Ziel dieser Studie war es, die Auswirkungen von Anmidan auf die Lern- und Gedächtnisstörungen bei schlafentzogenen (SD) Mäusen und die Expression von Proteinen des PI3K/Akt/mTOR-Signalwegs und der Zellautophagie zu untersuchen. Methoden: Es wurden 60 C57BL/6 Mäuse in eine normale Gruppe, eine Modellgruppe, eine niedrige, mittlere und hohe Dosis von Anmidan (6.57, 13.13, 26.26 g·kg-1·d-1) und eine Melatonin-Gruppe (1.3 mg·kg-1·d-1>), aufgeteilt und zur Erstellung eines Mausmodells wurde eine hausgemachte Schlafentzugskiste verwendet. Die Lern- und Gedächtnisleistung der Mäuse in den verschiedenen Gruppen wurde mittels des Morris-Wassertest-Labyrinths bewertet. Veränderungen in den pyramidialen Neuronen und der Zirbeldrüse wurden mittels Hämatoxylin-Eosin-Färbung (HE) und Nissl-Färbung beobachtet. Die Expression von Proteinen des Mikrotubuli-assoziierten Proteins 1A/1B leichte Kette 3 (LC3) und Protein p62 wurde mittels Immunfluoreszenz nachgewiesen, während Veränderungen in der Expression von Proteinen und der mRNA des PI3K/Akt/mTOR-Signalwegs und autophagierelevanter Proteine LC3, Beclin1, p62 mittels Western Blot und Echtzeit-PCR quantitativ nachgewiesen wurden. Ergebnisse: Im Vergleich zur normalen Gruppe beeinträchtigte Schlafentzug signifikant die räumliche Lern- und Gedächtnisleistung der Mäuse, was sich in einer Verlängerung der Latenzzeit auf der Plattform und der Gesamtstrecke des Schwimmens äußerte, während die Anzahl der Kreuze der Plattform und die Zeit der Aktivität im Zielsegment verringert wurden. Die histologische Untersuchung ergab, dass die Anzahl der Neuronen in der CA3-Region des Hippocampus bei den Mäusen der Modellgruppe abnahm und die zelluläre Degeneration zunahm, während die Konzentration der Pinealkörper abnahm. Die Ergebnisse des Western Blot, Echtzeit-PCR und der Immunfluoreszenz zeigten, dass der Schlafentzug die Phosphorylierung von PI3K (p)-PI3K, p-Akt/Akt, p-mTOR/mTOR, die Expression des Proteins p62 und der mRNA signifikant verringerte, gleichzeitig aber das Verhältnis von LC3Ⅱ/LC3Ⅰ und die Expression von Beclin1-Proteinen und -mRNA deutlich erhöhte (P<0,01), was auf eine erhöhte Autophagie hinweist. Verglichen mit der Modellgruppe verbesserte sich die Lern- und Gedächtnisleistung der Mäuse nach der Intervention mit Anmidan, was sich in einer Verkürzung der Latenzzeit auf der Plattform und der Gesamtstrecke des Schwimmens (P<0,05, P<0,01) sowie in einer Zunahme der Aktivitätszeit im Zielsegment und der Anzahl von Plattformübergängen zeigte (P<0,01). Die Schäden an den Neuronen des Hippocampus nahmen in den verschiedenen Dosisgruppen von Anmidan ab, und die Konzentration der Pinealkörper stieg an. Darüber hinaus erhöhte Anmidan die Phosphorylierung von PI3K (p)-PI3K, p-Akt/Akt, p-mTOR/mTOR, die Expression des Proteins p62 und der mRNA, reduzierte gleichzeitig aber das Verhältnis von LC3Ⅱ/LC3Ⅰ und die Expression von Beclin1-Proteinen und -mRNA (P<0,05, P<0,01), was die neuronale Autophagie hemmte.
