Resümee

Ziel ist es, den Wirkmechanismus von microRNA-155-5p (miR-155-5p) bei der Colitis ulcerosa (CU) zu untersuchen und den molekularen Mechanismus der Therapie von CU mit Paeonia-Tang durch Regulation von miR-155-5p zu erforschen. Methoden: Es wurden 48 männliche C57BL/6-Mäuse des SPF-Status ausgewählt und mittels zufälliger Zahlenlisten in 6 Gruppen zu je 8 Mäusen eingeteilt: Kontrollgruppe, Modellgruppe, Mesalazin-Gruppe (0,39 g/kg), Paeonia-Tang-Gruppe (31,08 g/kg), Januskinase-1-(JAK1)-Inhibitorgruppe (Baricitinib, 10 mg/kg) und Paeonia-Tang + Inhibitor-Gruppe (Baricitinib 10 mg/kg + Paeonia-Tang 31,08 g/kg). Mit Ausnahme der Kontrollgruppe wurden in den anderen Gruppen UC-Mausmodelle durch Trinklösung mit 3 % Dextran-Natriumsulfat für 7 Tage hergestellt. Nach Modellierung erfolgte eine 7-tägige Behandlung, wobei Paeonia-Tang und Mesalazin oral verabreicht wurden, Baricitinib intraperitoneal injiziert wurde. 24 Stunden nach der letzten Gabe wurden die Mäuse durch intraperitoneale Injektion von Pentobarbital-Natrium anästhesiert, Blut wurde zur Bestimmung der Leukozytenzahl und Blutsenkungsgeschwindigkeit entnommen, die Mäuse wurden getötet und die Kolonlänge gemessen; die Kolonpathologie wurde mittels Hämatoxylin-Eosin (HE)-Färbung beurteilt und bewertet; die Expression von miR-155-5p wurde mittels quantitativer Echtzeit-PCR gemessen; die Proteinspiegel von JAK1, phosphoryliertem (p)-JAK1, Zytokin-Signalinhibitor 1 (SOCS1), Signaltransduktions- und Transkriptionsaktivator 1 (STAT1) und p-STAT1 wurden mittels Western Blot bestimmt. Im Vergleich zur Kontrollgruppe zeigten die Modellmäuse eine signifikante Erhöhung des Krankheitsaktivitätsindexes (DAI), der pathologischen Bewertung, eine signifikante Verkürzung der Kolonlänge, eine signifikante Hochregulation der relativen miR-155-5p-Expression sowie der p-JAK1- und p-STAT1-Proteine, eine signifikante Herunterregulierung des SOCS1-Proteins, eine signifikante Verlängerung der Blutsenkungszeit und eine signifikante Zunahme der Leukozytenzahl (P<0,01); im Vergleich zur Modellgruppe verbesserten alle Behandlungsgruppen die Indikatoren signifikant (P<0,01), wobei die Paeonia-Tang-Gruppe eine signifikante Verbesserung der miR-155-5p-Expression, der zugehörigen Proteine sowie von DAI und pathologischer Bewertung gegenüber der Mesalazin-Gruppe aufwies (P<0,01). Fazit: Paeonia-Tang kann möglicherweise durch Herunterregulierung von miR-155-5p dessen Hemmung von SOCS1 aufheben, wodurch eine Überaktivierung des JAK1/STAT1-Signalwegs unterdrückt und letztlich die entzündlichen Darmverletzungen gemildert werden.
