dynasty, this herb was dried in the shade, and after the Ming dynasty, it was dried in the sun. The modern production processing of Fangji is to harvest it in autumn, wash it, remove the rough bark, dry it to half dry, cut it into sections, and then cut it longitudinally if it is large, and dry it. Based on the results, combined with current studies on the toxicity of aristolochic acid and influencing factors such as commercial circulation, it is suggested that S. tetrandra should be used as the origin of Fangji, the processed products are selected according to the prescription requirements, and those without specified requirements can be processed by referring to the raw products in the 2020 edition of Chinese Pharmacopoeia.

摘要:In this paper, the name, origin, quality evaluation, producing area and processing methods of Lablab Semen Album in the famous classical formulas were researched by reviewing the ancient materia medica, medical books, prescription books and modern literature. The results showed that the name of Lablab Semen Album in the past dynasties was mostly derived from its shape and color, called Biandou and Baibiandou. The mainstream origin used in the past dynasties was Lablab purpureus, the medicinal parts were mainly white mature seeds, with the addition of the leaves in the Song dynasty and the flowers in the Ming dynasty. Since modern times, the authentic producing areas of Lablab Semen Album are Suzhou, Zhejiang and other places, and now mainly produced in Chuxiong and Xinping, Yunnan and Panzhihua, Sichuan. The traditional quality evaluation of Lablab Semen Album is evaluated as large, solid, full and white. The harvesting time of this herb is recorded from the eighth to the ninth lunar month in related literature, the pods are picked when the seeds are ripe, and the seeds are dried in the sun. In ancient times, the processing of Lablab Semen Album mainly consisted of frying the seeds with skin and then pulverizing for use, or soaking and peeling seeds for raw use. Based on the conclusion of the textual research, it is recommended that the seeds or flowers of the white flowering plants of L. purpureus, a member of the leguminosae, should be used in the famous classical formulas, and the dried seeds or dried flowers of Lablab Semen Album can be used as medicine if the formula did not clearly indicate processing requirements.

摘要:ObjectiveTo explore the effect of Baitouweng Tang (BTWT) on the apoptosis of human colorectal cancer HCT116 cells and decipher the underlying mechanism based on the Hedgehog (Hh) signaling pathway.MethodHCT116 cells were treated with BTWT (25, 50, 100, 200, 500, 750, and 1 000 mg·L-1) for 24 h, and then the cell proliferation was detected by methyl thiazolyl tetrazolium (MTT) colorimetry. Five groups were designed for the treatment of HCT116 cells, including a blank control group, BTWT groups (125, 250, and 500 mg·L-1), and a positive control (5-fluorouracil, 5-FU, 40 mmol·L-1) group. The cell morphology was observed under an inverted microscope. The migration of the cells was detected by scratch test, and the apoptosis by Hoechest 33324/propidium iodide (PI) staining and flow cytometry. Western blot was employed to determine the protein levels of sonic hedgehog (SHh), GLI family zinc finger protein 1 (Gli1), smoothened (Smo), suppressor of fused (SuFu), cellular-myelocytomatosis viral oncogene (c-Myc), and the apoptosis-related proteins B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X protein (Bax). The quantitative real-time reverse transcription PCR (Real-time PCR) was employed to determine the mRNA levels of Bax, Bcl-2, SHh, Gli1, Smo, SuFu, and c-Myc.ResultCompared with the blank control group, BTWT changed the cell morphology (making the cell become round with dense nucleus), inhibited the proliferation of HCT116 cells in a dose-dependent manner, decreased the ability of migration (P<0.05, P<0.01), and increased apoptotic cells. Compared with the blank control group, BTWT (500 mg·L-1) treatment for 24 h up-regulated the protein and mRNA levels of Bax (P<0.05, P<0.01) and down-regulated the protein and mRNA levels of Bcl-2 in HCT116 cells (P<0.05, P<0.01). Moreover, the treatment down-regulated the mRNA and protein levels of SHh, Gli1, Smo, and c-Myc (P<0.05, P<0.01) and up-regulated the mRNA and protein levels of SuFu (P<0.05, P<0.01).ConclusionBTWT inhibited the proliferation and migration and induced the apoptosis of colorectal cancer HCT116 cells by down-regulating the Hh signaling pathway.
摘要:ObjectiveTo study the effect of Bushen Huoxuetang on the apoptosis and the expression of B-cell lymphoma (Bcl-2)-associated X protein (Bax)/ Bcl-2 and cleaved cysteine-containing aspartate proteolytic enzyme-3 (cleaved Caspase-3) in the nude mouse model of bone metastasis of breast cancer, and explore the mechanism of Bushen Huoxuetang in inhibiting bone destruction.MethodThirty BALB/c female nude mice were randomly assigned into blank group (n=6) and model group (n=24). The suspension of 4T1 breast cancer cells was injected into the tibia of mouse right lower limb to establish model of bone metastasis of breast cancer. The successfully modeled nude mice were randomly assigned into model group, Bushen Huoxuetang group, zoledronic acid group, and combined drug group, with 6 mice in each group. Bushen Huoxuetang was administrated at a dose of 36.67 g·kg-1, once a day, and zoledronic acid was administrated by subcutaneous injection at a dose of 100 μg·kg-1, twice a week. The combined drug group was administrated with the same doses of Bushen Huoxuetang group by gavage and zoledronic acid by subcutaneous injection. The mice in the blank group and the model group were administrated with the same volume of distilled water by gavage for 14 days. On the next day at the end of drug administration, the mice were sacrificed by cervical dislocation. The general situation and weight changes of the mice were examined. The right lower limb was collected, and X-ray scanning and hematoxylin-eosin (HE) staining methods were used for observation of pathological changes in the bone. The terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) was employed to detect the apoptosis of bone tissue in nude mice, and Western blot to determine the expression of Bax/Bcl-2 and cleaved Caspase-3 in the bone tissue.ResultCompared with the blank group, the modeling reduced the body weight (P<0.01) and increased the right lower limb weight of the nude mice (P<0.01). Compared with the model group, Bushen Huoxuetang, zoledronic acid, and their combination increased the body weight (P<0.01) and decreased the right lower limb weight (P<0.01). Compared with the blank group, the other groups showed obvious tumor cell atypia, deep nuclear staining, and clear bone metastasis, and the model group showed obvious osteolytic damage in right lower limb and loss of proximal tibia and knee joint. Compared with the model group, Bushen Huoxuetang, zoledronic acid, and their combination reduced the osteolytic lesions in the right lower limb and recovered part of the bone structure, demonstrating an inhibitory effect on bone destruction. The TUNEL assay showed that the model group had lower apoptosis rate of bone metastatic tumor cells than the blank group, Bushen Huoxuetang group, zoledronic acid group, and combined drug group (P<0.01). Compared with the blank group, the modeling down-regulated the expression of Bax and cleaved Caspase-3 (P<0.01) and up-regulated the expression of Bcl-2 (P<0.01). Compared with the model group, Bushen Huoxuetang, zoledronic acid, and their combination up-regulated the expression of Bax (P<0.01) and cleaved Caspase-3 (P<0.05, P<0.01) and down-regulated the expression of Bcl-2 (P<0.05, P<0.01).ConclusionBushen Huoxuetang may inhibit bone destruction in the nude mouse model of bone metastasis of breast cancer by up-regulating the expression of Bax, down-regulating the expression of Bcl-2, activating cleaved Caspase-3, and further inducing apoptosis.

摘要:ObjectiveTo explore the protective effect of Xielitang on ulcerative colitis (UC) mice induced by dextran sodium sulfate (DSS) and its possible mechanism.MethodSixty C57BL/6 mice were randomly divided into normal group, model group, sulfasalazine group and and low-, medium-, and high-dose Xielitang groups. Free drinking DSS solution to build the chronic UC model mice. Except for normal group, other groups were given 1.5% DSS for 3 cycles of drinking (days 1-7, days 22-28 and days 43-49) and distilled water for the rest of the time (days 8-21, days 29-42 and days 50-63). After the first cycle, corresponding drugs were given for 42 days. The changes of general condition, body weight and disease activity index (DAI) score of mice were daily recorded during the experiment. At the end of the treatment, serum and colon tissue samples were collected, colon length was measured, intestinal weight index and colonic mucosal injury (CMDI) score were calculated. The pathological status of colon tissue was observed by hematoxylin-eosin (HE) staining. The levels of interleukin-6 (IL-6), interleukin-10 (IL-10) and tumour necrosis factor-α (TNF-α) were measured by enzyme-linked immunosorbent assay (ELISA). The gene and protein expressions of Toll like receptor 4 (TLR4), nuclear transcription factor-κB (NF-κB) and hypoxia inducible factor-1α (HIF-1α) in colon tissue was detected by Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot.ResultCompared with the normal group, the body weight, colon length and IL-10 content in the model group were significantly decreased (P<0.01), DAI score, intestinal weight index, CMDI score, IL-6 and TNF-α contents, and mRNA and protein expression levels of TLR4, NF-κB and HIF-1α in the model group were significantly increased (P<0.01). Moreover, the structure of colonic mucosa was destroyed and inflammatory cells infiltrated in the model group. Compared with model group, body weight, colon length and IL-10 content in each dose group of Xielitang were significantly increased (P<0.05, P<0.01), DAI score, intestinal weight index and CMDI score, IL-6 and TNF-α contents, mRNA and protein expression levels of TLR4, NF-κB and HIF-1α were notably decreased (P<0.05, P<0.01). The pathological injury of colon was obviously alleviated.ConclusionXielitang can significantly improve the inflammatory response of UC mice induced by DSS, and its mechanism may be related to the regulation of TLR4/NF-κB/HIF-1α signaling pathway.
摘要:Baizhusan, derived from Key to Medicines and Patterns of Children's Diseases (《小儿药证直诀》) by QIAN Yi in Song dynasty, is a classical prescription that has been highly praised by doctors and widely used in clinical practice. It was included in the Catalogue of Ancient Classical Prescriptions (the second batch of Pediatrics) released by the National Administration of Traditional Chinese Medicine (TCM) in 2022. In this paper, the ancient documents related to Baizhusan were collected by bibliographical research, and the source, composition, herb origin and processing, dosage, decocting method, administration method, functions and indications of the prescription were analyzed and verified. A total of 211 ancient documents on Baizhusan were obtained, involving 116 ancient TCM books. It was found that there were many prescriptions named Baizhusan in ancient times, but the prescription named Baizhusan and composed of Ginseng Radix et Rhizoma, Poria, Atractylodis Macrocephalae Rhizoma, Pogostemonis Herba, Aucklandiae Radix, Glycyrrhizae Radix et Rhizoma, and Puerariae Lobatae Radix was first seen in Key to Medicines and Patterns of Children's Diseases, and is now generally known as Qiweibaizhusan. The composition and functions of Baizhusan were unified in past dynasties, and its herb origin and processing were basically clear. However, there was a dispute on the processing of Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix et Rhizoma, which was solved by recommended use of soil-fried Atractylodis Macrocephalae Rhizoma and raw Glycyrrhizae Radix et Rhizoma. Boasting the functions of tonifying spleen, replenishing Qi, harmonizing stomach and promoting fluid production, Baizhusan treated a variety of symptoms such as diarrhea, consumptive thirst and loss of appetite, which were consistent in ancient and modern literature, but attention should be attached to the basic pathogenesis of spleen deficiency and fluid inadequacy. In addition, considerable disagreements were also observed about the dosage and decocting method of Baizhusan in the documents, and the combination ratio of Baizhusan in another version of Key to Medicines and Patterns of Children's Diseases of Wuyingdian Zhenben was the dominant standard for drug use in past dynasties.

摘要:Jupi Zhurutang originated from Synopsis of the Golden Chamber (《金匮要略》), which consists of Pericarpium Citri Reticulatae, Caulis Bambusae in Taenia, Ginseng Radix et Rhizoma, Zingiberis Rhizoma Recens, Jujubae Fructus, and Glycyrrhizae Radix et Rhizoma and is used to treat retching. It has been put on the list of Catalogue of Ancient Classical Prescription (First Batch) released by National Administration of Traditional Chinese Medicine. With the bibliometric method, we searched the medical classics containing Jupi Zhurutang and systematically examined the information on the origin of the prescription, the indications, compatibility rule, medicinals in the prescription, dosage and usage, processing method, and decocting method. It was found that there are many versions of Jupi Zhurutang, and there are common grounds of main symptoms, pathogenesis, composition and dosage between the same prescription with different names and different prescriptions with the same name. The prescription which is closest to the original version in Synopsis of the Golden Chamber is mainly used for the treatment of stomach deficiency and qi counterflow without obvious cold or heat. According to the weights and measures, ratio of Pericarpium Citri Reticulatae, Caulis Bambusae in Taenia, Radix Ginseng, Rhizoma Zingiberis Recens, and Radix Ginseng in Synopsis of the Golden Chamber is approximately 6∶2∶8∶5∶1. The Jupi Zhurutang derived from other ancient classics such as Yanshi Jisheng Fang(《严氏济生方》) is a different prescription for hiccups caused by the stomach heat, and the ratio of Pericarpium Citri Reticulatae to Caulis Bambusae in Taenia in this prescription is about 1∶1. It is also found that cold herbs such as Red Poria, Eriobotryae Folium and Ophiopogon Japonicus are added to the formula in later generations. Therefore, the Jupi Zhurutang used in modern times is mostly modified and different from that in Synopsis of the Golden Chamber. This study summarizes the historical evolution of Jupi Zhurutang and identifies the key information, with a view to providing a reference for the rational modification of this prescription in clinical settings and further research.
摘要:Zhishi Xiebai Guizhi Tang, derived from Synopsis of the Golden Chamber (《金匮要略》, Han dynasty, ZHANG Zhongjing), is a famous classical prescription commonly used for chest impediment. By means of bibliometrics, the authors collected 63 ancient Chinese medical books related to Zhishi Xiebai Guizhi Tang and screened 36 effective books for statistical analysis of the historical origin, composition, main indications, dosage, processing, decocting method and other aspects of the prescription. The findings revealed that Zhishi Xiebai Guizhi Tang was composed of five medicinal herbs, namely, Aurantii Fructus Immaturus, Allii Macrostemonis Bulbus, Cinnamomi Ramulus, Magnoliae Officinalis Cortex, and Trichosanthis Fructus, with the function of activating Yang, dissipating mass, dispelling phlegm and lowering Qi. The prescription was mainly used to treat chest impediment, chest fullness, chest pain, wheezing, coughing and shortness of breath caused by suppressed Yang Qi, stagnant heart Qi, stagnant phlegm and stasis, and phlegm evil blocking heart, chest or lungs. Additionally, it was found that there were 70 modern literature recording the clinical applications of Zhishi Xiebai Guizhi Tang, and the main system diseases treated were circulatory system (51, 72.85%), endocrine system (4, 5.7%), respiratory system (9, 12.85%) and digestive system (6, 8.57%), of which circulatory system is dominated by coronary heart disease (chest impediment in traditional Chinese medicine). The involved medical syndrome types mainly included internal obstruction of phlegm heat and turbidity, obstruction of phlegm turbidity and stasis, congealing cold, phlegm, stasis and Qi stagnation, chest Yang depression and combined phlegm and stasis. Ancient medical records and modern clinical application are the keys to ensure the safety and effectiveness of famous classical prescriptions and compound preparations. Therefore, this paper sorted and mined ancient medical books of Zhishi Xiebai Guizhi Tang and statistically analyzed its modern clinical application, aiming to provide a literature reference for the research and development of new drugs and clinical application of the prescription.
摘要:MethodIn the experiment, 46% vol Red Star Erguotou (10 mL·kg·d-1) was used to establish the AONFH rat model, and the intervention effect of JPHGP at different doses (2.5, 5.0, 10.0 g·kg-1) was observed. Jiangusheng pill (JGS, 1.53 g·kg-1) was selected as the positive control. After 8 weeks of administration, the bone histomorphometry of the femoral head was analyzed by Micro-CT imaging, and the area of medullary microvessels in the femoral head was detected by ink perfusion. The pathological change was observed by hematoxylin and eosin (HE) staining. The protein expressions of Platelet endothelial cell adhesion molecule-1 (CD31), VEGF, VEGFR2, PI3K, phosphor-Akt (p-Akt) and phosphatase and Tensin homologue deleted on chromosome 10 (PTEN) in the femoral head were determined by immunohistochemistry and Western blot.ResultCompared with normal group, the model group presented the fracture and thinning of trabeculae in the femoral head, increased empty bone lacunae, and elevated number and diameter of adipocytes (P<0.01). Micro-CT imaging revealed a decrease in bone mineral density (BMD), bone volume fraction (BV/TV), trabecular thickness (Tb.Th) and trabecular number (Tb.N) (P<0.05, P<0.01) while an increase in bone surface-to-volume ratio (BS/BV) and trabecular separation (Tb.Sp) (P<0.01). The results of ink perfusion showed that the area of medullary microvessels in the femoral head was reduced (P<0.01). Compared with model group, JPHGP lowered the empty bone lacunae rate as well as the number and diameter of adipocytes in the femoral head of AONFH rats. Micro-CT imaging indicated that JPHGP low-dose group had elevated BV/TV, Tb.Th and Tb.N (P<0.05, P<0.01) while decreased BS/BV (P<0.01), and there was an upward trend in BMD while a downward trend in Tb.Sp, but without statistical difference. In addition, JPHGP medium- and high-dose groups had a rise in BMD, BV/TV, Tb.Th and Tb.N (P<0.05, P<0.01), a decrease in BS/BV and Tb.Sp (P<0.05, P<0.01) and enlarged area of medullary microvessels in the femoral head (P<0.05, P<0.01). The expressions of CD31, VEGF, VEGFR2, PI3K, p-Akt in the model group were lower than those in the normal group (P<0.01), and after medium and high doses of JPHGP treatment, the expressions of CD31, PI3K and p-Akt in the femoral head of rats were up-regulated (P<0.01) while the protein expression of PTEN was down-regulated (P<0.01). Moreover, JPHGP up-regulated the expressions of VEGF and VEGFR2 (P<0.05, P<0.01).ConclusionJPHGP can repair the vascular injury in AONFH, and its mechanism may be related to the activation of VEGF/VEGFR2/PI3K/Akt signaling pathway. This study provides certain scientific basis and reference for the clinical application of JPHGP.ObjecctiveTo observe the repair effect of Jianpi Huogu prescription (JPHGP) on vascular injury in experimental alcohol-induced osteonecrosis of femoral head (AONFH), and to explore its mechanism based on vascular endothelial growth factor (VEGF)/VEGFR2/phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway.

摘要:ObjectiveTo explore the amelioration of cognitive dysfunction in diabetes mellitus (DM) by Jianpi Qinghua prescription (JPQH) based on type 2 diabetes (T2DM) model rats.MethodFifty healthy male Wistar rats of SPF grade were randomly divided into control group (n=10) and experimental group (n=40). The rats in the control group were fed conventionally, while those in the experimental group were fed on a high-sugar, high-fat diet for six weeks and administered with streptozotocin (STZ) for the induction of the DM model. The model rats were randomly divided into model group, sitagliptin group (1.2 g·L-1), pioglitazone group (0.8 g·L-1), and JPQH group (1.3 g·mL-1), with 10 rats in each group. After six weeks of drug intervention, the changes in body weight, blood glucose, and other related indexes of each group were recorded. Enzyme-linked immunosorbent assay (ELISA) was performed to detect the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the peripheral blood and brain. The Morris water maze test was used to evaluate the cognitive function in rats. Hematoxylin-eosin (HE) staining was used to observe the pathological morphology of the hippocampal CA region. The amyloid β-protein 40 (Aβ40) level was detected by immunohistochemistry. The protein expression of t-tau and p-tau in hippocampal neurons of rats was detected by Western blot.ResultCompared with blank group, the body weight of model group was significantly decreased (P<0.05), blood glucose level was significantly increased (P<0.01), inflammatory cytokines TNF-α and IL-1β were increased (P<0.05), learning and spatial ability were significantly decreased (P<0.01), the arrangement of hippocampal cells was loose and disordered, and the intercellular space was significantly increased. The number of cells decreased significantly, and the expression of Aβ40 increased significantly. and increased t-tau and p-tau protein content in the hippocampus (P<0.01). Compared with model group, the JPQH group showed reduced blood glucose (P<0.01), decreased TNF-α and IL-1β levels in the peripheral blood and cerebrospinal fluid (P<0.05), a downward trend of IL-6 without a statistical difference, improved learning and spatial memory ability (P<0.01), densely arranged cells in the hippocampal CA1 area, increased cell number, reduced Aβ40 expression, and decreased p-tau protein expression (P<0.05).ConclusionJPQH can prevent cognitive dysfunction in DM by reducing inflammatory factor levels, decreasing neurotoxicity caused by Aβ40 deposition, and inhibiting hyperphosphorylation of tau protein in DM rats.

摘要:ObjectiveTo observe the effect of salvianolate on the protein expressions of adenosine monophosphate (AMP)-activated protein kinase (AMPK), silent information regulator 1 (SIRT1) and peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α), autophagy and apoptosis in kidney tissue of rats with membranous nephropathy (MN), and to explore its possible molecular mechanism against MN.MethodEighty male SD rats were randomly divided into normal group, model group, benazepril hydrochloride group (10 mg·kg-1), and salvianolate low-, medium-, and high-dose groups (16.7, 33.3 and 66.7 mg·kg-1). The rats were modeled by injection of cationized bovine serum albumin (C-BSA) into the tail vein. After successful modeling, rats in the administration groups were given corresponding doses of drugs for 4 consecutive weeks, and then 24-hour urine, serum and kidney tissue were collected for the detection of 24-hour urinary protein (UTP), blood urea nitrogen (BUN), serum creatinine (SCr), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), C reactive protein (CRP), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and malondialdehyde (MDA). The pathological changes of kidneys were observed by light microscope, electron microscope and immunofluorescence. Western blot was used to detect the protein expressions of phospho-AMPK (p-AMPK), AMPK, phospho-SIRT1 (p-SIRT1), SIRT1 and PGC-1α in rat kidney tissue. The protein expressions of autophagy-specific gene (Beclin-1), microtubule-associated protein 1 light chain 3 (LC3) Ⅱ, ubiquitin-binding protein (p62), B cell lymphoma (Bcl-2), Bcl-2-associated X (Bax), and cysteine aspartic protease-7 (Caspase-7) in rat kidney tissue were determined by immunohistochemistry (IHC).ResultCompared with the conditions in the normal group, the levels of UTP, IL-6, TNF-α, CRP and MDA in the model group were increased (P<0.05) while the levels of SOD and GSH-Px were decreased (P<0.05), and there was no difference in BUN and SCr. Compared with the model group, the administration groups had lowered UTP, IL-6, TNF-α, CRP and MDA (P<0.05) while elevated SOD and GSH-Px (P<0.05). It could be seen from hematoxylin and eosin (HE) staining, Masson staining, immunofluorescence and electron microscopy that the pathological damage of rat kidney tissue in the model group was significant, but after treatment with benazepril hydrochloride and salvianolate, the pathological damage of kidney cells was gradually improved. The expressions of p-AMPK/AMPK, p-SIRT1/SIRT1, PGC-1α, Bcl-2, Beclin-1 and LC3Ⅱ in rat kidney in the model group were lower than those in the normal group (P<0.05) while the expressions of Bax, Caspase-7 and p62 were higher (P<0.05). Compared with the model group, benazepril hydrochloride group and salvianolate groups had an up-regulation in the expressions of p-AMPK/AMPK, p-SIRT1/SIRT1, PGC-1α, Bcl-2, Beclin-1 and LC3Ⅱ in the kidney (P<0.05) while a down-regulation in the expressions of Bax, Caspase-7 and p62 (P<0.05).ConclusionThe protective effect of salvianolate on the kidneys of MN rats may be related to the activation of AMPK/SIRT1/PGC-1α signaling pathway, the up-regulation of autophagy and the reduction of apoptosis.

摘要:ObjectiveTo investigate effect of Maxing Shigantang and supplemented Guominjian decoction on symptoms and levels of inflammatory cytokines in induced sputum of children with cough variant asthma (CVA).MethodA total of 118 CVA children who were treated in our hospital from January 2020 to January 2021 were enrolled and randomized into the control group and the observation group with the random number table method. Control group received routine western medicine and the observation group was treated by routine western medicine, Maxing Shigantang, and supplemented Guominjian decoction. In the one-month follow-up, the scores of cough and accompanying symptoms, levels of inflammatory cytokines [interleukin-10 (IL-10), interleukin-5 (IL-5), tumor necrosis factor-α (TNF-α), neutrophil, eosinophil] in induced sputum, pulmonary function parameters [forced vital capacity (FVC), forced expiratory volume in one second (FEV1), FEV1/FVC], and treatment outcomes were compared between the two groups. Moreover, the frequency of acute asthma attacks during the three-month follow-up was also compared.ResultNo cases dropped out from this study. After treatment, the scores of cough and accompanying symptoms were decreased in both groups (P<0.05) and were lower in observation group than in control group (P<0.05). After treatment, FVC, FEV1, and FEV1/FVC were raised in both groups and were higher in observation group than in control group (P<0.05). The increase in the level of IL-10 along with the decrease in levels of IL-5, TNF-α, neutrophil, and eosinophil in induced sputum was found in both groups after treatment (P<0.05), and observation group had higher level of IL-10 and lower levels of IL-5, TNF-α, neutrophil, and eosinophil than the control group (P<0.05). The effective rate was 86.44% (51/59) in observation group, which was higher than the 69.49% (41/59) in control group (χ2=4.933, P<0.05). No serious adverse reaction occurred in either group. The frequency of acute asthma attacks during the three-month follow-up was (1.09±0.18) in observation group, which was lower than the (2.83±049) in the control group (P<0.05).ConclusionRoutine western medicine combined with Maxing Shigantang and supplemented Guominjian decoction can effectively and safely alleviate the airway inflammatory responses, control the clinical symptoms, improve pulmonary function, and reduce the frequency of acute recurrence in the treatment of CVA children.

摘要:ObjectiveTo analyze relevant literature on Lianhua Qingwen preparations and clarify the research advances and hot spots in this field, so as to provide references for clinical rational application and further research.MethodLiterature related to Lianhua Qingwen preparations in the recent 10 years was retrieved from six databases, including China National Knowledge Infrastructure(CNKI), VIP, Wanfang Data, PubMed, and Web of Science, followed by management and analysis by NoteExpress and CiteSpace.ResultFinally, 344 and 76 Chinese and English research articles were included, and the number of publications increased in recent years. The research articles were published in 162 Chinese and 48 English journals. Shijiazhuang Yiling Pharmaceutical Co., Ltd. and Guangzhou Medical University were institutions with the largest number of Chinese and English publications, respectively. LIU Minyan was the author who had published the most articles. Keywords with high frequency included clinical efficacy, Lianhua Qingwen, inflammatory factors, traditional Chinese medicine, and coronavirus disease-2019(COVID-19). Nineteen clusters, including clinical efficacy, Chinese medicine, Lianhua Qingwen, COVID-19, and influenza A virus, and 47 emergent keywords, including herpes zoster, pneumonia, inflammatory factors, influenza, and gut microbiota, were generated.ConclusionCooperation and exchanges in this field are insufficient. Research focuses on the clinical efficacy of Lianhua Qingwen in the treatment of COVID-19 and other diseases, pharmacological action and mechanism of antiviral drugs, and micro-mechanism research focuses on related pathways and target proteins, as well as the combination of Chinese and western medicines.
摘要:Ulcerative colitis (UC) is one of the chronic refractory inflammatory bowel diseases characterized by abdominal pain, diarrhea, and mucus, pus and blood in the stool. In recent years, with changes in human life style and improvements of the diagnosis, the incidence and prevalence of UC have been increasing. The pathogenesis of UC is closely related to intestinal mucosal immune dysfunction, intestinal flora disturbance, and abnormal bile acid secretion. Patients with UC have abnormal bile acid secretion and intestinal flora imbalance. A large number of studies have found that abnormal bile acid secretion inhibits immune function, affects signal transduction, and destroys the intestinal mucosal barrier. Intestinal flora disturbance has an important impact on the occurrence and development of inflammation, immune homeostasis, and stress. Bile acids indirectly or directly affect the structure and function of intestinal flora, and at the same time, they produce secondary bile acids under the modification of intestinal flora, entering the liver through enterohepatic circulation. Therefore, the complex dialogue mechanism of bile acid-intestinal flora axis is closely related to the occurrence and development of UC. Based on the basic theory of traditional Chinese medicine(TCM) and clinical research, it is found that emotion is an important factor that induces this disease, spleen and stomach weakness is the root of the disease, and liver depression and spleen deficiency are the key pathogenesis of UC. Combined with modern medicine and molecular biology research, it is believed that abnormal secretion of bile acids is a microscopic manifestation of liver depression in TCM, and intestinal flora disturbance is the biological basis of spleen deficiency. In the pathogenesis of UC, the imbalanced bile acid-intestinal flora axis is consistent with the pathogenesis of liver depression and spleen in TCM. The exploration of the biological connotation of the pathogenesis of UC with liver depression and spleen deficiency from the perspective of bile acid-intestinal flora axis can better explain the scientific nature of its pathogenesis, which provides new clinical solutions and reliable references for studying the pathogenesis of UC with liver depression and spleen deficiency and finding representative prescriptions to prevent and treat this disease.

摘要:Tongxie Yaofang, also known as Baizhu Shaoyaosan, was first recorded in Danxi's Experiential Therapy (《丹溪心法》) by ZHU Danxi in the Yuan dynasty. It is composed of Atractylodis Macrocephalae Rhizoma, Paeoniae Radix Alba, Citri Reticulatae Pericarpium, and Saposhnikoviae Radix, and serves as the representative prescription for the treatment of painful diarrhea. It has the functions of tonifying the spleen, emolliating the liver, relieving pain, and checking diarrhea, and is mainly used in the treatment of gastrointestinal diseases such as irritable bowel syndrome (IBS) and ulcerative colitis (UC). In addition, it is effective in treating gastrointestinal disorders with mental and psychological abnormalities, as well as obstinate anorexia in children, depression syndrome, and respiratory diseases. Experimental research and clinical practice have shown that Tongxie Yaofang has multi-component, multi-pathway, and multi-target characteristics in the treatment of diseases. The mechanism of Tongxie Yaofang in treating diseases is mainly attributed to anti-inflammation, immune function regulation, intestinal hypersensitivity improvement, emotion regulation, etc. Monoterpene glycosides, flavonoids, chromones, lactones, and other components contained play an important therapeutic role. The research on the systems biology of Tongxie Yaofang, such as metabolomics, proteomics, and network pharmacology, provides a scientific basis for clarifying its mechanism of action and expanding its clinical application. However, there are still some problems to be solved, such as difficulty in combining diseases and syndromes and lack of in-depth systematic research. Through the retrieval and collation of relevant literature, this paper systematically reviewed the material basis, pharmacological effects, and systems biology research of Tongxie Yaofang, aiming to lay a foundation for in-depth research on its mechanism in treating diseases and rational application in clinical practice.
摘要:Fibrosis refers to the disorder of repair response after tissue damage, which is mainly characterized by the increase of fibrous connective tissue and the decrease of parenchymal cells in organs. Progressive fibrosis can lead to the destruction of tissue structure and organ failure. Ferroptosis is a new type of programmed cell death, which is caused by the accumulation of iron dependent lipid peroxides and regulated by iron metabolism, iron autophagy, amino acid metabolism and lipid metabolism. Ferroptosis plays an important role in the pathological process of fibrosis, participating in many pathological processes such as organ parenchymal cell injury, macrophage inflammation, oxidative stress and myofibroblast transformation. Studies have shown that ferroptosis occurs in myofibroblasts during extracellular matrix deposition in the fibrous cascade reaction, and the targeted regulation of ferroptosis can effectively reduce chronic organ injury and tissue fibrosis. It has been confirmed that a variety of traditional Chinese medicine and its active components can regulate the ferroptosis parenchyma cells of organs and play an anti-fibrosis effect, showing a good research prospect. This article summarizes the role of ferroptosis in the pathological process of liver, lung, kidney and heart fibrosis and the intervention of related Chinese medicine, in order to provide a new target for Chinese medicine to prevent and treat fibrosis.

摘要:As a rare Chinese medicinal material, Paridis Rhizoma is mainly distributed in Yunnan, Guangxi, and Guizhou in southwestern China, with the effect of clearing heat and detoxifying, alleviating edema and relieving pain, cooling liver and tranquilizing mind. It is particularly effective for injuries from falls, fractures, contusions and strains, snake bites, cold wind-induced convulsion, and other diseases, which has been used for more than 2 000 years. According to modern research, polyphyllin Ⅱ, one of the main active components of Paridis Rhizoma, belongs to diosgenin in structure. It has the anti-tumor, anti-inflammatory, antiviral, antibacterial, immune-regulating, antioxidant, and multidrug resistance-reversing activities, showing good application prospect. Especially, the anti-tumor effect of polyphyllin Ⅱ has attracted wide attention, and the mechanism is inhibiting proliferation, migration, and invasion of tumor cells, inducing cell cycle arrest, apoptosis, and autophagy, suppressing angiogenesis, and modulating tumor microenvironment. However, the pharmacokinetic results show that polyphyllin Ⅱ has low bioavailability in vivo due to the low solubility, poor absorption, unsatisfactory distribution, and slow metabolism, which limit the clinical application. In recent years, there has been an explosion of research on the adverse reactions of polyphyllin Ⅱ, such as the strong hemolytic activity and obvious cytotoxicity to liver, kidney, myocardium and cardiovascular cells. Thus, papers were retrieved from "CNKI", "VIP", "Wanfang Data", "PubMed", "Web of Science", and "Elsevier SD" with "Paris saponin Ⅱ", "Polyphyllin Ⅱ" as the main keywords, and the pharmacological activities and mechanisms, pharmacokinetics, and adverse reactions were summarized. The findings are expected to serve as a reference for the in-depth research, development, and utilization of polyphyllin Ⅱ.
摘要:Traditional Chinese medicine dispensing granules(TCMDGs)is the new type of decoction pieces with the development of modernization of TCM, which has received mixed opinions since its practical application. In 2021, the national departments issued Announcement on Ending the Pilot Work of TCMDGs, marking the end of the 28-year pilot work of TCMDGs, and eligible TCM enterprises can produce TCMDGs after filing. However, this does not mean that the preparation process, quality standard and efficacy research of TCMDGs have been developed and matured, on the contrary, there are still some problems that need to be solved and gradually improved. For example, in the production process, there are problems such as unclear, unified and non-standardized preparation parameters. In terms of quality control, there are some problems such as lack of producing area regulation, variety selection and processing specification. In terms of consistency evaluation with traditional decoction, there are problems such as unclear relationship between the chemical constituents and pharmacological effects of the two. Therefore, in view of some prominent problems of TCMDGs at present, this paper takes the published literature as the main data source and combines the specific requirements of the code or technical standards such as the 2020 edition of Chinese Pharmacopoeia, Publicity of the Unified Standard on the Varieties of TCMDGs, Quality Control and Standard Formulation Technical Requirements of TCMDGs. The production process of TCMDGs, the origin and variety of raw materials, the processing of decoction pieces, the quality control standard and the consistency evaluation of formula granules and traditional decoction were sorted out and visualized by literature mining, data analysis and list comparison. Based on the analysis results, the following suggestions were made. In terms of preparation process, the completeness and standardization of process parameters should be strengthened. In terms of quality evaluation, attention should be paid to the relationship between the authenticity, variety, processing and quality of medicinal materials. In the consistency evaluation of formula granules and traditional decoction, the deep difference and mechanism between TCMDGs and traditional decoction were discussed by combining structural Chinese medicine, quality marker(Q-Marker) theory and physicochemical characterization, so as to provide reference for the application and development of TCMDGs.

摘要:Malignant tumor poses a threat to human health and life. The incidence and fatality rate of malignant tumor have been on the rise. The currently available therapies are radiotherapy and chemotherapy, which cause severe adverse reactions and irreversible damage and thus influence the quality of life of patients. Therefore, it is urgent to find new, safe and effective antitumor drugs. Chinese medicinals are safe with little adverse reactions and long-lasting effect in the treatment of tumor, which have attracted the attention of scholars. Amid the advancement of medical research, more and more anti-tumor components have been extracted from Chinese medicinals. Phellinus is a valuable Chinese medicinal material, and the chemical components mainly include polysaccharides, flavonoids, triterpenes, and polyphenols, which have anti-inflammatory, hypoglycemic, liver-protecting, and anti-tumor effect. The chemical components of Phellinus can inhibit various malignant tumors such as lung cancer, gastric cancer, colon cancer, and melanoma. It exerts the anti-tumor effect by inhibiting proliferation and metastasis of tumor cells, inducing apoptosis and autophagy of the cells, suppressing tumor angiogenesis, and regulating the immunity. In addition, it can enhance efficacy, reduce toxicity, and boost the sensitivity in the radiotherapy and chemotherapy. In this paper, articles were retrieved from China National Knowledge Infrastructure (CNKI), Web of Science, Pubmed, and Google Scholar with keywords such as "Phellinus, chemical components, and anti-tumor", and then the chemical components of Phellinus and the anti-tumor mechanisms were summarized. The findings are expected to lays a basis for further development and clinical application of this medicinal.
