摘要:ObjectiveTo explore the mechanism of Buzhong Yiqitang in ameliorating inflammatory injury in autoimmune thyroiditis (AIT) mice based on the Toll-like receptor 4(TLR4)/nuclear transcription factor-kappa B(NF-κB)/absent in melanoma 2(AIM2)inflammasome signaling pathway.MethodThe 120 genetically susceptible 8-week-old NOD.H-2h4 mice were selected and randomly divided into control group, model group, low, medium and high dose groups of Buzhong Yiqitang (4.78, 9.56, 19.12 g·kg-1), and western medicine group (selenium yeast tablets, 3.033×10-5 g·kg-1). The AIT model mice in each group drank ad libitum 0.05% sodium iodide aqueous solution for 8 weeks to establish the AIT model, and the control group drank ad libitum distilled water. Eight weeks later, the mice in each dosing group were divided into groups and gavage. The swelling of thyroid tissue was observed with the naked eye, and the weight of spleen was weighed. The content of serum inflammatory factors interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) was measured by enzyme-linked immunosorbent assay (ELISA), and Real-time PCR was used to detect the expression of HMGB1, TLR4, AIM2, NF-κB p65,apoptosis-associated speck-like protein(ASC),cysteinyl aspartate-specific protease-1(Caspase-1), IL-1β mRNA. Western blot was used to detect the expression of high motility group protein 1 (HMGB1), TLR4, AIM2, NF-κB p65, phosphorylation(p)-NF-κB p65, ASC, Caspase-1, and IL-1β proteins in thyroid tissue, and immunofluorescence staining was used to observe the protein expression of HMGB1, AIM2, and NF-κB p65 in thyroid tissue of mice.ResultCompared with the control group, the thyroid tissue of mice in the model group was significantly swollen, the spleen quality was significantly increased, and the expression of HMGB1, TLR4, NF-κB p65, AIM2, ASC, Caspase-1, IL-1β in thyroid tissue was significantly increased (P<0.01). Compared with the model group, the swelling of thyroid tissue in mice in each dose group of Buzhong Yiqitang was improved, the quality of spleen was significantly reduced, and the expression of HMGB1, TLR4, AIM2, NF-κB p65, p-NF-κB p65, ASC, Caspase-1, IL-1β in thyroid tissue was significantly reduced (P<0.05, P<0.01).ConclusionBuzhong Yiqitang can effectively improve the inflammatory injury of AIT, and regulating the abnormal activation of the TLR4/NF-κB/AIM2 inflammasome signal pathway may be one of its intervention mechanisms.
摘要:ObjectiveTo investigate the mechanism of Buzhong Yiqitang in ameliorating ferroptosis in autoimmune thyroiditis (AIT) mice based on the nuclear factor erythroid 2-related factor 2 (Nrf2)/peroxisome proliferator-activated receptor γ (PPARγ)/glutathione peroxidase 4 (GPX4) pathway.Method120 SPF-grade 7-8-week-old NOD.H-2h4 mice were randomly divided into control group, model group, low-, medium-, and high-dose Buzhong Yiqitang groups, and western medicine group, with 20 mice in each group. Except for the control group, all mice were fed with classic high-iodine water (0.05% NaI) to induce AIT models after 8 weeks. The low-, medium-, and high-dose Buzhong Yiqitang groups were administered 4.78, 9.56, 19.12 g·kg-1 of Buzhong Yiqitang, respectively, via gavage. The western medicine group was given 3.033×10-5 g·kg-1 selenium yeast tablet suspension via gavage, while the control and model groups were given an equal volume of distilled water via gavage. After 8 weeks of continuous treatment, samples were collected. The pathological morphology of mouse thyroid tissue was observed through hematoxylin-eosin (HE) staining,the content of serumantithyroid peroxidase autoantibody(TPOAb)and anti-thyroglobulin antibodies(TGAb)was measured by enzyme-linked immunosorbent assay (ELISA),the kit was used to detect the levels of superoxide dismutase (SOD), and malondialdehyde (MDA) in mouse serum. Immunofluorescence was used to detect the localized expression of GPX4 in thyroid tissue. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was used to detect the expression of Nrf2, PPARγ, solute carrier family 7 member 11 (SLC7A11), solute carrier family 3 member 2 (SLC3A2), lysolipid lecithin acyltransferase 3 (LPCAT3), and GPX4 mRNA in thyroid tissue. Western blot was used to detect the expression of Nrf2, PPARγ, SLC7A11, SLC3A2, LPCAT3, and GPX4 proteins in thyroid tissue.ResultCompared with control group, model group under light microscopy showed significant lymphocyte infiltration in the thyroid tissue, significantly increased levels of TGAb and TPOAb in serum (P<0.01), significantly increased MDA levels and decreased SOD levels in serum (P<0.01), significantly decreased expression of Nrf2, PPARγ, SLC7A11, SLC3A2, and GPX4 (P<0.01) in thyroid tissue, while the expression of LPCAT3 was significantly increased (P<0.01). Compared with model group, the Buzhong Yiqitang groups and the western medication group under light microscopy showed lymphocyte infiltration in the thyroid tissue of was decreased, significantly decreased levels of TPOAb and TGAb in serum (P<0.05,P<0.01), decreased MDA levels and increased SOD levels in serum(P<0.05,P<0.01),significantly increased expression of Nrf2, PPARγ, SLC7A11, SLC3A2, and GPX4, while the expression of LPCAT3 was significantly decreased (P<0.05,P<0.01) in the thyroid tissue. Compared with western medication group, Buzhong Yiqitang groups showed significant overall trends in the expression of Nrf2, PPARγ, SLC7A11, SLC3A2, GPX4, and LPCAT3 (P<0.05,P<0.01).ConclusionBuzhong Yiqitang can effectively improve the inflammatory injury of AIT, and its mechanism of action may be related to the regulation of Nrf2/PPARγ/GPX4 to inhibit ferroptosis.
摘要:ObjectiveTo explore the mechanism of Buzhong Yiqitang in improving autoimmune thyroiditis (AIT) by regulating helper T cell 17(Th17) cells through microRNA-155 (miR-155)/Nedd4 family interaction protein 1 (Ndfip1)/phosphatase and tensin homology (Pten) axis.MethodThe 100 SPF grade 8 week-old NOD.H-2h4 mice were fed with high iodine water (0.05% NaI) for 8 weeks, and AIT model was made. They were divided into model group, Buzhong Yiqitang low-,medium-,and high-dose groups (4.78,9.56,19.12 g·kg-1·d-1) and selenium yeast tablet group (3.033×10-5 g·kg-1) according to random number table method. There were 20 mice in each group and 20 mice in the control group. The control group and the model group were given the same amount of distilled water. After 8 weeks of continuous administration, Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was used to detect the expression of miR-155-5p, Ndfip1, Pten, protein tyrosine kinase 1 (Jak1), signaling and transcriptional activator 3 (Stat3) retinoic acid-associated orphan receptor γt (RORγt), and interleukin-17 (IL-17) mRNA in mouse thyroid tissue. Western blot was used to detect the expression of Ndfip1, Pten, Jak1, Stat3, RORγt, and IL-17 proteins in mouse thyroid tissue, immunohistochemical method was used to detect the expression of Ndfip1 and Pten proteins in mouse thyroid tissue; flow cytometry was used to detect the proportion of Th17 cells in mouse spleen.ResultCompared with the control group, the proportion of Th17 cells was increased (P<0.01). The expressions of miR-155-5p, Jak1, Stat3, RORγt and IL-17 were increased (P<0.01), while the expressions of Ndfip1 and Pten were decreased (P<0.01). Compared with model group, the proportion of Th17 cells was decreased (P<0.05,P<0.01). The expressions of miR-155-5p, Jak1, Stat3, RORγt and IL-17 were decreased (P<0.05,P<0.01), while the expressions of Ndfip1 and Pten were increased (P<0.05,P<0.01).ConclusionThe application of Buzhong Yiqitang can improve the autoimmune disorder of AIT mice, the mechanism of which may be related to the regulation of Ndfip1/Pten axis by miR-155 and then the regulation of Th17 cell differentiation.

摘要:ObjectiveTo investigate the effects of Buzhong Yiqitang on the tumor necrosis factor receptor superfamily member 6 (Fas)/Fas related death domain protein (FADD)/Caspase-8 cell apoptotic signaling pathway in mice model with autoimmune thyroiditis (AIT).MethodThere were 120 SPF grade NOD.H-2h4 mice aged 8 weeks, 100 of which were fed with high iodine water (0.05% NaI), and the AIT model was made after 8 weeks. According to random number table method, they were divided into model group, Buzhong Yiqitang low-dose, medium-dose and high-dose groups (4.78, 9.56, 19.12 g·kg-1), selenium yeast tablet group (3.033×10-5 g·kg-1), with 20 mice in each group and 20 control group. The apoptosis of thyroid cells was detected by in situ end labeling (TUNEL) after 8 weeks of administration. The real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression of Fas, FADD, Caspase-8, and Caspase-3 in thyroid tissue. Western blot was used to detect the protein expression of Fas, FADD, Caspase-8, Caspase-3, cleaved Caspase-8, and cleaved Caspase-3 in thyroid tissue, and the immunohistochemical method was used to detect the protein expression of Fas and Caspase-3 in thyroid tissue.ResultCompared with control group, there were more positive expressions of apoptotic cells in model group under fluorescence microscope, and the expressions of Fas, FADD, Caspase-8, Caspase-3, cleaved Caspase-8 and cleaved Caspase-3 were significantly increased (P<0.05, P<0.01). Compared with model group, the positive expression of thyroid apoptotic cells in each administration group was decreased under fluorescence microscope, and the expressions of Fas, FADD, Caspase-8, Caspase-3, cleaved Caspase-8, cleaved Caspase-3 were significantly decreased (P<0.05, P<0.01).ConclusionBuzhong Yiqitang can effectively improve thyroid cell apoptosis and inflammatory injury in AIT mice. The mechanism may be related to the inhibition of Fas/FADD/Caspase-8 signaling pathway.

摘要:ObjectiveTo investigate the inhibitory effect of Mahuang Xixin Fuzitang on the migration of dendritic cells (DCs) in mice and its underlying mechanism.MethodMouse bone marrow-derived DCs were isolated and cultured. The morphological changes of the cells at different stages were observed under a microscope, and the CD11c+ proportion was detected by flow cytometry to identify DC purity. Cells were treated with Mahuang Xixin Fuzitang (1, 2, 5, 10, 20, 40, 50, 100 g·L-1) for 24 hours, and the effect of Mahuang Xixin Fuzitang on cell proliferation was assessed using the cell counting kit-8 (CCK-8) assay to determine the appropriate concentrations for treatment. After modeling by lipopolysaccharide (LPS) induction, DCs were divided into a blank group, a model group, and Mahuang Xixin Fuzitang groups (2, 4, 8 g·L-1). The expression of surface molecules CD80, CD86, and major histocompatibility complex-Ⅱ (MHC-Ⅱ) were detected by flow cytometry. Transwell chamber assay was used to observe cell migration. The levels of chemokine C-C-primitive receptor 7 (CCR7) and chemokine C-X-C-primitive receptor 4 (CXCR4) on the cell surface were detected by enzyme-linked immunosorbent assay (ELISA). The expression of filamentous actin (F-actin) in the cell microfilament cytoskeleton was detected by immunofluorescence (IF) staining. Real-time quantitative polymerase chain reaction (Real-time PCR) was employed to determine the mRNA expression levels of Ras homolog family member A (RhoA) and Rho-associated coiled-coil containing protein kinase 1 (ROCK1). Western blot analysis was performed to detect the protein expression of RhoA and ROCK1.ResultCompared with the blank group, the model group exhibited significantly higher expression levels of CD80, CD86, and MHC-Ⅱ (P<0.01), a significantly increased number of cells migrating to the lower chamber (P<0.01), and significantly elevated levels of CCR7 and CXCR4 (P<0.05, P<0.01). Additionally, F-actin expression was significantly increased (P<0.01), and both RhoA and ROCK1 mRNA and protein expression levels were significantly higher (P<0.05, P<0.01). Compared with the model group, treatment with Mahuang Xixin Fuzitang (2, 4, 8 g·L-1) for 24 hours resulted in significantly lower expression levels of CD80, CD86, and MHC-Ⅱ (P<0.01), a significantly reduced number of cells migrating to the lower chamber (P<0.05), and significantly decreased levels of CCR7 and CXCR4 (P<0.05, P<0.01). Furthermore, F-actin expression was significantly reduced (P<0.01), and both RhoA and ROCK1 mRNA and protein expression levels were significantly decreased (P<0.05, P<0.01).ConclusionMahuang Xixin Fuzitang can inhibit the migration of DCs in mice, and its mechanism of action may be related to reducing the activity of the Rho/ROCK signaling pathway, thereby affecting changes in the cell cytoskeleton.

摘要:ObjectiveTo investigate the effects of Guizhi Fulingwan on oxidative stress and renal fibrosis in diabetic mice based on the nuclear factor E2-associated factor 2/antioxidant response element (Nrf2/ARE) pathway, and to explore its mechanism of action.MethodA diabetic mouse model was established by feeding high-fat diet combined with intraperitoneal injection of streptozotocin. The diabetic mice were divided into a model group, a metformin group (250 mg·kg-1), and Guizhi Fulingwan low, medium, and high dose groups (400.4, 800.8, 1 601.6 mg·kg-1), with 10 mice in each group. Another 10 mice were included in a normal group. The treatment was conducted continuously for 8 weeks. Body mass and blood glucose were recorded, and the renal index was calculated. Triglyceride (TG), total cholesterol (TC), urea nitrogen (BUN), serum creatinine (SCr), superoxide dismutase (SOD), and malondialdehyde (MDA) levels were measured. Pathological changes in renal tissues were observed using Hematoxylin-eosin, periodate-Schiff, and Masson staining. The protein levels of α-smooth muscle actin (α-SMA), fibronectin (FN), and transforming growth factor-β1 (TGF-β1) were detected by immunohistochemistry. The protein levels of Nrf2, Kelch-like ECH-associated protein 1 (Keap1), SOD1, NAD(P)H single-nucleotide oxidase 1 (NQO1), and heme oxygenase-1 (HO-1) were detected by Western blot.ResultCompared with the normal group, the model group showed elevated glucose, renal index, TG, TC, BUN, and SCr levels, thickened glomerular basement membrane, increased collagen fibers in renal tissues, decreased SOD activity, elevated MDA content, increased protein levels of FN, TGF-β1, α-SMA, and Keap1, and decreased protein levels of Nrf2, SOD1, NQO1, and HO-1 (P<0.01). Compared with the model group, the Guizhi Fulingwan and metformin groups showed reduced blood glucose, renal index, TG, TC, BUN, and SCr levels, improved renal histopathological injury, significantly increased SOD activity, significantly reduced MDA content, reduced protein levels of FN, TGF-β1, α-SMA, and Keap1, and increased protein levels of Nrf2, SOD1, NQO1, and HO-1 (P<0.05, P<0.01).ConclusionGuizhi Fulingwan can improve oxidative stress and inhibit renal fibrosis, and its mechanism may be related to the activation of the Nrf2/ARE signaling pathway.
摘要:ObjectiveTo investigate the therapeutic effect of Dayuanyin on acute lung injury induced by H1N1 infection and decipher the potential mechanism.MethodThe constituents in Dayuanyin were analyzed by ultra-high performance liquid chromatography-quadrupole-exactive orbitrap mass spectrometry (UHPLC-Q-Exactive Orbitrap MS). Forty-eight female BALB/c mice were randomized into normal, model, oseltamivir (19.5 mg·kg-1), and low-, medium-, and high-dose (2.73, 5.46, 10.92 g·kg-1) Dayuanyin groups. The normal and model groups were administrated with deionized water by gavage, and the other groups were administrated with the corresponding drugs by gavage. On day 3 of drug administration, the normal group received nasal inhalation of normal saline, and the other groups were inoculated intranasally with A/RP/8/34 (H1N1) for the modeling of influenza virus infection. Mice were administrated with drugs continuously for 7 days and weighed daily. Sampling was performed 12 h after the last administration, and the lung tissue was weighed to calculate the lung index. Hematoxylin-eosin staining was performed to observe the pathological and morphological changes of the lung tissue and bronchi. The cytometric bead array (CBA) was used to measure the serum levels of interferon-gamma (IFN-γ), C-X-C motif ligand 1 (CXCL1), tumor necrosis factor-alpha (TNF-α), chemokine ligand 2 (CCL2), interleukin-12p70 (IL-12p70), chemokine ligand 5 (CCL5), interleukin-1β (IL-1β), chemokine (C-X-C motif) ligand 10 (CXCL10), granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-10 (IL-10), interferon-beta (IFN-β), interferon-alpha (IFN-α), and interleukin-6 (IL-6). According to the results of mass spectrometry and network pharmacology, we analyzed the mechanism of Dayuanyin in treating acute lung injury caused by H1N1. The protein levels of extracellular signal-regulated kinase 1/2 (ERK1/2), p38 mitogen-activated protein kinase (p38 MAPK), nuclear factor-kappa B (NF-κB), and their phosphorylated forms were determined by Western blot. The mRNA levels of myeloid differentiation factor 88 (MyD88), Toll-like receptor 3 (TLR3), Toll-like receptor 7 (TLR7), and Toll-like receptor 8 (TLR8) in the lung tissue were measured by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR).ResultA total of 57 compounds, including paeoniflorin and baicalein, were detected in Dayuanyin. Compared with the normal group, the model group showed decreased body weight (P<0.01), lung edema and hemorrhage, increased lung index (P<0.01), and elevated levels of IFN-γ, IL-12p70, CCL5, IL-1β, CXCL10, GM-CSF, IFN-β, and IL-6 (P<0.01). Compared with the model group, Dayuanyin attenuated alveolar wall thickening, capillary congestion, and immune cell infiltration, reduced the alterations in body weight and lung index (P<0.01), and down-regulated the protein levels of IFN-γ, IL-12p70, CCL5, IL-1β, CXCL10, GM-CSF, IFN-β, and IL-6 (P<0.01). A total of 57 key genes were predicted by network pharmacological analysis, of which the MAPK signaling pathway was the main target signaling pathway. Compared with the normal group, the model group showed up-regulation in the protein levels of phosphorylation (p)-ERK1/2, p-p38 MAPK, and p-NF-κB (P<0.01) and the mRNA levels of TLR7, TLR8, MyD88, and TLR3 (P<0.05, P<0.01). Compared with the model group, Dayuanyin lowered the phosphorylation levels of ERK1/2, p38 MAPK, and NF-κB p65 in a dose-dependent manner (P<0.01) and down-regulated the mRNA levels of TLR3, TLR7, TLR8, and MyD88 (P<0.01).ConclusionDayuanyin can prevent and control H1N1 infection-induced acute lung injury by inhibiting the TLR/MAPK/NF-κB signaling pathway.

摘要:ObjectiveTo observe the effects of Agrimoniae Herba and Coptidis Rhizoma(XHC-HL) on the proliferation, migration, invasion, apoptosis, and autophagy of colorectal tumor cells and explore the mechanism of Agrimoniae Herba and Coptidis Rhizoma in inhibiting colorectal cancer by regulating chaperone-mediated autophagy(CMA).MethodXHC-HL-containing serum was prepared, and human colorectal cancer cell lines HT29 and LOVO were cultivated and divided into blank group (20% blank serum), low XHC-HL groups (5%, 10%, 20% drug-containing serum), and a positive control group using 5-fluorouracil (5-FU). Cell viability was measured using the cell counting kit-8 (CCK-8) method. Cell proliferation ability was assessed through 5-ethynyl-2′-deoxyuridine (EdU) staining. Scratch assays and Transwell migration tests were conducted to evaluate cell migration ability, and Transwell invasion assays were used to assess cell invasion ability. Apoptosis rates were determined by flow cytometry, and the impact of XHC-HL on yeast Atg6 homologous 1 (Beclin-1), microtubule-associated protein1 light chain3 Ⅱ (LC3 Ⅱ), and p62 was analyzed via Western blot. The influence of XHC-HL on CMA-related proteins and mRNA was examined using Western blot and real-time fluorescence quantitative polymerase chain reaction (Real-time PCR).ResultCompared to that in the blank group, the vitality of colorectal cancer cells HT-29 and LOVO was significantly inhibited in XHC-HL groups and the 5-FU group (P<0.01). Compared to that in the blank group, the proliferation of colorectal cancer cells HT-29 and LOVO was significantly inhibited in XHC-HL groups and the 5-FU group (P<0.05, P<0.01). Compared to the blank group, there was a significant reduction in cell migration rates (P<0.01), the number of cells migrating to the lower chamber (P<0.01), and the number of invasive cells (P<0.01), as well as and an increase in cell apoptosis (P<0.01) in XHC-HL groups and the 5-FU group (P<0.01). Western blot results indicated that compared to that in the blank group, the expression levels of Beclin-1 and LC3 Ⅱ were increased(P<0.05,P<0.01)in XHC-HL groups and the 5-FU group, while the p62 levels were decreased(P<0.05,P<0.01). Furthermore, the protein expression levels of lysosomal associated protein 2A (LAMP2A), heat shock cognate protein 70 (HSC70), and heat-shock protein 90 (HSP90) in XHC-HL groups and the 5-FU group were decreased to varying extents, whereas the expression levels of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) were elevated (P<0.01). The Real-time PCR results showed that compared to those in the blank group, the mRNA levels of LAMP2A, HSC70, and HSP90 were downregulated in XHC-HL groups and the 5-FU group, and the mRNA expression level of GAPDH was upregulated (P<0.01).ConclusionXHC-HL can inhibit the proliferation, migration, and invasion of colorectal cancer cells by suppressing CMA activity, thus inducing autophagy and promoting apoptosis.

摘要:ObjectiveTo compare the effects of different doses and withdrawal time of 4-vinylcyclohexene diepoxide (VCD) on the reproductive endocrine levels of female rats, and to explore the effective, stable, and safe dosage of VCD for constructing a premature ovarian insufficiency (POI) rat model.MethodSD rats with regular estrous cycles were randomly divided into three groups: blank group, low-dose VCD group (80 mg·kg-1·d-1), and high-dose VCD group (160 mg·kg-1·d-1), with 24 rats in each group. After drug intervention, samples were collected on the 15th day (D15) and the 45th day (D45) after intervention. The general condition, rate of estrous cycle disturbance, serum hormone levels, ovarian histomorphology, follicle count, pregnancy outcome, and the protein and mRNA expression of transforming growth factor (TGF)-β and Smad2/3 were assessed.ResultCompared with the blank group, the low-dose VCD group showed no significant differences in the rate of estrous cycle disturbance or serum follicle-stimulating hormone (FSH), luteinizing hormone (LH), and estradiol (E2) levels. Ovarian tissue was damaged. Specifically, the number of primordial and primary follicles decreased on D15 (P<0.01), and the number of secondary follicles (P<0.01) and antral follicles (P<0.05) further decreased on D45. The litter number decreased on D15 (P<0.05), but there was no significant difference on D45. Furthermore, TGF-β protein levels increased on D15 (P<0.05) and D45 (P<0.01). The Smad2/3 levels increased on D45 (P<0.01), and TGF-β and Smad2/3 mRNA levels increased on D45 (P<0.05). Compared with the results in the blank group, the disturbance rate of the estrous cycle increased on D45 in the high-dose VCD group (P<0.01). The serum of FSH and LH increased (P<0.01), while E2 decreased (P<0.05). Ovarian tissue was damaged, and the downward trend of follicles at all levels was similar to that in the low-dose VCD group. The litter number significantly decreased on D15 and D45. TGF-β and Smad2/3 protein levels increased (P<0.05,P<0.01), and TGF-β mRNA increased on D45 (P<0.05).ConclusionHigh-dose VCD is an ideal method for constructing a POI rat model, being effective, stable, and safe.
摘要:ObjectiveTo explore the antioxidant and anti-human cervical cancer HeLa cell effect and mechanisms of Andrographis paniculata polysaccharide (APP).MethodCell function assays were conducted to assess the effects of APP (400, 450, 500 mg·L-1) on the proliferation, migration, and invasion of HeLa cells using the cell counting kit-8 (CCK-8) assay, scratch assay, and Transwell assay. Molecular mechanism experiments were conducted to detect the effects of APP on HeLa cell apoptosis and cell cycle-related mRNA and protein expression using flow cytometry, real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR), and Western blot analysis. The antioxidant activity of APP was tested using DPPH+, OH-, and reducing power assays.ResultCompared with the blank group, APP (400, 450, 500 mg·L-1) significantly inhibited the migration, proliferation, and invasion abilities of HeLa cells in a time- and dose-dependent manner (P<0.05, P<0.01). Flow cytometry with propidium iodide (PI) single staining was used to detect cell cycle. The results showed that compared with the blank group, the proportion of HeLa cells in the G2/M phase increased after 48 hours of treatment with APP (400, 450, 500 mg·L-1), indicating that APP can arrest HeLa cells in the G2/M phase. Flow cytometry with fluorescein isothiocyanate (Annexin V-FITC)/PI apoptosis kit was used to detect cell apoptosis. Compared with the blank group, the proportion of early and late apoptotic HeLa cells increased in a dose-dependent manner after 48 hours of APP (400, 450, 500 mg·L-1) treatment (P<0.05, P<0.01), indicating that APP promotes HeLa cell apoptosis. The results of Real-time PCR and Western blot assay showed that compared with the blank group, after 48 hours of APP (400, 450, 500 mg·L-1) treatment resulted in decreased mRNA and protein expression of cell cycle-dependent kinase-1 (CDK-1), Cyclin B1, and B-cell lymphoma-2 (Bcl-2), and increased mRNA and protein expression of cysteine aspartate protease (Caspase)-3, Caspase-8, Caspase-9, and Bcl-2-associated X protein (Bax) (P<0.05, P<0.01). These findings were consistent with the flow cytometry results and showed a dose-dependent effect. In vitro antioxidant tests demonstrated that different concentrations of APP (50-1 000 mg·L-1) were able to scavenge DPPH+ and OH- radicals, indicating certain antioxidant activity.ConclusionAPP possesses antioxidant activity and can inhibit the viability of HeLa cells while promoting their apoptosis.

摘要:ObjectiveSerum metabolomics of acute lung injury(ALI) in rats was conducted using ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS) to explore the similarities and differences in the mechanism of Qingqiao(harvested when the fruits of Forsythiae Fructus were initially ripe and still green in color) and Laoqiao(harvested when the fruits of Forsythiae Fructus were ripe) in the treatment of ALI.MethodA total of 24 SD male rats were acclimatized and fed for 1 week, 6 of them were randomly selected for the blank group and 18 for the experimental group. The ALI model was induced in the experimental group by tracheal intubation with lipopolysaccharide(LPS). After successfully constructing the ALI model, these rats was randomly divided into model group, Qingqiao group and Laoqiao group, with 6 rats in each group. The Qingqiao and Laoqiao groups were administered orally once a day at a dose of 1.5 g·kg-1, while the blank and model groups received an equivalent volume of saline for 3 consecutive days. The pathological conditions of rat lung tissues were comprehensively assessed by hematoxylin-eosin(HE) staining, wet-to-dry mass ratio(W/D) of lung tissues, and protein concentration in rat bronchoalveolar lavage fluid(BALF). The levels of interleukin(IL)-6, IL-1β and tumor necrosis factor(TNF)-α in BALF were quantified using enzyme-linked immunosorbent assay(ELISA). UPLC-Q-TOF-MS was used to identify and analyze the chemical compositions of Qingqiao and Laoqiao, and serum metabolomics of rats in each group was analyzed, combined with multivariate statistical analysis with variable importance in the projection(VIP) value>1, P<0.05 from t-test, and fold change(FC)≥1.5 or FC≤0.5 to screen the differential metabolites Qingqiao and Laoqiao for the treatment of ALI. The Kyoto Encyclopedia of Genes and Genomes(KEGG) database was used in combination with MetaboAnalyst for the metabolic pathway analysis of the screened differential metabolites.ResultCompared with the blank group, rats in the model group exhibited enlarged alveolar lumen, ruptured alveoli, interstitial hemorrhage, bronchial exudation of a large number of neutrophils and erythrocytes, and a significant increase in the protein concentration in the BALF and the W/D value of the lung tissues(P<0.01). In contrast, compared with the model group, rats in the Qingqiao group and the Laoqiao group showed reduced bronchial hemorrhage in the lungs, and the protein concentration in the BALF and the W/D value of the lung tissues were significantly decreased(P<0.01), the lung injury was significantly alleviated, but more obvious in the Qingqiao group. Compared with the blank group, the expression levels of IL-6, IL-1β and TNF-α in the BALF of the model group were significantly higher(P<0.01). Additionally, compared with the model group, the expression levels of IL-6, IL-1β and TNF-α in the Qingqiao and Laoqiao groups were significantly lower(P<0.01). The chemical composition analysis of Qingqiao and Laoqiao revealed that 63 components were detected in Qingqiao and 55 components were detected in Laoqiao, with 47 common components, 16 components unique to Qingqiao and 8 components unique to Laoqiao. Characterizing the differences in serum metabolomics in rats, 19 and 12 metabolites were called back by Qingqiao and Laoqiao, respectively. The metabolic pathway enrichment analysis showed that Qingqiao exerted its therapeutic effects by affecting 6 key metabolic pathways, including linoleic acid metabolism, phenylalanine metabolism, phenylalanine, tyrosine and tryptophan biosynthesis, glycerophospholipid metabolism, α-linolenic acid metabolism, and arachidonic acid metabolism, and Laoqiao exerted therapeutic effects by affecting 6 key metabolic pathways, including linoleic acid metabolism, arachidonic acid metabolism, sphingolipid metabolism, phenylalanine metabolism, ascorbate and aldarate metabolism, and glycerophospholipid metabolism.ConclusionQingqiao and Laoqiao have therapeutic effects on ALI, and Qingqiao is more effective. Both of them can play a therapeutic role in ALI by regulating amino acid metabolism and lipid metabolism, but the metabolic pathways affected by them are different.

摘要:ObjectiveTo explore and verify the key pathway of Zuogui Jiangtang Jieyu prescription in the treatment of diabetes with depression by means of gene set enrichment analysis (GSEA) and short time-series expression miner (STEM).MethodSD rats were randomly divided into six groups, including a normal group, a model group, high, medium, and low dose groups of Zuogui Jiangtang Jieyu prescription, and a positive drug group. The model of diabetes with depression was established by high-fat feeding, streptozotocin (STZ) injection, and chronic mild unpredictable stress. The high, medium, and low dose groups of Zuogui Jiangtang Jieyu prescription were orally administered at 20.52, 10.26, and 5.13 g·kg-1 respectively. The positive drug group was orally administered 0.18 g·kg-1 metformin and 1.8 g·kg-1 fluoxetine. The rats in the normal group and model group were administered with an equal volume of distilled water. After 28 days, the animals were tested for depressive-like behaviors and cognitive function using the forced swimming test and Morris water maze. Fasting blood glucose was measured using blood glucose test strips. Cholesterol and triglyceride levels were measured using enzyme-linked immunosorbent assay (ELISA). Three hippocampus samples were randomly selected from the normal group, the model group, the high dose group of Zuogui Jiangtang Jieyu prescription for high-throughput transcriptome sequencing. Differential gene analysis, GSEA analysis, and STEM analysis were used to screen the key pathways and target genes of Zuogui Jiangtang Jieyu prescription in the treatment of diabetes with depression. Key target genes were validated using real-time fluorescence quantitative PCR (Real-time PCR). The expression of the signal protein mediated by the target genes was detected by Western blot.ResultCompared with the results in the normal group, the fasting blood glucose, cholesterol, and triglyceride levels in the model group were significantly increased (P<0.01). Moreover, the immobility time in the forced swimming test was significantly increased, while the time to climb the platform was significantly prolonged, and the search distance in the target quadrant was significantly reduced in the Morris water maze test (P<0.05,P<0.01). Compared with the model group, the high dose of Zuogui Jiangtang Jieyu prescription significantly reduced the levels of blood glucose, cholesterol, and triglycerides in rats with diabetes with depression (P<0.05,P<0.01), reduced the immobility time in the forced swimming test, shortened the stage time in the Morris water maze test, and increased the search distance ratio in the target area (P<0.05,P<0.01). Transcriptome sequencing differential analysis showed that the normal group had 1 366 differentially expressed genes compared to the model group, while the model group had 1 149 differentially expressed genes compared to the high dose group of Zuogui Jiangtang Jieyu prescription, with 581 intersecting genes. The GSEA results showed that there were 9 sets of differentially expressed genes between the normal group and the model group, and 43 sets of differentially expressed genes between the model group and the high dose group of Zuogui Jiangtang Jieyu prescription, with 7 intersecting gene sets. STEM analysis showed that according to the analysis order of the normal group, model group, and high dose group of Zuogui Jiangtang Jieyu prescription, two significantly different trend clustering groups were obtained. One key gene set for axonal guidance, as well as key target signal elements Sema3c, Sema7a, Robo3, Epha8, and Epha7, were identified through synthesizing the three analysis results. Real-time PCR validated that compared with the results in the normal group, the mRNA expression of Robo3, Sema7a, and Epha7 in the hippocampus of the model rats was significantly reduced (P<0.05, P<0.01). Compared with the model group, the high dose of Zuogui Jiangtang Jieyu prescription significantly increased the mRNA expression of Robo3, Sema7a, and Epha7 (P<0.05, P<0.01). Western blot results showed that compared with the results in the normal group, the Sema7a, ITGB1, and FAK protein expression in the hippocampus of the model group was significantly reduced (P<0.01). Compared with the model group, the high dose of Zuogui Jiangtang Jieyu prescription significantly increased the protein expression of Sema7a, ITGB1, and FAK in the hippocampus (P<0.05,P<0.01).ConclusionZuogui Jiangtang Jieyu prescription may treat diabetes with depression by regulating axonal guidance based on the Sema7a/ITGB1 signaling pathway.

摘要:ObjectiveBased on cyclic adenosine monophosphate (cAMP)/cAMP-regulated guanine nucleotide exchange factor 1 (Epac1)/Ras-homologous protein 1 (Rap1) signaling pathway to explore the myocardial protective mechanism of Yiqi Huoxue prescription on coronary heart disease (CHD) rats with Qi deficiency and blood stasis syndrome.MethodEighty-eight specific-pathogen-free (SPF) grade male Sprague-Dawley (SD) rats were divided into a sham operation group (n=12) and an experimental group (n=76) according to the random number table. The experimental group underwent a restricted diet and exhaustive swimming combined with left anterior descending (LAD) coronary artery ligation to construct a model of CHD with Qi deficiency and blood stasis syndrome, and electrocardiograms (ECGs) of rats before and after the LAD operation were collected. After the model was successfully established, the rats were randomly divided into model group, Yiqi Huoxue prescription low-dose group (4.28 g·kg-1), medium-dose group (8.55 g·kg-1), high-dose group (17.1 g·kg-1), and Western medicine group (isosorbide mononitrate tablets, 3.6 mg·kg-1). Rats were intragastrically administered assigned drugs for 4 weeks consecutively, while the sham operation group and the model group were administered with equal volumes of double distilled water. Twenty-four hours after the final administration, the rats were anesthetized with isoflurane to detect the left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD), left ventricular fractional shortening (FS, %), and ejection fraction (EF, %) by echocardiography. Blood samples were collected from the abdominal aorta for hemorheological measurements, and plasma cAMP levels were determined using enzyme-linked immunosorbent assay (ELISA). Myocardial tissue was collected for hematoxylin-eosin (HE) staining and Masson staining to observe myocardial pathological damage, and a transmission electron microscope was used to observe ultrastructural changes of myocardial tissue, fluorescent quantitative real-time polymerase chain reaction (Real-time PCR) was used to detect the expressions of myocardial Epac1, Rap1 GTPase activating protein (Rap1GAP) and Rap1 mRNA, and Western blot was used to detect the expressions of myocardial Epac1, Rap1GAP and Rap1 protein.ResultCompared with those in the sham operation group, LVEDD and LVISD of rats in the model group significantly increased (P<0.01), and the ratios of EF and FS significantly decreased (P<0.01), indicating symptoms of heart function decline, referred to as "heart Qi deficiency". The viscosity of whole blood and plasma significantly increased (P<0.01), and the content of cAMP significant increased (P<0.01). In addition, there was a significant proliferation of collagen fibers in myocardial tissue (P<0.01), and the ultrastructure of the myocardial tissue was severely damaged, indicating pathological changes consistent with "blood stasis". Real-time PCR results showed that Epac1 and Rap1 mRNA levels in the model group were significantly reduced (P<0.01), while Rap1GAP mRNA levels were significantly increased (P<0.01). Western blot analysis showed a significant decrease in Epac1 protein expression (P<0.01) and a significant increase in Rap1GAP protein expression (P<0.05). Compared with the model group, Yiqi Huoxue prescription improved cardiac function, reduced blood viscosity, lowered plasma cAMP levels, decreased collagen fiber proliferation, and improved myocardial ultrastructure damage in CHD rats with Qi deficiency and blood stasis syndrome. The high-dose group showed the most significant effects. In the high-dose group, Epac1 mRNA and protein expression levels significantly increased (P<0.01), Rap1 mRNA expression significantly increased (P<0.01), and Rap1GAP mRNA and Rap1GAP/Rap1 protein expression levels significantly decreased (P<0.05, P<0.01).ConclusionYiqi Huoxue prescription can improve cardiac function, reduce blood viscosity and plasma cAMP levels, improve myocardial damage, and reduce collagen fiber proliferation in CHD rats with Qi deficiency and blood stasis syndrome. The myocardial protection mechanism may be related to the regulation of the cAMP/Epac1/Rap1 signaling pathway.
摘要:ObjectiveTo investigate the therapeutic effect and mechanism of coptisine on chronic atrophic gastritis (CAG) in rats based on the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR) signaling pathway.MethodA CAG rat model was induced by multiple factors, including sodium salicylate, N-methyl-N′-nitro-N-nitrosoguanidine (MNNG), and irregular feeding. The successfully modeled rats were randomly divided into the model group, folic acid group, and high- and low-dose coptisine groups. The high- and low-dose coptisine groups were given coptisine (50, 10 mg·kg-1, respectively), and the folic acid group was given folic acid at 2 mg·kg-1 for 60 days. The pathological changes were detected by hematoxylin-eosin (HE) staining. The ultrastructure of gastric mucosal cells was observed by electron microscopy. Serum pepsinogen Ⅰ (PGⅠ), pepsinogen Ⅱ (PGⅡ), and PGⅠ/PGⅡ ratio (PGR) were detected by immunoturbidimetry. Serum gastrin-17 (G-17) level was detected by radioimmunoassay. The content of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in serum of rats was detected by enzyme-linked immunosorbent assay (ELSIA). Western blot analysis was used to detect the expression levels of TGF-β1, PI3K, phosphorylated-Akt (p-Akt), mTOR, and phosphatase and tensin homolog deleted on chromosome 10 (PTEN) in gastric mucosa. The mRNA levels of TGF-β1, PI3K, Akt, mTOR, PTEN, microtubule-associated protein light chain 3Ⅱ (LC3Ⅱ), and Beclin-1 were detected by real-time quantitative polymerase chain reaction (Real-time PCR).ResultCompared with the normal group, the model group showed atrophy and reduced number of intrinsic glands in the gastric mucosal tissues, as well as inflammatory cell infiltration. The ultrastructure of gastric mucosal cells in the model group displayed nuclear condensation, reduced and swollen mitochondria, and abnormal structure. The serum levels of G-17, PGⅠ, PGR, and the protein and mRNA levels of PTEN in gastric tissues were significantly lower in the model group (P<0.01), while serum levels of IL-6, IL-1β, TNF-α, and the protein and mRNA levels of TGF-β1, PI3K, Akt, and mTOR in gastric tissues were significantly higher (P<0.01). Compared with the model group, various drug intervention groups showed different degrees of improvement in pathological damage and gastric mucosal cell ultrastructure, significantly increased serum levels of G-17, PGⅠ, and PGR (P<0.05,P<0.01), and significantly decreased levels of IL-6, IL-1β, and TNF-α (P<0.05,P<0.01). The high-dose coptisine group significantly downregulated the protein and mRNA levels of TGF-β1, PI3K, Akt, and mTOR (P<0.05,P<0.01).ConclusionBerberine has a therapeutic effect on CAG in rats, possibly exerting a protective effect on gastric mucosa by inhibiting inflammation and blocking the PI3K/Akt/mTOR signaling pathway.

摘要:ObjectiveTo evaluate the clinical efficacy and safety of Tripterygium wilfordii polyglycoside tablets (TWP) in the treatment of rheumatoid arthritis (RA) in the real world.MethodDiagnosis and treatment data of patients with RA in Chinese medicine rheumatology registration research information platform information database (CERTAIN) from January 1,2019 to January, 2024 were collected. According to the inclusion and exclusion criteria, data were screened. The included data were divided into an exposure group and a control group according to the use of TWP or not. Propensity score matching (PSM) was used in both groups to keep the baseline balanced and comparable. The disease activity score (DAS28-ESR) of 28 joints based on the erythrocyte sedimentation rate (ESR)before and after treatment was compared between the two groups, as well as health assessment questionnaire (HAQ),visual analogue scale (VAS),tender joint count (TJC),swollen joint count (SJC), patient's global assessment (PGA),evaluator's global assessment (EGA),laboratory indexes, clinical curative effect, and adverse reactions.ResultA total of 3 978 patients were included,including 929 in the exposure group and 3 049 in the control group. Before PSM,there were significant differences in demographic information,DAS28-ESR score,PGA,EGA,HAQ,VAS scores,SJC, and TJC between the two groups (P<0.05). After successful PSM matching,922 patients in the exposure group and 922 patients in the control group were included. There was no significant difference in demographic information and DAS28-ESR between the two groups before treatment,and the differences in other indexes between the two groups decreased to varying degrees. After treatment,the DAS28-ESR,PGA,EGA,HAQ,SJC,TJC,VAS scores, ESR,and IgG immune index of the two groups were significantly lower (P<0.01). Compared with those in the control group after treatment,the DAS28-ESR,PGA,EGA,HAQ,VAS scores, and ESR in the exposure group after treatment decreased more significantly (P<0.05,P<0.01). There was no significant difference in TJC in the exposure group after treatment. However, TJC in the exposure group was significantly higher than that in the control group before treatment (P<0.05). In terms of TJC reduction,the exposure group performed better than the control group. There was no significant difference in SJC and IgG between the exposure group and the control group after treatment. After treatment,the clinical symptoms of poor appetite,insomnia and many dreams,upset,fatigue,and fear of wind and cold in the two groups were improved. Except that the proportion of women in the exposure group was higher than that in the control group (P<0.01),there was no significant difference in the incidence of other adverse reactions between the two groups after treatment.ConclusionTWP to treat RA can effectively reduce DAS28-ESR,PGA,EGA,HAQ,TJC,and VAS scores and improve the general symptoms. Except for the women at childbearing age with fertility requirements that TWP is not applicable,it shows good security.
摘要:ObjectiveTo observe the clinical characteristics and influencing factors of adverse events of Osteoking and provide a basis for its rational use in clinical practice.MethodA prospective and multicenter Cohort study with large samples was conducted to observe the effects of Osteoking in the treatment of 922 patients with knee osteoarthritis from 20 hospitals from May 1, 2020 to December 31, 2021. Patients who were treated with Osteoking were set as the exposed cohort, and those who were not treated with Osteoking were set as the non-exposed cohort. The gender, age, body mass index (BMI), occupation, allergy history, past medical history, hospital information, medication, and the occurrence of adverse events of the patients were recorded, and the incidence of adverse events was analyzed, as well as its characteristics and factors.ResultA total of 922 patients with knee osteoarthritis were involved, including 274 males (29.72%) and 648 females (70.28%), from which 617 cases were in the exposed cohort, and 305 cases were in the non-exposed cohort. A total of 25 adverse events occurred in both cases, accounting for 2.71% of the total number of cases, with 17 cases in the exposed cohort (2.76%) and eight cases in the non-exposed cohort (2.62%). There was no difference in the incidence rate between the two groups (P=0.907). The age group with the highest incidence of adverse events was between 50 and 59 years old in the exposed cohort (4.61%). The incidence rate in women was 3.49%, slightly higher than 1.07% in men, but there was no difference (P=0.156). According to the systematic classification of adverse events, five cases were respiratory, thoracic, and mediastinal diseases, with an incidence rate of 0.81%. There were two cases of infection and infection diseases, two cases of skin and subcutaneous tissue diseases, two cases of heart-related diseases, two cases of symptoms and signs (not otherwise classified), and two cases of eye organ diseases, and the incidence rate was 0.32%. There was one case of systemic disease, one case of neuropathy, one case of heart organ disease, and one case of vascular hypotension disease, and the incidence rate was 0.16%. During the trial, a total of seven adverse reactions occurred. Among them, there were two cases of dry pharynx, two cases of dizziness, one case of drowsiness, one case of hypotension, and one case of eye discharge, with an incidence rate of 1.13%. Through binary Logistic regression analysis, it was found that among the factors that may affect the occurrence of adverse events in the exposed group, traditional Chinese medicine hospitals were the protective factors for the occurrence of adverse events (OR=0.200, P=0.002), while gender, age, BMI, occupation, allergy history, past medical history, and hospital level cannot be considered to have an impact on the occurrence of adverse events.ConclusionOsteoking can be used to treat knee osteoarthritis of patients of all ages and genders by doctors from hospitals of different levels with higher safety, with occasional and mild adverse events, and seeing a doctor in a traditional Chinese medicine hospital can reduce the occurrence of adverse reactions.
摘要:ObjectiveTo observe the clinical efficacy of Qushi Huayu granules in treating non-alcoholic fatty liver disease (NAFLD) with dampness-heat accumulation.MethodSixty NAFLD patients with the syndrome of dampness-heat accumulation treated in the outpatient and inpatient departments of Yueyang Hospital of Integrated Traditional Chinese and Western Medicine Affiliated to Shanghai University of Traditional Chinese Medicine from July 2018 to May 2020 were selected according to the diagnostic criteria and inclusion criteria of both traditional Chinese medicine (TCM) and Western medicine. The patients were assigned into a control group and an observation group by a random, controlled, double-blind, and double simulated method. The observation group was treated with Qushi Huayu granules combined with the mimetic agent of Dangfei Liganning capsules, and the control group was treated with Dangfei Liganning capsules combined with the mimetic agent of Qushi Huayu granules. The treatment course of both groups was 24 weeks. The TCM symptom scores, liver imaging parameters [controlled attenuation parameter (CAP) in Fibroscan and liver-to-spleen ratio in upper abdominal computerized tomography (CT)], serum levels of alanine aminotransferase (ALT) and γ-glutamyl transpeptidase (γ-GT), and safety indicators of the two groups were measured.Result① The total response rate in terms of TCM symptoms in the observation group was 89.29% (25/28), which was higher than that (48.15%, 13/27) in the control group (Z=-3.582,P<0.01). The total score of the primary and secondary symptoms in both groups of patients declined after treatment (P<0.05), and the observation group outperformed the control group in decreasing the total score of the main and secondary symptoms as well as the scores of right rib swelling pain, abdominal fullness and distension or pain, sticky stool, and yellow urine (P<0.05). ② The response rate in terms of Fibroscan CAP of the liver in the observation group was 75.00% (21/28), which was higher than that (48.15%, 13/27) in the control group (Z=-1.968,P<0.05). ③ The response rate in terms of the serum enzyme levels in the observation group was 75.00% (21/28), which was higher than that (44.44%, 12/27) in the control group (Z=-2.018,P<0.05). The serum levels of ALT and γ-GT in the two groups declined after treatment (P<0.05, P<0.01) and were lower in the observation group than in the control group (P<0.05, P<0.01). ④ The response rate in terms of liver CT in the observation group was 67.86% (19/28), which was higher than that (30.77%,8/26) in the control group (Z=-2.507,P<0.05).ConclusionQushi Huayu granules were safe and effective in the clinical treatment of NAFLD patients with the syndrome of dampness-heat accumulation, which improved the evidence in TCM treatment of NAFLD and was worthy of in-depth clinical research and promotion. Qushi Huayu granules outperformed Dangfei Liganning capsules in terms of TCM symptoms, serum levels of ALT and γ-GT, and liver imaging parameters.
摘要:Objectiveto investigate the differential expression of intestinal flora and serum metabolites and potential biomarkers in patients with pre-diabetic sputum syndrome.MethodA total of 34 patients with pre-diabetic sputum syndrome were included as the phlegm syndrome group,and 37 healthy people were selected as the normal group. Serum and fecal samples of the two groups were collected,and liquid chromatography-mass spectrometry (LC-MS) non-targeted metabolomics and 16S rRNA high-throughput sequencing technology were used to detect serum metabolites and different intestinal flora of the two groups and explore the relationship among pre-diabetic sputum syndrome,serum metabolites,and intestinal flora.ResultIn the distribution of disease syndrome elements in the phlegm syndrome group,the first five disease syndrome elements in terms of frequency and proportion were dampness (73.53%),Qi stagnation (58.82%),Yin deficiency (50.00%),blood stasis (41.18%),and heat (35.29%). According to the frequency and proportion of disease location syndrome elements,the first three main disease location syndrome elements were spleen (100.00%),liver (41.18%),and kidney (23.53%). The results of 16S rRNA high-throughput sequencing showed that there were 44 different intestinal flora between the two groups. In order genus,there were significant differences in Bifidobacterium,Veillonococcus,and Roseococcus between the two groups (P<0.05). The diversity,abundance,and evenness of intestinal flora in the phlegm syndrome group were lower than those in the normal group,with the difference not statistically significant. There was no significant difference in the community structure between two groups. The results of serum metabolomics showed that there were 13 differential metabolites in the two groups,which were mainly concentrated in amino acid metabolism,bile secretion,bile acid biosynthesis,and lipid metabolism (P<0.05). The correlation among differential metabolites,intestinal flora,and syndrome elements was analyzed,and the results showed that ① lysine was positively correlated with spleen,Yin deficiency,and blood stasis,while taurocholic acid was positively correlated with liver,kidney,blood stasis,and dampness,and there was a positive correlation between taurocholic acid and yin deficiency and heat. The taurochenodeoxycholic acid was positively correlated with liver and dampness,and there was a negative correlation between arachidonic acid and dampness,as well as a positive correlation between glucose and spleen and blood stasis. ② Clostridium was positively correlated with spleen,kidney,Yin deficiency,and Qi stagnation. Rosepiella was negatively correlated with spleen,and Sutterella was negatively correlated with dampness. Bacteroides was negatively correlated with the spleen and kidney,and Bifidobacterium was negatively correlated with the spleen and dampness. ③ Bifidobacterium was positively correlated with glycine,threonine,lysine,and deoxycholic acid significantly,negatively correlated with cholic acid significantly,and positively correlated with taurochenodeoxycholic acid and pyruvic acid. Clostridium was positively correlated with glycine significantly and positively correlated with threonine and lysine. Lachnospira was negatively correlated with glycine,threonine,and pyruvic acid. Lysine was also negatively correlated with Faecalibacterium and Eubacterium ventriosum and positively correlated with Megamonas. There was a positive correlation between taurocholic acid and glycine bile acid and Campylobacter,between taurochenodeoxycholic acid and Veillonococcus,and between glucose and Rosepiella and Eubacterium ventriosum. There was a negative correlation between pyruvic acid and Escheria-Shigella and between taurochenodeoxycholic acid and Prevotella.Conclusionthere are differences in intestinal flora and serum metabolites between patients with pre-diabetic sputum syndrome and healthy people. The intestinal flora and metabolites have been disturbed in the stage of pre-diabetes,Bifidobacterium,Clostridium,Lachnospira,glycine,threonine,and lysine may be the breakthrough to explore the development of pre-diabetic sputum syndrome.
摘要:ObjectiveTaking the oligosaccharides in Rehmanniae Radix(RR) as the research object, the content determination method based on high performance liquid chromatography-evaporative light scattering detection(HPLC-ELSD) and thin layer chromatography(TLC) identification method were established to explore the content and distribution of oligosaccharides in different RR herbs and decoction pieces.MethodA total of 10 batches of fresh and raw RR, 12 batches of RR decoction pieces and Rehmanniae Radix Praeparata(RRP) were collected. A TLC identification method for fructose, sucrose, manninotriose, raffinose and stachyose in RR was established by using silica gel G thin-layer plates with ethyl acetate-water-anhydrous formic acid-glacial acetic acid(12∶6∶5∶4) as the developing agent and 10% sulfuric acid-ethanol solution as chromogenic agent. A HPLC-ELSD was used to determine the contents of fructose, glucose, sucrose, melibiose, raffinose, manninotriose and stachyose in different RR herbs and decoction pieces. Then principal component analysis(PCA) and partial least squares-discriminant analysis(PLS-DA) were used to analyze the contents of 7 kinds of saccharides in RR herbs and decoction pieces, and the differential components were screened with the value of variable importance in the projection(VIP)>1.ResultThe results of TLC identification showed that fresh RR, raw RR and its decoction pieces showed spots of the same color on the corresponding positions with the control products of stachyose, raffinose and sucrose, while the TLC of RRP showed spots of the same color at corresponding positions to manninotriose and fructose controls. The results of methodological investigations of 7 analytes met the requirements of determination. Only glucose, sucrose, raffinose and stachyose were detected in 10 batches of fresh RR and 10 batches of raw RR herbs, the average contents of which were 0.84%, 4.62%, 2.42% and 57.90% in fresh samples, while those were 3.16%, 9.36%, 7.05% and 38.10% in raw samples, respectively. In 12 batches of RR decoction pieces, the contents of the above seven sugars(fructose, glucose, sucrose, melibiose, raffinose, manninotriose and stachyose) were 1.68%, 4.27%, 9.96%, 0.53%, 6.85%, 3.05% and 37.52%, respectively. In 12 batches of RRP, the contents of the above seven sugars were 10.62%, 11.01%, 1.25%, 3.35%, 1.12%, 28.16% and 6.39%, respectively. The results of multivariate statistical analysis showed that fresh RR, raw RR and RRP could be distinguished from each other by the contents of the 7 sugars, and the main differential components were stachyose, sucrose, raffinose and manninotriose.ConclusionIn terms of oligosaccharides, the contents and types of saccharides in different herbs and decoction pieces of RR are quite different, and the TLC identification method based on this can be used to distinguish raw RR from RRP, which can lay a foundation for improving the quality standard of RR and developing and applying oligosaccharides in different processed products of RR.

摘要:ObjectiveTo establish the fingerprint of seven commercially available Yangyin Qingfei preparations, to quantitatively analyze the index components, to evaluate their quality uniformity with multivariate statistical analysis, and to explore the quality differences among different dosage forms.MethodA total of 33 batches of commercially available 7 kinds of Yangyin Qingfei preparations were analyzed by high performance liquid chromatography(HPLC), the fingerprints were established and the common peaks were identified. Paeoniflorin, verbascoside, harpagoside, glycyrrhizic acid and paeonol were selected as the indicators of quality attributes to quantitatively analyze 33 batches of preparations. Based on the administration methods of Yangyin Qingfei preparations, the daily intake was calculated and the radar charts were poltted, and cluster analysis and principal component analysis were used to explore the quality differences of 7 kinds of Yangyin Qingfei preparations and the quality uniformity among different batches of the same dosage form.ResultThe similarity of fingerprints of 7 dosage forms was 0.248-0.956, suggesting that there were significant differences among different dosage forms of Yangyin Qingfei preparations, and a total of 15 common peaks were calibrated, of which peak 7, peak 8, peak 11, peak 13 and peak 15 were paeoniflorin, verbascoside, harpagoside, glycyrrhizic acid and paeonol, respectively. The radar plots showed that the average total daily intake of large honeyed pills and water honeyed pills was the highest, and the uniformity of pill components was better. The quality of 33 batches of samples was divided into poor quality and high quality by cluster analysis. Principal component analysis showed that the uniformity and dosage form of different dosage forms were significantly different, the oral liquid had the best quality homogeneity with the minimum dispersion. And the content of paeonol in different dosage forms was significantly different, which was the key component of quality control of Yangyin Qingfei preparations.ConclusionYangyin Qingfei large honeyed pills and water honeyed pills show high content and good uniformity, which are relatively preferred dosage forms. Different preparation processes have a great influence on the content of paeonol, and its quality control should be emphasized during production. This study provides a scientific method for the comparison of product quality of different dosage forms of traditional Chinese medicine prescriptions, which is helpful for the development of preferred dosage forms of different prescriptions, and provides a reference for efficient use of medication in the clinical practice.
摘要:ObjectiveTo explore the genetic relationship and reveal the genetic variations of 45 germplasm accessions of Artemisia argyi.MethodGenotyping-by-sequencing (GBS) was employed to mine single nucleotide-polymorphisms (SNPs) from the 45 germplasm accessions. Principal component analysis, phylogenetic analysis, population genetic structure analysis, and genetic variation analysis were conducted based on the SNPs.ResultA total of 111.91 Gb of data were obtained, with the Q20, Q30, and average GC content of 96.39%, 90.33%, and 39.37%, respectively. The comparison rate between clean reads and the reference genome was 70.24%-98.97%. A total of 22 399 Indels and 170 539 SNPs were obtained, and the 10th pair of chromosomes had the most variation sites. The results of principal component analysis, cluster analysis, and genetic diversity analysis classified the 45 germplasm accessions into three groups. Group Ⅰ contained three germplasm accessions from Qichun County. The germplasm accessions in group Ⅱ were all wild. Group Ⅲ contained 31 germplasm accessions, with the most complex sources. Moreover, the 45 germplasm accessions can be classified into 3 subtypes, containing the genetic information from three ancestors. The results indicated rich genetic diversity of A. argyi from different sources, especially the germplasm accessions from Qichun County, Hubei province.ConclusionThis study provides theoretical support for breeding new varieties, developing specific SNP markers, and revealing the genetic relationship of A. argyi.
摘要:ObjectiveThe potential suitable area for ecological planting, key ecological factors, and suitable range of Polygonatum odoratum in China were analyzed to provide theoretical and scientific guidance for the artificial planting of P. odoratum.MethodA total of 454 geographical distribution records of P. odoratum in China and 118 ecological factors were used in this study. The maximum entropy model (MaxEnt) was adopted to predict the suitable areas of P. odoratum. The key ecological factors and their suitable ranges were analyzed by the jackknife method, contribution rates of ecological factors, and response curves.ResultThe suitable areas of P. odoratum were mainly located in the northwest, north, and northeast of China, the highly suitable areas of which were concentrated in Shaanxi, Shanxi, Gansu, etc. Solar radiation in November (Srad11), precipitation in July (Prec7), percentage of evergreen/deciduous needleleaf trees (Class1), silt content (2-50 μm) mass fraction (SLTPPT), and annual average temperature (Bio1) were found to be the key ecological factors affecting the suitable distribution of P. odoratum in China. The cumulative contribution rate of solar radiation factors (31.29%)>vegetation factors (25.61%)>soil factors (19.52%)>precipitation factors (11.38%)>temperature factors (8.57%)>topography factors (3.63%).ConclusionIt is suggested to carry out ecological planting of P. odoratum mainly in Shaanxi (such as Baoji and Ankang Cities and Ningshan, Liuba, and Hua Counties), Gansu (such as Tianshui City, Gannan Tibetan Autonomous Prefecture, and Liangdang and Huating Counties), and Shanxi (such as Yangquan, Taiyuan, Fenyang, and Jinzhong Cities, as well as Xingxian County) of China. Solar radiation factors should be given priority in the planting process, followed by vegetation, soil, precipitation, temperature, and topography factors. The range of key ecological factors, namely Srad11, Prec7, Class1, SLTPPT, and Bio1 should be controlled within 8 095.21-10 334.98 (optimum 8 787.50) kJ·m-2·d-1, 109.99-223.60 (146.91) mm, 1.00%-9.45% (6.76%-10.68%), 41.73%-50.35% (46.53%), and 3.29-16.33 (13.57) °C, respectively.
摘要:This article systematically analyzes the historical evolution of the name, origin, scientific name evolution, medicinal parts, producting area, harvesting and processing, clinical efficacy of Paridis Rhizoma by consulting the ancient materia medica, medical books and prescription books, combined with modern literature, in order to provide a reference for the development of famous classical formulas containing Paridis Rhizoma. According to research, it can be verified that Paridis Rhizoma was first recorded in Shennong Bencaojing with the name of Zaoxiu, and this name has been used for ancient materia medica in the past generations. The name of Chonglou was first appeared in Xinxiu Bencao of the Tang dynasty, and has gradually become the rectification of name, with it being the correct name in modern legal names at all levels. The mainstream source of Paridis Rhizoma used in ancient times was the dried rhizomes of Paris polyphylla var. chinensis, but due to the similarity plant morphology of the genus Paris, there was much confusion, and the same genus of plants with thick rhizomes were also used as Paridis Rhizoma, such as P. polyphylla, P. polyphylla var. stenophylla. Since modern times, P. polyphylla var. yunnanensis has also been included as one of the mainstream sources of Paridis Rhizoma. The origin recorded in ancient materia medica are mainly in Shandong, Anhui, Jiangsu, Jiangxi, Yunnan and other regions, which are collected in the wild for medicinal purposes, modern cultivation is more prevalent in southwestern regions, such as Yunnan, Sichuan, Guizhou and Guangxi. In ancient times, the harvest time was mostly root harvesting in April and May of the lunar calendar, while in modern times, they are mostly harvested throughout the year or in autumn. Autumn harvesting is the best choice, with roots removed from the production area, washed, and dried in the sun. In ancient times, Paridis Rhizoma was processed by baking, grinding and other methods, but in modern times, sliced raw products were mainly used as medicine. Paridis Rhizoma has a slightly cold nature, a bitter taste, and a small toxicity. It can regulate the liver meridian, clear heat and detoxify, reduce swelling and pain, cool the liver and calm convulsions. It is used for symptoms such as boils and carbuncles, throat swelling and pain, snake and insect bites, and its flavor and efficacy are basically consistent in ancient and modern records. Based on the results of the textual research, it is recommended that the dried rhizomes of P. polyphylla var. chinensis or P. polyphylla var. yunnanensis should be used in the development of famous classical formulas containing Paridis Rhizoma, the processing method is selected according to the requirements of the formula, and the raw product is recommended to be used as medicine if not specified.

摘要:Wuhutang is the 51st Chinese medicine prescription of Han ethnic group on in the catalogue of ancient classical prescriptions (the second batch) ,which originated from Renzhai' Straight Diections Prescriptions (《仁斋直指方》) by YANG Shiying in the Song dynasty,it is composed of 5 medicines, including Ephedrae Herba,Armeniacae Semen Amarum, Glycyrrhizae Radix et Rhizoma, tea and Gypsum Fibrosum. It treats shortness of breath and phlegm. Following the principle of textual research on the key information of ancient classical famous prescriptions, the author collected and recorded the ancient books of traditional Chinese medicine of Wuhutang with the method of Bibliometrics, and screened out 53 valid data, involving 41 ancient books. Based on the historical evolution, drug composition, main treatment of disease and syndrome, drug dosage, method of preparation and usage, drug origin and processing, the author conducted a dig and a systematic study of Wuhutang. The results show that:in the later generations, besides inheriting the original prescription, the composition of medicine was added or subtracted, and the frequency of addition of Mori Cortex was the most;in the treatment of disease and syndrome, most of them inherited the original prescription mainly treating asthma syndrome, also see in the treatment of asthma, cough and other lung disease, in the drug dosage and method of use , it is suggested to inherit the original prescription, the dosage of each medicine is: Ephedrae Herba 2.89 g, Armeniacae Semen Amarum 4.13 g, Glycyrrhizae Radix et Rhizoma 1.65 g, tea 3.30 g, Gypsum Fibrosum 6.20 g, decoction, take warm clothes before eating;on the source of medcine,ephedra is made from the dried herbaceous stems of Ephedra sinica (Ephedraceae),bitter almond is made from the dried and mature seeds of Prunus armeniaca (Rosaceae),Licorice is made from the dried roots and rhizomes of Glycyrrhiza uralensis(Legumes), tea is made from the buds leaves of Camellia sinensis(Camellia),plaster is made from Gypsum Fibrosum of sulfate mineral gypsum family. At the same time, we collected and sorted the modern clinical application literature of Wuhutang, and obtained 73 effective literatures, all of which are for the treatment of respiratory system diseases, especially for children's pneumonia and asthma, and mostly belong to"phlegm-heat obstructing the lung syndrome". Through the analysis and study of ancient books and modern applied literature, the historical evolution and prescription evolution of Wuhutang are clarified, and its key information is determined, in order to provide more accurate reference for the research and development of the classic Wuhutang and the rational addition and subtraction of the application.

摘要:ObjectiveThis study conducted a "6 + 1" clinical comprehensive evaluation of the existing research on Zhichuanling oral liquid (ZOL) in the treatment of asthma,so as to clarify the clinical advantages and precise clinical positioning of ZOL in the treatment of asthma, lay a foundation for further research and academic promotion of ZOL, and provide new directions for patent cultivation.MethodAn evaluation method featuring a qualitative and quantitative combination was used, which considered the dimensions of safety,effectiveness,economy,innovation,suitability,accessibility, and traditional Chinese medicine (TCM) characteristics. According to Expert Meeting Law, relevant weights were obtained through voting. CSC_v2.0 software was used to calculate each dimension and convert it into the corresponding grade score.ResultBased on the existing materials,① ZOL instruction indicates the adverse reactions,taboo, and notes. Multiple data of clinical research before and after marketing and spontaneous reporting system shows that ZOL has controllable risk and good safety. Safety is rated as B grade. ② Multiple data of clinical research before marketing, systematic evaluation of clinical effectiveness, and Meta-analysis shows that ZOL has good effectiveness and clinical significance. Effectiveness is rated as a B grade. ③ Analysis of the cost-effectiveness of ZOL combined with conventional treatment shows that the economy of the drug is good and rated as a B grade. ④ ZOL has better innovation, which is rated as an A grade. ⑤ ZOL can basically meet the clinical drug needs based on the result of the questionnaire survey and has good suitability, which is rated as a B grade. ⑥ ZOL has better accessibility, and accessibility is rated as A grade. ⑦ ZOL involves a rich theory of TCM but insufficient experience of human usage. It is thus rated as a C grade in terms of TCM characteristics. Based on the results of "6 + 1" dimension,the clinical comprehensive evaluation of ZOL in the treatment of asthma (cold syndrome and heat syndrome)is rated as B category.ConclusionZOL has good clinical value and outstanding innovation and accessibility in the treatment of asthma (cold syndrome and heat syndrome). It is recommended that ZOL be transformed into the relevant policy results of basic clinical drug management procedurally. At the same time,it is recommended to actively cultivate patents with TCM characteristics.
摘要:"State-target medicine" is a traditional Chinese medicine (TCM) diagnosis and treatment theoretical system proposed by Academician Tong Xiaolin based on the current development of modern medicine. The active stage of gastric ulcer, as a precancerous state of gastric cancer, has a great impact on people's health. Prof. ZHOU Xuewen, a master of TCM, innovatively put forward the theory of "toxicity-heat" etiology for the active stage of gastric ulcer, which plays an important guiding role in clinical diagnosis and treatment. The article took the theoretical system of "state-target medicine" as the framework to explain the rationale, method, formula, and medicine of Prof. ZHOU Xuewen, who applied the Xiaoyong Kuidekang based on the "toxicity-heat" theory to treat the gastric ulcer in the active stage. The Chinese medical name of gastric ulcer, "gastric carbuncle", was established, and it was believed that gastric ulcer is born due to "toxicity" and is based on "toxicity and heat". In the course of the disease, "toxicity", "heat", "deficiency", and "stasis" coexisted, and its pathogenesis was divided into three phases, namely, toxicity-heat accumulation phase, toxicity-heat affecting the health phase, and weakened body resistance and strengthened toxicity phase. According to the positioning of gastric ulcer as an "internal carbuncle", Prof. ZHOU Xuewen proposed the treatment of gastric ulcer in the active stage with "carbuncle theory" and introduced the surgical methods of "elimination", "support", and "tonifying" into the treatment of gastric ulcer in the active stage. Prof. ZHOU Xuewen took "clearing heat and removing toxins, eliminating carbuncle and generating muscle" as the basic treatment of the disease. For different stages of the disease, Prof. ZHOU Xuewen emphasized the use of the methods of clearing heat and removing toxins, supporting rot and muscle growth, and strengthening the spleen and harmonizing the stomach and created the representative formula for the treatment of gastric ulcer in the active stage with "carbuncle theory", namely "Xiaoyong Kuidekang", which could regulate state and targets.

摘要:Cassiae Semen is a substance with both edible and medicinal uses approved by the National Health Administration, and it is widely used in health food products in China. In this study, an information database of Cassiae Semen-based health food products was constructed, and the common patterns of functional claims, major raw materials, and dosage forms were analyzed. The Apriori algorithm and network method were used to analyze the association rules of high-frequency ingredients with the three major functional claims: Maintaining blood lipid (cholesterol/triglyceride) level, controlling fat, and relaxing bowel. Secondly, based on the positioning and regulatory requirements of health food and the recent research progress in Cassiae Semen, this paper analyzed the problems in technology review and product development from safety, function claim, and quality controllability. Finally, suggestions were put forward to promote the inheritance and carry forward China's traditional healthcare culture and the achievement of the Healthy China Initiative. These suggestions included classifying and optimizing the access management of health food, innovating the traditional healthcare function claims, and strengthening the quality system construction and information disclosure of post-marketing evaluation.

摘要:Baihe Dihuangtang is a famous classical formula that has been respected by physicians in the past and is still used today. It was first recorded in ZHANG Zhongjing's Synopsis of the Golden Chamber, and is composed of Lilii Bulbus and Rehmanniae Radix juice. This paper systematically reviewed the research progress of historical evolution, pharmacological activities and clinical applications of Baihe Dihuangtang in recent years, and found that there was no major changes in the composition, decoction method and indications of this formula since the Han dynasty. According to the herbal textual research, the fleshy scaly leaves of Lilium brownii var. viridulum should be selected for Lilii Bulbus in this formula, and the tuberous roots of Rehmannia glutinosa were selected for Rehmanniae Radix. According to the dosage conversion of ancient and modern times, the dosage is 245 g of fresh Lilii Bulbus and 400 g of fresh Rehmanniae Radix, and the ratio of their juice is 1∶1. The quality marker(Q-Marker) of Baihe Dihuangtang were predicted according to the principles of ingredient specificity, component validity, component measurability, formula compatibility, and quality transmissibility and traceability, and it was determined that catalpol, rhmannioside D, regaloside A, regaloside B, regaloside C, and acteoside could be selected as potential Q-Markers of Baihe Dihuangtang, which could provide scientific reference for the establishment of the quality control system and the development of compound preparation of this famous classical formula.

摘要:Stroke, including ischemic stroke and hemorrhagic stroke, has the characteristics of high morbidity, high disability rate, high mortality rate, high recurrence rate, and high economic burden. Ischemic stroke is the most common type of stroke, which is mainly caused by intracranial artery occlusion. The clinical manifestations of patients are hemiplegia, aphasia, sensory disturbance, and other neurological deficits, accompanied by gastrointestinal symptoms such as constipation and gastrointestinal bleeding. Intestinal flora plays an important role in the pathogenesis of ischemic stroke, and its potential biological effects have received extensive attention. Intestinal flora can not only affect intestinal barrier function but also regulate gastrointestinal immunity and affect host homeostasis. In recent years, traditional Chinese medicine (TCM) has shown remarkable effects and small adverse reactions in the prevention and treatment of cerebral ischemia. The research on the effect of TCM in improving cerebral ischemia injury by regulating the structure and metabolism of intestinal flora and maintaining the function of intestinal flora has gradually become a hot topic. Based on the interaction between TCM and intestinal flora in relevant literature in recent years, this review investigated the mechanism of anti-cerebral ischemic injury of TCM via regulating intestinal flora structure, affecting intestinal flora metabolism, and regulating body immunity and made a summary, proving a basis for further elucidating the role of intestinal flora in cerebral ischemia and the mechanism of TCM in prevention and treatment of ischemic stroke.

摘要:Stroke is a neurological disease characterized by cerebral ischemia or hemorrhagic injury, leading to death and disability worldwide. At present, there is still a lack of neuroprotective drugs for the treatment of stroke. Niuhuang Qingxin pills are Chinese patent medicine recommended by guidelines and expert consensus for preventing and treating stroke. Studies have shown that Niuhuang Qingxin pills have sedative, anticonvulsant, antipyretic, and other pharmacological effects. Moreover, the main components, such as flavonoids, phenolic acids, and saponins, also exhibit strong pharmacological activities which can improve stroke-induced nerve damage. Studies have confirmed that representative ingredients such as baicalin and ginsenosides can interfere with multiple pathological events, including blood-brain barrier destruction, oxidative stress, inflammatory response, apoptosis, and autophagy, and the mechanisms of the essential ingredients are related to the action on important targets such as Kelch-like ECH-associated protein 1 (Keap1), matrix metalloproteinase-9 (MMP-9), and high-mobility group box 1 (HMGB1), and involvement in the regulation of Toll-like receptor 4 (TLR4)/nuclear factor-κB (NF-κB), nuclear factor E2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1), and phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling pathways. This article summarized the research status of Niuhuang Qingxin pills and the experimental pharmacological progress of common ingredients in the prevention and treatment of stroke to provide clues for further research into Niuhuang Qingxin pills and the development of active ingredients of traditional Chinese medicines.
摘要:Bronchial asthma (asthma for short) is a common clinical respiratory disease mainly characterized by chronic airway inflammation, with complicated pathogenesis and a long treatment cycle. It is lingering and difficult to be cured, and lack specific drugs. Oxidative stress is a new focus in the research on the pathogenesis of asthma and a potential key target for the treatment. Under physiological conditions, the oxidative and antioxidative systems in the body are in a dynamic balance, and the two antagonize each other to maintain normal life activities. In the case of asthma attack, oxidation products such as reactive oxygen species (ROS), malondialdehyde (MDA), and nitric oxide (NO) are produced excessively, while the content of antioxidants such as superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) is reduced. As a result, the oxidation exceeds the removal of oxidation products, which aggravates oxidative stress. In addition, the overproduction of ROS activates oxidative stress-related signaling pathways to produce pro-inflammatory factors, exacerbating inflammation, which leads to lung and airway tissue damage. In recent years, traditional Chinese medicine has garnering increasing attention because of the unique advantages in the treatment of asthma, especially in regulating redox balance, alleviating oxidative stress in asthma patients, and reducing inflammation. On the one hand, by inhibiting the mitogen-activated protein kinase (MAPK) and transcription factor (NF)-κB signaling pathways, traditional Chinese medicine can reduce the content of oxidation products and pro-inflammatory factors from the source. On the other hand, by activating the nuclear factor-erythroid 2-related factor 2 (NrF2) signaling pathway, traditional Chinese medicine can elevate the levels of antioxidant enzymes and enhance the antioxidant system to neutralize the excessive accumulation of oxidation products. Therefore, the adjustment of redox balance state by traditional Chinese medicine may be a new means and a new direction for the prevention and treatment of asthma in the future. This paper summarizes the oxidative stress-related pathways in the pathogenesis of asthma and reviews the latest research progress in the regulation of oxidative stress-related pathways by Chinese medicine extracts and prescriptions in the treatment of asthma, with a view to providing a fuller, more solid, and more scientific theoretical basis for the clinical and basic research on the prevention and treatment of asthma by traditional Chinese medicine.

摘要:Lung cancer is the leading cause of cancer-related death, characterized by high invasiveness, early metastasis and poor prognosis. It has the highest incidence and mortality rates among cancers in China. Currently, the treatment of lung cancer is still dominated by the classic therapies such as surgery, radiotherapy, chemotherapy and targeted therapy. However, the classic therapies are not suitable for all patients with lung cancer, so new treatment strategies and methods are needed to prevent and treat lung cancer. In recent years, Ginseng Radix et Rhizoma has attracted wide attention in the field of anti-lung cancer research, and traditional Chinese medicine has a substantial theoretical and practical foundation in treating lung cancer. Ginseng Radix et Rhizoma , as a commonly used Chinese herbal medicine, has the functions of replenishing vital Qi, replenishing veins, strengthening the spleen and lung, and nourishing body and blood. The main active components of Ginseng Radix et Rhizoma , known as ginsenosides, exhibit anti-cancer and anti-inflammatory activities. Therefore, the mechanism and pharmacological activity of ginsenosides in the intervention of lung cancer have been extensively studied by researchers worldwide. The results show that ginsenosides can effectively inhibit the proliferation, invasion, migration, epithelial-mesenchymal transition and angiogenesis of lung cancer cells. Additionally, they inhibit drug resistance, enhance chemotherapy sensitivity and efficacy, and promote apoptosis and autophagy of lung cancer cells. Ginsenosides also modulate the tumor microenvironment and regulate immunity, thereby delaying the occurrence and development of lung cancer. The rapid advancements in related research have outpaced previous literature review, creating challenges for scholars seeking the latest information. Based on this, this article summarizes recent findings on the mechanism and pharmacological activities of ginsenosides in lung cancer intervention, aiming to provide new insights for the development of molecular biology, drug treatment research and clinical new drug research in lung cancer. It also provides the reference for further mechanism research.

摘要:Nonalcoholic fatty liver disease (NAFLD) is one of the most common complications of type 2 diabetes mellitus (T2DM). Cell death caused by iron-lipid disorder is a new mode of regulating programmed cell death, which is characterized by lipid peroxidation induced by the accumulation of lipid reactive oxygen species and excessive accumulation of iron ions induced by iron metabolism disorders. Among them, iron homeostasis disorder and metabolic pathway disorder are the main causes of iron-lipid disorder, which play an important role in a variety of pathological processes related to cell death. Because the liver is an important organ for iron storage and lipid metabolism, iron-lipid disorder is an ideal target for liver disease, and inhibition of iron-lipid disorder may become a new strategy for the treatment of NAFLD. However, the pathogenic relationship and mechanism between NAFLD and iron-lipid disorder have not been fully elucidated. Based on the complex molecular regulation mechanism of iron-lipid disorder, by expounding the role of iron-lipid disorder in NAFLD and its related mechanism, this paper summarizes the research status of traditional Chinese medicine on the target treatment of NAFLD in recent years, so as to provide a new perspective and point out a new direction for the treatment of NAFLD in the future.

摘要:Ovarian aging is a reproductive endocrine disease caused by a variety of factors leading to a gradual decline in ovarian function until ovarian failure, which seriously affects women's physical and reproductive health and is a major factor leading to female infertility. Mitochondria, the energy metabolism centers of cells, are critical for ovarian functions. Their structural and functional abnormalities are key pathological factors leading to the declined ovarian function. Mitochondrial quality control is an important endogenous regulatory mechanism for the maintenance of mitochondrial homeostasis and the improvement of mitochondrial functions. Abundant studies have shown that the dysregulation of mitochondrial quality control, characterized by mitochondrial oxidative damage, abnormal mitochondrial biogenesis, abnormal mitochondrial dynamics, abnormal mitochondrial autophagy, and dysregulated calcium homeostasis, is closely associated with the occurrence of ovarian hypofunction. Traditional Chinese medicine (TCM) is a treasure of China's medicine, demonstrating remarkable efficacy in the clinical treatment of ovarian aging-related diseases. In recent years, research progress has been achieved in the TCM treatment of ovarian aging by regulating mitochondrial quality control disorders in a multi-target and multi-pathway manner. However, systematic research remains to be carried out regarding the research progress in this field. Therefore, this article reviews the research progress in the TCM treatment of ovarian aging based on mitochondrial quality control, with a view to providing a theoretical basis for studying the clinical efficacy of TCM in the treatment of ovarian aging and a new strategy for the in-depth research on the prevention and treatment of ovarian aging by TCM.
