摘要:ObjectiveTo observe the mechanism of chronic psychological stress aggravating idiopathic pulmonary fibrosis (IPF) in rats and the regulatory effect of Xiaoyaosan.MethodSixty SD rats were acclimatized for one week and then randomly divided into five groups: sham operation group, IPF group, IPF and depression model group, pirfenidone group, and pirfenidone + Xiaoyaosan group, with 12 rats in each group. The IPF group was induced by intratracheal instillation of bleomycin (5 mg·kg-1) and administered 0.9% sodium chloride solution intragastrically. The model group was induced with bleomycin (5 mg·kg-1) combined with chronic unpredictable mild stress to establish a rat model of IPF and depression and administered 0.9% sodium chloride solution intragastrically. Concurrently, the pirfenidone group was administered pirfenidone aqueous solution (50 mg·kg-1) intragastrically, and the pirfenidone + Xiaoyaosan group was administered pirfenidone aqueous solution (50 mg·kg-1) and Xiaoyaosan decoction (19.27 g·kg-1) intragastrically. The experiment lasted for four weeks. Various parameters, including body weight, food intake, sucrose consumption rate, open field behavior, lung function, lung coefficient, pathological changes in lung tissue, and hydroxyproline (HYP) content were compared among the groups. Enzyme-linked immunosorbent assay (ELISA) was used to measure serum cortisol (CORT), corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH) levels, and serotonin (5-HT) levels in serum and hippocampus. The levels of interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) in serum and lung tissue were also measured. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was employed to determine the mRNA expression of IL-1β, TNF-α, IL-6, p38 mitogen-activated protein kinase (p38 MAPK), and nuclear transcription factor-κB p65 (NF-κB p65) in lung tissue. Western blot was used to detect the expression of proteins related to the p38 MAPK/NF-κB signaling pathway.ResultCompared with the normal group, IPF and depression model group showed slow weight gain, reduced food intake, decreased sucrose consumption rate, reduced total distance and average speed of movement in the open field test, weakened lung function, increased lung coefficient (P<0.01), significant inflammatory cell infiltration in lung tissue by hematoxylin-eosin (HE) staining, collagen fiber deposition by Masson staining, and increased HYP content (P<0.01). There were elevated levels of serum CORT, CRH, and ACTH (P<0.05, P<0.01), increased protein and mRNA levels of IL-1β, IL-6, and TNF-α in serum and lung tissue, decreased 5-HT levels in serum and hippocampus, and increased relative expression of p-p38 MAPK and p-NF-κB p65 proteins in lung tissue (P<0.01). Compared with the IPF and depression model group, the depression and IPF of rats in the pirfenidone + Xiaoyaosan group were effectively ameliorated, as evidenced by faster weight gain, increased food intake and sucrose consumption rate, increased total distance and average speed of movement in the open field test, enhanced lung function, reduced lung coefficient (P<0.01), decreased inflammatory cell infiltration by HE staining, reduced collagen fiber deposition by Masson staining in lung tissue, and decreased HYP content (P<0.01). Decreased serum CORT, CRH, and ACTH levels (P<0.05, P<0.01), decreased protein and mRNA levels of IL-1β, IL-6, and TNF-α in serum and lung tissue (P<0.05, P<0.01), increased 5-HT levels in serum and hippocampus (P<0.05, P<0.01), and decreased relative expression of p-p38 MAPK and p-NF-κB p65 proteins in lung tissue were also noted (P<0.05, P<0.01), with the effects of Xiaoyaosan + pirfenidone being more significant.ConclusionChronic unpredictable stress exacerbates the progression of IPF in rats. The combination of Xiaoyaosan and pirfenidone not only improves depressive-like behavior but also alleviates pulmonary fibrosis, potentially through the regulation of the p38 MAPK/NF-κB signaling pathway and inhibition of excessive expression of inflammatory factors.
摘要:ObjectiveTo explore the mechanism of Taohong Siwutang in regulating the levels of inflammatory mediators, cysteine-dependent aspartate-specific protease Caspase-14, and recombinant envoplakin (EVPL) in the mouse model of psoriasis.MethodForty-eight BALB/c mice were randomized into blank, model, methotrexate tablets(3.6×10-4 g·kg-1), and low-, medium-, and high-dose Taohong Siwutang groups(7.14,14.28,28.56 g·kg-1). The mouse model of psoriasis-like skin lesion was induced by applying 5% imiquimod cream on the back of mice, and drug intervention was carried out at the same time. The psoriasis area and severity index (PASI) was scored by photographing. Hematoxylin-eosin staining was employed to observe the pathological changes of the skin, which was evaluated based on the Baker score. The levels of interleukin (IL)-2, IL-17, IL-22, and IL-23 in the peripheral blood of mice were measured by the enzyme-linked immunosorbent assay. Immunohistochemical staining was adopted to detect the positive expression of Caspase-14 and EVPL in mouse skin lesions. Western blot and real-time polymerase chain reaction were employed to determine the protein and mRNA levels, respectively, of IL-2, IL-17, IL-22, IL-23, Caspase-14, and EVPL in mouse skin lesions.ResultCompared with the blank group, the model group showed increased PASI score and Baker score, elevated levels of IL-2, IL-17, IL-22, and IL-23 in the peripheral blood, up-regulated mRNA and protein levels of IL-2, IL-17, IL-22 and IL-23, and down-regulated mRNA and protein levels and positive expression of Caspase-14 and EVPL in skin lesions (P<0.05). Compared with the model group, drug interventions reduced the PASI score and Baker score, lowered the levels of IL-2, IL-17, IL-22, and IL-23 in the peripheral blood, down-regulated the mRNA and protein levels of IL-2, IL-17, IL-22, and IL-23, and up-regulated the mRNA and protein levels and positive expression of Caspase-14 and EVPL in skin lesions (P<0.05).ConclusionTaohong Siwutang demonstrates good anti-inflammatory effect and skin barrier repair function in the treatment of psoriasis. It can significantly reduce the levels of inflammatory mediators in the peripheral blood and skin lesions and promote the expression of Caspase-14 and EVPL involved in skin barrier repair in the mouse model of psoriasis.

摘要:ObjectiveTo observe the effect of Coptidis Rhizoma and Ophiopogonis Radix on renal tissue injury and epidermal growth factor receptor (EGFR)/phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway in rats with diabetic nephropathy (DN) and explore its possible mechanism of delaying DN.MethodThirty-six male Wistar rats were randomly divided into a normal group (6 rats) and a model group (30 rats). The model group was fed with a high-fat and high-sugar diet combined with streptozotocin (STZ) to establish a rat model of type 2 diabetes. After the successful preparation of the model, the rats were randomly divided into the model group, low, medium, and high dose groups of Coptidis Rhizoma and Ophiopogonis Radix (100, 200, 400 mg·kg-1), and metformin group (200 mg·kg-1). After administration, the levels of fasting blood glucose (FBG), 24 h urine protein (24 h-UTP), creatinine (SCr), urea nitrogen (BUN), and uric acid (UA) were detected. Hematoxylin-eosin (HE) staining and Masson staining were used to observe the pathological changes of renal tissue in rats. Western blot and Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) were used to detect the related protein expression of EGFR, PI3K, and Akt and their mRNA expression levels in the renal tissue of rats in each group.ResultsCompared with the normal group, the levels of FBG, SCr, BUN, UA, 24 h-UTP, and kidney index in the model group were significantly increased (P<0.01), most renal tubular epithelial cells were necrotic, and the content of collagen in glomeruli was significantly increased (P<0.01). Compared with the model group, the above indexes of rats in each administration group were improved to varying degrees. The FBG, SCr, BUN, UA, 24 h-UTP, and kidney index of rats in each dose group and metformin group were significantly decreased (P<0.01, P<0.05). The necrosis degree of renal tubular epithelial cells was reduced, and the fibrosis area was decreased (P<0.01). There related protein and mRNA expressions of EGFR, PI3K, and Akt were significantly increased (P<0.05, P<0.01).ConclusionCoptidis Rhizoma and Ophiopogonis Radix can alleviate renal tissue injury in rats with DN, and their mechanism may be related to the regulation of the EGFR/PI3K/Akt signaling pathway.
摘要:ObjectiveTo observe the effects of Coptidis Rhizoma-Fermentum Rubrum on non-alcoholic fatty liver disease (NAFLD) in mice and explore its possible mechanisms.MethodSixty male SPF C57BL/6J mice were randomly divided into six groups: control group, model group, low-, medium-, and high-dose Coptidis Rhizoma-Fermentum Rubrum group (0.75, 1.5, 3 g·kg-1), and metformin group (0.075 g·kg-1), with 10 mice in each group. NAFLD mouse models were induced by high-fat diet feeding for 24 weeks. The low, medium, and high-dose Coptidis Rhizoma-Fermentum Rubrum groups were administered corresponding doses of Coptidis Rhizoma-Fermentum Rubrum by gavage, while the control and model groups received an equivalent amount of saline for four weeks. Serum total cholesterol (TC), triglycerides (TG), free fatty acids (FFA), and liver function markers including alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured using an automatic biochemical analyzer. Hematoxylin-eosin (HE) and oil red O staining were used to detect liver lipid deposition, and Prussian blue staining was used to measure liver ferrous ion levels. Western blot was performed to detect the expression of key proteins in the nuclear factor erythroid-2-related factor 2 (Nrf2)-glutathione peroxidase 4 (GPX4) axis.ResultAfter 24 weeks of high-fat feeding, compared with the control group, the model group showed significant increases in body weight, liver weight and liver index, and serum lipid levels (P<0.01), as well as substantial hepatic lipid deposition with marked steatosis. Compared with the model group, Coptidis Rhizoma-Fermentum Rubrum intervention reduced body weight (P<0.01), liver weight and liver index (P<0.01), and serum lipid levels (P<0.05, P<0.01), improved liver function (P<0.01), and decreased hepatic lipid deposition, with the low-dose Coptidis Rhizoma-Fermentum Rubrum group showing the best effect. Western blot results showed that compared with those in the control group, the expression levels of Nrf2, heme oxygenase-1 (HO-1), kelch-like ECH-associated protein 1(Keap1), and GPX4 proteins in the model group were decreased (P<0.05, P<0.01). Compared with the model group, Coptidis Rhizoma-Fermentum Rubrum increased the expression levels of these proteins (P<0.05, P<0.01).ConclusionCoptidis Rhizoma-Fermentum Rubrum can alleviate fatty liver in mice, improve liver function, and reduce hepatic lipid deposition, possibly by regulating the Nrf2/GPX4 ferroptosis axis.
摘要:ObjectiveTo investigate the mechanism by which Liuwei Dihuang Erzhiwan combination inhibit the lung metastasis of spontaneous breast cancer in mice by regulating the recruitment of myeloid-derived suppressor cells (MDSCs).MethodThree hundred and eighty SPF-grade 10-month-old female breeders of Kunming mouse were palpated at the mammary gland site once every 3 days. Mice that have not had a lump touched after being raised for 6 months are used as control group. After tumor development, the mice were randomized into model, positive control (paclitaxel, intraperitoneal injection at 0.01 g·kg-1 every other day for 22 d), Liuwei Dihuangwan (0.65 g·kg-1·d-1 by gavage), Erzhiwan (5.41 g·kg-1·d-1 by gavage), and Liuwei Dihuang Erzhiwan combination (6.05 g·kg-1·d-1 by gavage) groups. The mice were euthanised when the tumor reached a diameter of about 15 mm, and the tumor and lung tissues were collected. The survival time, tumor mass, and lung metastasis rate of tumor-bearing mice were recorded. Hematoxylin-eosin (HE) staining was used to observe the histopathological and morphological changes of mouse tumor and lung tissues. Immunofluorescence (IF) was used to detect the distribution of MDSCs in tissues of mice in each group by double-staining of MDSCs cells with lymphocyte antigen 6 complex site G6D (Ly6G) and CD11 antigen-like family member B (CD11b). Western blot was employed to determine the protein levels of matrix metalloproteinase-9 (MMP-9), transforming growth factor-β (TGF-β), zinc finger transcription factor 1 (Snail1), and E-cadherin in the tumor tissue and CC motif chemokine 9 (CCL9) and CC motif chemokine receptor 1 (CCR1) in the lung tissue.ResultDuring the modelling period, the paclitaxel group and Chinese medicine intervention groups had longer median number of days of survival and lower tumor weight, lung metastasis rate, and lung nodule than the model group (P<0.05, P<0.01). HE staining showed an increase in tumor cell necrosis in the paclitaxel group and the Liuwei Dihuang Erzhiwan combination group. The paclitaxel group and Chinese medicine intervention groups had lower fluorescence intensity of MDSCs in the tumor tissue than the model group (P<0.05, P<0.01). Compared with the normal control group, the model group showed increased fluorescence intensity of MDSCs in the metastatic lung tissue (P<0.01), which, however, was decreased in the paclitaxel group and Chinese medicine intervention groups (P<0.01). The model group showed higher protein levels of MMP-9, TGF-β, and Snail1 and lower protein level of E-cadherin in the tumor tissue than in the normal control group (P<0.01). Compared with model group, paclitaxel and Chinese medicine interventions downregulated the protein levels of MMP-9, TGF-β, and Snail1 (P<0.05, P<0.01) and upregulated the protein level of E-cadherin in the tumor tissue (P<0.01). Moreover, the Liuwei Dihuang Erzhiwan combination group had lower protein levels of TGF-β and Snail1 than the Liuwei Dihuangwan group and Erzhiwan group (P<0.05). In the metastatic lung tissue, the expression of CCL9 and CCR1 was higher in the model group than in the normal control group, paclitaxel group, and Chinese medicine intervention groups (P<0.05, P<0.01).ConclusionLiuwei Dihuang Erzhiwan combination inhibit tumor growth, prolong survival time, and reduce the occurrence of lung metastasis in the mouse model of spontaneous breast cancer by reducing the recruitment of MDSCs in the tumor and lung tissues and modulating the phenotypes of epithelial-mesenchymal transition (EMT)-related molecules and the expression of CCL9/CCR1.
摘要:ObjectiveTo study the effect and mechanism of Curculiginis Rhizoma in ameliorating kidney-Yang deficiency in castrated rats.MethodThe targets of Curculiginis Rhizoma and male reproductive diseases due to kidney-Yang deficiency were screened from relevant databases by network pharmacology, and key targets were screened out according to topological eigenvalues. After that, gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed to obtain the pathways of Curculiginis Rhizoma in treating kidney-Yang deficiency. The rat model of kidney-Yang deficiency was established by castration. The rats were assigned into model, testosterone propinate (2 mg·kg-1), and low-, medium-, high-dose (0.14, 0.28, 0.56 g·kg-1, respectively) Curculiginis Rhizoma groups. Another 8 healthy male rats with first incising and then suturing were used as the sham group. The rats were administrated with corresponding agents by gavage once a day for 6 consecutive weeks. During the experiment, the general conditions of rats in each group were observed, and their body mass was recorded. At the end of the experiment, the indexes of accessory sex organs were calculated, and the pathological changes of the seminal vesicle glands and prostate glands were observed by hematoxylin-eosin (HE) staining. The serum levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone (T), and interleukin-6 (IL-6) were determined by enzyme-linked immunosorbent assay. The results of the network pharmacological prediction were verified by animal experiments, and the expression levels of related genes and proteins were determined by real-time polymerase chain reaction and Western blot, respectively.ResultThe biological functions enriched mainly involved four overexpression modules including regulation of cell responses to various stimuli, metabolic responses, regulation of intracellular signal transduction, and regulation of reproduction. Phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway was a significantly enriched pathway for the core target and also a pathway with the highest enrichment factor in the biological process of regulating cell responses to stimuli. The results of animal experiments showed that compared with the model group, low-dose Curculiginis Rhizoma group increased the body weight gain (P<0.05). In addition, high-dose Curculiginis Rhizoma group increased the seminal vesicle gland index and epididymis index (P<0.05). The administration with Curculiginis Rhizoma ameliorate epithelial cell hyperplasia of the seminal vesicle glands, did not attenuate the vacuolar degeneration, mitigated the enlarged vesicular lumen of the prostate gland, changing of epithelial cells of the glands from flattened to cubic and columnar shapes, and cellular disarrangement, and reduced the mesenchymal stroma thickness. Compared with the model group, all the intervention measures elevated the levels of FSH, LH, T, and IL-6 (P<0.05, P<0.01) and up-regulated the mRNA and protein levels of PI3K and the mRNA levels of Akt in the epididymis tissue (P<0.05).ConclusionCurculiginis Rhizoma can alleviate the T level reduction and accessory sex organ atrophy in castrated rats by regulating the PI3K/Akt signaling pathway.

The protein expression levels of ATF4 and CHOP were increased in the combined group (P<0.05), while the expression level of Bcl-2 protein gradually decreased (P<0.05).ConclusionGuiqi Yiyuan ointment combined with cisplatin can exert anti-tumor effects in mice with Lewis lung cancer, reduce tumor mass, increase tumor inhibition rate, and induce apoptosis of lung cancer cells. Its mechanism may be related to the regulation of the endoplasmic reticulum stress pathway and mitochondrial apoptosis pathway.
摘要:ObjectiveTo examine the protective mechanism of wogonin in SH-SY5Y cells cultured in the conditioned media with lipopolysaccharide (LPS)-induced BV-2 microglia.MethodBV-2 microglia were divided into the blank group, LPS group, low concentration group of wogonin (4 μmol∙L-1), medium concentration group of wogonin (8 μmol∙L-1), and high concentration group of wogonin (16 μmol∙L-1). The LPS group was given 1 mg·L-1 LPS, and the other three groups were treated with the corresponding concentration of wogonin for 4 h and then given 1 mg·L-1 LPS. The conditioned media from these groups were used to cultivate SH-SY5Y cells. Cell counting kit-8 (CCK-8) was used to assess the vitality of BV-2 cells in the above groups. Compared with the blank group, NF-κB p65 protein gradually accumulated into the nucleus, and the fluorescence expression intensity was significantly enhanced (P<0.01). Compared with the LPS group, the NF-κB p65 protein was gradually dispersed outside the nucleus, and the fluorescence expression intensity was gradually weakened in all concentration groups of wogonin. The fluorescence intensity in the high concentration group of wogonin was significantly reduced (P<0.01). Compared with those in the blank group, the expression levels of TLR4 protein, phosphorylated(p)-NF-κB p65 protein, and MyD88 protein in the LPS group were significantly increased (P<0.05, P<0.01). Compared with those in the LPS group, the expressions of TLR4 protein, p-NF-κB p65 protein, and MyD88 protein in the medium concentration group of wogonin were all significantly decreased (P<0.05, P<0.01). The expressions of TLR4 protein, and MyD88 protein in the high concentration group of wogonin were significantly decreased (P<0.05, P<0.01).ConclusionWogonin may regulate the TLR4/MyD88/NF-κB signaling pathway to inhibit the release of LPS-induced inflammatory factors in BV-2 microglia and protect SH-SY5Y cells, thereby reducing inflammation and achieving neuroprotective effects.

摘要:ObjectiveTo explore the protective effect and mechanism of Yangxin Dingji capsules on isoproterenol (ISO)-induced ventricular arrhythmia in SD rat cardiomyocytes based on the transforming growth factor-β activated kinase 1 (TAK1)-mitogen-activated protein kinase kinase 3 (MKK3)-p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway.MethodFifty male SPF-grade SD rats were randomly divided into a normal group, a model group, a propranolol group, a low-dose Chinese medicine group, and a high-dose Chinese medicine group. The ventricular arrhythmia model was constructed using the ISO "6+1" method. The propranolol group received propranolol at 0.015 g·kg-1·d-1. The Chinese medicine groups received Yangxin Dingji capsules at doses of 0.5、 2 g·kg-1·d-1, respectively. The normal and model groups were given an equal volume of 0.9% NaCl solution. Electrocardiogram (ECG) changes in SD rats were recorded using the BL-420F biological function experimental system. Hematoxylin-eosin (HE) staining was used to observe the pathological changes in the heart. Serum levels of cardiac troponin I (cTnI), creatine kinase-MB (CK-MB), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and transforming growth factor-β1 (TGF-β1) were measured using enzyme-linked immunosorbent assay (ELISA). The mRNA expression of IL-1β and TNF-α was assessed using real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Reactive oxygen species (ROS) expression was detected using immunofluorescence. Protein expression levels of TAK1, phosphorylated TAK1 (p-TAK1), MKK3, phosphorylated MKK3 (p-MKK3), p38 MAPK, phosphorylated p38 MAPK (p-p38 MAPK), nuclear factor-κB (NF-κB), phosphorylated NF-κB (p-NF-κB), IL-1β, and TNF-α were measured using Western blot or immunohistochemistry.ResultCompared with normal group, the model group showed significant ventricular arrhythmia in ECG, with an increased arrhythmia score (P<0.01). Pathological damage to myocardial tissue was evident, and serum levels of cTnI, CK-MB, IL-1β, TNF-α, and TGF-β1 were elevated (P<0.01). The mRNA and protein expression of IL-1β and TNF-α in myocardial tissue was also increased (P<0.01). ROS level and protein expression of p-TAK1, p-MKK3, p-p38 MAPK, and p-NF-κB were elevated in myocardial tissue (P<0.01). In the propranolol and Chinese medicine groups, the incidence of sustained ventricular tachycardia (SVT) and arrhythmia scores were significantly reduced compared to model group (P<0.05, P<0.01). Pathological damage to cardiomyocytes was alleviated, and levels of myocardial injury markers and inflammatory factors in serum and myocardial tissue were decreased. The ROS level in myocardial tissue was also reduced (P<0.01), with a noticeable reduction in related molecules in the p38 MAPK pathway (P<0.05, P<0.01).ConclusionThe expression of p38 MAPK pathway molecules was up-regulated in myocardial tissue of ISO-induced ventricular arrhythmia rats. Yangxin Dingji capsules may inhibit cardiac inflammation damage by regulating the expression of related molecules in the p38 MAPK pathway, thereby exerting a protective effect on myocardial cells, with TAK1 being a potential target.

摘要:This study analyzed the outcome index and related design elements of randomized controlled trials (RCTs) of traditional Chinese medicine (TCM) in the treatment of children with tic disorder (TD) in the past ten years, so as to provide a basis for the construction of the core index set of TCM in the treatment of children with TD. Eight databases were searched, including four English databases (PubMed, Web of Science, Embase, and Cochrane Library) and four Chinese databases, including China National Knowledge Infrastructure (CNKI), Wanfang Database, VIP Database, and China Biology Medicine disc (CBMdisc), as well as ClinicalTrials.gov and China Clinical Trial Registry. The search time was limited to from January 1, 2013 to October 29, 2023. RCTs on the TD in children treated with TCM were collected. Two researchers independently conducted literature screening, data extraction, and literature quality evaluation and summarized clinical outcome indexes and related trial design elements through qualitative analysis. A total of 67 RCTs were included, including 63 outcome indexes, with a total frequency of 348 times. The related outcome indexes could be divided into six categories: 12 symptom/sign indexes with a frequency of 134 (38.5%), seven TCM symptom/syndrome indexes with a frequency of 31 (8.9%), 33 physical and chemical examination indexes with a frequency of 97 (27.9%), four safety indexes with a frequency of 67 (19.3%), three long-term prognostic indexes with a frequency of 14 (4.0%), and one kind of quality-of-life evaluation index (0.3%). Currently, the RCTs research design of TCM in the treatment of TD in children has not yet formed a unified standard, and there are many problems in the quality of methodology, which reduces the authenticity and reliability of clinical conclusions. There are problems with clinical outcome indexes, such as significant quantity differences, unclear primary and secondary outcome indexes, unreasonable alternative indexes, non-standard TCM syndrome types and TCM evaluation indexes, lack of economic evaluation indexes, and less attention to long-term prognostic indexes and safety indexes. It is suggested that the researchers should design a more rigorous trial scheme and reasonably design the outcome index which is in line with the clinical trial efficacy evaluation of TCM, so as to construct the core index set with the characteristics of TCM for the treatment TD in children.

摘要:ObjectiveTo reveal the influence of Jianchangbang characteristic processing method on the change process of volatile components and the processing mechanism of reducing toxicity and increasing efficiency of Magnoliae Officinalis Cortex(MOC) by studying the changes in the composition and content of volatile components during the processing of ginger processed Xingpo pieces.MethodSamples of raw products, ginger juice moisturized products and stir-fried and heap moisturized products of MOC were taken according to the set time points, and headspace gas chromatography-mass spectrometry(HS-GC-MS) was used to determine the contents of volatile components in the samples, and the relative content of each component was obtained by peak area normalization. Principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA) were performed on the sample data using SIMCA 14.1 software, and the differential components during the processing were screened with variable importance in the projection(VIP) value>1 as the indicator.ResultA total of 68 volatile components were identified in the samples, among which some of the chemical components with similar structures showed similar trends of changes, and there was also the phenomenon of interconversion between compounds. Compared with the raw products, the contents of 42 components in ginger juice moisturized products increased, while the contents of 25 components decreased, 19 components were unique, and 4 components were unique to the raw products. Compared with ginger juice moisturized products, MOC in the early stage of piling had three unique components, and the contents of 11 components such as cyclosativene and (+)-α-pinene increased, and the contents of 5 components such as tricyclic terpene and α-curcumene decreased, and ginger juice moisturized products had four unique components. Compared with the early stage of piling, in the later stage, the contents of 8 components such as (+)-α-pinene and camphene significantly increased, while the contents of 6 components such as linalool and α-selinene significantly decreased. During the processing of MOC, there were significant changes in the chemical composition of the samples before and after 20 days. The differences between ginger juice moistening and the early stage of piling, the early stage and the later stage of piling could be clearly distinguished.ConclusionDuring the preparation process of ginger processed Xingpo pieces, the addition of ginger juice can reduce the contents of stimulating components, and the contents of active components continue to increase in several stages, such as the addition of ginger juice, frying and heap moisturizing, the quality of the decoction pieces may change significantly at about 20 d of processing. This study can provide a research basis for exploring the processing mechanism of ginger processed Xingpo pieces.

摘要:ObjectiveTaking Shengxuebao as the model traditional Chinese medicine(TCM), this study aims to explore the methodologies and transfer patterns in the composition changes of TCM compound preparations from liquid to solid dosage forms.MethodBased on the previously established ultra-high performance liquid chromatography(UPLC) qualitative and quantitative methods for Shengxuebao, the fingerprint profiles of three preparations of Shengxuebao(homemade decoction, commercially available granules and mixtures) were established, and nineteen components were quantitatively analyzed. The similarities of fingerprint profiles among these three preparations were assessed using the Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012 edition) and the total quantum statistical moment similarity method, respectively. The molecular connectivity index (MCI) and the average molar MCI of the chemical components in the three preparations were calculated. Seventy-two MCI values across 0-8 orders, including 8 subclasses, were calculated. The average molar MCI of 0-3 orders was used to study the "imprint template" structural characteristics and transfer patterns of the medicinal component groups.ResultThe similarities of the fingerprint profile of the decoction with those of the control, granules, and mixtures were 0.971, 0.888 and 0.799, respectively, indicating that the chemical composition of the granules and mixtures differed from that of the decoction, and that the 19 components were able to basically respond to the overall situation of the component group of Shengxuebao decoction. The total mass fractions of the detectable major components in Shengxuebao decoction, granules and mixtures was measured to be 5.498 6, 2.282 0, 1.416 6 mg·g-1, respectively. The average molar MCI of 0-3 orders showed a gradual decrease across the three preparations, with the trend of decoction>granules>mixtures, indicating subtle changes in the overall structural characteristics of the chemical components in the samples of the three preparations.ConclusionAmong the three formulations of Shengxuebao, the traditional decoction contained the highest variety and quantity of components and carried the most substantial amount of component information, with a decreasing order observed in the granules and mixtures. This study further validates that traditional decoction is a reliable and fundamental dosage form, providing a reference for the modernization of TCM dosage forms and the quality consistency evaluation of compound preparations.

摘要:ObjectiveTo investigate the correlation between the color, odor and volatile components of Paeoniae Radix Alba(PRA) and its processed products, and to examine the effects of different processing methods on the odor and color formation of PRA.MethodThe odor and chromaticity information of PRA, honey chaff-fried PRA and honey bran-fried PRA were identified by electronic nose and colorimeter, and the volatile components in the different processed products of PRA were identified and relatively quantified by headspace gas chromatography-mass spectrometry(HS-GC-MS), and analyzed using principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA), then the differential key flavor components among the three were screened according to the principles of variable importance in the projection(VIP) value>1 and relative odor activity value(ROAV)≥1. Pearson correlation analysis was used to investigate the association between the differential flavor components and the colorimetric values and electronic nose sensors, respectively.ResultAfter being fried with honey chaff and honey bran, the lightness value(L*) of PRA decreased, and red-green value(a*) and yellow-blue value(b*) increased significantly(P<0.05, P<0.01). The odor differences were mainly reflected in the S1, S2, S4, S5, S6, S8 and S11 sensors, and the results of PCA of the electronic nose indicated that the odor differences among PRA, honey chaff-fried PRA and honey bran-fried PRA were obvious, among which the overall odor intensity of honey bran-fried PRA was higher than that of honey chaff-fried PRA. A total of 47 volatile components were identified from PRA and its processed products, including 21 for PRA, 36 for honey chaff-fried PRA, and 37 for honey bran-fried PRA. Odor analysis revealed that 12, 24 and 22 volatile components may be the key flavor components in PRA, honey chaff-fried PRA and honey bran-fried PRA, respectively. Correlation analysis showed that the L* of the decoction pieces was negatively correlated with the content of the Maillard reaction products, the a* and b* were positively correlated with the content of the Maillard reaction products, the S1 and S8 sensors were negatively correlated with the content of the Maillard reaction products, and the S2, S4, S5, S6 and S11 sensors were positively correlated with the Maillard reaction products.ConclusionThe color of PRA is deepened after being stir-fried with honey chaff and honey bran, and 5-hydroxymethylfurfural, furfural and other components are generated at the same time, which is in line with the theory of burning aroma strengthens the spleen of stir-fried with honey chaff and honey bran. Honey bran-fried PRA has a stronger Maillard reaction than honey chaff-fried PRA, which makes honey bran-fried PRA with a burnt flavor and a dark yellow color, while honey chaff-fried PRA has a sweet flavor and a bright yellow color.

摘要:ObjectiveHost mitochondrial DNA (mtDNA) and chloroplast DNA (cpDNA) contamination severely affects high-throughput sequencing of endophytic bacteria in plants. This study aims to explore and evaluate a novel strategy of inhibiting host gene amplification in high-throughput sequencing of endophytic bacteria in medicinal plants.MethodGreen Shield Sequencing (GSS) was introduced in the 16S rDNA polymerase chain reaction (PCR) of endophytic bacteria to shield the non-target amplification of genes in the host (Ligusticum chuanxiong). The performance was compared between GSS-PCR and conventional PCR in the high-throughput sequencing of endophytic bacteria and rhizosphere soil bacteria.ResultCompared with conventional PCR, GSS-PCR significantly reduced the amplification of mtDNA and cpDNA in L. chuanxiong in high-throughput sequencing, decreasing the non-target genes by more than 60%. Moreover, this strategy significantly increased the diversity of endophytic bacteria and multiplied the species without compromising the extraction of the information about the dominant bacteria. The GSS amplification of 16S rDNA V4 region of L. chuanxiong showed lower host contamination rate and higher endophytic bacterial diversity than that of V3-V4 regions.ConclusionGSS can significantly reduce host gene contamination in the high-throughput sequencing of L. chuanxiong endophytic bacteria and improve the accuracy of endophytic bacterial diversity analysis at the same sequencing depth, thus improving the high-throughput analysis quality of endophytic bacteria in plants. Accordingly, this strategy improves the feasibility and reliability of high-throughput sequencing for the 16S rDNA V3-V4 and V4 regions of endophytic bacteria. GSS used in this study provides a method reference for studying the endophytic bacteria in other medicinal plants.
摘要:Zuoguiyin, which first recorded in Jingyue Quanshu written by ZHANG Jiebin in the Ming dynasty, was included in the Catalogue of Ancient Famous Classical Formulas(The Second Batch). This study followed the Principles of Textual Research on Key Information of Ancient Famous Classical Formulas to determine the key information of Zuoguiyin in ancient and modern literature, such as the formula origin, the composition of the formula and the origin of the drugs. It was found that the composition, dosage, preparation and processing methods of Zuoguiyin were basically the same as the original formula. The original dosage of this formula is 41.03 g of Rehmanniae Radix Praeparata(the fresh or dried tuberous roots of Rehmannia glutinosa, processed by wine stewing or wine steaming), 7.46 g of Dioscoreae Rhizoma(the dried rhizomes of Dioscorea opposita), 7.46 g of Lycii Fructus(the dried mature fruit of Lycium barbarum), 3.73 g of Glycyrrhizae Radix et Rhizoma Praeparata cum Melle(the dried roots and rhizomes of Glycyrrhiza uralensis, processed by honey-roasted method), 5.595 g of Poria(the sclerotium of Poria cocos), 5.595 g of Corni Fructus(the dried mature fruit pulp of Cornus officinalis). The method of administration is to add 600 mL of water to all the herbs, decoct to 140 mL and take before meals. The function of Zuoguiyin is to nourish Yin and tonify the kidney, and it is often used in the treatment of lumbar soreness and ejaculation, night sweating, dry mouth and throat, thirst and desire to drink, glossy red tongue, thin and rapid pulse, etc. Since ancient times, Zuoguiyin has been used to treat a variety of internal and gynaecological diseases as well as diseases of the nervous, circulatory and reproductive systems that are predominantly caused by kidney Yin deficiency. However, there is not much research on the modern application and therapeutic mechanism of this formula, and there is no standardized preparation in the market, so the degree of development and utilization is not high, and there is still a lot of room for research.

摘要:Basic research and interdisciplinary application of traditional Chinese medicine (TCM) have been continuously extended and rapidly developed with the continuous funding and promotion of basic research on TCM supported by the National Natural Science Foundation of China (NSFC), and the overall level of basic research on TCM has been significantly improved, which has made important contributions to the diagnosis and treatment mechanism of TCM by "clarifying, identifying, and interpreting", providing benchmark guarantees for basic research and industry development of TCM. This article focused on the application and funded projects of NSFC in 2023 under the H31 application code of TCM. It analyzed the project types, branch codes, research question categories, supporting units and regions, research directions, and keywords and summarized the characteristics of the currently funded projects in H31. At the same time, it analyzed the relevant problems in the project application process and put forward several suggestions, so as to provide a reference for scholars in the field of TCM. In 2023, a total of 8 334 applications for three types of projects (general projects, projects for young scholars, and regional projects) under the H31 code have been received by NSFC, which is the highest number in the past five years. However, the proportion of funding is still on a downward trend, and there is fierce competition among applicants. Finally, 804 projects for the three categories under the H31 code have been funded. The total funding decreased in 2022, and the average funding amount of the general project and the regional project has dropped to the lowest in the past five years. It is found that the development of branch research and the research questions under the H31 code is unbalanced. Few projects have focused on the original theory of TCM. Some projects are detached from the characteristics and research laws of TCM. Most researchers lack innovative research ideas, and the applications they write are filled with routines and patterns. In addition, the issue of scientific integrity remains a primary principle for project applicants.

摘要:Chronic obstructive pulmonary disease (COPD) is susceptible to systemic complications. Especially, sarcopenia is an independent risk factor for COPD patients that can exacerbate respiratory muscle fatigue, lead to a higher risk of falls and fractures, and lower the quality of life. The pathogenesis of sarcopenia involves such aspects as mitochondrial dysfunction, insulin resistance, changes in skeletal muscle fiber types, and an imbalance in protein synthesis and breakdown. As nutrition support and exercise rehabilitation therapy frequently have limited effectiveness,it is urgent to find a way to slow the progression of COPD with sarcopenia. Numerous studies conducted in recent years have discovered a potential link between pulmonary microbiome disorders and the gut microbiota of COPD patients. Furthermore,a wide range of functions of gut microbiota and its metabolites have been demonstrated, such as regulating inflammation and immunity, glucose and lipid metabolism, and mitochondrial function. According to the Zangxiang theory in Chinese medicine, there are intimate connections between the spleen, intestine, and muscle, and gut microbiota may be the most essential part of ameliorating "spleen governing muscle" and increasing muscle mass and strength. This study aims to expound on the effect mechanism of gut microbiota in slowing the progression of COPD with sarcopenia, thereby furnishing novel perspectives and recommendations for forthcoming investigations and therapeutic applications.
摘要:The prevalence of dilated cardiomyopathy (DCM) has been increasing year by year, which seriously endangers the physical and mental health of patients. Establishing an animal model that conforms to the clinical characteristics of the disease is a prerequisite for conducting experimental research on DCM. This study sorted out and summarized the commonly used DCM animal models and evaluated the degree of consistency between the models and clinical practice based on the etiology and pathogenesis of traditional Chinese and western medicine and clinical diagnostic criteria. The models with high consistency with western medicine include immune models, drug toxicity models, and tachycardia models, while the models with high consistency with traditional Chinese medicine include drug toxicity models, gene correlation models, and tachycardia models. With consideration of the characteristics of the symptoms, consistency, and their respective advantages and disadvantages, the more ideal models are drug toxicity models, tachycardia models, and gene correlation models. In addition to evaluating the clinical consistency of traditional Chinese and western medicine in DCM animal models, this study also analyzed the advantages and disadvantages of existing DCM animal models, existing problems, and observation indicators of the models and pointed out the lack of standardized and unified syndrome model differentiation standards. Due to the complexity of the disease and the unclear pathogenesis, there are few studies on DCM models, and there is room for improvement. On this basis, four improvement directions are proposed to make the animal models as close to the clinical pathological and physiological processes as possible,so that the combination of disease and syndrome in animal models is more practical. These animal models can provide an experimental basis for the pathogenesis, drug intervention, efficacy evaluation, and scientific research of DCM and better serve scientific research and clinical practice.

摘要:Cardiometabolic disease is a clinical syndrome with a causal relationship between metabolic abnormalities and cardiovascular damage. With its global incidence and related mortality rates continually rising, it has become a major health concern worldwide. The role of the gut microbiome and its metabolic products in cardiovascular metabolic health has received widespread attention, with gut microbiota dysbiosis considered a key factor in promoting the development of cardiometabolic disease. Dysbiosis disrupts the balance of the "heart-spleen-intestine" axis, leading to dysfunction of the spleen and intestines, which triggers metabolic disorders and accelerates the progression of cardiometabolic disease. Cardiac dysfunction can also negatively affect spleen and intestinal function, leading to imbalances in the heart and spleen, disharmony in Qi and blood, and exacerbating metabolic anomalies and further dysbiosis, thus forming a vicious cycle. From a modern biological perspective, the gut microbiome and its metabolic products can influence disease progression by modulating inflammatory responses and immune imbalances, leading to endothelial dysfunction and metabolic disorders, thereby increasing the risk of cardiometabolic disease. Additionally, the article proposes strategies for managing cardiometabolic disease by regulating the gut microbiome through a combination of Chinese and western medicine approaches. Traditional Chinese medicine(TCM) treatment starts from the gut microbiome, using the "heart-spleen-intestine" axis as a mediator to regulate cardiovascular metabolic health, highlighting the unique advantages of TCM in targeting the gut microbiome to treat cardiometabolic disease. This article takes the TCM theory of the "heart-spleen-intestine" axis as a starting point, discusses the pivotal role played by this axis in the connection between gut microbiome dysbiosis and the development of cardiometabolic disease, aiming to provide a new perspective for the integrated traditional Chinese and western medical research on cardiometabolic disease, offering scientific evidence and practical guidance to improve the prognosis and quality of life for patients.

摘要:This study, anchored in the traditional Chinese medicine (TCM) syndrome differentiation and treatment principles alongside the clinical characteristics of sepsis in Western medicine, extensively gathers and meticulously dissects the latest research findings on sepsis animal models from both Chinese and international sources. Adhering strictly to TCM syndrome diagnostic criteria for sepsis, the study conducts a thorough evaluation of various animal models across multiple dimensions, including clinical manifestations, pathological changes, and biomarker expressions, so as to reflect the degree of resemblance these models have to human sepsis TCM syndromes. The results reveal that the colon stent implantation model exhibits a higher degree of congruence with both TCM and Western medicine standards, particularly aligning with the diagnosis of the "Fu-Qi obstruction syndrome". Conversely, the extraperitoneal sepsis model shows a higher degree of congruence with TCM, fitting more closely with the diagnosis of "acute deficiency syndrome" and emphasizing the core pathogenesis of Qi deficiency in sepsis. These findings not only augment the diversity of sepsis animal models but also highlight the necessity and potential of integrated TCM and western medicine research. Current sepsis animal models predominantly focus on western pathophysiological mechanisms, with limited direct incorporation of TCM syndrome differentiation elements. This underscores the need, in future study designs, to actively explore integrating TCM syndrome classification and intervention principles into model development. This could be achieved by manipulating model-inducing factors and observing more TCM-specific symptoms and signs among other strategies, so as to establish sepsis models that more closely resemble clinical reality and incorporate both TCM and western medical perspectives.

摘要:Polycystic ovary syndrome(PCOS) is a common reproductive endocrine and metabolic disorder, affecting about 6%-10% of women of childbearing age worldwide, and has always been a major challenge in clinical treatment. Guizhi Fulingwan, derived from the Synopsis of the Golden Chamber(《金匮要略》), has the effect of promoting blood circulation, removing blood stasis, and alleviating blockages. In recent years, many studies have found that it has a good therapeutic effect on PCOS. By sorting out the research literature on the mechanism of Guizhi Fulingwan treating PCOS, it is found that this formula can decrease the autophagy and apoptosis of ovarian granulosa cells, correct endocrine hormone disorder, improve the inflammatory environment, alleviate oxidative stress. Moreover, it regulates phosphatidylinositide 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR), lncRNA H19/miR-29b-3p, nuclear factor E2-related factor 2(Nrf2)/heme oxygenase-1(HO-1), PI3K/Akt/nuclear factor-κB(NF-κB), and other signaling pathways. And also effects the regulator expression of matrix metallopeptidase-9(MMP-9), matrix metalloproteinase inhibitor-1(TIMP-1), cytochrome P45019A1 enzyme(CYP19A1), glucose transporter protein 4(GLUT4), regulated on activation normal T cell expressed and secreted(RANTES), monocyte chemoattractant protein-1(MCP-1). Thus, Guizhi Fulingwan is confirmed to treat PCOS based on these molecular mechanisms. Besides, the main active ingredients of the formula such as cinnamic aldehyde, (+)-catechin, stigmasterol, also have the effect of anti-oxidant, anti-inflammatory, regulating mitochondrial function, regulating endocrine, and regulating metabolism. According to the summary of clinical literature on Guizhi Fulingwan, it is found that the use of the formula alone or in combination with other drugs can significantly alleviate the clinical symptoms and complications of PCOS. Thus, it is safe with no adverse reactions in long-term use, which is worthily further promoted in clinical application. This paper comprehensively analyzed the current research status of Guizhi Fulingwan for the treatment of PCOS through the mechanism and clinical studies, summarized the limitations in current research, and put forward suggestions, aiming to provide the strong literature support for the future in-depth research on and clinical application of the formula.

摘要:Chronic constipation (CC) is one of the common and frequent functional gastrointestinal diseases in clinic. At present, the overall therapeutic effect of CC is still not satisfactory in China and abroad, which seriously affects the quality of life of patients. The pathophysiological mechanism of constipation has not been fully elucidated, and studies have mainly focused on the dysfunction of the enteric nervous system (ENS), abnormalities of Cajal interstitial cells (ICCs), alterations in mucosal immune function (low-grade intestinal inflammation, intestinal barrier damage, and immune activation), abnormalities of gastrointestinal hormones, dysregulation of intestinal flora, and dysfunctions of intestinal dynamics. Traditional Chinese medicine (TCM) is a unique and precious resource in China with good efficacy and safety. TCM and its active ingredients can effectively treat constipation, and its mechanism of action needs to be further investigated. Signaling pathways are involved in the development of constipation, which is one of the main targets in the study of pathological mechanisms of constipation and the pharmacological study of therapeutic drugs. Therefore, the authors searched PubMed, China National Knowledge Infrastructure (CNKI), and other literature databases with the keywords of ''constipation'', ''signaling pathway'', and ''traditional Chinese medicine'' to systematically summarize the signaling pathways related to constipation regulated by TCM. The comprehensive analysis shows that TCM mainly plays a role in regulating autophagy, apoptosis, and proliferation and ensuring anti-oxidative stress, anti-inflammation, and protection of intestinal barrier of ICCs by modulating signaling pathways such as stem cell factor/tyrosine kinase receptor (SCF/c-kit), phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt), 5-hydroxytryptophan (5-HT), mitogen-activated protein kinases (MAPK), and Toll-like receptors (TLRs). TCM regulates neurotransmitter secretion and intestinal water metabolism to improve constipation. This demonstrates the advantages of multi-level, multi-target, and multi-level effects of TCM, and in-depth research can provide new ideas for exploring the pathogenesis of constipation and developing new drugs.

摘要:Diabetic nephropathy (DN) is a chronic microvascular complication in diabetic patients and the main cause of end-stage renal disease (ESRD). Studies have shown that nuclear factor kappa-B (NF-κB) signaling pathway is involved in the pathological process of DN by activating pathological mechanisms such as inflammation, oxidative stress, fibrosis, apoptosis, autophagy, and pyroptosis. Therefore, blocking the transduction of NF-κB signaling pathway can help prevent and treat DN. Currently, western medical treatment involves strategies such as lowering blood sugar, blood pressure, and lipids, as well as using endothelin receptor antagonists, mineralocorticoid receptor antagonists, aldosterone synthase inhibitors, and other drugs, but they still cannot block the pathological process of DN. Traditional Chinese medicine (TCM) offers a simpler and more cost-effective approach that addresses both the symptoms and underlying causes of DN. Recent research has shown promising results in managing DN with TCM, and NF-κB, as a key factor, plays an important role in the prevention and treatment of DN. This article summarized the research results of TCM based on the NF-κB signaling pathway for the treatment of DN in the past five years. It described a variety of TCM extracts, such as polysaccharides, terpenes, phenols, flavonoids, saponins, and alkaloids, as well as TCM compound prescriptions such as Huaiqihuang granules, Astragalus mongholicus Bunge and Panax notoginseng formula, and Danzhi Jiangtang capsules, which regulated the NF-κB signaling pathway and its upstream and downstream factors to block the pathological process of DN. This inhibition aims to prevent renal pathological damage caused by DN and slow down the deterioration of renal function. The article aims to provide new ideas and references for the research and development of drugs for the prevention and treatment of DN.

摘要:Ischemic stroke(IS) is a common cerebrovascular disease in clinics, with a high disability rate. In recent years, the incidence of IS has gradually increased, especially in young people, which seriously affects the physical and mental health and quality of life of patients. Western medicine has made some progress in the treatment of IS but still faces some limitations. Therefore, choosing a measure that can assist in the treatment of IS is particularly important. A large number of studies have confirmed that traditional Chinese medicine can treat IS based on the effective ingredients and extracts of traditional Chinese medicine monomers and formulas, utilizing advantages such as multiple targets, multiple bioactive ingredients, multiple systems, and fewer adverse reactions to ensure the safety and effectiveness of the treatment pathway. At present, the mechanism of action of traditional Chinese medicine in treating IS can be achieved by intervening in various signaling pathways, mainly including secretory glycoprotein/β-catenin(Wnt/β-catenin) signaling pathway, phosphatidylinositol 3-kinase/protein kinase B(PI3K/Akt) signaling pathway, nuclear factor kappa-B(NF-κB) signaling pathway,Toll-like receptor 4/NOD-like receptor protein 3(TLR4/NLRP3) signaling pathway,mitogen-activated protein kinase(MAPK) signaling pathway, and neurogenic site-gap homologous protein(Notch) signaling pathway. This article summarized the relationship between IS and related signaling pathways in recent years,as well as the regulation of IS-related pathways and corresponding mechanisms by traditional Chinese medicine,so as to provide a reference for future clinical and experimental research on IS.

摘要:Gastroesophageal reflux disease (GERD) is a prevalent chronic digestive disorder, affecting about 20% of the population. In addition to the typical symptoms of acid regurgitation and pyrosis, GERD can lead to chronic esophageal mucosal inflammation, thereby increasing the risk of esophageal carcinoma. Proton pump inhibitors (PPIs) have emerged as the primary pharmacological intervention due to their efficacy in reducing gastric acid production. However, the long-term use of PPIs is associated with various complications, including malabsorption of nutrients, heightened susceptibility to enteric infections, and an elevated risk of fractures. These concerns have driven the pursuit of safer and more efficacious therapeutic alternatives by both patients and healthcare providers. Recent studies have highlighted the role of persistent inflammation triggered by irritation of the esophageal epithelial cells in the pathogenesis of GERD. Such inflammation not only exacerbates mucosal damage but also contributes to the chronicity and complexity of the disease. Consequently, the development of novel anti-inflammatory therapeutic strategies has become a focus in contemporary research. Chinese herbal medicine prescriptions, with distinctive anti-inflammatory and antioxidant properties, offer a promising avenue for the management of GERD. These prescriptions, composed of a variety of herbal medicines, are prescribed in accordance with the traditional Chinese medicine (TCM) principle of harmonizing Qi, blood, and Yin-Yang, thereby addressing the underlying disease processes. Among the numerous Chinese herbal medicine prescriptions for GERD, ingredients such as Coptidis Rhizoma, Bupleuri Radix, Euodiae Fructus, Glycyrrhizae Radix et Rhizoma, and Atractylodis Macrocephalae Rhizoma are commonly utilized for their effects of inhibiting inflammation, modulating immune functions, and alleviating oxidative stress. Despite the potential in treating GERD, concrete evidence-based medical proof of these prescriptions remains limited. This review summarizes the anti-inflammatory mechanisms and prospects of Chinese herbal medicines in treating GERD, providing an outlook on the value of these medicines in the anti-inflammatory management of GERD.

摘要:Alzheimer's disease (AD) is a chronic neurodegenerative disorder characterized by the progressive loss of cognitive and memory functions. Its pathological features mainly include neurofibrillary tangles formed by the aggregation of hyperphosphorylated tau proteins and amyloid plaques formed by the accumulation of β-amyloid. The exact pathogenesis of AD has not been fully elucidated, and there are currently no effective specific drugs or radical treatments available in clinical practice. In recent years, the incidence of AD has been on the rise, severely affecting life and health, making the search for effective drugs and therapeutic components for AD treatment crucial. Modern medical research has found that the brain-derived neurotrophic factor (BDNF)/tropomyosin receptor kinase B (TrkB) signaling pathway is closely related to neurogenesis, apoptosis, neuroinflammation, synaptic plasticity, and oxidative stress, playing a vital role in the pathophysiological development of AD. Additionally, Chinese medicine has a long history of treating neurodegenerative diseases with few adverse reactions and features a multi-target, multi-link, and multi-pathway approach to treatment. Therefore, the author reviewed the latest research reports in China and abroad to elaborate on the role of the BDNF/TrkB signaling pathway in the onset and progression of AD, and summarized the research progress on the regulation of the BDNF/TrkB pathway by Chinese medicine compounds and monomers in AD intervention. This study is expected to provide references for the development of clinical drugs for the prevention and treatment of AD and to broaden the perspective on Chinese medicine treatment of AD.
摘要:In vitro experiments are an indispensable method in the pharmacological research of Chinese materia medica. However, the development of in vitro experiments is hindered by the various sources, complex composition, and unstable physical and chemical properties of Chinese materia medica. Therefore, it has become an urgent problem to develop reasonable methods for in vitro pharmacological research. At present, the commonly used in vitro experimental methods in the pharmacological research of Chinese materia medica include direct administration and indirect administration. Indirect administration methods include drug-containing serum/plasma, drug-containing intestinal absorption solution, and drug-containing liver incubation solution. With the development of computer technology and systems biology, the combination of high performance liquid chromatography-mass spectrometry (HPLC-MS), omics, network pharmacology, computer/artificial intelligence-assisted drug design and other technologies with in vitro experiments of Chinese materia medica has not only greatly reduced the research cost but also significantly improved the efficiency of pharmacological exploration, effective ingredient screening, and drug discovery. Therefore, the combined methods have been gradually recognized and widely used. This paper reviewed the in vitro experimental methods in pharmacological research of Chinese materia medica and their progress, aiming to provide help for selecting reasonable research methods.

摘要:Gastric cancer is a common malignant tumor with complex pathological mechanisms, a low early diagnosis rate, and a high mortality rate. However, surgical treatment, targeted therapy, and chemotherapy have their treatment limitations and toxic side effects. Therefore, exploring the pathogenesis and mechanism of gastric cancer and finding effective treatment methods are important. At present, researches has found that tumor epithelial cells exhibit individual differences in molecular characteristics and exhibit metabolic heterogeneity that affects cell phenotype and function. The interaction between metabolites and cytokines can inhibit the formation of the tumor immune microenvironment and promote malignant progression. Therefore, metabolic reprogramming is regarded as a key feature of tumors and plays an important role in the process of tumor occurrence and development. However, the continuous deterioration of gastric cancer may be closely related to changes in the energy metabolism of cancer cells. Gastric cancer cells may regulate the dysregulation of synthesis or decomposition pathways such as glucose metabolism, amino acid metabolism, lipid metabolism, and nucleotide metabolism and activate associated signaling pathways, key proteins, and genes, leading to proliferation, invasion, and metastasis of cancer cells. In recent years, there has been a close relationship between the effective intervention by traditional Chinese medicine in gastric cancer and the regulation of metabolic reprogramming. There has been some progress in the intervention research on effective ingredients and formulas of traditional Chinese medicine for cancer. This article summarized existing Chinese and foreign literature on how gastric cancer cells affect disease progression by regulating their related metabolic networks, such as glucose metabolism, amino acid metabolism, lipid metabolism, and nucleotide metabolism, as well as how effective ingredients and formulas of traditional Chinese medicine enhance anti-tumor effects through targeted metabolism. It reviewed metabolic reprogramming intervention in gastric cancer, providing a reference for research on metabolic reprogramming regulation by traditional Chinese medicine and new targets and strategies for the treatment and prognosis of gastric cancer.
