      摘要:The quality-control of Chinese herbal materials,pesticide residues,heavy metals and other problems have caused great challenges to their effectiveness and safety.Through summarizing the developing process and characteristics of environmental standard of planting pollution-free agricultural products in China,considering the planting characteristics of Chinese medicinal materials and current standards available,we have established an environmental standard for pollution-free Chinese herbal medicines.The aim is to reduce pesticide residues and heavy metals by employing this standard to guide the environmental selection of Chinese herbal medicines.Meanwhile,the Geographic Information System for Global Medicinal Plants(GMPGIS) is proposed to reasonably and scientifically regulate Chinese herbal medicines by predicting the distribution area,authentic habitats,climate and soil conditions of producing regions so as to guide the herbal production layout.In this article,a scientific planting plan for Chinese medicinal materials by geographic information system is proposed to obtain high-quality Chinese medicinal materials.

    • Discussion on Pollution-free Planting Technology of Species

      摘要:Panax species is an Araliaceae perennial herb,which is a very famous Chinese medical herb and had huge demand in market.However,the issues of disordering production and non-standard pesticide application are common in the production of Panax species,and the non-polluted production technology would be the mainly development direction in the future and the key to solve the issues.Aimed to above problems,the research team built the pollution-free cultivation technique system of Panax species.The key technologies were established based on the research results of field investigation in the cultivated regions.The system includes suitable planting regions selecting based on Geographic Information System for Global Medicinal Plants(GMPGIS),comprehensive guiding the soil improvement,modern molecule marker assisted breeding,etc.In all,these strategies were considered to largely developing of the pollution-free Panax industry in the green and sustainable way,and in order to achieve the goal of producing high quality Chinese herbal medicines.

      摘要:The issues of traditional breeding of Chinese herbal medicines is long period and low efficiency.Aimed to above problems,our research team puts forward that the herbgenomics assists the breeding of high-quality new cultivars of Chinese medicinal materials.Through the analysis of the genome,transcriptome,simplified genome and others,a large of single nucleotide polymorphism(SNP),simple sequence repeat(SSR) and other makers were obtained,combined with high-yield,high-quality,resistance and other target traits,and then asssisted the breeding of new cultivars,such as high-yield cultivars "Zhongyan Feisu No.1" and anti-disease cultivars "Miaoxiang Kangqi No.1".However,the cultivars of Chinese herbal medicines are various and the research foundation is weak.It is necessary to provide molecular markers for the screening of high-quality genes by strengthening the study of herbgenomics.At the same time,a comprehensive breeding technique combining molecular breeding with traditional breeding should be established to achieve the linkage of molecular markers and excellent traits,and accelerate the breeding process of high-quality Chinese herbal medicines,so as to ensure the safety and effectiveness of Chinese herbal medicines.

    • Preparation of Oral Film and Investigation of Its Antioral Ulcer Action

      摘要:Objective:Based on the anti-ulcer effect of Periplaneta americana,to prepare P. americana oral film(PAOF) and investigate its pharmacodynamics. Method:Adhesion time,adhesion and dissolution time were used as the comprehensive score index to screen the best prescription of oral film with freeze-dried powder of P. americana extract as raw material.The performance evaluation and the determination of drug loading of PAOF were carried out.Subsequently,model of oral ulcer in rabbits burned with 20% glacial acetic acid was established.The histopathological changes of rabbit oral ulcer tissues were observed under light microscope with hematoxylin-eosin(HE) staining and proliferating cell nuclear antigen(PCNA) staining,the concentrations of superoxide dismutase(SOD),malondialdehyde(MDA),interleukin(IL)-2,IL-6,tumor necrosis factor-α(TNF-α) and basic fibroblast growth factor(bFGF) were measured by biochemical test and enzyme linked immunosorbent assay(ELISA),the effect of PAOF on oral ulcer was evaluated. Result:Taking 4.0 g of ployvinyl alcohol 17-88(PVA 17-88),0.3 g of hydroxypropyl cellulose(HPC),0.15 g of carbomer 934P as film forming materials,they were dissolved in 25,20,10 mL of ultrapure water,respectively.These three matrices after fully swelling were mixed,and 1 g of freeze-dried powder of P. americana extract and 0.6 mL of glycerine were added in the mixture.The PAOF had good performance for its long adhesion time[(152±2) min],long dissolution time[(173±3) min],strong adhesion force[(184±3) N] and low expansion degree(20%).Drug loading of each oral film was 2.09 mg(with glycine as index).Compared with the model group,SOD activity in oral mucosal ulcer tissues increased significantly in the PAOF high-dose group and dexamethasone acetate adhesive tablets group(P<0.01),and MDA content decreased significantly(P<0.01).IL-2 level was significantly increased(P<0.05) and IL-6 level was significantly decreased(P<0.05,P<0.01) in the PAOF high and medium dose group and dexamethasone acetate adhesive tablets group.TNF-α level was significantly decreased(P<0.05) and bFGF level was significantly increased(P<0.05,P<0.01) in the PAOF high and medium dose group and dexamethasone acetate adhesive tablets group;oral mucosa of rabbits in model group was damaged and then edema phenomenon appeared.There were different levels of tissue repair and no edema in the oral cavity ulcer in rabbits,which was significantly different from the model group.It suggested that the PAOF could promote the healing of oral ulcer in rabbits. Conclusion:This optimized preparation technology of PAOF is simple and feasible,and it has a significant effect on oral ulcer in rabbits.

      摘要:Objective:To optimize the processing technology of Alismatis Rhizoma with salt,and clarify the processing parameters,in order to provide the basis for standardized production and quality control of Alismatis Rhizoma. Method:With comprehensive score of total content of five ingredients(alismoxide,23-acetyl alisol C,alisenol,alisol B and 23-acetyl alisol B),content of alcohol soluble extract and appearance score as indicator,single factor tests and orthogonal test were adopted to optimize processing technology of salt-processed Alismatis Rhizoma by taking moistening temperature,moistening time,slice thickness and others as factors.UPLC was employed to determine contents of these five ingredients with mobile phase of acetonitrile-water for gradient elution and detection wavelength at 208 nm. Result:Optimum processing technology of salt-processed Alismatis Rhizoma was as followings:moistening temperature of 75℃,slice thicknesses of 4 mm,moistening time of 1.0 h,drying temperature of 65℃,2 kg salt dissolved with 13 kg water for each 100 kg of Alismatis Rhizoma,moistening for 3 h,stir-frying for 8 minutes under 160℃. Conclusion:This optimized production process of salt-processed Alismatis Rhizoma has good feasibility and controllability,which is suitable for industrial production of this decoction pieces.

      摘要:Objective:To systematically study on the pharmacognosy characteristics of Wahlenbergiae Marginatae Radix seu Herba,in order to provide references for its identification,quality standards and further development and utilization. Method:Under Digital-Camera 6.0 software environment combined with digital microscope and electronic eyepiece,microscopic character observation was used to observe surface characteristics of the whole plant.Tissue sections were prepared by frozen section method,then microscopic observations,photographs,measurements,and related data were recorded.The superposition of depth of focus was treated by Adobe Bridge CS6 and Photoshop CS6 softwares. Result:Roots of Wahlenbergiae Marginatae Radix seu Herba had lots of laticifer groups and irregular-crystal inulins in cortex and phloem;its stems had many infinitive stomata in flat axis,collenchymas,single-cell non-glandular hairs,endodermis,single distributed laticifers and fiber bundles.The main identificational points in its leaves were squarish epidermal cells with cuticle,infinitive stomata,single-cell non-glandular hairs,collateral vascular bundle and obvious endothelium ring.And also there were lots of other points in the crude herb,such as protuberances in the petals margins,pollen morphology,the size of fruits and seeds,wrinkles and longitudinal stripes in the surface of seeds.Hence,the microscope characteristics of the powder of Wahlenbergiae Marginatae Radix seu Herba contained lots of multi-hole fibers,infinitive stomata in flat axis,single-cell type non-glandular hairs,inulins in irregular crystals;and also there were leaf epidermal cells of shallow waves,oblong seeds of longitudinal stripes,scattered colorless transparent oil drops,round pollen grains of three germinating holes in the medicinal powder;the above aspects could be used as the main identificational points of its powder. Conclusion:The main identificational points of Wahlenbergiae Marginatae Radix seu Herba include the characteristics of the transverse section of its roots,stems and leaves,and the surface characteristics of its flowers,fruits and seeds.

    • Microscopic Identification of Medicinal Material from Six Different Species

      摘要:Objective:To study the microscopic characteristics of Uncaria medicinal material from six different species, and provide basis for fundamental identification of Uncaria medicinal material. Method:Transection characteristics and powder microscopic characteristics of stem, hook and leaf from 6 species of Uncaria were observed under electron microscope. Result:The tissues and structures of different species of Uncaria were basically the same composed of epidermis,cortex,phloem,xylem and medulla from outside to inside. Most of the cortex,phloem and pith parenchyma cells of Uncaria contained brown inclusion, but there were different features in different species,such as the shape of main veins on the back of abdomen,the number of columns of the palisade tissue,the number of vascular bundles,and the type of crystal of calcium oxalate visible in parenchyma cells. The comparative microscopic characteristics of six species of Uncaria medicinal material showed that characters such as the transverse section profile of stem,the presence or absence of epidermal hair,the proportion of phloem and xylem,the presence of clear ring in cambium,the width of radiation ray,the scattered or radially connected catheters,the proportion of pith as well as the protruding shape of main veins (of leaves) in ventral and dorsal surface,the column number of palisade tissue,the type of crystals in parenchyma cells and the shape of hook,were used as the fundamental identification characteristics among various species. Powder identification included the shape of epidermal cells and the presence of oil droplets,ladder vessel and reticulate vessel within those cells. Conclusion:The above characteristics can be used as the identification basis for the six species of Uncaria medicinal material.

      摘要:Objective:To provide scientific basis for the quality evaluation and identification of Belamcanda chinensis,Iris tectorum and I. lactea var. chinensis seeds by studying the morphological and anatomical structure of seeds. Method:The morphology was observed under stereo microscope,and paraffin sections were made to observe the anatomical structure. Result:Seeds of B. chinensis,and I. tectorum were spherical and enveloped with black lustrous arils, with reticulate grain outside. The hilum of B. chinensis was round or oval,tawny,adn low arched. The hilum of I. tectorum was round,hoary or yellowish white. Seeds of I. lactea var. chinensis were irregularly spherical,dark brick-red,tan or atropurpureus,and lusterless, with episperm and endopleura closely combined. The hilum of I. lactea var. chinensis was yellowish white and linear. The microscopic structure showed that the testa cells of B. chinensis were heliotrope,and of I. tectorum were dark purple. I. lactea var. chinensis seeds had no arils,but with a complex testa structure. All of seeds of the three species possessed a compact endosperm,and the embryos were usually central. I. lactea var. chinensis seeds were different from B. chinensis and I. tectorum seeds in shape,color,the presence or absence of aril,shape and color of hilum. Based on the above characteristics, I. lactea var. chinensis seeds could be distinguished from the latter two species. B. chinensis and I. tectorum seeds were similar in appearance,but they were different in shape and color of hilum and color of testa cells,which can be regarded as identification characters. Conclusion:All of the above mentioned characteristics can be used to distinguish B. chinensis,I. tectorum and I. lactea var. chinensis seeds and provide basis for identifying them.

      摘要:Objective:To evaluate the factors that can affect the quality from harvest to decoction pieces of Fritillariae Cirrhosae Bulbus (FCB) and to provide reference for its standardized processing,reasonable storage,and modern packaging by investigating the present situation about post-harvest processing,storage,and packaging of FCB from different habitats. Method:On the basis of present researches,various factors which can affect the quality of FCB were analyzed comprehensively by field investigation in medicine markets and genuine producing areas, supplemented by literature retrieval and analysis. Result:Through investigation,the present situation and problems of the initial processing,storage condition,and packaging after harvest were summarized in the present paper. Conclusion:FCB is harvested in unconcentrated wild mainly. The harvest time is scattered. The traditional technology of drying process is primitive,which is a key step in production of FCB. The phenomenon of Youzi (oily) and Huangzi (yellow) was easy to appear in storage process. Damages by worms can appear in the packaging section. All sections still lack scientific and unified standards,affecting the quality of FCB products. In order to ensure the quality of FCB and safety of clinical uses,we need to use a variety of techniques to control the whole process from multiple sections,establish a reasonable quality evaluation method and indicators,and formulate a scientific standard operating procedure.

    • Chemical Compositions from Stems and Their Anti-fungal Activities

      摘要:Objective:To investigate the chemical compositions from the stems of Murraya microphylla and their anti-fungal activity against eight plant fungal pathogens in vitro. Method:The compounds were isolated and purified by the normal phase silica gel,Sephadex LH-20 HPLC column chromatography and other methods. Their structures were identified on the basis of spectral analysis such as 1H-NMR,13 C-NMR and MS. Their anti-fungal activity was evaluated by the mycelium growth rate method. Result:Eleven compounds were obtained and identified as girinimbine (1),mahanimbine (2),koenimbine (3),koenidine (4),mukonal (5),murrayanine (6),murrayaquinone-B (7),β-sitosterol (8),daucosterol (9),3β-hydroxystigmast-5-en-7-one (10),and tetracosanoic acid,methyl ester (11),respectively. Even at the high concentration of 200 mg·L-1,carbazoles 1-7 exhibited different anti-fungal activity against these fungi in vitro,especially for M. grisea. Compounds 6 and 1 showed a broad spectrum and potent activity against these fungal pathogens, exhibiting a high activity against M. grisea,with IC50 values of 65.81,161.61 mg·L-1,respectively. Conclusion:Compounds 2-3,5-7 were isolated from the plant for the first time. Compounds 1 and 6 had great potentials for further research and development.

      摘要:Objective:A great deal of researches have shown that Populus tomentosa male inflorescence,as our traditional Chinese medicine,has a variety of chemical constituents and strong anti-bacterial, anti-inflammatory and other pharmacological effect. The aim of this study was to investigate the chemical constituents of P. tomentosa systemically to enrich its research materials and lay foundations for the development of new drugs and its clinical application. Method:The male anthotaxy of P. tomentosa male inflorescence(30 kg)was extracted for three times with 70% ethanol,concentrated under reduced pressure,and then the extract was suspended in water. Then the solvent was partitioned with petroleum ether,EtOAc and n-BuOH successively. The components of EtOAc extract were isolated and purified by ODS,silica gel,Sephadex LH-20 column chromatography and recrystallization. Their structures were identified based on the physiochemical properties and spectroscopy techniques such as ESI-MS and nuclear magnetic resonance (NMR). Result:Twelve compounds were isolated from the ethyl acetate fraction,and they were identified as quercetin(1),taxifolin(2),luteolin(3),eriodictyol(4),myricetin(5),dihydromyricetin(6),(2S)-5,7-dihydroxyflavanone-7-O-β-D-glucopy-ranoside(7),protocatechuic acid(8),caffeic acid(9),2-acetyl-1,3-dicaffeoylglycerol(10),salicortin-6'-benzoate(11),and daucosterol(12),respectively. Conclusion:Compounds 2,5-7 were isolated from the Populus genus for the first time,and Compounds 2,4-8,10,11 were isolated from the plant for the first time,providing certain chemical basis for comprehensive utilization of Populus tomentosa.

      摘要:Objective:To explore the method of multi-index quality control for Aolanti Weikang tablets,and establish its HPLC characteristic spectrum and determine the contents of main flavonoid glycosides in Aolanti Weikang tablets. Method:Neoeriocitrin,narirutin,naringin,hesperidin and neohesperidin were selected as index components,HPLC analysis was applied on an YMC-Pack ODS-A column(4.6 mm×250 mm,5 μm),the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution(18:82),the detection wavelength was set at 283 nm,the flow rate was 1.0 mL·min-1,and the column temperature was 30℃. Result:HPLC characteristic spectrum was established,and five characteristic peaks in ten batches of Aolanti Weikang tablets were simultaneously determined.Five components(neoeriocitrin,narirutin,naringin,hesperidin and neohesperidin) all showed good linear relationship within the ranges of 0.266 5-10.66 mg·L-1(r=0.999 9),1.055-42.20 mg·L-1(r=1.000 0),7.070-282.8 mg·L-1(r=1.000 0),0.797 8-31.91 mg·L-1(r=0.999 9),6.955-278.2 mg·L-1(r=1.000 0),respectively.The average recoveries of these five components were 99.4%(RSD 1.0%),100.5%(RSD 0.6%),97.8%(RSD 0.9%),96.7%(RSD 0.7%) and 98.3%(RSD 0.8%).The similarity of characteristic spectrum between Aolanti Weikang tablets and the total flavonoid glycosides extract of Aurantii Fructus Immaturus was 1.000. Conclusion:The proposed HPLC characteristic spectrum and determination method of five flavonoid glycosides can be used as the quality control method of Aolanti Weikang tablets with multitude indexes.

      摘要:Objective:To establish a ultra-high performance liquid chromatography-triple quadrupole tandem mass spectrometry (UPLC-QqQ-MS/MS) method for the simultaneous determination of ten kinds of stilbene glycosides in Polygoni Multiflori Radix praeparata,and compare their contents in samples from different origins. Method:The determination was performed on the ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.8 μm);with 0.1% formic acid water(A) and 0.1% formic acid acetonitrile(B) as the mobile phase for gradient elution. The flow rate was 0.5 mL·min-1and the column temperature was 40℃. The extracted ion chromatograms were obtained with ESI- ion source under negative ion detection method, and the peak area was used for quantitative analysis. Result:The resveratrol,polydatin,2,3,5,4'-tetrahydroxysilbene-2-O-glucopyranoside,2,3,5,4'-tetrahydroxysilbene-2,3-di-O-glucopyranoside,2,3,5,4'-tetrahydroxysilbene-2-O-glucopyransyl-(1→6)-glucopyranoside,2,3,5,4'-tetrahydroxysilbene-2-O-(2″-O-galloyl)-glucopyranoside,2,3,5,4'-tetrahydroxystilbene-2-O-(6″-O-acetyl)-glucopyranoside,2,3,5,4'-tetrahydroxy silbene-2-O-(2″-O-ferulicacyl)-glucopyranoside,2,3,5,4'-tetrahydroxysilbene-2-O-(2″-O-coumaryl)-glucopyranoside and 2,3,5,4'-tetrahydroxystilbene-2-O-(2″-O-acetyl)-glucopyranoside in Polygoni Multiflori Radix showed good linearity relationship,r ≥ 0.998 7.The average recovery rate was 96.5%-106.3%,RSD ≤ 3.5%. Conclusion:The established method is simple, accurate and reproducible for the determination and quality control of Polygoni Multiflori Radix.

      摘要:Objective:To explore the correlation between high performance liquid chromatography (HPLC)fingerprint and free radical-scavenging activity of Rhamnus heterophylla leaves. Method:Fingerprint of eight batches of R. heterophylla leaves from different regions and harvest times was established by HPLC. Origin Pro 2017 software was used for hierarchical cluster analysis(HCA) and principal component analysis (PCA) to evaluate the common peaks in the fingerprint on the basis of traditional similarity evaluation method. The free radical-scavenging activity was determined by 1,1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl (DPPH) assay. Partial least squares regression (PLSR) and grey relational analysis (GRA) were used to study the relationship between HPLC fingerprint and free radical-scavenging activity. Result:The HPLC fingerprint of eight batches of R. heterophylla leaves was established,and 11 common peaks were determined,with the similarity of 0.948-0.976.Six peaks among common peaks were identified by comparing to reference substances:x1,x2,x5,x8,x10,x11 referred to protocatechic acid,luteoloside,quercetin,kaempferol,emodin-8-O-α-L-rhamnoside,and emodin,respectively. The samples were clustered into four categories. All of the samples had different degrees of scavenging free radical activity. The result of GRA showed that the order of correlation was x11 > x10 > x8 > x1 > x9 > x4 > x3 > x2 > x6 > x5 > x7.The regression equation established by PLSR analysis was Y=99.769 17-0.357 49x1-0.001 36x2-0.002 65x3+0.059 63x4+0.011 81x6+0.010 63x7-0.006 99x8-0.009 14x9+0.054 27x10+0.022 75x11. Conclusion:The free radical-scavenging activity of R. heterophylla leaves was the result of the combined effects of multiple components.

      摘要:Objective:To establish the methods of content determination of stachydrine hydrochloride in Leonurus granule by high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD). Method:The samples were treated with 732 sodium cation exchange resin. The determination was performed on Venusil HILIC column(4.6 mm×250 mm,5 μm);column temperature:room temperature;with acetonitrile and 0.2% acetic solution as mobile phase (80:20) at the flow rate 1 mL·min-1;sample amount 10 μL;drift tube temperature 79℃;air volume flow 2 L·min-1. Result:The stachydrine hydrochlorideshowed a good linear relationship within the range of 0.498-4.980 μg(r=0.999 5,n=6);and the average recovery was 99.7% and 99.3% respectively for sugar free Leonurus granule and sugar containing Leonurus granule. Conclusion:The method is specific, accurate and simple,which can be applied as a new way for content determination of Leonurus granule,and provide reference for content determination of stachydrine hydrochloride in other unilateral preparations of Leonuri Herb.

      摘要:Objective:To analyze the chemical constituents and molecular mechanism of pharmacological action of essential oil from Xinjiang Carthami Flos. Method:Essential oil was extracted from Carthami Flos with the hydrodistillation (HD) method and its components were analyzed by gas chromatography and mass spectrometry (GC-MS). The contents of the chemical components were determined by using the peak area normalization method. The structure of active component (relative content>1) MOL2 of the essential oil was downloaded and Ligand library was established based on TCSMP. Target prediction based on Swiss Target Prediction was used to construct the major chemical components-target network model in order to screen key targets. Prediction of drug metabolism active was based on admetSAR and SwissADME. The diseases with involvement of key targets were screened by comparative toxicogenomics database (CTD) to construct key target-disease network model. Key protein validation was based on SystemsDock online software,docking the volatile oil from Carthami Flos with carboxylesterase 1 (CESI),carbonic anhydrase 1 (CA1),and CA2. Result:The 48 compounds were identified by mass spectrometry and literature retrieval. Main chemical compositions (7,9-docosanedione,caryophyllene oxide,heptacosane,hexacosane,hexahydrofarnesyl acetone,nonacosane,pentacosane,tetracosane,and tricosane) accounted for 32.787% of the total essential oil. 135 target proteins were predicted from 9 chemical compositions,and CES1,CES3,CA1,CA2 were key targets, mainly playing a role in diseases such as cancer,liver disease,and lung disease. The key target proteins verification based on SystemsDock showed a strong binding activity of key components and important targets. Conclusion:The present study has preliminarily predicted the target proteins with pharmacological action for the essential oil from Xinjiang Carthami Flos, providing a theoretical basis for the development and application of its products.

      摘要:Objective:To explore the mechanism of Buyang Huanwu Tang in intervening hypoxia preconditioned cardiac fibroblasts and promoting mesenchymal stem cell migration. Method:The 6.5,13,26 g·kg-1 Buyang Huanwu Tang and the same volume of distilled water were used to continuously administer SD(Sprague Dawley)rats for 7 days. Then, serum containing Buyang Huanwu Tang and blank serum were prepared. After different doses of serum containing Buyang Huanwu Tang and intervening hypoxia preconditioned cardiac fibroblasts(CFs)for 24 h, enzyme-linked immunosorbent assay(ELISA), real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot were used to detect the protein levels of hypoxia-inducible factor-1α(HIF-1α), monocyte chemotactic protein-1(MCP-1)and hepatocyte growth factor (HGF) in CF culture medium. After culture medium intervened the mesenchymal stem cells (MSC), the MSC were tested for cell migration. Result:Compared with normoxic group, hypoxia could significantly promote the secretion of HIF-1α, MCP-1 and HGF of CFs (P<0.01). At the same time, the levels of HIF-1α, MCP-1 and HGF were significantly increased after intervention of MSC with hypoxia-pretreated CFs (P<0.01). When different doses of serum containing Buyang Huanwu Tang were given, the expressions were further increased compared with the hypoxic group (P<0.05, P<0.01). After HIF-1α inhibitor YC-1 was given, the expressions of MCP-1 and HGF were increased in Buyang Huanwu Tang group(P<0.05). Conclusion:Buyang Huanwu Tang combined with hypoxia preconditioning CFs paracrine effect can promote the migration of MSC, which may be achieved by up-regulating the expression of HIF-1α and increasing the expression of the migration factors MCP-1 and HGF.

      摘要:Objective:To investigate effect of Wuzi Yanzong Fang (WYF) on proliferation of spermatogonial stem cells in aging rats through phosphoinositol 3-kinase(PI3K)/protein kinase(Akt) signaling pathway. Method:The 18-month-old male SD rats were randomly divided into aging model group, and low, middle and high-dose WYF groups, with 10 rats in each group. Ten 2-month-old rats were used as young control group. low, middle and high-dose WYF groups were given WYF 0.4, 0.8, 1.6 g·kg-1 by gavage for 4 months, respectively. Young control group and aging model group were given normal saline, and the administration was suspended for 2 days a week. The administration lasted for 4 months. The rats were put to death after the final administration of WYF, and then the testes were quickly collected. The numbers of Sertoli cells were detected by immunofluorescence. The relative mRNA expression levels of glial cell line-derived neurotrophicfactor (GDNF), promyelocyte Zinc finger protein (PLZF), bone morphogenetic protein4 (BMP4) and stem cell factor (SCF) in testicular tissues were detected by Real-time quantitative PCR (Real-time PCR). The numbers of spermatogonial stem cells and the expression levels of PCNA were detected by immunofluorescence. The expressions and localizations of PI3K, Akt, p-Akt were detected by Western blot and immunofluorescence. Result:Compared with young control group, there was no significant difference in the numbers of Sertoli cells in each group, the mRNA expression levels of GDNF, PLZF, BMP4 and SCF in testis of aging model group were significantly decreased. The number of spermatogonial stem cells and levels of PI3K, p-Akt, PCNA expression decreased(P<0.01). After WYF treatment, the levels of GDNF, PLZF, BMP4 and SCF were significantly increased, the numbers of spermatogonial stem cells were increased, and PCNA, PI3K, p-Akt expressions were up-regulated(P<0.05,P<0.01). Conclusion:WYF can effectively promote the proliferation of spermatogonial stem cells in aging rats through PI3K/Akt signaling pathway.

      摘要:Objective:To observe the effect of Juanbi Lijieqing formula on Toll-like receptors 4(TLR4), nuclear factor-kappa B (NF-κB), peroxisome proliferator-activated receptor γ(PPARγ)in synovial tissue of modified gouty arthritis rats, in order to explore the possible mechanism of Juanbi Lijieqing formula in the treatment of acute gouty arthritis with local tissue inflammation. Method:Among 42 male SD rats, 6 were selected as blank group according to the random number table method, 30 were included in the modeling group. The modeling group was randomly divided into model group, Juanbi Lijieqing formula group (4 400, 2 200, 1 100 mg·kg-1), celecoxib group (11 mg·kg-1), and pioglitazone group (20 mg·kg-1), with 6 mice in each group. Rats in normal control group and model group were given normal saline (20 mL·kg-1). Rats in each group were administrated with drugs for 2 times a day for 2 days; after that, the rats were put to death, the right ankle joints were collected, and synovial tissues were isolated. The tissues were divided into two parts, one part was used for the observation of pathomorphology, while the other part was used to detect mRNA expressions of TLR4, NF-κB p65 and PPARγ by reverse transcription-polymerase chain reaction (RT-PCR). Western blot was used to detect protein expressions of TLR4, NF-κB p65 and PPARγ. Result:Compared with the normal control group, mRNA and protein expressions of TLR4 and NF-κB increased significantly (P<0.01) in the model group, and mRNA and protein expression of PPARγ in the model group increased (P<0.05). Compared with the model group, the expressions of TLR4 and NF-κB in the pioglitazone group and low, middle and high-dose Juanbi Lijieqing formula groups decreased significantly (P<0.01), and mRNA and protein expression of PPARγ increased significantly(P<0.05). Conclusion:Juanbi Lijieqing formula may inhibit local inflammation in acute gouty arthritis by up-regulating the expression of PPARγ and inhibiting the expressions of TLR4 and NF-κB.

      摘要:Objective:To explore the effect and mechanism of olivine (OL) on glucose metabolism of liver in diabetic mice. Method:Totally 40 db/db mice of spontaneous diabetes were randomly divided into model group, metformin group (0.13 g·kg-1), and high and low-dose olivine group (0.05, 0.025 g·kg-1), with 10 mice in each group, and 10 C57BL/6J mice were set as normal group. After 6 weeks of treatment, fasting blood glucose(FBG), Fins, HOMA insulin resistance index(HOMA-IR), oral glucose tolerance test (OGTT), body weight, serum creatinine (SCr), blood urea nitrogen(BUN), aspartate aminotransferase(AST) and liver glycogen content were measured. Real-time PCR was used to detect the mRNA expressions of glucose-6-phosphatase (G-6-P) and phosphoenolpyruvate carboxykinase (p-EPCK). Western blot was used to detect the protein expressions of forehead box-containing protein of the O subfamily 1 (FoxO1) and protein kinase B (Akt). Result:Compared with model group, body weight, FBG, Fins and HOMA-IR in OL group decreased significantly (P<0.01); liver glycogen content increased significantly in OL group (P<0.05, P<0.01); blood glucose significantly decreased at 120 min of OGTT test (P<0.01); the mRNA expressions of p-EPCK,G-6-P in mouse liver decreased significantly (P<0.01), and the phosphorylation levels of Akt, FoxO1 increased significantly (P<0.05, P<0.01). Conclusion:OL can inhibit gluconeogenesis, decrease FBG, and alliviate insulin resistance, which may be related to the increase of phosphorylation of Akt-FoxO1 and the decrease of transcription of p-EPCK and G-6-P.

      摘要:Objective:To discuss the anti-tumor mechanism of Xiaoai Jiedu decoction(XAJD) through the genome-wide expression profile chip of transplanted tumor in H22 tumor-bearing mice and the expression of related proteins. Method:ICR mice were randomly divided into different groups after inoculating ascites cells from H22 tumor-bearing mice, including blank control group, XAJD low, middle, and high dose groups (10, 30, 90 g·kg-1) and cisplatin group (1 mg·kg-1). After drug administration, all the animals were killed and transplanted tumor tissues were harvested, washed by sterile saline and weighed to calculate the tumor inhibition rate. Some fresh tumor tissues were used to conduct genome-wide expression profile chip detection and some were used to detect their relevant mRNA expression and protein expression by Real-time polymerase chain reaction (Real-time PCR) and Western blot assay respectively. Result:As compared with blank control group, all of XAJD low, middle and high dose groups could significantly reduce transplanted tumor weight (P<0.05), and the effect of XAJD middle dose group and cisplatin group were better than those in XAJD low and high dose groups (P<0.01), reaching an inhibitory rate of 42.8% and 58.6% respectively. The genome-wide expression profile chip analysis showed that matrix metalloproteinases(MMPs) and tissueinhibitor of metalloproteinase(TIMPs) related proteins had obvious changes in different dose groups of XAJD. Real-time PCR and Western blot assay showed that as compared with blank control group, mRNA and protein expression levels of MMP9 were significantly lower in XAJD low, middle, high dose groups and cisplatin group (P<0.01); and the mRNA and protein expression levels of MMP12 were significantly lower in XAJD middle, high dose groups and cisplatin group (P<0.01); while the mRNA and protein expression levels of TIMP2 were significantly higher in XAJD middle, high dose groups and cisplatin group (P<0.05). Conclusion:XAJD can inhibit tumor growth of H22 tumor-bearing mice, and the anti-tumor mechanism might be associated with inhibiting the expression of MMPs related proteins and reducing tumor metastasis and recurrence.

      摘要:Objective:The in vitro study of different doses of polysaccharides from Curcumae Kwangsiensis Radix on the protective effect and mechanism of H2O2-induced injury of HUVECs. Method:Protective effects of polysaccharides from Curcumae Kwangsiensis Radix on H2O2-induced injury of human umbilical vein endothelial cells (HUVECs) and its mechanism were studied. HUVECs were cultured in vitro, HUVECs were injured with different concentrations of H2O2, and the optimal concentration of H2O2 was 500 μmol·L-1 by cell counting kit-8(CCK-8) method. Then, different concentrations of polysaccharides from Curcumae Kwangsiensis Radix were used to protect the damaged HUVECs. The CCK-8 method was used to determine 62.5, 125, 250 mg·L-1 as the low, medium and high-dose groups. Effect of CCK-8 method was used to detect the polysaccharides from Curcumae Kwangsiensis Radix H2O2 on the injury of HUVECs cell activity. Lactic dehydrogenase (LDH), superoxide dismutase (SOD), glutathione-peroxidase (GSH-Px), malondialdehyde (MDA) were determined by kit. The levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in cells were detected by enzyme-linked immunosorbent assay (ELISA). Hoechst 33258 fluorescence staining was used to observe the cell apoptosis. The protein expressions of B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X (Bax) and cysteine aspartate-specific protease-3 (Caspase-3) genes were detected by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Result:The result of CCK-8 assay showed after administration for 24 h, polysaccharides from Curcumae Kwangsiensis Radix within the concentration range of 15.625-500 mg·L-1 can promote cell proliferation; as the concentration increased, polysaccharides from Curcumae Kwangsiensis Radix had a certain inhibitory effect on cells. Compared with the model group, polysaccharides from Curcumae Kwangsiensis Radix could significantly reduce the content of LDH, MDA, IL-6, TNF-α(P<0.05,P<0.01), while significantly increased the contents of SOD and GSH-Px(P<0.05,P<0.01). Hoechst 33258 fluorescence staining results showed that compared with the normal group, the nucleus of HUVECs showed bright blue fluorescence in the model group, dense and dense granular lumps of fluorescence were observed in the cytoplasm, and the cell density was decreased. Compared with the model group, the blue fluorescence intensity decreased, and the typical morphological apoptosis decreased. The Real\|time PCR results showed that compared with the normal group, the model group decreased the expression of Bcl-2 and increased the expression of Bax and Caspase-3(P<0.05,P<0.01). Compared with the model group, polysaccharides from Curcumae Kwangsiensis Radix group could increase the expression of Bcl-2(P<0.05,P<0.01), while reduce Bax, Caspase-3 expression(P<0.05,P<0.01). Conclusion:polysaccharides from Curcumae Kwangsiensis Radix have a protective effect on human umbilical vein endothelial cells injured by H2O2 by resisting oxidation and inhibiting cell apoptosis.

      摘要:Objective:To investigate the therapeutic effect of polysaccharides from Eucommiae Cortex on liver fibrosis, in order to provide the experimental basis for the development of new anti-liver fibrosis drugs. Method:The 60 healthy SD rats were randomly divided into 2 groups, namely the normal control group (n=10) and the hepatic fibrosis(HF) group (n=50). The model of liver fibrosis was prepared through intraperitoneal injection with 40%CCl4 in the model group. The model was randomly divided into 5 groups:the model control group, the high, middle and low-dose polysaccharides from Eucommiae Cortex groups and the colchicine group, with 10 rats in each group. After successful modeling, colchicine group (1×10-4g·kg-1), and high, medium and low-dose polysaccharides from Eucommiae Cortex groups (0.14, 0.07, 0.035 g·kg-1) were given drugs by gavage for 8 weeks, and then samples were collected to determine the mass of rats and calculate the liver coefficient. The levels of serum interleukin-6 (IL-6), high-mobility group box 1 (HMGB1), lipopolysaccharide (LPS) and the levels of matrix metalloproteinse-1(MMP-1) and tissue inhibitors of metalloproteinase-1(TIMP-1) in liver tissues were detected by enzyme-linked immunosorbent assay(ELISA). The expressions of Ⅰ, Ⅲ type collagen, MMP-1, TIMP-1 and transforming growth factor-β1(TGF-β1) in liver tissues of rats were analyzed by reverse transcription-polymerase chain reaction(RT-PCR) technique. Result:CCl4 can significantly reduce the body mass (P<0.01) of experimental rats and increase the liver coefficient (P<0.01). Polysaccharides from Eucommiae Cortex can significantly increase the body mass of the HF model (P<0.01), and significantly lower the HF model liver coefficient (P<0.01). ELISA detection showed that polysaccharides from Eucommiae Cortex can significantly reduce IL-6, HMGB1 and LPS content (P<0.01) in the serum of liver fibroid rats. Polysaccharides from Eucommiae Cortex and colchicine could significantly reduce the mRNA content of Ⅰ, Ⅲ type collagen, TIMP-1 and TGF-β1 (P<0.05, P<0.01) in rat liver, and significantly increase the content of MMP-1 (P<0.05, P<0.01), with a dose-effect relationship. Conclusion:Polysaccharides from Eucommiae Cortex had a significant anti-hepatic fibrosis effect. The mechanism may be related to the inhibition of the expressions of I, type Ⅲ collagen, TIMP-1 and TGF-β1 in HF liver.

      摘要:Objective:To study the effect of total flavonoids from Puerariae Lobatae Radix on oxidative stress injury induced by H2O2 in human umbilical vein endothelial cells (HUVECs). Method:With HUVECs as the study object, the experiment was divided into blank group, model group, positive drug group (10 μmol·L-1, Edaravone) and total flavone from Pueraria Lobatae Radix groups (10, 20, 40, 80, 160 mg·L-1). Methyl thiazolyl tetrazolium(MTT) assay was used to detect the protective effect of total flavonoids of Puerariae Lobatae Radix on HUVECs. The contents of superoxide dismutase (SOD), malondialdehyde (MDA) and nitric oxide (NO) were detected by spectrophotometry, and the content of endothelin-1(ET-1) in the cell supernatant was detected by enzyme linked immunosorbent assay (ELISA). The level of reactive oxygen(ROS) was detected by DCFH-DA, and Western blot was used to detect the expressions of endothelial nitric oxide synthase (eNOS) and vascular cell adhesion molecule-1 (VCAM-1). Result:Compared with normal group, model group showed a significantly lower cell survival rate, activity of SOD, level of NO and eROS protein expression (P<0.01), and a significantly higher MDA, ET-1 level, cell ROS level and VCAM-1 protein expression (P<0.01). Compared with model group, total flavone from Pueraria Lobatae Radix (40, 80, 160 mg·L-1) could significantly increase the survival rate of HUVECs with oxidative stress injury (P<0.05, P<0.01), and total flavone from Pueraria Lobatae Radix (20, 40, 80 mg·L-1) could significantly increase NO level and eNOS, and significantly decrease MDA and ET-1 levels, cell ROS level and VCAM-1 expression (P<0.05, P<0.01). Conclusion:Total flavonoids from Puerariae Lobatae Radix can protect human umbilical vein endothelial cells from oxidative stress injury induced by H2O2.The mechanism may be related to the regulation of oxidative stress-related factors of SOD, MDA, ET-1 and NO, and the expressions of endothelial cell function-related proteins eNOS and VCAM-1.

    • Mechanism of in Regulating Tumor Angiogenesis by NF-B/VEGF Signal Pathway

      摘要:Objective:To observe the mechanism of Humifuse euphorbia on tumor angiogenesis in H22 liver cancer mice by regulating signal pathway of nuclear factor kappa-B/vascular endothelial growth factor (NF-κB/VEGF). Method:The models of SPF grade Kunming mice with H22 liver cancer were established through armpit injection. The animals with successful modeling were randomly divided into five groups:model control group, cyclophosphamide group (ip, 50 mg·kg-1), high-dose group (ig, 4.50 g·kg-1), middle-dose group (ig, 2.25 g·kg-1) and low-dose group (ig, 1.13 g·kg-1), with 10 mice in each group. Their weights and tumor weights were measured after the 14-day drug administration. Index of tumor was calculated. Tumor micro-vessel density, which was marked as platelet endothelial cell adhesion molecule-31, was detected by immunohistochemitry. Tumor tissue NF-κB, VEGF, tumor necrosis factor-α(TNF-α)mRNA expressions were detected by polymerase chain reaction and protein expression by immunohistochemitry. Result:Compared with model group, H. euphorbia could significantly reduce tumor weigh (P<0.01). High-dose group showed an inhibition rate of 59.1% for H22 liver cancer in mice. CD31 expression of the high-dose group was significantly lower than that of the model group, while the expressions of NF-κB, TNF-α, VEGF mRNA and protein in tumor tissue were significantly decreased (P<0.01). Conclusion:H. euphorbia can inhibit the growth of H22 liver cancer mice, and its mechanism is probably related to the inhibition of the growth of tumor angiogenesis, which may activate NF-κB/VEGF signaling pathway.

      摘要:Objective:To explore the clinical efficacy of modified Dingxianwan combined with levetiracetam on syndrome of phlegm-fire disturbing mind in children with epilepsy. Method:The 114 children with epilepsy of phlegm-fire disturbing mind type admitted in our hospital from September 2015 to September 2017, were evenly divided into two groups on the basis of random number table. The patients in control group (57 cases) were given with levetiracetam tablets orally, while the patients in observation group (57 cases) received modified Dingxianwan in addition to what was applied to control group. All children from the two groups were treated for 6 months. The clinical efficacy, changes of Chinese revised Wechsler Intelligence Scale for children (C-WISC) and the Quality of Life in Childhood Epilepsy (QOLCE) scores before and after treatment, and adverse reactions, interleukin-6 (IL-6), interleukin-1β(IL-1β), tumor necrosis factor-α (TNF-α), and superoxide dismutase (SOD), reduced glutathione(GSH), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) levels were detected and compared between two groups. Result:After 6 months of treatment, the overall effective rate was 93.0% (53/57) in observation group, much higher than 77.2% (44/57) in control group (χ2=5.600, P<0.05). After treatment, both groups showed a remarkable increase in scores for their intellectual function[performance intelligence quotient (PIQ), verbal intelligence quotient (VIQ)]and full intelligence quotient (FIQ) as compared with those before treatment (P<0.05), and the increase in observation group was more obvious (P<0.05). As compared with those before treatment, the scores for all QOLCE dimensions (including physical function, emotional well-being, and behavioral function, etc.), general health, self-rating life quality, and the total score for QOLCE were obviously increased in both groups after treatment (P<0.05), and observation group showed a more significant increase (P<0.05). As compared with control group[5.3%(3/57)], the adverse reaction rate was 8.8% (5/57) in observation group, with no statistically significant difference. The levels of IL-6, IL-1β, TNF-α and MDA in observation group were lower than those in control group (P<0.05); the levels of serum SOD, GSH, and GSH-Px were higher than those in control group (P<0.05). Conclusion:In treating children with epilepsy (syndrome of phlegm-fire disturbing mind), the combination of modified Dingxianwan and levetiracetam can significantly relieve the symptom in the children, mitigate epileptic seizures, and improve cognitive function and life quality, proving to be effective and safe.

      摘要:Objective:To investigate the clinical efficacy of Tangshen prescription combined with Guanchang enema for patients with type 2 Ⅳ-stage diabetic kidney disease (DKD) and observe its effect on clinical indices and inflammatory factors. Method:The 120 eligible patients with type 2 Ⅳ-stage DKD were randomly divided into three groups:western medicine group, Tangshen prescription (TS) group and Tangshen prescription combined with Guanchang enema (TG) group. A parallel control study was conducted according to certain medication methods. The clinical effective rate and Serum creatinine(SCr), urinary albumin excretion rate(UAER), 2 h-postprandial plasma glucos(2 h-PPG), 24 h urinary protein quantitation (24 h UPr), transforming growth factor-β1(TGF-β1), and plasminogen activator inhibitor-1(PAI-1) levels in each group were observed both before and after treatment. Result:After treatment, the total effective rate in TG group was higher than that in western medicine group (P<0.05). As compared with those before treatment,all the indexes except SCr and 2 h PPG were significantly reduced after treatment in all groups (P<0.05, P<0.01).After treatment, as compared with western medicine group, levels of UAER, 24 h UPr, TGF-β1, PAI-1 were decreased in TS group(P<0.05, P<0.01), and levels of SCr, UAER, 2 h PPG, 24 h UPr, TGF-β1, and PAI-1 were significantly decreased in TG group (P<0.05,P<0.01). After treatment, as compared with TS group, levels of SCr, UAER, 24 h UPr, TGF-β1, and PAI-1 were decreased in TG group (P<0.05, P<0.01). Conclusion:The combination of Tangshen prescription and Guanchang enema in treatment of DKD can improve total clinical effective rate, reduce urinary protein excretion and protect renal function, and effect in reducing SCr, UAER and 24 h UPr is more obvious than Tangshen prescription alone. The efficacy of Tangshen prescription combined with Guanchang enema for kidney function may be associated with regulation of TGF-β1 and PAI-1 expression in serum.

      摘要:Objective:To explore and analysis the value and significance of the combined application of Danshen injection on vasodilatation and inflammation in patients with diabetes mellitus combined with lower extremity vascular disease. Method:A total of 136 patients with diabetes mellitus combined with lower extremity vascular disease treated in our hospital from January 2015 to December 2016 were selected as clinical research subjects. By the random number table method, the patients were randomly divided into observation group (69 cases) and control group (67 cases). The patients with diabetes mellitus combined with lower extremity vascular disease in control group were treated with α-lipoic acid. The patients with diabetes mellitus combined with lower extremity vascular disease in observation group were treated with Danshen injection in addition to the therapy of control group. The clinical treatment, changes in vasodilation, hemodynamic changes, changes in inflammatory markers, adverse reactions of the patients with diabetes mellitus combined with lower extremity vascular disease in the two groups were compared and analyzed. Result:Compared with control group, the effective rate of clinical treatment in observation group was 50.72% (35/69), and the total effective rate was 95.65% (66/69), indicating a significant increase, while the inefficiency rate was 4.35% (3/69), indicating a significant decrease. There were statistically significant differences (P<0.05). In observation group, flow-mediated dilatation (FMD) was (5.31±0.64)% and ankle brachial index (ABI) was (0.99±0.12), with statistically significant differences (P<0.05); the intravascular diameter was (1.70±0.28) mm, the blood flow velocity was (52.29±6.34)min·s-1, and the blood flow volume was (24.39±3.54) mL·min-1, with statistically significant differences (P<0.05); C-reactive protein (CRP) was (5.46±0.65) mg·L-1, interleukin (IL)-6 was (81.84±9.29) μg·L-1 and tumor necrosis factor (TNF) -α was (11.53±2.26) μg·L-1, with statistically significant differences (P<0.05); total incidence of adverse reactions was 8.70% (6/69), including nausea and vomiting, headbilges and headache, abdominal pain and diarrhea, with statistically significant differences. Conclusion:In the process of the adjuvant treatment for the patients with diabetes mellitus combined with lower extremity vascular disease with Danshen injection, Danshen injection could effectively alleviate the patients' vasodilatation and hemodynamic indicators, and regulate the patient's inflammatory cytokines in an all-round way, so as to enhance the patients' clinical treatment.

      摘要:Objective:To discuss the clinical efficacy of modified Erlong Zuoci Wan combined with acoustic therapy for tinnitus (deficiency of kidney essence), and investigate its effects on superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC). Method:Seventy-nine patients were randomly divided into control group (39 cases) and observation group (40 cases) by random number table. Patients in control group got acoustic therapy. Based on the treatment in control group, patients in observation group additionally received modified Erlong Zuoci Wan. The treatment course was 3 months in both groups. Before and after treatment, scores of tinnitus severity, tinnitus handicap inventory (THI), self-rating anxiety scale (SAS), self-rating scale of sleep (SRSS) and deficiency of kidney essence were graded. Levels of SOD, MDA, GSH-Px and T-AOC were detected both before and after treatment. Result:In rank sum test, the clinical efficacy in observation group was superior to that in control group(Z=1.936,P<0.05). The degree of tinnitus disability in observation group was lighter than that in control group (P<0.05). Scores of tinnitus severity, THI, SAS, SRSS and deficiency of kidney essence symptoms in observation group were lower than those in control group (P<0.01). After treatment. Levels of MDA was lower and levels of SOD, GSH-Px and T-AOC were higher than those in control group (P<0.01). Conclusion:Based on acoustic therapy, modified Erlong Zuoci Wan can relieve tinnitus severity and the degree of tinnitus disability, ameliorate clinical symptoms, improve anxiety and sleeping, and it can also resist oxygen free radical damage, with better clinical efficacy than pure acoustic therapy.

      摘要:Objective:To observe the efficacy of modified Simiao Wan and Shixiaosan combined with Qianlie Beixi capsule in treating type Ⅲ chronic prostatitis with syndrome of damp-heat stagnation, and investigate its effect on levels of Th1/Th2 and cytokines. Method:One hundred and fifty-two type Ⅲ prostatitis patients were selected and randomly divided into control group (76 cases) and observation group (76 cases) by random number table. The patients in control group were given with Qianlie Beixi capsules (6 capsules/time before meals, tid). Based on the treatment in control group, the patients in observation group were additionally treated with modified Simiao pills and Shixiao San (1 dose/day, tid). Both groups were treated for 6 weeks. Then the scores of National Institutes of Health-chronic prostatitis symptom index (NIH-CPSI) and symptoms of damp-heat stagnation syndrome (frequent urination, urine pain, perineum and other local pain, scrotum wet, dripping urine, yellow turbidity, hot and dampness pain) were compared between two groups. The clinical efficacy and syndrome efficacy were analyzed for both groups. Peripheral blood levels of Th1 and Th2 and serum levels of interleukin(IL) -2, IL-4, IL-6, IL-10, tumor necrosis factor (TNF) -α and interferon (IFN) -γ were detected in two groups. Adverse reactions were also monitored in both groups. Result:After treatment, scores of pain discomfort, urinary symptoms, quality of life, NIH-CPSI total score and damp-heat stagnation symptoms in observation group were obviously lower than those in control group (P<0.01). Total clinical efficacy rate was 96.05% in observation group, higher than 84.21% in control group (P<0.05). Total syndrome efficacy rate was 97.37% in observation group, higher than 84.21% in control group (P<0.05). After treatment, peripheral blood levels of Th1 and Th1/Th2 in observation group were lower, while Th2 level was higher than those in control group (P<0.01). Serum levels of IL-2, IL-4, and IL-10 in observation group were significantly higher, while serum levels of IL-6, TNF-α and IFN-γ were lower than those in control group (P<0.01). Adverse reaction rate was 3.95% in control group and 5.26% in observation group, with no significant difference between two groups. Conclusion:Modified Simiaowan and Shixiaosan combined with Qianlie Beixi capsule in treating type Ⅲ chronic prostatitis with syndrome of damp-heat stagnation can improve clinical symptoms and syndrome indexes, increase the efficacy with a good safety, and regulate levels of Thl/Th2 and cytokines to restore balance.

      摘要:Diabetic nephropathy (DN) is one of the most important microvascular complications of diabetes, and the mortality caused by end-stage renal failure is very high. Current studies have discovered many factors affecting the development of DN, and the pathogenesis is complicated. Among them, oxidative stress caused by hyperglycemia, abnormal expression of cytokines, increased renal inflammatory reaction, renal hemodynamic changes and disorders of glycolipid metabolism are the main causes for DN. Western medicine used in clinical practice has a certain effect on the treatment of DN. Its main therapeutic mechanism is to reduce glycolipid, which however cannot fundamentally prevent the occurrence and development of DN. In addition, the treatment of DN is a long process, which requires long-term use of western medicine, and cause toxic side effects and economic burden on patients. In this case, traditional Chinese medicines(TCM) show great advantages in the treatment of multi-factor-induced DN. Many TCM have a variety of pharmacological properties, such as anti-oxidation, immune regulation, cardiovascular nutrition, lipid-lowering and hypoglycemic effects. With respect to the advantages of TCM, there have been many studies on the treatment of DN with TCM, and many drugs with good effects have been discovered. Clinical studies have shown that TCM for promoting blood circulation and removing phlegm can significantly reduce urinary protein, improve clinical symptoms, delay the occurrence and development of DN, and improve the quality of life of patients. To this end, this paper reviews the mechanisms of TCM for promoting blood circulation and removing blood stasis in the treatment of DN, and provides a scientific theoretical basis for the treatment of DN with TCM for promoting blood circulation and removing blood stasis from the perspective of molecular biology.

      摘要:Adriamycin (ADR), an anthracycline drug, is widely used and clearly effective in the treatment of acute lymphoblastic leukemia, lung cancer, breast cancer and other diseases, with a broad anti-tumor spectrum. However, ADR shows extensive and severe toxicity. Especially, the accumulation of cardiac myocytes causes myocardial tissues more vulnerable to ADR, demonstrating an irreversible dose-dependent injury. Thus, the dosage of ADR is enormously limited in clinical applications. Meanwhile, the incidence and mortality of cardiovascular diseases in cancer survivors has been increased. It is believed that adriamycin-induced cardiotoxicity is the result of a variety of factors, such as the mass formation of reactive oxygen free radicals, calcium overload, mitochondrial damage, apoptosis and autophagy. These factors are interrelated and mutually causative, forming a vicious circulating network to jointly promote the development of their cardiotoxicity. So far, no single drug or method has been particularly effective in preventing adriamycin-induced cardiotoxicity, without reducing its anticancer efficacy. Therefore, it is still a severe challenge to find a safer and more effective drug or method to fight against adriamycin-induced cardiotoxicity. Many traditional Chinese medicines(TCM) with the characteristics of multiple targets and multiple effects, can antagonize adriamycin-induced cardiotoxicity by antioxidant stress, scavenging free radicals, regulation of calcium metabolism, protection of mitochondrial damage and antagonism of myocardial cell apoptosis. In recent years, lots of researches on TCM and their effective components in protection and reduction of adriamycin-induced cardiotoxicity have been reported and great progresses have been made for the researches on mechanism of action. This article reviews the research progresses on the application and mechanism of TCM and the effective components in the prevention and treatment of adriamycin-induced cardiotoxicity in the last ten years, in order to provide certain reference for safer and more reasonable clinical use of adriamycin combined with TCM.

      摘要:The emergence of antibiotics has made great contribution to the treatment of infectious diseases and the improvement of human health. However,with the intensive use of antibiotics, especially their abuse, bacterial resistance becomes increasingly serious. The increase and spread of drug-resistant bacteria seriously threaten human health. Bacterial resistance has been a global health problem. On one side, more and more bacteria become resistant to the commonly used antibiotics, and there have been even "super bacteria"; on the other side, it takes a long time to develop new chemically synthetic drugs with antibacterial activity. Chinese herbal medicine has a history of several thousand years in China, with many advantages, such as abundant resources, less adverse effects and rare drug resistance. Therefore, developing new antibacterial Chinese herbal medicine, especially those against bacterial resistance, has increasingly become a research hotspot. The authors made a review of the research progress on antimicrobial resistance of Chinese herbal medicine, including the recent reports of antibacterial activity of Chinese herbal medicine and their mechanisms, the factors affecting antibacterial activity and the prospect in this field, in the hope of providing a reference for further study of exploring traditional Chinese herbal medicines as sources of antimicrobial resistance drugs.

      摘要:Cerebral ischemia reperfusion injury (CIRI) is a multiple-link, multiple-factor and multiple-pathway enzyme cascade reaction. Its occurrence and development are closely related to inflammation, oxidative stress, vascular dysfunction, apoptosis and other mechanisms. At present, there are many studies on the changes of single structure and function of specific neurons or blood brain barrier after CIRI, but a few studies on the overall structure and function of brain tissue. The neurovascular unit contains interactions of blood vessels, neurons, blood brain barriers and glial cells. After CIRI, there are a series of complex pathological changes in neuron degeneration and necrosis, blood brain barrier permeability and microglia proliferation, which eventually causes secondary brain injury. Therefore, in the treatment strategy, the signal connection between neuron-cells and non-neuronal cells was strengthened to regulate the neuro-glial cell-vascular endothelial cells in a two-way manner, promote neuronal survival and improve the repair process of synaptic remodeling and nerve regeneration. This is the material basis to guarantee the normal function and blood flow between neurovascular units, and can be used as a new therapeutic target. The concept of neurovascular unit provides a new direction for the basic research and treatment of ischemic brain injury. Traditional Chinese medicine (TCM) has advantages of multiple components, multiple targets and overall regulation, which generally play a protective role to neurovascular units after injury protection. This article reviews the changes of neurovascular unit structure after CIRI and the mechanism of TCM regulation on it.
