      摘要:The pharmacological mechanisms of traditional Chinese medicine (TCM) are very complicated. At the molecular level, the Chinese medicine emphasizes multi-component and multi-target synergistic effects. However, the composition of the TCM is complex and it is difficult to reveal the action mechanisms of TCM. At present, there are many methods to study the pharmacological mechanisms of TCM, which are roughly divided into three models:chemistry-focused study, target-directed study, and systems-biology-based study. To some extent, these methods may illustrate mechanism of action of TCM. However, there are still some limitations. Network-based analysis is still difficult and deficient, including the biological network is extremely complex, optimization problem for module identification, the lack of dynamic and continuous quantitative researches. To effectively find the relationship among multiple targets, we propose a modular analysis framework, including module identification, optimization, similarity calculation of module structure and functional enrichment analysis. The exploration of modular structure has been proposed as a key factor to understand the complexity of drug-target networks. The method of modular analysis provides a new method for pharmacological study in future.

      摘要:Objective:To analyze the saponins in Yiqi Fumai freeze-dried powder for injection by HPLC-Q-TOF-MS,and provide reference for quantitative study of saponins in the preparation and its pharmacodynamical material basis. Method:The fragmentation patterns of ginsenoside Rg1,Re and Rb1 were analyzed in detail,and the saponins in Yiqi Fumai freeze-dried powder for injection were identified by HPLC-Q-TOF-MS. Result:There were two main pathways for mass spectrometric cleavage of ginsenoside Rg1,Re and Rb1:①loss of all aglycones until forming a stable deprotonated parent nucleus,②aglycones fragmentation and the formation of characteristic fragment ions.On the basis of the fragmentation law of mass spectrometry,21 saponins in Yiqi Fumai freeze-dried powder for injection were identified,in addition to ginsenoside Rg1,Rb1,Re,Rc,Rd,Rf,Rg3 and notoginsenoside R1,the other 13 saponins were first reported in this preparation.At the same time,deoxyschizandrin was initially identified by literature comparison. Conclusion:Most of the saponins with high content in Yiqi Fumai freeze-dried powder for injection are identified by HPLC-Q-TOF-MS,which can provide material foundation for further investigation of this preparation.

      摘要:Objective:To analyze the chemical components of volatile oil in different processed products of Aurantii Fructus by GC-MS and briefly discuss the change mechanism of Aurantii Fructus processed by Lingnan special processing technology. Method:According to different processing methods,different processed products of Aurantii Fructus were prepared.Volatile oil in Aurantii Fructus was extracted by steam distillation,and its components were identified by GC-MS,the amount of these components were determined by peak area normalization method.The components and contents of volatile oil were analyzed by cluster analysis and principal component analysis. Result:Fifty-four components were separated and identified from the raw Aurantii Fructus,forty-four for Aurantii Fructus processed by Lingnan special processing method (first fermentation and then decoction),thirty-two for processed products of stir-frying with bran,thirty-one for the steamed Aurantii Fructus and twenty-five for purely fermented Aurantii Fructus.Among these processed products,the one using Lingnan special processing method contained the most newly-add compounds. Conclusion:Lingnan special processing method is significant for enhancing the quality and clinical efficacy of processed products of Aurantii Fructus,this method is worth further research and promotion.

      摘要:Objective:To evaluate the feasibility of residual SO2 and sulfite-ligustilide used for the safety control of herbal dispensing granules that contained sulfur-fumigated Angelicae Sinensis Radix. Method:Residual SO2 was determined by the method documented in the 2015 edition of Chinese Pharmacopoeia,while sulfite-ligustilide was analyzed semi-quantitatively by HPLC-MS/MS.The mimic Angelicae Sinensis Radix dispensing granules were self-made,and the content transfer rules of residual SO2 and sulfite-ligustilide in sulfur-fumigated Angelicae Sinensis Radix during preparation procedures of dispensing granules,including extraction,concentration and drying,were systematically investigated. Result:Residual SO2 and sulfite-ligustilide could all be detectable in the samples of extract,concentrate and dry extract,their final transfer ratios in dry extract were 8.13% and 60.75%,respectively.In addition,the stability of sulfite-ligustilide in the preparation procedures of the dispensing granules and the specificity,sensitivity of the detection method were all better than residual SO2. Conclusion:As the well-recognized harmfulness of SO2 and potential toxicity of sulfite-ligustilide,it is suggested that sulfite-ligustilide combined with residual SO2 could be used as indicators for the safety control of sulfur-fumigated Angelicae Sinensis Radix dispensing granules.

      摘要:Objective:To study on the powder properties,compressibility and moldability of sugar alcohols,and to provide reference for the application of sugar alcohols in solid preparation. Method:Five kinds of sugar alcohols were chosen,after determining the powder properties,such as water content,particle size and particle size distribution,angle of repose,flowability,bulk density and tap density,the powder was tableted at the same filling height,and the compressibility and moldability were evaluated by the tablet weight difference,ejection force,disintegration time,friability,tensile strength and their change rules as indexes. Result:The water contents of 5 kinds of sugar alcohols were below 0.3%,particle size of erythritol was maximum and particle size distribution was the most uniform,particle size of maltose alcohol was minimum and particle size distribution was the most nonuniform.The Carr index of xylitol was 15.68%,and the others were between 5%-15%,indicating that the 5 kinds of sugar alcohols had good flowability.Tablet weight variation of the 5 kinds of sugar alcohols within the prescribed limits.For the ejection force,sorbitol could meet the needs of production,mannitol and erythritol could basically meet the needs of production.Among 5 kinds of sugar alcohols,the tensile strength of sorbitol was the largest,which showed its moldability was good.The disintegration times of all sugar alcohols under different pressures were within 650 s.The friability of sorbitol at various pressures was <1%,it met the requirements of the 2015 edition of Chinese Pharmacopoeia,but friabilities of the other 4 kinds of sugar alcohols were not in accordance with the requirements of the 2015 edition of Chinese Pharmacopoeia. Conclusion:The regularity of compressibility and moldability of sugar alcohols under different pressures is obtained.Different kinds of sugar alcohols have different advantages in direct compression process,this study can provide a theoretical basis for the application of sugar alcohols in solid preparations.

      摘要:Objective:To investigate the influence of temperature,light and pH on stability of cinobufagin,resibufogenin and total bufotenine in Bufonis Venenum extract. Method:The contents of cinobufagin and resibufogenin were determined by HPLC,the detection wavelength was 296 nm and the mobile phase was acetonitrile-water(58:42).The content of total bufotenine was determined by UV,the detection wavelength was set at 559 nm.The effect of high temperature (60,40℃),strong light irradiation[(4 500±500) Lx] and different pH (40℃,pH of 4.32,4.99,5.73,6.47,6.97,7.75) on stability of cinobufagin,resibufogenin and total bufotenine in Bufonis Venenum extract placed 0,5,10 d was investigated. Result:The degradation of the three indicators was greater than 5% under 60℃ for 10 d.The degradations of cinobufagin and resibufogenin were both <5% under the conditions of 40℃ and (4 500±500) Lx,the degradation of total bufotenine was >5% under the above conditions.Degradation of cinobufagin at pH 5.73 was the slowest,degradation of resibufogenin was the slowest at about pH 6.47,and degradation of total bufotenine at pH 6.47 was the slowest. Conclusion:High temperature and strong light irradiation have small effect on cinobufagin and resibufogenin in Bufonis Venenum extract,but total bufotenine is affected by high temperature and strong light irradiation.The pH is the main factor affecting the stability of three index components in Bufonis Venenum extract,but different pH conditions have different effects on the stability of these three index components.

    • Analysis on L-F and K Genetic Relationship of var. in Guizhou Province

      摘要:Objective:To explore the genetic relationship of the Valeriana officinalis var. latifolia in Guizhou province at trnL-F and matK sequence. Method:The trnL-F and matK sequences of V. officinalis var. latifolia were analyzed. The genetic distance was calculated by Bioedit, Mega 5.0, and then the clustering analysis was conducted to establish the phylogenetic tree. Result:In the different populations of the V. officinalis var. latifolia,the lengths of trnL-F sequence ranged from 947-985 bp, and the guanine and cytosihe mass fraction was between 35.92%-36.55%, including 8 differential sites and 5 variable sites. The genetic distance ranged from 0.000 0-0.005 4.The branch type of phylogeny trees was constructed by ML,NJ,ME and UPGMA methods and showed that samples No.15 and 18 could be distinguished from the most samples. In the different populations of the V. officinalis var. latifolia, the lengths of matK sequence ranged from 1 080-1 088 bp,the guanine and cytosine mass fraction was between 35.19%-36.64%, including 2 differential sites and 1 variable site. The genetic distance ranged from 0.000 0-0.000 9.The branch type of phylogeny trees was constructed by ML,NJ,ME and UPGMA methods, and showed that samples No.2, 5, 6 and 20 were clustered as a class and the remained 19 samples were clustered into another group. Conclusion:The trnL-F and matK sequences of V. officinalis var. latifolia in Guizhou province are highly conservative.

    • Development of EST-SSR Markers and Genetic Background Analysis for in China

      摘要:Objective:To explore the genetic differentiationof Polygonum sibiricum in different areas in China, in order toprovide basis for its population subdivision, evolution, protection and utilization, and quality control. Method:Fiftysix samples were collected from 13 sampling sites in four provinces of China. EST-SSR primers were developed and screened, and the genetic differentiation was evaluated with both EST-SSR and sequence related amplified polymorphism(SRAP) markers. Result:Among 45 pairs of EST-SSR primers, 38 pairs amplified 660 distinguished bands, and 17 pairs amplified 303 polymorphic bands, with a polymorphism rate of 100%. Among 96 pairs of SRAP primers, 67 pairs amplified 1 347 distinguished bands, and 58 pairs amplified 1 147 polymorphic bands, with a polymorphism rate of 100%. Genetic distance and cluster analysis revealed both inter-and inner-population have genetic differentiation. Conclusion:Genetic variation and geographic differentiation areobserved among P. sibiricum populations in China, which were affected by geographic distances, altitudes and possibly mankind activities.

      摘要:Objective:To establish a two-dimensional gel electrophoresis(2-DE) protocol for theproteomic study of Hippocampus trimaculatus, in order to lay the foundation for further research on Hippocampus in the late stage. Method:The methods of protein extraction, lysis, purification, and sample loading quantity were optimized. Result:The optimized protein extraction method was the liquid nitrogen grinding method. The best lysate contained 7 mol·L-1 Urea, 2 mol·L-1 Thiourea, 4% Chaps, 1 mmol·L-1 PMSF,65 mmol·L-1 DTT and 0.2% Bio-Lyte. The best protein purification method was ultrafiltration+TCA/acetone precipitation.The best sample loading quantity was 300 μg.A total of 236 protein spots were detected by image analysis, all of which were distributed within the range between 14.4-97.4 kDa, with unobvious differences. Protein isoelectric pointswere mainly distributed between pH 4-9, among which alkaline protein was the most abundant. Conclusion:The two-dimensional electrophoretic map obtained by the method mentioned above shows clear spots, uniform separation and a good quality. The method is suitable for the proteomic analysis of H. trimaculatus. This study can lay the foundation for further studying the proteomics of H. trimaculatus and screening characteristic proteins.

      摘要:Objective:To study the chemical constituents of Stachys geobombycis. Method:S. geobombycis crushed into coarse powder,extracted by 95% ethanol,concentrated under reduced pressure,and then its ethyl acetate and n-butanol extract was gained by fractional extraction. Chemical constituents were isolated and purified by column chromatography with silica gel,Sephadex LH-20,and semi-preparative HPLC. Their structures were elucidated by spectral data and physicochemical properties. Result:Twelve compounds were obtained and identified as:acteoside (1), isoverbascoside (2),p-hydroxy-benzonic acid (3),latifolicinin C (4),papuline (5),linoleic acid methyl ester (6), α-linoleic acid (7),3,4-dihydroxybenzaldehyde (8),uridine (9),methyl 3,4-dihydroxy-benzoate (10),β-sitosterol (11) and stigmasterol (12). Conclusion:Compounds 5,10 and 11 were isolated from this genus for the first time, compounds 1-4, 6-9 and 12 were isolated from S. geobombycis for the first time.

      摘要:Objective:To establish a new type method for Separation-determination of stachyose in Leonuri Herba and its preparation by ion chromatography,which provides a new idea for the study of stachyose. Method:The CS12A cation exchange column (4 mm×250 mm),CSRS 500 (4 mm) self circulation electric control mode to suppress the background conductance signal of eluent,and the suppression current is 16 mA,6 mmol·L-1 mesylate solution as eluent of isocratic eluent by isocratic elution,the flow rate is 0.8 mL·min-1,the column temperature is 30℃,and the detection method is conductivity detection. Result:According to the above chromatographic conditions,the effective separation was achieved and the rapid detection was achieved in Leonuri Herba of stachydrine.The mass concentration of stachyose is 0.41-10.23 mg,the scope of insider relations is good (r=0.999 9), the detection limit is 0.11 mg·L,the limit of quantitation was 0.41 mg·L,reproducible test results for 7.88-8.38 mg·g-1,RSD was 2.3% (n=6),the average recovery rate of sample recovery test results was 98.74%-101.82%,RSD was 1.2%(n=6),the average content of stachyose in the 8 batches of leonurus japonicus from different habitats was 7.16-15.63 g·L-1,the content of in Guizhou province Xiuwen county up to 15.63 mg·g-1, and the content of Pingyi Liuyu for a minimum of 7.16 mg·g-1. Conclusion:The method has good reproducibility,high accuracy,strong specificity and simple operation in this paper.It can be used for quantitative and qualitative analysis of the stachydrine in Leonuri Herba and its preparations,and also provides a basis for the experimental study on the determination of quaternary ammonium compounds by ion chromatography.

      摘要:Traditional famous prescription Liuwei Dihuang prescription (LWDH) has a good effect in treating type 2 diabetes. α-Glucosidase (α-GC) is the key target of development of type 2 diabetes. In this paper, we analyzed active α-GC inhibitors from LWDH by using virtual screening technique. Method:We obtained structures of 21 known main chemical components and metabolites in LWDH from literatures to compose the molecular ligand library. Ligands were combined with α-GC at terminal N and terminal C by using molecular docking. After docking, the lowest binding energy (LBE) of the complexes was calculated to predict potential active α-GC inhibitors, with LBEs of α-GC inhibitor acarbose (ACR) as the threshold (-38.38, -36.38 J·mol-1 at terminals N and C, respectively). Result:Two components in LWDH, which belong to tetracyclic triterpenoids of Alismatis Rhizoma, had lower LBEs with α-GC than ACR at terminals N and C, respectively, with hydrogen bonds and Van der Waals force as the main acting forces. Conclusion:Virtual screening based on molecular docking could help us to predict active α-GC inhibitors in LWDH, and provide scientific evidence for further studies on the mechanism of LWDH in the treatment of type 2 diabetes.

      摘要:Objective:To analyze the differences of volatile components in various parts of Trifolium pratense. Method:The volatile oils were extracted by the method of headspace solid phase micro-extraction. The chemical components were determined by GC-MS, quantified by the area normalization method, and analyzed by the principal component analysis. Result:Totally 155 volatile oil compounds were identified from stems, leaves and flowers of T. pretense, including 55 hydrocarbons, 29 alcohols, 23 ketones, 13 esters, 21 aldehydes, 2 acids, 3 phenols, and 9 heterocyclic and other compounds. Totally 95 compounds were identified from stems, accounting for 94.15% of volatile components, mainly containing 35 kinds of hydrocarbons (39.14%), 18 alcohols (10.86%), 16 ketones (19.61%), 6 esters (4.06%) and 11 aldehydes (7.48%). Totally 89 compounds were identified from leaves, taking up 95.17% of volatile components, mainly containing 37 kinds of hydrocarbons (41.58%), 16 alcohols (14.88%), 12 ketones (18.18%), 3 esters (9.27%) and 14 aldehydes (4.69%). Totally 78 compounds were identified from flowers, occupying 91.66% of volatile components, mainly containing 28 kinds of hydrocarbons (45.24%), 12 alcohols (4.81%), 10 ketones (11.57%), 8 esters (6.65%) and 11 aldehydes (9.24%). The total peak areas of volatile oil obtained from the 3 samples of stems, leaves and flowers were respectively 0.56×109, 1.14×109 and 0.55×109, with the largest total peak area in volatile oil of leaves. There were 41 constituents in 3 parts in total; stems, leaves and flowers contained 29, 41 and 14 specific components, respectively. The 2 principal components could represent 155 kinds of volatile oil components of the 3 samples. The principal component analysis showed great differences among the samples, with the highest comprehensive score for leaves samples. Conclusion:There are great differences between species and content of volatile oil from stems, leaves and flowers of T. pratense.

      摘要:Objective:To clarify the analgesic effects of Wutoutang (WTT) on neuropathic pain (NP) mice and explore the mechanism of its effect on the descending inhibitory system. Method:ICR mice were randomly divided into the Sham operation group, spinal cord ligation (SNL) group, pregabalin (PGB) group, WTT high, middle and low dose (WTT-H, WTT-M and WTT-L) groups. The L5 spinal nerve was stripped in mice of Sham group, and was stripped and then tightly ligated in other groups. After that, the mice were gavaged with saline,12.60, 6.30 and 3.15 g·kg-1WTT, and 0.025 g·kg-1 prebahrain respectively for 21 days. The analgesic efficacy of WTT was identified by the Von Frey method; the protein expression levels of brain derived neurotrophic factor (BDNF) and 5-hydroxytryptarnjne (5-HT) in key brain regions of descending inhibitory system, the anterior cingulate cortex (ACC)and the rostral ventromedial medulla (RVM) for example, were detected in each group by enzyme-linked immunosorbent assay (ELISA); and the protein expression levels of phosphorylated-extracellular signal-regulated kinase (p-ERK) and postsynaptic density protein-95 (PSD-95) were analyzed by Western blot assay. Result:At day 21 of administration, the mechanical pain threshold value was significantly decreased in model group as compared with the Sham operation group (P<0.01), and the expression levels of 5-HT and BDNF in RVM and ACC were also decreased (P<0.05), while the proteinexpression levels of p-ERK and PSD-95 were elevated(P<0.05). Themechanical pain threshold valuein WTT groups was significant higher than that in SNL group (P<0.01), and both the mentioned 5-HT and BDNF expression levels were increased (P<0.05), while the protein expression levels of p-ERK and PSD-95 were inhibited by WTT. Conclusion:WTT can alleviate NP in mice, and the mechanismmay be associated with regulating the expression levels of 5-HT, BDNF, p-ERK and PSD-95 proteins in RVM and ACC of descending inhibitory system.

      摘要:Objective:To observe the effect of hydroxysafflower yellow A (HSYA) combined with astragaloside-Ⅳ (As-Ⅳ) on the apoptosis of human umbilical vein endothelial cells EA.hy926 induced by human recombinant tumor necrosis factor-α (TNF-α). Method:The effects of different concentrations of TNF-α (0, 10, 20, 50 μg·L-1), HSYA (0, 1×10-6, 5×10-6, 1×10-5, 1×10-4 mol·L-1), As-Ⅳ (0, 1×10-6, 5×10-6, 1×10-5, 1×10-4mol·L-1) on the cell viability of EA.hy926, as well as the effect of HSYA combined with As-Ⅳ(1×10-5 mol·L-1) on the viability of EA.hy926 cells induced by TNF-α (20 μg·L-1) were measured by thiazole blue method(MTT) assay; Western blot assay was used to detect the effect of different concentrations of TNF-α on the expressions of B-cell lymphoma-2(Bcl-2), Bcl-2 associated X protein(Bax), cleaved cysteine asparate protease-3(Caspase-3) and cleaved Caspase-9 proteins, and the effect of HSYA combined with As-Ⅳ in improving expressions of the above proteins. Real-time PCR assay was used to observe the effect of HSYA combined with As-Ⅳ on the expression of Bcl-2 gene induced by TNF-α. Immunofluorescence technique was used to analyze the effect of HSYA combined with As-Ⅳ on the expressions of Bcl-2 and Bax proteins induced by TNF-α. Result:Compared with the blank group, TNF-α could induce the activity of EA.hy926 cells at 20 μg·L-1, up-regulate the expressions of Bax, cleaved Caspase-3 and cleaved Caspase-9, and down-regulate the expression of Bcl-2.Compared with TNF-α group, HSYA combined with As-Ⅳ pretreatment group could decrease the cell viability induced by TNF-α, inhibit the expressions of Bax, cleaved Caspase-3 and cleaved Caspase-9, and increase the expression of Bcl-2 induced by TNF-α. The effect of HSYA combined with As-Ⅳ pretreatment group was better than that of single use of HSYA or As-Ⅳ pretreatment group. Conclusion:HSYA combined with As-Ⅳ has an inhibitory effect on the apoptosis of EA.hy926 cells induced by TNF-α, with a better effect than that of single drug. The mechanism may be related to the down-regulation of expressions of Bax, cleaved Caspase-3 and cleaved Caspase-9 and the up-regulation of Bcl-2 expression.

      摘要:Objective:To investigate the pathogenesis of gastrointestinal dysfunction (GID) with spleen deficiency syndrome and the intervention effect of Si Junzitang. Method:Totally 36 rats were randomly divided into control group, model group, Si Junzitang group and mosapride group. Iodoacetamide gavage+small platform standing+irregular feeding were used to establish the rat model with spleen deficiency and gastrointestinal dysfunction. After modeling, Si Junzitang (6.3 g·kg-1) and mosapride (0.45 mg·kg-1) were given to intervention groups by gavage; subsequently, gastric emptying detection, Western blot, immunohistochemical staining, Real-time PCR and Mg2+-ATPase activity detection were performed to detect the changes in Calmodulin (CaM)-myosin light chain kinase (MLCK) pathway. Result:Compared with normal group, weight and gastric emptying rate were lower (P<0.01), and CaM and MLCK protein and mRNA expressions and Mg2+-ATPase activity of gastric antral smooth muscle were higher in model group (P<0.05, P<0.01). Compared with model group, weight and gastric emptying rate were higher (P<0.05), and CaM and MLCK protein and mRNA expressions and Mg2+-ATPase activity were lower in Si Junzitang group and mosapride group (P<0.05, P<0.01). Conclusion:Rats with spleen deficiency and gastrointestinal dysfunction show CaM-MLCK pathway disorder. Si Junzitang may indirectly improve gastric motility by regulating CaM-MLCK pathway.

      摘要:Objective:To discuss the molecular mechanism of ‘lung disease transferring to large intestine’ and the intervention effect of Maxing Shigan Tang thought the influenza virus infection lung model and the protein expression level of monocyte chemotactic protein-1 (MCP-1) in lung tissue and colon tissue. Method:Nasal inoculation with influenza virus was performed to establish influenza virus-infected animal models. The experiment included the normal control group, the model control group, the Maxing Shigan Tang group and the oseltamivir group. The groups were given the corresponding drugs or saline by gavage for 3 days and 7 days respectively. The mice's body weight, lung index, pathological changes in lung tissue and colon tissue were detected by routine method; MCP-1 protein expressions in lung tissue and colon tissue were detected by Western blot technology and immunohistochemistry. Result:① Compared with the normal control group, the lung index of the model control group increased, with significant differences (P<0.01). ② Compared with the normal control group, the model control group's lung tissue and colon tissue showed serious inflammation; after drug treatment for 3 days and 7 days, Maxing Shigan Tang group showed obvious decrease in pathological damage of lung tissue and colon tissue. ③ Compared with the normal control group, MCP-1 protein expression in lung tissue of the model control group increased, with significant differences (P<0.01); after drug treatment for 3 days and 7 days, Maxing Shigan Tang group showed decrease in MCP-1 protein expression, with significant differences (P<0.01); there was a similar trend in the detection result of Western blot and immunohistochemistry. ④ Compared with the normal control group, MCP-1 protein expression in colon tissue of the model control group increased, with significant differences (P<0.05), after drug treatment for 3 days and 7 days, Maxing Shigan Tang group showed no statistically significant difference in MCP-1 protein expression. ⑤ The lung index inhibiting effect of Maxing Shigan Tang in the influenza virus lung infection model had a positive correlation with the regulating effect in MCP-1 protein expression level of lung tissue and colon tissue. Conclusion:The influenza virus lung infection model has a serious colon tissue pathological injury, and the MCP-1 protein expression levels in lung tissue and colon tissue increased. Maxing Shigan Tang can play a role in anti-flu virus by regulating MCP-1 protein expression.

      摘要:Objective:To investigate the glucose and lipid-lowering effects and mechanisms of Lianmei granule. Method:Experimental rat models of type 2 diabetes were induced by high-fat diets combined with small-dose streptozotocin, and treated with Lianmei granule. Fasting blood glucose (FBG), urine glucose, glycosylated hemoglobin (GHb), total cholesterol (TC), triglyceride (TG), serum insulin (Ins), lactoperoxidase(LPO), superoxide dismutase(SOD) and pancreatic histopathological findings were determined. Result:Compared with control group, blood glucose was significantly decreased by Lianmei granule at low, middle and high doses (P<0.05, P<0.01). At week 9, blood glucose (P<0.01) and urine glucose (P<0.05, P<0.01) were significantly decreased by Lianmei granule at low, middle and high doses. Middle and high doses could notably reduce glycosylated hemoglobin (P<0.01), and the middle dose could lower the insulin resistance index (P<0.05). Lianmei granule at low, mid and high doses could significantly increase the superoxide dismutase activity (P<0.05), and reduce the plasma peroxidative lipid (P<0.05). Pathological results showed that pancreatic tissue structure score, cell vacuolation score of rats in low, middle and high-dose Lianmei granule groups had decreased; the data of islet score and proportions of β-cell score had no significant difference; total score of islet had decreased substantially. Conclusion:Lianmei granule could effectively lower blood glucose, urine glucose, blood fat and glycosylated hemoglobin of diabetic rats by alleviating insulin resistance, elevating insulin susceptibility and resisting oxygen free radicals to achieve the modulating effect on blood glucose and blood lipids.

      摘要:Objective:To investigate the effect of Yangyin Runchang decoction on the expressions of aquaporin 3 (AQP3) and aquaporin 9 (AQP9) in constipated mice. Method:Fifty male ICR mice were randomly divided into normal group, model group, low-dose group, high-dose group and prucalopride group, with 10 mice in each group. The mice were gavaged with compound diphenoxylateat (20 mg·kg-1) for 15 days, then treated with Yangyin Runchang decoction at the doses of 29.6,59.2 g·kg-1 for 15 days. The colon tissues was collected, the locations and semi-quantified expressions of AQP3 and AQP9 were detected by immunohistochemistry, the mRNA and protein expressions of AQP3 and AQP9 were detected by Real-time PCR and Western blot. Result:Compared with the normal group, the fecal water content of the model group decreased significantly (P<0.01), AQP3 expression increased (P<0.01), the mRNA and protein expressions of AQP3 also increased (P<0.05), AQP9 expression decreased, and the mRNA and protein expressions of AQP9 were also decreased, but with no significant difference. Compared with the model group, the fecal water content in treated groups increased (P<0.05), AQP3 expression decreased (P<0.05), and the mRNA and protein expressions of AQP3 were also decreased (P<0.05). The expression of AQP9 in the low-dose group increased (P<0.01), and the mRNA and protein expressions of AQP9 increased (P<0.05), while there was no significant difference in the high-dose group. Conclusion:Yangyin Runchang decoction can increase the water content of feces in constipated mice. Its mechanism may be correlated with the decreased expression of AQP3 and increased expression of AQP9 in colon tissues, but the effect of high dose is not obvious on the contrary.

      摘要:Objective:To investigate the effect of serum containing Shenqi Yiliu prescription on inhibiting cell proliferation and blocking cell cycle of gastric cancer cell lines BGC-823, and explore its possible mechanism. Method:After being treated with 3%, 5% and 10% serum containing Shenqi Yiliu prescription, gastric cancer cell lines BGC-823 were tested by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide for proliferation; flow cytometry was used to detect the cell cycle; Real-time PCR technique was used to detect the mRNA expression of cyclin-dependent kinase inhibitor 1B (CDKN1B) and cyclin-dependent kinase inhibitor 1C (CDKN1C) genes; and Western blot was employed to test the protein expression levels of p27 and p57. Result:After 24, 48, 72 hours treatment with different concentrations of serum containing Shenqi Yiliu prescription, the cell viability was decreased significantly (P<0.01) in a time and dose dependent manner. Flow cytometry analysis showed, when treated by different concentrations of serum containing Shenqi Yiliu prescription for 24h, BGC-823 cells count in S phase was decreased significantly and increased in G0/G1 phase. Real-time PCR results showed, as compared with blank control group and solvent group, CDKN1B and CDKN1C mRNA expression levels were increased in each Shenqi Yiliu prescription group (P<0.05, P<0.01). Western blot results showed that as compared with blank control group, p27 and p57 protein expression levels were increased significantly in each Shenqi Yiliu prescription group (P<0.05,P<0.01). Conclusion:The serum containing Shenqi Yiliu prescription can inhibit the BGC-823 cell proliferation and the mechanism may be associated with regulating CDKN1B, CDKN1C mRNA and protein expression to intervene in the cell cycle.

      摘要:Objective:To investigate the curative effect and the mechanism of Xinmaixian powder in ulcerative colitis(UC) rats by observing its regulatory effect on protein kinase R like endoplasmic reticulum kinase (PERK)/eukaryotic translation initiation factor-2α(eIF-2α)/nuclear factor-κB(NF-κB) signaling pathway. Method:Sixty SD rats were randomly divided into six groups:normal group,model group,mesalazine group,Xinmaixian powder low,middle and high dose groups, with 10 rats in each group.After UC models were successfully prepared by dextran sodium sulfate(DSS), the rats in mesalazine group received mesalazine sustained-release granules suspension in the dose of 0.42 g·kg-1·d-1 for 14 days; the rats in Xinmaixian powder low, middle and high dose groups were treated with Xinmaixian powder suspension in the dose of 1.5, 3, and 6 g·kg-1·d-1 respectively for 14 days.The other groups were treated with same volume of normal saline for 14 days.Finally, the scores of disease activity index (DAI) were evaluated; and expression levels of NF-κB in serum were tested by enzyme-linked immunosorbent assay(ELISA).In addition,protein expression levels of PERK and eIF-2α in colonic tissues were detected by Western blot, while mRNA expression levels of PERK and eIF-2α in colonic tissues were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Result:DA-I scores, expression levels of NF-κB in serum and the protein and mRNA expression levels of PERK and eIF-2α in colonic tissues in the model group were significantly higher than those in normal group(P<0.05). As compared with the model group,Xinmaixian powder groups can reduce DAI scores and expression levels of NF-κB in serum(P<0.05), and the protein and mRNA expression levels of PERK and eIF-2α were also decreased in Xinmaixian powder middle and high dose groups(P<0.05). Conclusion:Xinmaixian powder has a good curative effect on UC rats,and its therapeutic mechanism may be related to inhibiting the activation of PERK/eIF-2α/NF-κB signaling pathway.

      摘要:Objective:To observe the effect of Yifei Zhuji prescription on cells activity, apoptosis, and the protein expression of epidermal growth factor receptor (EGFR) and G protein-coupled receptor 30 (GPR30) of human lung adenocarcinoma A549 cells, investigate its mechanism on inhibiting the activity of lung cancer cells and inducing apoptosis of lung cancer cells. Method:Human lung adenocarcinoma A549 cells were cultured to a logarithmic phase; 2, 1.5 g·L-1 drug culture medium was added in Yifei Zhuji prescription high and low dose groups; 2.5 g·L-1 5-FU drug culture medium was added in blank group; RPMI 1640 culture medium containing 10% fetal bovine serum was added in solvent group. After treatment for 24, 48 h, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to detect the effect of Yifei Zhuji prescription on the activity of human lung adenocarcinoma A549 cells. Acridine orange/ethidium bromide (AO/EB) was used to observe the morphological changes of apoptosis of human lung adenocarcinoma A549 cells. Flow cytometry (FCM) was used to detect the apoptosis rate of A549 cells, and the morphological changes of A549 cells were observed by inverted microscope. In addition, Western blot was used to determine the apoptosis-related protein expression of EGFR and GPR30. Result:Compared with solvent group, both 1.5 and 2 g·L-1 Yifei Zhuji prescription could inhibit the activity of A549 cells(P<0.05), and induce the apoptosis morphological changes of A549 cells; FCM results showed that A549 cells were induced to have late stage apoptosis after Yifei Zhuji prescription treatment for 24 h and 48 h(P<0.05). Western blot results showed that Yifei Zhuji prescription could down-regulate the apoptosis-related protein expression of EGFR and GPR30 to different degrees(P<0.05). Conclusion:Yifei Zhuji prescription could inhibit the activity of A549 cells, induce the apoptosis of A549 cells and induce the apoptosis morphological changes. The mechanism might be associated with down-regulating the protein expression of EGFR and GPR30.

      摘要:Objective:To Study the effect of Guizhou zedoary oil on death factor receptor (FAS) protein expression, Toll like receptor 2 (TLR2), Toll like receptor 4 (TLR4) and interleukin-10 (IL-10), C-Raf, transforming growth factor beta 1 (TGF-β1) in rectal cancer cells SW1463, and investigate the immune effect of Guizhou zedoary oil on tumor cells. Method:Guizhou zedoary oil extracted by steam distillation was divided into 80, 120, 160, 200 mg·L-1 groups, and another blank group was also set up. After 24 h treatment on rectal cancer cell SW1463, enzyme linked immunosorbent assay (ELISA) was used to detect the different doses of Guizhou zedoary oil on the expression of IL-10; and Western blot method was used to detect Fas, TLR2, TLR4, C-Raf and TGF-β1 protein expression levels and investigate its inhibitory effect on rectal cancer cells SW1463 proliferation. Result:ELISA results showed that as compared with blank control group, IL-10 expression was down-regulated with the increase of concentration in Guizhou zedoary oil groups (F=23.11, P<0.01). Western blot results showed that the protein expression levels of TLR2, TLR4 and C-Raf were significantly decreased with the increase of drug concentration (P<0.01), and the protein expression levels of Fas and TGF-β1 were also decreased (P<0.05). Conclusion:The immune mechanism of Guizhou zedoary oil in inhibiting SW1463 cells may be associated with down-regulation of Fas/FasL pathway, down-regulation of Toll like receptor TLR2 and TLR4 protein expression, leading to the down-reulation of relate factor TGF-β1, so that cell apoptosis is achieved to enhance the immune function.

      摘要:Objective:To explore the effect of Wangbipian on the histopathological lesion of knee-joint in experimental animal with osteoarthritis. Method:The therapeutical effect of Wangbipian on the osteoarthritis was detected in the rat osteoarthritis model induced by transection of anterior cruciate ligament on the knee-joint and the rabbit osteoarthritis model induced by injection of papain in knee-joint.SD rats and rabbits were divided into sham operation group,model group,glucosamine hydrochloride group and three Wangbipian-treated groups.For the rabbit osteoarthritis model,the rabbits were intragastric administrated with glucosamine hydrochloride(dose of 0.26 g·kg-1) and Wangbipian at doses of 10.0,5.0,2.5 g·kg-1 for 33 days.For the rat osteoarthritis model,the rats were intragastric administrated with glucosamine hydrochloride(0.36 g·kg-1) and Wangbipian at doses of 14.0,7.0,3.5 g·kg-1 for 31 days.Animals in the sham operation group and model group were administrated with the same volume of water.Range of motion(ROM),circumference and histopathological changes in the knee-joint were observed after the administration.The lesions of cartilago articularis,synovium and the inflammation of soft tissue were comprehensive evaluated. Result:As compared with the model group,administration of Wangbipian could significantly improve the ROM and decrease the circumference of the knee-joint.Histopathological analyses demonstrated that Wangbipian could inhibit the hyperplasisa of synoviocyte and reduce the infiltration of inflammatory cells in both rat and rabbit models. Conclusion:Wangbipian can alleviate the damage of cartilage and synovium of knee-joint in the experimental animal models of osteoarthritis,which suggests that Wangbipian has therapeutic effect on osteoarthritis.

      摘要:Objective:To observe the clinical effects and safety of Qushi Qingchang recipe for treating mild-to-moderate ulcerative colitis with syndrome of dampness-heat in large intestine, and investigate the changes in expression levels of cytokines interleukin (IL)-17, IL-6, IL-10 and transforming growth factor-beta 1 (TGF-β1) in peripheral blood. Method:The 70 patients with mild-to-moderate ulcerative colitis were assigned to experiment group (35 cases) and control group (35 cases) randomly. Patients in experiment group took Qushi Qingchang recipe orally, combined with Chinese medicine by gavage administration. Those in control group took mesalazine enteric-coated tablet. The treatment course was 3 months in both groups. The Truelove and Witts score, sutherland disease activity index and the remission rate were compared between two groups. Meanwhile, expressions of cytokines IL-17, IL-6, IL-10 and TGF-β1 in peripheral blood were compared and the medication safety was monitored. Result:By the end of the treatment, the frequency of hemafecia and mucosanguineous stools in experiment group was less than that in control group (P<0.05); erythrocyte sedimentation rate(ESR) and C-reactive protein(CRP) levels were significantly lower than those in control group (P<0.01), but with no difference in body temperature, pulse, and hemoglobin. There was no difference between two groups in Sutherland DAI scores, and there were no differences in the number of defecation, bloody stool and endoscopic mucosal performance, but the score of physician evaluation index in experiment group was higher than that in control group (P<0.05). The experiment group can improve the clinical symptoms more effectively than control group (P<0.01). The remission rate was 51.43% (18/35) in control group, lower than 74.29% (26/35) in experiment group (P<0.05). The levels of IL-17 and IL-10 in experiment group were significantly lower than those in control group (P<0.01), and the level of TGF-β1 was lower than that of the control group (P<0.05), but there was no significant difference in the level of IL-6. Patients in experiment group had adverse reactions (nausea, perianal pruritus), and 3 patients in control group had adverse reactions (ALT increased to more than 80 U·L-1. Conclusion:Qushi Qingchang recipe combined with herbal enema was effective and safe in treating mild-to-moderate ulcerative colitis with syndrome of dampness-heat in large intestine. Besides, its effect on improving the symptoms was better than that of mesalazine enteric-coated tablet.

      摘要:Objective:To investigate the efficacy of Jiawei Tongguan Tang combined with massage manipulation in treating uroschesis after ischemic stroke and observe its effect on serum levels of substance P (SP), calcitonin-gene related peptide (CGRP) and the antioxygenation. Method:One hundred and four patients with ischemic stroke were selected as object, study and randomly divided into observation group(52 cases)and control group(52 cases)by referring to random number table method. All cases were given with conventional measures according to ‘Chinese Stroke Rehabilitation Treatment Guidelines’. The patients in control group also received bladder function training and routine urinary retention therapy. On basis of the treatment in control group, the patients in observation group were also treated with modified Tongguan Tang combined with massage manipulation. Both groups were treated for 4 weeks. A single urine output, urinary catheter indwelling time, main traditional Chinese medicine symptom scores, and clinical efficacy were compared between two group. Serum levels of SP, CGRP, xanthine oxidoreductase (XOR), malondialdehyde (MDA) and superoxide dismutase (SOD) were detected in both groups. Result:After treatment, the single urine output, maximum urine flow, the detrusor pressure at the maximum urine rate, and maximum urinary bladder capacity in observation group were obviously higher than those in control group, while the catheter indwelling time was significantly shorter than that in the control group (P<0.01). Scores of acraturesis, abdominal distension, fatigue and weakness in observation group were lower than those in the control group after treatment (P<0.01). The total effective rate was 90.2% in the observation group, higher than 72% in control group (P<0.05). After treatment, serum levels of SP and CGRP in observation group were remarkably higher than those in control group (P<0.01). As compared to control group, the decrease in serum levels of XOD and MDA and the increase in SOD in observation group were more obvious, in observation group after treatment (P<0.01). Conclusion:Jiawei Tongguan Tang combined with massage manipulation has a definite curative effect in treating uroschesis after ischemic stroke, which may be related to up-regulating levels of SP and CGRP and enhancing the antioxygenation.

      摘要:Objective:To discuss the clinical efficacy of dialectical therapy of Professor Fu Rumei's Xiao Qinglongtang combined with the therapy of applying ginger at urinary bladder meridian on pulmonary distention with excessive dyspnea with syndrome of external cold and internal fluid, and its effect on serum inflammatory factors. Method:One hundred and sixteen patients were randomly into control group (58 cases) and observation group (58 cases) by random number table. Both groups' patients were treated with infection control, bronchodilator, oxygen therapy, expectorant and cough medicine, and symptomatic supportive treatment. Patients in control group got treatment of ginger acting at urinary bladder meridian. And patients in observation group got Xiao Qinglongtang combined with the therapy of applying ginger at urinary bladder meridian. A course of treatment was 14 days. Before and after treatment, self-assessment test of chronic obstructive pulmonary disease (CAT), excessive dyspnea with syndrome of external cold and internal fluid, six-minutes' walk distance (6 MWD) and BODE were scored. Levels of partial pressure of oxygen (PaO2), partial pressure of carbon dioxide (PaCO2), blood oxygen saturation (SaO2), the 1st-second forced vital capacity (FEV1), percentage of 1st-second forced vital capacity in estimated value (FEV1%) and ratio of FEV1 to forced vital capacity (FEV1/FVC%) were calculated. And levels of procalcitonin (PCT), interleukin-8 (IL-8), tumor necrosis factor-α (TNF-α), serum amyloid A (SAA) were detected. Result:The total effective rate in observation group was 96.55%, which was higher than 84.48% in control group (χ2=4.921, P<0.05). After treatment, scores of CAT, excessive dyspnea with syndrome of external cold and internal fluid, and BODE in observation group were all lower than those in control group (P<0.01), while 6 MWD in observation group was longer than that in control group (P<0.01). Levels of PaO2, SaO2, FEV1, FEV1% and FEV1/FVC% in observation group were higher than those in control group, whereas PaCO2, PCT, SAA, IL-8 and TNF-α in observation group were lower than that in control group (P<0.05, P<0.01). Conclusion:Dialectical therapy of Xiao Qinglongtang combined with the therapy of applying ginger at urinary bladder meridian can ameliorate clinical symptoms, improve pulmonary function and activity, relieve limited degree of airflow and control inflammation, with an obvious clinical effect.

      摘要:Objective:To observe the clinical efficacy of Chaiqin Yiwei decoction on chronic superficial gastritis (CSG) with liver-stomach disharmony and to discusses its mechanism of treatment. Method:The 130 CSG patients with liver-stomach disharmony treated in Department of Spleen and Stomach of the Hebei Province Hospital of Traditional Chinese Medicine(TCM) from January, 2015 to January, 2017 were divided into 2 groups according to the random number table, with 65 cases in each group. Patients in treatment group were given with Chaiqin Yiwei decotion by oral administration, 1 dose/day. The patients in control group were given with conventional western medicine such as hydrotalcite tablets, rabeprazole sodium enteric-coated capsules and mosapride citrate tablets, and all patients were treated for 4 weeks in both groups. The main TCM syndrome scores, peripheral blood T lymphocyte subsets, gastric mucosa cyclooxygenase-2 (COX-2) expression and gastroscope observation before and after treatment were compared in two groups, and the effective rate of gastroscope observation, effective rate of pathology and total clinical effective rate were compared between these two groups. Result:There was not statistically significant difference in the main TCM syndrome score, peripheral blood T lymphocyte subsets and gastric mucosa COX-2 expression before treatment. As compared with before treatment, the main TCM syndrome score was decreased, CD3+, CD4+ and CD4+/CD8+ ratio were increased, and gastric mucosa COX-2 expression was reduced in both groups after treatment (P<0.01). As compared with the control group, the scores in stomach duct pain, abdominal fullness and distention, chest distress and eructation were lower, peripheral blood CD3+, CD4+, CD4+/CD8+ were higher, and gastric mucosa COX-2 expression was lower in treatment group (P<0.01). The effective rates of gastroscope observation, pathology and total clinical effective rate in treatment group were all higher than those of control group (P<0.05). Conclusion:Chaiqin Yiwei decoction can relieve patients' symptoms, with clear efficacy for CSG with liver-stomach disharmony, and its mechanism may be related to regulating the body's immune system and inhibiting inflammation of gastric mucosa.

      摘要:Objective:To study the clinical efficacy of Bushen Tiaochong decoction combined with routine western medicine in treating patients with kidney deficiency and blood stasis type recurrent spontaneous abortion (RSA), and its effect on Th1/Th2 cytokines balance. Method:Totally 122 cases of RSA patients treated in our hospital from March 2015 to August 2016 were selected and divided into two groups according to the group sequential design, with 61 cases in clinical group and 61 in control group. Patients in control group were treated with Dydrogesterone tablets; in addition, those in clinical group were also given Gexia Zhunyu decoction. Then the clinical efficacy, β-human chorionic gonadotropin (β-HCG), progesterone (P), interleukin-2 (IL-2), interleukin-10 (IL-10), IL-2/IL-10 and blood rheology were compared. Result:The total effective rate of the control group was 80.33%, while that of the clinical group was 91.80%. There was a significant difference (P<0.05). Compared with before treatment, β-HCG increased after treatment (P<0.05). Compared with control group, β-HCG and P of clinical group were higher than those of control group, with statistically significant differences (P<0.05). Compared with before treatment, IL-10 of the two groups after treatment increased, while IL-2 and IL-2/IL-10 decreased (P<0.05). Compared with control group, the value of IL-10 in patients of the experimental group was higher after treatment, while IL-2 and IL-2/IL-10 were lower (P<0.05). After treatment, both groups of CPID patients showed significant reduction in blood viscosity at high/low shear rate, erythrocyte aggregation index and hematocrit, with statistically significant differences (P<0.05). Conclusion:Bushen Tiaochong decoction is very effective in treating RSA. It can significantly improve β-HCG and P, balance Th1/Th2 cytokines and improve microcirculation, thus is worth promotion in clinic.

      摘要:Objective:To investigate the effect of nourishing spleen andkidney method combined with prednisone acetate tablets on bone metabolism, urine β2-microglobulin (β2-MG) and α1-microglobulin (α1-MG) of pediatric nephrotic syndrome (NS). Method:The 68 children with NS in our hospital from January 2014 to January 2016 were collected and divided into research group and control group in accordance with the admission number and random number table, 34 cases in each group. The control group was given with basic treatment and prednisone treatment, and the research group additionally received nourishing kidney and spleen method treatment on the basis of treatment in control group. The treatment course was 3 months in both groups. Before and after treatment, the serum albumin, osteocalcin N-terminal molecular fragment(N-MID), bone glaprotein(BGP), porathroid hormone(PTH), bone specific alkaline phosphatase(BALP), and 24 h urinary protein were measured; the efficacy of two groups was observed; the adverse reactions were recorded; and the recurrence was recorded in one year follow up. Result:There was no significant difference in total effective rate between two groups. After treatment, traditional Chinese medicine(TCM) symptom scores were significantly reduced in both groups (P<0.05), and the TCM symptom scores in research group were lower than those in control group(P<0.05). Calcitonin N-MID and BGP levels were significantly increased in research group (P<0.05), and the BALP and PTH levels were decreased obviously (P<0.05), and N-MID, BGP levels in research group were significantly higher, while BALP, PTH levels were significantly lower than those in control group after treatment (P<0.05). The urine β2-MG and α1-MG levels were significantly decreased in both groups (P<0.05), and those levels in research group were higher than those in control group (P<0.05). The serum albumin was increased significantly in both groups (P<0.05), and 24 h protein content was significantly decreased (P<0.05). The serum albumin level in research group was higher than that in control group after treatment (P<0.05), and 24 h protein content was lower than that in control group (P<0.05). There was no significant difference in incidence of adverse reactions between two groups. In 1 year follow-up, the recurrence rate was 17.65% (6/34) in research group, lower than 47.06% (16/34) in the control group (P<0.05). Conclusion:Nourishing spleen and kidney method combined with prednisone acetate tablets is effective in treating pediatric nephrotic syndrome. It can relieve clinical symptoms, improve bone metabolism, regulate urine β2-MG and α1-MG, protect kidney injury and reduce the recurrence rate.

      摘要:Objective:To observe the clinical efficacy of Yupingfeng granules in preventing and treating the recurrent common cold and recuperating the Qi-deficient constitution. Method:A total of 229 subjects with Qi-deficient constitution were recruited in this trial and randomly divide into treatment group (174 cases) and control group (55 cases). The subjects in treatment group orally took Yupingfeng granules, while the subjects in control group took Yupingfeng granules placebo. Each group was treated for 3 months, and the follow-up period was 1 year. Clinical efficacy was observed before treatment, 3, 6, 9, 12 months after treatment. Result:The study was finished well with a comparable baseline. The number of the recurrent common cold in treatment group was lower than that in control group after treatment (P<0.05); the total score of clinical features of the Qi-deficient constitution in treatment group was lower than that in control group (P<0.05), and the reductive ratio of total score in treatment group was 49.82%, and each of the reductive ratio of clinical features in treatment group was higher than that in control group. After 3 treatment courses, the appearance of the first time of colds in treatment group was significantly expanded as compared with control group (P<0.05), and the scores of clinical features of the Qi-deficient constitution in treatment group were significant lower than those in control group at different time points (P<0.01) Conclusion:Yupingfeng granules can effectively reduce the numbers of the recurrent common cold of the Qi-deficient constitution, with some preventive effect on recurrence of colds and recuperate the Qi-deficient constitution.

      摘要:Objective:To study the clinical efficacy of Bushen Tiaochong decoction in the obesity patients with polycystic ovarian syndrome (PCOS) and investigate its effect on endometrial receptivity. Method:The 147 obesity patients with PCOS treated in our hospital from June 2014 to July 2016 were selected in the study and divided into two groups according to the random number table, with 73 cases in treatment group and 74 cases in control group. Patients in control group were treated with ethinylestradiol cyproterone combined with letrozole; on this basis, patients in treatment group received additional Bushen Tiaochong decoction. Both groups were treated for 3 menstrual cycles, then the treatment efficacy, reproductive endocrine hormone levels, body mass index (BMI), fat cell factors and endometrial receptivity were compared. Result:The total effective rate was 90.41% in treatment group, higher than 79.73% in control group (P<0.05). After treatment, the levels of luteotropic hormone (LH), follicle-stimulating hormone (FSH), and LH/FSH were significantly decreased while Estradiol (E2) was increased in both groups, and the effect in treatment group was more obvious (P<0.05). After treatment, te levels of visfatin (VF), leptin (LEP) and BMI were significantly decreased while adiponectin (APN) was increased in both groups, and the effect was more obvious in treatment group (P<0.05). As compared with conditions before treatment, pulsatility index (PI) and resistance index (RI) were decreased in both groups after treatment, while endometrial thickness was increased, and the effect was more obvious in treatment gorup (P<0.05). Conclusion:As a safe and potential method for PCOS, Bushen Tiaochong decoction has better effect on obesity PCOS and can significantly improve reproductive endocrine hormone levels, reduce body mass, and improve endometrial receptivit.

      摘要:Objective:To observe the efficacy of Bushen Qingre Paishi decoction for the post-operative prevention and treatment of the renal deficiency and humid heat type infectious renal calculus. Method:Totally 70 patients were randomly divided into two groups:treatment group and control group, 35 cases in each group. The treatment group orally took Bushen Qingre Paishi decoction that was modified from Bazhengsan and Shenqiwan, while the control group orally took levofloxacin, ceftizoxime and ammonium chloride. Calculus recurrence and changes in negative rate of urine culture, urine pH value, 24 h urine biochemical analysis, serum calcium, serum phosphorus, serum creatinine were observed 4 weeks after treatment. Result:The total effective rate of the treatment group was 88.6%, which was significantly higher than 68.6% of control group (P<0.05). The improvement in 24 h urine citric acid and the reduction in 24 h urine phosphorus of control group were more significant than those of control group after administration (P<0.05), but with no obvious difference in infection control and reduction of urine pH value between two groups. The two groups showed no statistically significant difference in 24 h urine oxalate, 24 h urine calcium and serum calcium, serum phosphorus, serum creatinine after administration. Conclusion:Bushen Qingre Paishi decoction has a satisfactory effect for the post-operative treatment of renal deficiency and humid heat type infectious renal calculus, and can also prevent the recurrence of infectious renal calculus and provide new ideas for treating infectious kidney stones, with a good study prospect.

      摘要:Nineteen components from Qingxin Jieyu formula in blood were imported in BATMAN-TCM platform to make the network pharmacological analysis.Sixty target genes were significantly enriched in 27 Kyoto Encyclopedia of Genes and Genomes(KEGG) signaling pathways,which were related with inhibiting inflammatory reaction,antiplatelet,regulating calcium homeostasis,inhibiting sympathetic nervous system,regulating glucose and lipid metabolism,et al.In addition,the disease items enrichment analysis showed that not only cardiovascular diseases,such as ischemic heart disease,arrhythmia,hypertension and heart failure,but also other common diseases,such as Parkinson's disease,chronic obstructive pulmonary disease,depression and so on,may be treated by Qingxin Jieyu formula.

      摘要:Brown algae is an important source of marine bioactive substances, and fucoidans is a kind of water-soluble heteropolysaccharide mainly in the cell wall matrix of brown algae, which has been proved to have a variety of biological activities, including anti-oxidation, anticoagulant, anti-inflammation, anti-viral, anti-tumor, immune regulation, hyperglycemic, and other activities. However, the structure of algal polysaccharide sulfate in brown algae is complex and diverse, presenting the diversity of physiological activity. Natural fucoidan has a large molecular weight, complex structure, and the research on the structural analysis and structure-effect relationship of diversity is not deep enough, so that the research progress is slow, with low international recognition, and the application and popularization are severely restricted. Therefore, it is an indispensable key step in drug development to deduce the functional structure of fucoidan by integrating the existing research results and systematical research and investigation. Based on these considerations above, the research status of structures, analytical methods of structures, structural modification, and structure-activity relationship were reviewed through CNKI, Pubmed, ScienceDirect databases, and the deep research on structure-activity relationship was prospected, aiming to provide a reference for the further study in functional structures and action mechanism of fucoidan structure, as well as for development of medicine and health care product.

      摘要:Heart failure (HF) is a common clinical cardiovascular syndrome. The rising morbidity and mortality of HF has threatened human health. The causes for HF are complicated. Hemodynamic abnormalities and ventricular overload lead to myocardial hypertrophy, and excessive activation of neuroendocrine cell factor causes ventricular remodeling, all of which can lead to HF symptoms. Because the existing studies on the pathogenesis and treatment in HF don't meet clinical requirements, it urgently needs to further study the pathogenic mechanism and find more effective drugs or therapeutic approaches. HF animal models can simulate disease in human by various means, and provide scientific and theoretical basis for pathogenesis, treatment and prevention of HF. Due to higher requirements of large animal model of HF for experiment equipment and technical operation, as well as a high cost, small animal models of heart failure with a low cost and simple operation have often been used in screening drug efficacy and studying mechanisms. According to the principle, commonly used small animal models of HF can be divided into pressure overload type, heart ischemia type and cardiac inhibition type, and the modeling methods mainly include aorta coarctation, pulmonary hypertension, salt load, coronary artery occlusion, adriamycin, isopropyl adrenaline, pentobarbital sodium and propafenone. Animal models of HF made by various methods have various principle and characteristics. Therefore, purpose of experiment, infrastructure and funds shall be considered in choosing animal model for study.

    • Mechanism of Drugs in Brain Region of PMDD Liver-Qi Stagnation

      摘要:The increasing pressure of life as well as complex and changeable of economic and social environment have led to high incidence of emotional illness, such as social anxiety disorders, depression. Premenstrual syndrome (PMS) and premenstrual dysphoric disorder (PMDD) cause serious physical and psychological distress, disrupt interpersonal communication and reduce quality of life and happiness index. Suicide is also possible in severe cases, so this disease prevention is imminent. Related research proves that many biochemical factors are involved in the PMS(PMDD) disease process, and personality, physical and many other psychological and social factors are also involved although the researches are not deep enough and unclear in mechanism exploration. In recent years, Chinese medicine has achieved remarkable efficacy in the prevention and treatment of emotional diseases, and has been recognized by the academic circles. In traditional Chinese medicine (TCM), stagnation of liver Qi is PMS(PMDD) pathogenesis, caused by liver-Qi depression, characterized by depressive mood, manifested as depression, down in spirits and negative attitude. There is a cohort association with depression. With the gradual deepening of the mechanism research, the brain center is considered to be the ultimate mechanism. The brain is the center of the body, which plays an important role in the occurence, development and treatment of the disease, and can be used for precise positioning of the brain center abnormalities. Therefore, it is very important to identify the exact pathogenesis. At the same time, the mechanisms of the prescriptions widely used in the market are still not clear, and whether they have reached the lesion is still unknown. However, in the context of ‘precision medicine’, accurate alignment of the brain center is achieved through the modern scientific and technological means, with great significance in clarifying the drug mechanism and efficacy, developing new drugs with precise targets, and completely curing the disease.

      摘要:Asperosaponin Ⅵ also known as Akebia saponin D(ASD),it is the main active ingredient of Dipsaci Radix.Studies have shown that ASD has a wide range of pharmacological effects,such as neuroprotection,myocardial protection,prevention of osteoporosis,anti-apoptotic,hepatoprotective effect and lipid-lowering,and has broad prospects for developing.In recent years,with the development of science and technology and the deepening of research,some new pharmacological activities have been discovered.ASD can play its pharmacological effects with multiple targets,its neuroprotection,myocardial protection,hepatoprotective effect and lipid-lowering are related to suppress cell apoptosis,and it can promote tumor cell apoptosis.Although the application of pharmacological effects of ASD has a wide range of prospects,poor bioavailability limits its development,the following study can through the nano-drug delivery system,sustained and controlled release drug delivery system to further improve.Based on the literatures of ASD reported in recent years,this paper summarizes the extraction,determination,pharmacological action and non-clinical pharmacokinetics of ASD,it can provide the basis for the follow-up clinical application of ASD.
