Das Ziel der vorliegenden Studie ist es, den Mechanismus der synergistischen Wirkung von Arsen-Trioxid (ATO) in Kombination mit seinem Metaboliten Dimethylarsen (DMAV) auf die Induktion von Apoptose bei akuten promyelozytären Leukämiezellen NB4 zu studieren. Methoden: Ratten erhielten entsprechende Dosen steriler physiologischer Kochsalzlösung, ATO (1,6 mg/kg), DMAV (4,8,16,32 mg/kg) in Kombination mit ATO (1,6 mg/kg) zur entsprechenden serösen Therapie; der Zellproliferations- und Aktivitätstest-8 (CCK-8) wurde verwendet, um die Wirkung der serösen Therapie auf die Zellproliferation der NB4-Zellen zu bewerten; die Durchflusszytometrie, die Doppel-Färbung von Annexin-V-FITC/Propidiumiodid (PI) wurde verwendet, um die zelluläre Apoptose zu bewerten; der Einsatz des fluoreszierenden Sondes DCFH-DA zur Analyse der intrazellulären reaktiven Sauerstoffakkumulation; der Einsatz des fluoreszierenden Sondes JC-1 zur Analyse der Veränderungen des mitochondrialen Membranpotenzials (Δψm); Western Blot wurde verwendet, um die Veränderungen in der Expression von Apoptose-assoziierten Proteinen Bcl-2-assoziierter X-Protein (Bax)/Bcl-2, Cytochrom C, geschnittener Caspase-9 und geschnittener Caspase-3 sowie das Phosphorylierungsniveau von JNK zu bewerten. Ergebnisse: 1) Die Kombination der beiden Medikamente führt zu einer höheren Hemmungsrate der Proliferation und mehr Apoptose im Vergleich zur einzelnen Anwendung von ATO. 2) Die Kombination der beiden Medikamente erhöht signifikant den ROS-Spiegel. 3) Die Kombination der beiden Medikamente reduziert signifikant Δψm im Vergleich zur einzelnen Anwendung. 4) Die Kombination der beiden Medikamente führt zu einer signifikanten Freisetzung von Cytochrom C, einer signifikanten Verringerung der Bcl-2-Expression, einer Erhöhung der Bax-Expression, geschnittener Caspase-3 und geschnittener Caspase-9. 5) Die Kombination der beiden Medikamente führt zu einer signifikanten Erhöhung des Phosphorylierungsniveaus von JNK im Vergleich zur einzelnen Anwendung. Fazit: Die Kombination von DMAV und ATO induziert wahrscheinlich die Apoptose durch eine synergistische oxidativen Stress vermittelte ROS-Induktion, Δψm-Auflösung, die Freisetzung von Cytochrom C, die Aktivierung von Caspase-9/Caspase-3 und das Phosphorylierungsniveau von JNK, die Regulierung der Apoptose-assoziierter Proteine der Bcl-2-Familie (Bax/Bcl-2), die die Apoptose der NB4-Zellen stimulieren.
