      摘要:ObjectiveTo investigate the effects of Chaihu and Longgu Mulitang (CLMT) on hippocampal neural damage in the mouse model of depression via the extracellular signal-regulated protein kinase (ERK)/cAMP-response element-binding protein (CREB) signaling pathway.MethodsSeventy-eight male C57BL/6 mice were randomly allocated into normal control, model, low/medium/high-dose (2.89, 5.78, and 11.56 g·kg-1, respectively) CLMT, and paroxetine (10 mg·kg-1) groups. A depression model was established by chronic unpredictable mild stress (CUMS) combined with social isolation. Behavioral tests were carried out to evaluate depressive-like behaviors. Hematoxylin-eosin staining and Nissl staining were performed to assess hippocampal morphology and neuronal damage. Immunofluorescence was employed to detect glial fibrillary acidic protein (GFAP) and ionized calcium-binding adapter molecule 1 (Iba1). Real-time PCR was employed to measure the mRNA levels of ERK and CREB. Western blot was employed to determine the expression of ERK/CREB pathway proteins and brain-derived neurotrophic factor (BDNF) in the hippocampal tissue. Molecular Operating Environment (MOE) software was used for molecular docking to evaluate the interactions between CLMT components and target proteins.ResultsCompared with the normal control group, the model group showed decreased sucrose preference (P0.01), increased tail-suspension immobility time (P0.01), decreased activity in the central region of the open field test (P0.01), and decreased activity in the middle and open-arm region of the elevated plus maze test (P0.01). The hippocampal area in the model group showed wrinkled cells and a reduction in the number of cells, neurons with reduced sizes and Nissl bodies, enhanced fluorescence intensity of GFAP and Iba1 (P0.01), and down-regulated expression of phosphorylated (p)-ERK, p-CREB, and BDNF (P0.05, P0.01) and mRNA levels of ERK and CREB (P0.01). Compared with the model group, the CLMT group showed increased body weight (P0.05, P0.01), restored cell morphology, with only a small number of ruptured cells, normal neuronal structure and morphology with obvious nuclei and abundant Nissl bodies, weakened fluorescence intensity of GFAP and Iba1 (P0.05, P0.01), up-regulated mRNA levels of ERK and CREB (P0.05, P0.01) and protein levels of phosphorylated (p)-ERK, p-CREB, and BDNF in the hippocampal tissue (P0.05, P0.01). The results of molecular docking indicated that nine active ingredients in CLMT had good binding affinity with ERK and CREB.ConclusionCLMT may ameliorate the hippocampal nerve injury in the mouse model of depression by regulating the ERK/CREB pathway.

      摘要:ObjectiveTo investigate the mechanism of action of Huangqi Guizhi Wuwutang (HGWT) against cerebral ischemia-reperfusion injury (CIRI) based on bioinformatics and experimental validation.MethodsBiological informatics methods were used to screen for active components of HGWT and their targets. The GEO database was utilized to obtain CIRI-related differentially expressed genes (DEGs), and platforms such as GeneCards were used to identify disease targets. Venn diagram analysis was conducted to identify overlapping targets, followed by protein-protein interaction (PPI), gene ontology (GO), and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, as well as immune infiltration and immune cell differential analysis. Core genes (Hub genes) were screened using LASSO regression and ROC curves, and molecular docking was used to validate the binding efficiency between the active components of the drug and the core targets. A rat CIRI model was established, with rats randomly divided into five groups (n=10): Sham surgery group (Sham), model group (MG), and low-dose (LD,5.3 g·kg-1), medium-dose (MD,10.6 g·kg-1), and high-dose (HD,21.2 g·kg-1) HGWT groups. From 3 days before modeling to 7 days after surgery, oral administration was performed daily: Sham and MG groups received physiological saline, while each drug group received the corresponding dose of HGWT. Hematoxylin-eosin (HE) staining, Nissl staining, and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL staining) were used to assess the repair effects of HGWT on neural damage. Western blot analysis was used to detect B-cell lymphoma-2 protein (Bcl-2), Bcl-2-associated X protein (Bax), signal transducer and activator of transcription 3 (STAT3), phosphorylated STAT3 [p-STAT3 (Tyr705)], protein kinase B1 (Akt1), and phosphorylated Akt1 [p-Akt1 (Ser473)], among other target proteins.ResultsAfter screening, 56 common target points of DEGs-disease-drug were obtained. GO and KEGG analyses indicated that HGWT primarily functions in pathways such as apoptosis, oxidative stress, and inflammatory responses. Immune infiltration analysis revealed a significant association between HGWT's anti-CIRI activity and immune cells such as Th17 cells and myeloid-derived suppressor cells (MDSCs) (P0.01). LASSO-ROC analysis identified Akt1, Caspase-3, glycogen synthase kinase-3β (GSK-3β), and STAT3 as core genes. Molecular docking confirmed that Hub genes exhibit significant binding affinity with the active components of HGWT (binding energy ≤ -5 kJ·mol-1)(1 cal≈4.186 J). Animal experiment results showed that compared with the sham group, the MG group exhibited significant neuronal necrosis, nuclear condensation, and vacuolar degeneration in rat brains, with a significant decrease in Nissl body density (P0.01) and increased neuronal apoptosis in rat brains as indicated by TUNEL staining (P0.01). Compared with the MG, the LD, MD, and HD groups showed reduced neuronal necrosis, nuclear condensation, and vacuolar degeneration in rat brain neurons, increased Nissl body density, and reduced apoptosis (P0.01), with significant differences among the drug groups (P0.01). Western blot results showed that compared with the sham group, the MG group had reduced Bcl-2 and p-Akt1 (P0.01) and increased Bax and p-STAT3 (P0.01). Compared with the MG group, the drug groups showed increased Bcl-2 and p-Akt1 (P0.01) and decreased Bax and p-STAT3 (P0.01). There were no significant changes in total Akt1 and STAT3 protein levels among the groups.ConclusionBased on network pharmacology and experimental verification, HGWT may exert its neuroprotective effects by regulating the phosphorylation levels of Akt1 and STAT3, thereby alleviating cell apoptosis, inflammatory responses, and oxidative stress in rat brain tissue following CIRI. This provides theoretical support for the clinical treatment of CIRI.

      摘要:ObjectiveTo investigate the mechanism of Huangqin Tang (HQT) in regulating metabolic reprogramming during the inflammation-cancer transformation in colitis-associated colorectal cancer (CAC).MethodsCAC mouse model was established using the carcinogen azoxymethane (AOM) combined with the inflammatory agent dextran sulfate sodium (DSS). HQT treatment was adopted. Serum metabolomics analysis was performed at three stages (inflammation, proliferation, and tumor formation) using liquid chromatography-tandem mass spectrometry (LC-MS/MS) untargeted metabolomics coupled with multivariate statistical analysis to explore the mechanism of HQT intervention in metabolism in CAC.ResultsThe results revealed that HQT significantly reversed the disturbance of key metabolites in CAC mice. A total of 52, 67, and 45 differential metabolites were identified in the model group, compared to the normal group, during inflammation, proliferation, and tumor stages, respectively. Lactate, linoleic acid, oleic acid, elaidic acid, and betaine were characteristic metabolites persistently enriched throughout colitis-cancer transformation. Pathway enrichment analysis of differential metabolites showed that linoleic acid metabolism and arachidonic acid metabolism were the most significantly disturbed in CAC pathogenesis. The proliferation stage featured expanded amino acid metabolic networks, while the tumor stage uniquely exhibited two new pathways of nicotinate and nicotinamide metabolism and phosphoinositide metabolism. HQT exerted stage-specific regulatory effects: targeting arachidonic acid metabolism in the inflammation stage, correcting the dysregulation of choline-carnitine metabolism in the proliferation stage, and rescuing nicotinamide and tryptophan metabolic collapse in the tumor stage.ConclusionHQT exerts regulatory effects on metabolic disorders at various stages of the colitis-cancer transformation process, thereby effectively slowing the progression from colitis to cancer. The study also reveals the dynamic metabolic characteristics of colorectal "inflammation-cancer transformation,"providing new insights for research on the targeted mechanisms of traditional Chinese medicine in anti-tumor therapy based on metabolic reprogramming.

      摘要:ObjectiveTo compare the protective effects of water extracts from fresh and dried Dendrobium huoshanense on gastric mucosa in chronic atrophic gastritis (CAG).MethodsMale SD rats (n=72) were randomly divided into 9 groups, with 8 rats in each group, which were normal group, model group, Yangwei Shu (4 g·kg-1) group, low-, medium-, and high-dose fresh D. huoshanense (3.5, 7, and 14 g·kg-1) groups, and low-, medium-, and high-dose dried D. huoshanense (0.7, 1.4, 2.8 g·kg-1) groups. The CAG rat model was successfully established by inducing with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and other factors for a total of 11 weeks. Then, the rats were intervened with fresh and dried D. huoshanense for 4 weeks. The serum and gastric tissues of the rats were collected. The changes in gastric juice secretion volume and gastric acid pH value in each group were observed. The gastric mucosal injury was observed by naked eyes and hematoxylin-eosin(HE) staining. The gastric mucus secretion level was determined by Alcian blue and periodic acid-Schiff staining(AB-PAS) staining. The expression levels of tight junction proteins Occludin and ZO-1 in gastric tissues were determined by immunofluorescence. The expression levels of serum pepsinogen Ⅰ (PG Ⅰ), pepsinogen Ⅱ (PG Ⅱ), gastrin 17 (G-17), interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) were determined by enzyme-linked immunosorbent assay (ELISA). The expression levels of aquaporin 1 (AQP1), aquaporin 3 (AQP3), and aquaporin 4 (AQP4) in gastric tissues were determined by Western blot.ResultsCompared with the normal group, the model group showed an obviously reduced gastric juice secretion volume (P0.05), significantly increased gastric acid pH value (P0.01), gastric mucosa with obvious atrophy, and a significantly reduced gastric mucus secretion volume (P0.01). The expression of Occludin and ZO-1 in the gastric mucosal barrier was significantly decreased (P0.01). The levels of PG Ⅰ and PG Ⅱ in the serum were obviously decreased (P0.05, P0.01), and the levels of G-17, IL-1β, IL-6, and TNF-α were significantly increased (P0.01). The expression level of AQP1 in the gastric tissue was significantly upregulated (P0.01), and the expression levels of AQP3 and AQP4 were significantly downregulated (P0.01). Compared with the model group, each drug administration group could improve the gastric mucosal atrophy of CAG model rats to varying degrees, obviously increase the gastric juice secretion volume of the model rats (P0.05, P0.01), significantly decrease the gastric acid pH value (P0.01), obviously increase the gastric mucus secretion volume (P0.05, P0.01), obviously decrease the expression levels of G-17, IL-6, IL-1β, and TNF-α (P0.05, P0.01), obviously increase the expression levels of Occludin, ZO-1, PG Ⅰ, and PG Ⅱ (P0.05, P0.01), obviously upregulate the expression levels of AQP3 and AQP4 (P0.05, P0.01), and obviously downregulate the expression level of AQP1 (P0.05, P0.01).ConclusionThe water extracts of fresh and dried D. huoshanense can exert therapeutic effects on CAG by improving gastric mucosal injury, reducing inflammation, and regulating water metabolism. Moreover, the dried D. huoshanense has a better effect.

      摘要:ObjectiveTo explore the mechanism of Huoxue Rongluo prescription (HXRLP) in repairing the blood-brain barrier (BBB) after ischemic stroke (IS).MethodsMale C57BL/6 mice were randomly divided into sham operation (Sham) group, cerebral infarction model (MCAO) group, environmental circadian disruption with cerebral infarction model (ECD-MCAO) group, low-, medium-, and high-dose HXRLP (HXRLP-L, M, and H) groups (8.5, 17, 34 g·kg-1·d-1, respectively), and positive drug butylphthalide (NBP) group (0.23 mL·d-1). In the Sham group, only the exposed blood vessels were isolated without suture insertion. In the other groups, the middle cerebral artery occlusion (MCAO) model of mice was prepared. In the ECD-MCAO group, HXRLP groups, and NBP group, the environmental circadian disruption (ECD) model was prepared. The mice in the Sham group, MCAO group, and ECD-MCAO group were given the same volume of soybean oil by gavage, while those in the other groups were given the corresponding drugs by gavage. Samples were collected after 7 consecutive days of administration. The mNSS score was used to evaluate the repair effect of HXRLP on neurological deficits after IS. Hematoxylin-eosin (HE) staining was used to assess the impact of HXRLP on the pathological damage of brain tissue after IS. 2,3,5-Triphenyltetrazolium chloride (TTC) staining and cerebral blood perfusion status were used to evaluate the repair effect of HXRLP on brain tissue damage after IS. Evans blue staining and transmission electron microscopy were used to evaluate the improvement effect of HXRLP on the permeability injury of BBB after IS. Immunofluorescence (IF) staining was used to observe the expression of von Willebrand Factor (vWF), brain and muscle Arnt-like 1 (Bmal1), and Occludin in brain tissue. Western blot was used to detect the protein expression of Bmal1, Occludin, tight junction protein (Claudin-5), vascular endothelial growth factor (VEGF), and angiopoietins(Ang), and related analysis was conducted.ResultsCompared with the Sham group, the MCAO group exhibited significantly aggravated neurological deficits, cerebral infarction volume, brain pathological damage, and BBB leakage (P0.01) and significantly reduced cerebral blood perfusion (P0.01). The expression of Bmal1, vWF, vascular endothelial growth factor A (VEGFA), and Ang in brain tissue was significantly enhanced (P0.01), while the expression of Occludin and Claudin-5 was significantly weakened (P0.01). Compared with the MCAO group, the ECD-MCAO group showed significantly aggravated neurological deficits, cerebral infarction volume, and BBB leakage (P0.01), obviously worsened brain pathological damage (P0.05), significantly reduced cerebral blood perfusion (P0.01), and significantly decreased expression of Bmal1, vWF, VEGFA, Ang, Occludin, and Claudin-5 in brain tissue (P0.01). Compared with the ECD-MCAO group, the HXRLP groups of all doses presented significantly improved neurological deficits, cerebral infarction volume, brain pathological damage, and BBB leakage (P0.01), significantly increased cerebral blood perfusion (P0.01), and enhanced expression levels of Bmal1, vWF, VEGFA, Ang, Occludin, and Claudin-5 in brain tissue (P0.01).ConclusionHXRLP can regulate the clock protein Bmal1 and promote the expression of VEGFA, Ang, Occludin, and Claudin-5, thereby improving BBB damage after IS.

      摘要:ObjectiveTo explore the regulation of hypothalamic-pituitary-gonadal (HPG) axis by modified Erxian decoction in rats with perimenopausal syndrome (PMS) and to further analyze the expression of proteins related to the silent information regulator 1 (SIRT1)/hypothalamic kisspeptin (Kisspeptin)/gonadotropin-releasing hormone (GnRH) signaling pathway in the arcuate nucleus region (ARC) of the hypothalamus, so as to reveal the potential target of action and molecular biological mechanism of modified Erxian decoction for the treatment of perimenopausal syndrome.MethodsAn animal model was established via the incomplete castration method, with successful modeling confirmed by the exfoliated cervical cell smear method. The 48 rats were divided into six groups based on the randomization principle after successful modeling, including a sham operation group, a model group, an estradiol valerate group (0.09 mg∙kg-1∙d-1), high-, medium-, and low-dose modified Erxian decoction groups (7.614, 3.807,1.903 5 g∙kg-1∙d-1), with 8 rats in each group. The estradiol valerate group and the high-, medium- and low-dose modified Erxian decoction groups were continuously administered by gavage for 28 days, and the indicators were detected 24 hours after the last administration. Body weights and uterine indices were measured. The pathological changes of the uterus were observed by hematoxylin-eosin (HE) staining. Enzyme-linked immunosorbent assay (ELISA) was performed to measure the levels of estradiol (E2), follicle-stimulating hormone (FSH), luteinizing hormone (LH), and gonadotropin-releasing hormone (GnRH). Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot were used to determine the expression levels of SIRT1, Kisspeptin, kisspeptin receptor (GPR54), and GnRH in the ARC region of the hypothalamus and gonadotropin-releasing hormone receptor (GnRH-R) in pituitary.ResultsCompared with the sham operation group, rats in the model group had a significantly increased body weight (P0.01), reduced wet weight and index of uterus (P0.01), endometrial thinning or atrophy, glandular atrophy, and a decreasing number of glands. Additionally, serum levels of E2 and the expression of SIRT1 in the ARC region of the hypothalamus significantly decreased (P0.01). Serum levels of FSH, LH, and GnRH, the expression of Kisspeptin, GPR54, and GnRH in the ARC region of the hypothalamus, and GnRH-R in pituitary significantly increased (P0.01). Compared with the model group, the estradiol valerate group and the high-, medium-dose modified Erxian decoction groups had significantly reduced body weight, serum levels of FSH, LH, and GnRH, and expression of Kisspeptin, GPR54, and GnRH in the ARC region of the hypothalamus and GnRH-R in pituitary (P0.05, P0.01) and significantly increased wet weight and index of uterus, serum level of E2, and expression of SIRT1 in the ARC region of the hypothalamus (P0.05, P0.01). In addition, they showed thickened endometrium, increased number of endometrial glands, and improved glandular atrophy.ConclusionModified Erxian decoction regulates the function of the HPG axis through multi-targets, and its mechanism of action may be related to the up-regulation of the expression of SIRT1 in the ARC region of the hypothalamus, the inhibition of the over-activation of the Kisspeptin/GnRH signaling pathway, the regulation of the expression of GnRH-R in the pituitary, the restoration of secretion balance of gonadotropins, and the elevation of the estrogen level. This study provides an experimental basis for the interpretation of the scientific connotation of modified Erxian decoction in the treatment of perimenopausal syndrome and a theoretical reference for the development of a novel therapeutic strategy based on the SIRT1/Kisspeptin/GnRH pathway.

      摘要:ObjectiveTo investigate the intervention effects and mechanisms of the flavonoid hyperoside (Hyp) on lipopolysaccharide (LPS)-induced inflammation in the zebrafish model.MethodsZebrafish larvae were either microinjected with 0.5 g·L-1 LPS or immersed in 1 g·L-1 LPS for the modeling of inflammation. The larvae were then treated with Hyp at 25, 50, and 100 mg·L-1 through immersion for four consecutive days. The inflammatory phenotypes were assessed by analyzing the mortality rate, malformation rate, body length, and yolk sac area ratio. Behavioral tests were conducted to evaluate the inflammatory stress responses, and macrophage migration was observed by fluorescence microscopy. Additionally, the mRNA levels of inflammation-related genes, including interleukin-1β (IL-1β), interleukin-6 (IL-6), chemokine C-C motif ligand 2 (CCL2), chemokine C-X3-C motif receptor 1 (CX3CR1), chemokine C-C motif receptor 2 (CCR2), and genes associated with the Toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)/nuclear factor-kappa B (NF-κB) signaling pathway, were measured by Real-time quantitative polymerase chain reaction(Real-time PCR).ResultsCompared with the pure water injection group, the model group exhibited increased mortality, malformation rates and yolk sac area ratio (P0.01), reduced body length (P0.01), increased total swimming distance and high-speed swimming duration (P0.01), and up-regulated mRNA levels of TLR4, MyD88, NF-κB, IL-1β, IL-6, CCL2, CX3CR1, and CCR2 (P0.01). Hyp at low, medium and high doses, as well as aspirin, reduced the mortality and malformation rates (P0.05,P0.01), increased the body length (P0.05,P0.01), decreased the yolk sac area ratio (P0.01), reduced the high-speed swimming duration (P0.01), and down-regulated the mRNA levels of TLR4, MyD88, NF-κB, IL-1β, IL-6, CCL2, CX3CR1, and CCR2 (P0.05,P0.01) compared with the model group.ConclusionHyp may modulate the TLR4/MyD88/NF-κB pathway to ameliorate inflammatory phenotypes and alleviate stress conditions in zebrafish, thereby exerting the anti-inflammatory effect.

      摘要:ObjectiveTo evaluate the antipyretic effect of the Qingwen Baiduling prescription in a dry yeast-induced rat fever model and to investigate its antipyretic mechanism, providing a theoretical basis for its clinical application.MethodsSPF-grade male SD rats were randomly assigned to six groups (n=6 per group): control, model, aspirin (20 mg·kg-1), and high-, medium-, and low-dose Qingwen Baiduling prescription groups (14.40, 7.20, 3.60 g·kg-1). The fever model was established by subcutaneous injection of dry yeast suspension on the back. After model induction, body temperature was recorded every 1 hour. Drugs were administered at 6 hours after modeling, and body temperature was continuously recorded hourly for 12 hours. Record the body temperature of rats to observe the trend of changes in body temperature. Serum interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) levels, as well as hypothalamic prostaglandin E2 (PGE2) and cyclic adenosine monophosphate (cAMP), were measured using enzyme-linked immunosorbent assay (ELISA). Hypothalamic tissue morphology was examined by hematoxylin and eosin (HE) staining. Western blot was used to detect hypothalamic expression of Toll-like receptor 4 (TLR4), myeloid differentiation factor 88 (MyD88), nuclear factor-κB p65 (NF-κB p65), inhibitor κBα (IκBα), phosphorylated IκBα (p-IκBα), IκB kinase α (IKKα), IKKβ, phosphorylated IKKα/β (p-IKKα/β), and cyclooxygenase-2 (COX2).ResultsCompared with the control group, the model group showed a significant increase in body temperature 6 hours after modeling (P0.01), confirming successful fever induction. Serum IL-6, IL-1β, TNF-α, and hypothalamic cAMP and PGE2 levels were significantly elevated (P0.01). Hypothalamic neurons exhibited irregular morphology and disordered distribution, accompanied by inflammatory cell infiltration and microglial aggregation. Expression levels of TLR4/NF-κB pathway-related proteins and phosphorylated proteins were significantly increased (P0.05, P0.01). Compared with the model group, 2-3 hours after administration, all Qingwen Baiduling prescription dose groups significantly reduced body temperature (P0.01). All dose groups significantly decreased serum IL-6, IL-1β, TNF-α, and hypothalamic cAMP and PGE2 levels (P0.05, P0.01). Neuronal morphology was markedly improved in the high- and medium-dose groups, with narrowed intercellular spaces and reduced inflammatory infiltration. The prescription effectively inhibited hypothalamic expression of TLR4, MyD88, NF-κB p65, p-IκBα, p-IKKα/β, and COX2 proteins (P0.05, P0.01).ConclusionQingwen Baiduling prescription effectively reduces body temperature in rats by mitigating the further effects of inflammatory cytokines on the hypothalamic thermoregulatory center through the blood-brain barrier. Its antipyretic mechanism may be related to inhibition of NF-κB pathway activation.

      摘要:ObjectiveTo investigate the effects and mechanism of the combination of arsenic trioxide (ATO) and its dimethylarsinic acid (DMAV) metabolite on apoptosis of human acute promyelocytic leukemia NB4 cells by affecting the balance of metabolic reaction.MethodsThe rats were injected with the same amount of sterile normal saline, ATO(1.6 mg·kg-1), and DMAV(4, 8, 16, and 32 mg·kg-1) combined with ATO(1.6 mg·kg-1), respectively, to obtain the corresponding drug-containing serum. The effect of drug-containing serum on the proliferation of NB4 cells was detected by the cell counting kit-8 (CCK-8 )method. Apoptosis was detected by flow cytometry with Annexin-V-FITC/PI double staining. Intracellular reactive oxygen species (ROS) accumulation was detected by flow cytometry using fluorescent probe DCFH-DA. The changes of mitochondrial membrane potential (Δψm) were detected by the fluorescence probe JC-1. Western blot detected the expression of apoptosis-related proteins, namely B-cell lymphoma-2(Bcl-2)-associated X protein(Bax)/Bcl-2, Cytochrome C, cleaved Caspase-9, and cleaved Caspase-3, as well as c-Jun N-terminal kinase(JNK) phosphorylation levels.Results①The Combination of the two drugs had a higher proliferation inhibition rate and more apoptosis than ATO alone. ②The combination of two drugs can significantly increase the production of ROS compared with any single treatment group. ③The Δψm was significantly reduced in the two-drug combination group compared with any single treatment group. ④Compared with either group, the combination group significantly released Cytochrome C, significantly down-regulated the expression of Bcl-2, and up-regulated the expression of Bax, cleaved Caspase-3, and cleaved Caspase-9. ⑤The phosphorylation level of JNK was significantly up-regulated in the two-drug combination group compared with any single treatment group.ConclusionThe combination of DMAV and ATO may synergistically induce mitochondrial apoptosis through ROS-mediated oxidative stress, triggering Δψm dissipation and Cytochrome C release. By activating Caspase-9/Caspase-3 and the phosphorylation level of JNK, the Bcl-2 family protein (Bax/Bcl-2) was regulated to promote the apoptosis of NB4 cells.

      摘要:ObjectiveBased on the AMP-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR) signaling pathway, this study aimed to observe the effect of the Huazhuo Jiedu prescription on renal fibrosis in 5/6 nephrectomy rats and explore its underlying mechanism.MethodsA total of 67 SPF-grade male SD rats were used, of which 11 were randomly selected as the normal group. A chronic renal failure (CRF) model was established using 5/6 nephrectomy. The successfully modeled rats were randomly assigned to the model group, losartan potassium group (4.5 mg·kg-1), and low- (1.175 g·kg-1), medium- (2.35 g·kg-1) and high-dose (4.7 g·kg-1) Huazhuo Jiedu prescription groups, with 9 rats per group. Each group received an equivalent volume of saline or the corresponding concentration of Huazhuo Jiedu prescription by gavage once daily for 8 weeks. Hematoxylin-eosin (HE) and Masson staining were used to observe renal tissue pathological changes. Transmission electron microscopy examined renal ultrastructure. Immunohistochemistry (IHC) detected expressions of α-smooth muscle actin (α-SMA) and transforming growth factor-β1 (TGF-β1). Western blot analyzed expression levels of microtubule-associated protein Ⅰ light chain 3Ⅱ (LC3Ⅱ), Beclin1, p62, AMPK, phosphorylated AMPK (p-AMPK), mTOR, and phosphorylated mTOR (p-mTOR).ResultsCompared with the normal group, the model group exhibited glomerular shrinkage, mesangial and interstitial thickening, and tubular vacuolar degeneration, with no evident autophagosomes or autophagolysosome structures. Expression levels of α-SMA and TGF-β1 were significantly increased (P0.01), while p-AMPK/AMPK, Beclin1, and LC3Ⅱ were significantly decreased (P0.01), and p-mTOR/mTOR and p62 were significantly increased (P0.01). Compared with the model group, the medium- and high-dose Huazhuo Jiedu prescription groups and the losartan potassium group showed varying degrees of pathological improvement. Autophagosomes with double- or multiple-layer membranes and autophagolysosomes with monolayer membranes containing undegraded organelles were observed. Renal α-SMA and TGF-β1 protein expression levels were markedly reduced (P0.05, P0.01), p-mTOR/mTOR and p62 were significantly decreased (P0.05, P0.01), and p-AMPK/AMPK, Beclin1, and LC3Ⅱ expression levels were significantly increased (P0.05, P0.01).ConclusionHuazhuo Jiedu prescription may improve renal fibrosis in 5/6 nephrectomy rats by regulating the AMPK/mTOR signaling pathway and enhancing autophagy.

      摘要:ObjectiveTo observe the efficacy of Sangmei Pingfeng granules combined with western medicine in the treatment of patients with allergic asthma in the chronic persistent stage, diagnosed with lung-spleen Qi deficiency syndrome in traditional Chinese medicine (TCM).MethodsA retrospective analysis was conducted on 420 patients with allergic asthma in the chronic persistent stage and TCM-diagnosed lung-spleen Qi deficiency syndrome, treated at the Third Affiliated Hospital of Beijing University of Chinese Medicine from January 2017 to September 2024. Patients were divided into an exposed group (320 cases) and a non-exposed group (100 cases) based on whether they used Sangmei Pingfeng granules. The non-exposed group received conventional western medicine treatment, while the exposed group took Sangmei Pingfeng Granules in addition to conventional western medicine. The following indicators were observed: recurrence rate per 100 patients per year, pulmonary function tests, fractional exhaled nitric oxide (FeNO) and fractional nasal nitric oxide (FnNO) tests, serum total IgE (TIgE) detection, TCM syndrome score, and changes in asthma control test (ACT) scores between the two groups.ResultsThe annual recurrence rate per 100 people was 8.4 times/100 person-years in the exposed group, lower than the 13 times/100 person-years in the unexposed group. After treatment, the exposed group showed increases in forced vital capacity percentage predicted (FVC%pred), forced expiratory volume in one second percentage predicted (FEV1%pred), the ratio of forced expiratory volume in one second to forced vital capacity (FEV1/FVC), and peak expiratory flow percentage predicted (PEF%pred) (P0.05). Lung function indicators in the non-exposed group showed no statistically significant differences after treatment. Residual volume as a percentage of total lung capacity (RV%TLC) showed no statistically significant difference after treatment in either group. Compared with the non-exposed group, the exposed group had increased FVC%pred, FEV1%pred, FEV1/FVC, and PEF%pred, without statistically significant differences. After treatment, fractional exhaled nitric oxide (FeNO), fractional nasal nitric oxide (FnNO), and total immunoglobulin E (TIgE) decreased in the exposed group. FeNO decreased in the non-exposed group (P0.05). FnNO decreased, and TIgE increased in the non-exposed group, without statistically significant differences. After treatment, compared with the non-exposed group, the treatment group had decreased FeNO (P0.05). After treatment, traditional Chinese medicine (TCM) syndrome scores decreased, and asthma control test (ACT) scores increased in both groups (P0.01). After treatment, compared with the non-exposed group, the exposed group had decreased TCM syndrome scores and increased ACT scores (P0.01).ConclusionCombining Sangmei Pingfeng granules with Western medicine significantly reduces recurrence rates, enhances clinical efficacy, improves lung function, lowers serum TIgE and FeNO levels, and reduces reliance on Western medications in patients with allergic asthma. This integrated approach is worthy of clinical promotion and application.

      摘要:ObjectiveTo analyze the correlation between the clinical symptoms, signs, syndrome characteristics and laboratory indicators of cough variant asthma (CVA) and deepen the understanding of the treatment of this disease based on the theory of "Fu Feng".MethodsAn observational study was conducted. A total of 207 CVA patients who visited the respiratory department of the Third Affiliated Hospital of Beijing University of Chinese Medicine from September 2022 to November 2023 were included. The information from the four diagnostic methods and the laboratory test results of patients were collected. Factor analysis was conducted on the information obtained through the four diagnostic methods in TCM, and the nature and location of CVA were extracted. Sample cluster analysis (Q clustering) and the K-means method were used for data clustering analysis to determine the syndrome types of CVA and analyze the syndrome characteristics and differences in laboratory indicators among different syndrome types.ResultsThe main symptom of CVA patients was cough, accompanied by symptoms such as itchy throat, foreign body sensation in the throat, dry throat, shortness of breath, dry mouth, chest tightness, hoarseness, bitter mouth, poor appetite, and skin itching. Factor analysis showed that the disease was located in the lung, involving the liver, spleen, and kidney. The pathological factors involved Yin deficiency, Yang deficiency, wind factor, dampness factor, Yin factor, and Qi stagnation. Cluster analysis revealed four syndrome types: Fengfu Yinshang syndrome, Shixie Neiyun syndrome, Tanyin Zufei syndrome, and Ganhuo Fanfei syndrome. Fengfu Yinshang syndrome accounted for the highest proportion, followed by Tanyin Zufei syndrome. There were no significant differences in eosinophil count and percentage, fractional nasal nitric oxide (FnNO) level, and pulmonary function indexes among the four syndromes. The levels of serum total IgE and fractional exhaled nitric oxide (FeNO) in patients with Fengfu Yinshang syndrome were significantly higher than those in patients with Shixie Neiyun syndrome and Tanyin Zufeisyndrome.ConclusionCough is the main symptom of CVA, accompanied by pharyngeal itching, foreign body sensation in the throat, dry throat, shortness of breath, dry mouth, and allergic manifestations. The disease involves the lung, liver, spleen, and kidneys. The essence of the pathogenesis lies in a latent dormant pathogen and a disorder of the pivot mechanism. The four common syndrome types are Fengfu Yinshang syndrome, Shixie Neiyun syndrome, Tanyin Zufei syndrome, and Ganhuo Fanfei syndrome. The TCM syndrome types are correlated with laboratory indexes. The serum total IgE and FeNO of patients with Fengfu Yinshang syndrome are worse.

      摘要:ObjectiveTo observe the regulation effect of Sangmei Zhike granules on airway inflammation and cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA)-cAMP response element binding protein (CREB) pathway in cough variant asthma (CVA) rats with Fengfu Yinshang syndrome.MethodsEight-week-old male rats were randomly divided into a blank group, model group, western medicine group, and traditional Chinese medicine (TCM) group. Except for the blank group, Fengfu Yinshang models of CVA were established in the other groups. On the 16th day of the experiment, the western medicine group was treated with budesonide aerosol inhalation (dosage of 0.5 g·L-1), and the TCM group was given Sangmei Zhike granules by gavage (dosage of 4.19 g·kg-1·d-1), with each group treated once daily for 14 days. On the 30th day of the experiment, lung function parameters were determined. Whole blood inflammatory cell detection, hematoxylin-eosin (HE) staining of lung tissue, and immunohistochemistry were used to detect the expression of CREB, phosphorylated CREB (pCREB), and phosphorylated vasodilator-stimulated phosphoprotein (pVASP) proteins in rat lung tissue, and Reverse transcription quantitative polymerase chain reaction (Real-time PCR) was used to determine the protein expressions of cAMP, CREB, and PKA in rat lung tissue. Western blot (WB) was used to detect the expression of PKA, VASP, p-VASP, CREB, and p-CREB proteins.ResultsCompared with the model group, the average dynamic lung compliance (CDynaverage) in the TCM group increased (P0.05). Compared with the model group, the TCM group showed a decrease in absolute and percentage values of neutrophils (NEUT%), absolute values of lymphocytes (LYMPH), absolute and percentage values of basophils (BASO%), absolute values of monocytes (MONO), absolute and percentage values of eosinophils (EOS%), and white blood cells (WBC) (P0.05). Compared with the western medicine group, the TCM group showed a decrease in MONO, EOS%, and LYMPH% and an increase in MONO% (P0.05). ELISA results showed that compared with the model group, the TCM group had increased interferon-gamma (IFN-γ) and decreased interleukins (IL)-4, IL-5, IL-13, and total serum IgE, while the Western medicine group had increased IFN-γ and decreased IL-5, IL-13, and total serum IgE (P0.05). Compared with the western medicine group, the TCM group had decreased IL-5 (P0.05). HE staining results of rat lung tissue showed that the ciliated epithelium of the D bronchi in the TCM group recovered well. Mucous edema secretion decreased, and a small number of bronchiolar epithelial cells were shed, with reduced infiltration of inflammatory cells and the degree of epithelial injury. Immunohistochemical (IHC) results showed that compared with the model group, positive expressions of CREB, pCREB, and pVASP (brownish-yellow) were significantly increased in the TCM and western medicine groups. Real-time PCR results showed that compared with the model group, the relative expression levels of cAMP, CREB, and PKA in the TCM group were significantly increased (P0.05), and compared with the western medicine group, the TCM group also showed significantly increased relative expression levels of cAMP, CREB, and PKA (P0.05). WB results showed that compared with the model group, the expressions of PKA, VASP, p-VASP, CREB, p-CREB, and p-CREB/CREB proteins were increased in the TCM group, while p-VASP/VASP protein expression decreased (P0.05). Compared with the western medicine group, the TCM group had increased expressions of PKA, VASP, and p-CREB proteins and decreased p-VASP/VASP expression (P0.05).ConclusionSangmei Zhike granules have the effect of improving lung function and inhibiting airway inflammation in rats with CVA due to Fengfu Yinshang syndrome, possibly related to the activation of the cAMP/PKA/CREB pathway.

      摘要:Cough variant asthma (CVA) is a special type of asthma and the most common cause of chronic cough in China. It has a relatively high incidence, seriously affects patients' quality of life, and in some cases may even progress to typical asthma, posing an important challenge in the field of health economics. At present, first-line treatment in Western medicine mainly consists of inhaled corticosteroids combined with bronchodilators. However, there remain many problems such as poor efficacy in some patients, frequent relapse after drug withdrawal, and poor compliance, making it urgent to improve treatment strategies. In-depth research on the pathogenesis is helpful for a comprehensive understanding and prevention of CVA. The pathogenesis of CVA is complex and involves multiple pathophysiological links. While similar to typical asthma, it still presents some differences. Traditional Chinese medicine (TCM) has unique advantages in the treatment of CVA. Guided by the theory of syndrome differentiation and treatment, TCM can not only improve the clinical symptoms of CVA but also prevent recurrence. In recent years, with the continuous deepening of research on the pathogenesis of CVA and on its treatment with TCM based on syndrome differentiation, related academic achievements have been updated year by year. A review of literature in China and abroad shows that the current pathogenesis of CVA can be summarized into six aspects: airway inflammation, airway remodeling, airway hyperresponsiveness, cough hypersensitivity, genetic factors, and imbalance of intestinal flora. TCM often differentiates and treats CVA from four perspectives: cause, disease location, disease nature, and disease tendency. This paper systematically summarizes the progress in the study of CVA pathogenesis, comprehensively collates the experience and clinical research evidence on its treatment with TCM based on syndrome differentiation, and discusses the problems existing in current research. On this basis, it puts forward suggestions and prospects for future research and development of TCM in the treatment of CVA, with a view to providing a theoretical basis and therapeutic approaches for its prevention and treatment.

      摘要:ObjectiveTo investigate the therapeutic effects of Chaipo decoction on bronchial asthma in rats and its regulatory effects on the nucleotide-binding oligomerization domain-like receptor pyrin domain-containing protein 3 (NLRP3)/cysteinyl aspartate-specific protease-1 (Caspase-1)/Gasdermin D (GSDMD) pathway, aiming to elucidate its mechanism in ameliorating pyroptosis.MethodsSixty male Sprague-Dawley (SD) rats were randomly divided into six groups (n=10 per group): normal control, asthma model, Chaipo decoction low-dose (5.0 g·kg-1), medium-dose (10.0 g·kg-1), high-dose (20.0 g·kg-1), and dexamethasone (1.0 mg·kg-1). The asthma model was established in all groups except the normal control group via ovalbumin (OVA) sensitization and challenge. Rats in the Chaipo decoction groups received intragastric administration of Chaipo decoction at the corresponding doses, while the dexamethasone group was treated with dexamethasone. The normal and model groups were administered equivalent volumes of saline. After 14 days of intervention, asthma symptom scores were assessed. Dynamic lung compliance (Cdyn), lung resistance (RL), and functional residual capacity (FRC) were measured using a small animal pulmonary function testing system. Lung tissue pathology was evaluated by hematoxylin-eosin (HE), Masson's trichrome, and periodic acid-Schiff (PAS) staining. Levels of interleukin (IL)-6, IL-1β, and IL-18 in bronchoalveolar lavage fluid (BALF) were determined by enzyme-linked immunosorbent assay (ELISA). Expression of NLRP3 and apoptosis-associated speck-like protein (ASC) in lung tissues was assessed by immunohistochemistry (IHC). Protein levels of NLRP3, Caspase-1, GSDMD, and other pyroptosis-related proteins were measured by Western blot.ResultsCompared with the normal group, the model group exhibited significantly increased asthma symptom scores, inflammatory scores, collagen deposition, PAS scores, RL, FRC, levels of IL-6, IL-1β, and IL-18 in BALF, and expression levels of NLRP3, ASC, and other pyroptosis-related proteins in lung tissue (P0.01), while Cdyn was significantly decreased (P0.01). Compared with the model group, all doses of Chaipo decoction markedly improved asthma symptoms, with significantly reduced symptom scores (P0.05, P0.01). Pulmonary function analysis showed that medium and high doses of Chaipo decoction significantly increased Cdyn (P0.05, P0.01) and decreased RL and FRC (P0.05, P0.01). Histopathological evaluation indicated that high-dose Chaipo decoction significantly reduced inflammatory scores, collagen deposition, and goblet cell hyperplasia in lung tissue (P0.05, P0.01). ELISA results showed that all doses of Chaipo decoction significantly decreased IL-6, IL-1β, and IL-18 levels in BALF (P0.05, P0.01). IHC and Western blot analyses demonstrated that medium and high doses of Chaipo decoction markedly downregulated NLRP3, ASC, and other pyroptosis-related proteins in lung tissue (P0.05, P0.01).ConclusionChaipo decoction effectively improves pulmonary function and pathological damage in asthmatic rats, potentially by inhibiting the NLRP3/Caspase-1/GSDMD pathway and reducing pyroptosis.

      摘要:ObjectiveTo explore the main mechanism of self-precipitation formed during the decoction of Sinitang(SNT), and to provide a research basis for exploring the differences in the toxic and effective components of this compound.MethodsThe average precipitation yields of SNT, Glycyrrhizae Radix et Rhizoma(GRR)-Aconiti Lateralis Radix Praeparata(ALRP) decoction(GF), ALRP-Zingiberis Rhizoma(ZR) decoction(FJ), GRR-ZR decoction(GJD), ALRP decoction(FZ), ZR decoction(GJ) and GRR decoction(GC) were determined. The four main self-precipitation samples of SNT, GF, FZ and GC were physically characterized by particle size, scanning electron microscopy(SEM), pH, total dissolved solids(TDS), conductivity, and Fourier transform infrared spectroscopy(FT-IR) analysis. The chemical compositions of SNT decoction and its different phases was identified by ultra-performance liquid chromatography-quadrupole-electrostatic field orbitrap high-resolution mass spectrometry(UPLC-Q-Exactive Orbitrap-MS) for SNT, SNT self-precipitation and SNT supernatant, and the contents of its main toxic and effective components were determined by high performance liquid chromatography(HPLC).ResultsPrecipitation yield results of the 7 samples of SNT decoction and single decoction showed that SNT had the highest self-precipitation yield. The formation of SNT self-precipitation was mainly related to the reaction between ALRP and GRR components to form complexes, and FT-IR showed that GRR had the greatest influence on the formation of self-precipitation. A total of 110 components were identified in the SNT decoction, including 100 components in the SNT self-precipitation and 106 components in the SNT supernatant. And quantitative results of the main toxic and effective components revealed that the reaction between ALRP and GRR components formed complexes, resulting in the following content hierarchy for free components:SNT decoctionsupernatantself-precipitation, these components included free liquiritin, benzoylmesaconine, benzoylaconitine, benzoylhypacoitine, liquiritigenin, aconitine, hypoaconitine, isoliquiritigenin and ammonium glycyrrhizinate.ConclusionSNT exhibits spontaneous precipitation during compound decoction, with GRR exerting the greatest influence on its formation. This suggests GRR plays a significant role in the detoxification of SNT. The differences in the self-precipitated toxic-effective components of SNT compound decoction primarily manifest as changes in component content, reflecting the characteristics of SNT "deposition in vitro and sustained release in vivo" and the importance of "administered at draught" in the clinical application of SNT.

      摘要:Xiao Chaihutang,an ancient classic prescription,originated from the Treatise on Febrile Diseases and Miscellaneous Diseases (hereinafter referred to as Treatise on Febrile Diseases) written by ZHANG Zhongjing in the Eastern Han Dynasty. It is a representative prescription for reconciling Shaoyang. It can strengthening the body resistance to eliminate pathogenic factors,clear away the evil heat,and harmonize the stomach Qi and can be used to treat various diseases caused by insufficient vital energy,lingering evil energy,dysregulation of ascending and descending,disharmony between the exterior and interior,and dysfunction of the liver,gallbladder,and stomach,such as liver-gallbladder damp heat syndrome,disharmony between the exterior and interior (intermittent chills and fever,malaria),abnormal ascending and descending (vomiting,thirst,irritability,constipation, and depression),and disharmony between Yin and Yang. The core point of the pathogenesis is that the lack of positive energy and the invasion of evil energy,with an unfavorable Shaoyang cardinal system,which is consistent with the pathophysiological characteristics of women. In gynecology,on the one hand,women are prone to Qi and blood deficiency,which affects the body's ability to fight off evil influences. On the other hand,women's liver function is the most important,and they are prone to the pathological manifestations of catharsis dysfunction,Qi stagnation,and loss of function. Therefore,Xiao Chaihutang is used in the treatment of various gynecological diseases such as menstruation,leukorrhea,childbirth,and lactation. The key to using ancient formulas lies in flexible application. As long as the additions and subtractions are appropriate,different diseases can be treated in the same way. If the clinical syndrome can be adjusted according to the changes,the ratio of monarch drug and ministerial drug group,the nature of cold and warm,and the ability to tonify and diarrhea,it can be widely used in the treatment of a variety of gynecological diseases based on the reconciliation of the pathogenesis of Shaoyang,the essence of the same treatment of different diseases,and flexible tailoring. This article aims to study the application of the classic prescription Xiao Chaihutang from the Treatise on Febrile Diseases in gynecology,summarize its characteristics of pathological mechanism integration,and help peers in the treatment of gynecological diseases.

      摘要:ObjectiveTo multidimensionally analyze the clinical effects of Qianyang Yuyin granules on elderly hypertensive patients through an "energy-inflammation-aging" network.MethodsRelevant datasets were retrieved from the GEO database. Gene set enrichment analysis (GSEA) was performed on the gene expression profiles of peripheral blood cells from patients with essential hypertension in dataset GSE24752. The GSEA referenced "GO gene sets" and "KEGG gene sets" to identify significantly enriched gene sets. A clinical trial was conducted using a randomized controlled study design. A total of 40 patients meeting the inclusion criteria were enrolled. The control group received standard antihypertensive treatment with angiotensin receptor blockers (ARBs) or combined calcium channel blockers (CCBs). In contrast,the treatment group received Qianyang Yuyin Granules in addition to the standard treatment for 12 weeks. Blood pressure levels and clinical efficacy were observed,and changes in energy metabolism indicators,DNA damage markers,and senescence-associated secretory phenotype (SASP) in blood were measured using ELISA before and after treatment.ResultsGSEA results indicated significant energy metabolism dysregulation in hypertensive patients. Clinical findings showed that both groups achieved blood pressure control without significant intergroup differences. In terms of clinical efficacy,the treatment group had a significantly higher effective rate compared to the control group (95% vs 65%,P0.05). After treatment,the treatment group showed a significant increase in NAD+ levels (P0.01),with higher levels compared to the control group (P0.05). The treatment group also exhibited a greater reduction in DNA damage marker 8-OHdG (P0.01) and cell adhesion factors ICAM-1 and VCAM-1 (P0.01) compared to the control group. Pro-inflammatory cytokines IL-1β and IL-6 were significantly reduced in the treatment group (P0.01),with greater reductions compared to the control group (P0.05,P0.01). Anti-inflammatory cytokines IFN-α,IL-4,and IL-10 were significantly elevated in the treatment group (P0.01),with higher levels compared to the control group (P0.01). No significant adverse reactions were reported in either group.ConclusionThe "energy- inflammation- aging" network plays an important role in the pathological mechanism of hypertension patients. Qianyang Yuyin granules may delay the aging process by increasing patients' energy metabolism levels,reducing DNA oxidative damage,and maintaining the balance of inflammatory factors.

      摘要:ObjectiveTo explore the medication patterns of Professor Zhang Farong in treating type 2 diabetes mellitus (T2DM) and the clinical efficacy of his core prescription.MethodsClinical case records of T2DM treated by Professor Zhang Farong were collected to establish a prescription database. Frequency statistics, visual analysis, and factor analysis were employed to investigate the characteristics and principle of the prescriptions, and a core prescription was derived. A randomized controlled trial was conducted, enrolling 60 T2DM patients with the dampness-heat syndrome. The patients were allocated into an observation group (core prescription + metformin) and a control group (metformin alone), with both groups undergoing a 12-week treatment course. Changes in TCM symptom scores, glucose metabolism indicators [fasting plasma glucose (FPG), 2-hour postprandial blood glucose (2 hPG), and glycated hemoglobin (HbA1c)], pancreatic function indicators [fasting C-peptide (FCP), 2-hour postprandial C-peptide (2 hCP), and area under the C-peptide curve (AUCcp)], and lipid profiles were measured before and after treatment. The adverse reactions were observed and recorded.ResultsA core prescription named modified Gegen Qinlian Decoction was formulated, comprising Puerariae Lobatae Radix, Coptidis Rhizoma, Scutellariae Radix, Astragali Radix, Lycii Cortex, Mori Cortex, Jineijin Endothelium Corneum Gigeriae Galli, Rehmanniae Radix Praeparata, Atractylodis Rhizoma, Polygonati Rhizoma, and Pogostemonis Herba. The clinical trial results showed that both groups had significantly decreased FPG, 2 hPG, and HbA1c (P0.05), and the observation group outperformed the control group in recovering the level of HbA1c (P0.05). After treatment, both groups had declined TCM symptoms scores (P0.05), and the declines in the observation group were larger than those in the control group (P0.05). After treatment, the TC and LDL-C levels declined in the observation group (P 0.05), while the lipid levels showed a decreasing trend with no statistically significant difference in the control group. After treatment, both groups showed increases in FCP and AUCcp (P0.05), and the 2 hCP in both groups presented a recovering trend with no statistically significant difference. There was no statistically significant difference in the incidence of adverse reactions between the two groups.ConclusionModified Gegen Qinlian Decoction embodies Professor Zhang Farong's academic philosophy of treating consumptive thirst by tonifying the spleen and kidney, replenishing Qi and Yin, clearing deficiency and heat, unblocking stasis in collaterals, and addressing both deficiency and stasis. The combination of the core prescription with metformin alleviates clinical symptoms in T2DM patients with the dampness-heat syndrome, demonstrating potential effects in restoring pancreatic islet function, regulating blood glucose, and improving lipid profiles. It serves as a therapeutic option for T2DM in the patients with the dampness-heat syndrome under syndrome differentiation, meriting broader clinical application.

      摘要:ObjectiveTo investigate the modifiable areal unit problem (MAUP) in the spatial pattern of Pseudostellaria heterophylla production regions and reveal the impact of statistical scales on the spatial distribution characteristics of this medicinal plant species.MethodsUsing multi-source data (literature records, field surveys, and statistical data), we systematically analyzed the spatial patterns across three administrative levels (provincial, prefectural, and county scales). Spatial autocorrelation (Moran's I) analysis, high-low clustering (Getis-Ord General G), and hot/cold spot analysis (Getis-Ord Gi*) were employed.ResultsThe literature-based analysis showed that the production regions of P. heterophylla presented random distribution on the provincial scale and significant aggregation on the prefectural scale. The field survey data showed that the production regions displayed random distribution on the provincial scale but significant aggregation on both prefectural and county scales. The statistical data revealed that the production regions lacked spatial autocorrelation on the provincial scale but demonstrated significant aggregation on prefectural and county scales.ConclusionMAUP effects have substantive implications for understanding and decision-making in the arrangement of medicinal plant production regions. The county scale proves to be the most sensitive and explanatory level for analyzing the spatial pattern of P. heterophylla production regions, providing a critical foundation for habitat modeling, suitability evaluation, and ecological cultivation planning of medicinal plants.

      摘要:ObjectiveTo investigate the quality markers of Xiaoqinglong granules(XQLG) for treating bronchial asthma using the analytic hierarchy process(AHP)-entropy weight method(EWM), network pharmacology and high performance liquid chromatography(HPLC) content determination.MethodsEffectiveness, testability and peculiarity component data of XQLG in treating bronchial asthma were constructed through database retrieval, literature review, and network pharmacology. Subsequently, AHP-EWM was used to quantitatively identify and weight the control layer and element layer, the relevant compounds were selected as candidate quality markers based on comprehensive scores. Further comparison of reference substances and establishment of HPLC content determination method were used to determine the potential quality markers of XQLG, which were verified by molecular docking with disease targets.ResultsA total of 13 components, including glycyrrhizic acid, paeoniflorin, schisandrol A, isoliquiritigenin, 6-gingerol, ephedrine, liquiritin, albiflorin, liquiritigenin, 6-shogaol, pseudoephedrine, cinnamic acid and cinnamaldehyde, were identified as potential quality markers of XQLG by AHP-EWM. Quantitative analysis indicated that all aforementioned quality markers could be detected in 13 batches of XQLG, indicating that it had stable testability as a quality marker. Among these 13 batches of samples, ephedrine and paeoniflorin exhibited good consistency in content, while pseudoephedrine and cinnamaldehyde showed poor consistency. Molecular docking analysis revealed that the 13 compounds exhibited binding energies with the core targets -2.11 kcal·mol-1, indicating that the 13 compounds could spontaneously bind to the disease targets, which may be the material basis for the treatment of bronchial asthma with XQLG.ConclusionIn this study, 13 compounds were screened by AHP-EWM combined with network pharmacology and HPLC as quality markers for the treatment of bronchial asthma by XQLG, laying the foundation for enhancing the quality standards of this preparation.

      摘要:As a mercury-containing elixir, Hongshengdan has been known as a sacred medicine for surgery by ancient medical practitioners because of its precise curative effects. It originated from Yizong Shuoyue in the Qing dynasty, Qing dynasty and modern medical practitioners have adapted and modified its formula for clinical application. Employing bibliometric methods, the authors systematically organized relevant ancient literature of the Qing dynasty and modern literature, and analyzed the composition and dosage, preparation method, and clinical application. Among the 25 ancient books concerning Hongshengdan, a total of 12 medicinal formulas, 15 refining methods and 9 clinical applications were obtained. Research confirms that Hongshengdan consisted of mercury, saltpeter, alum, soap alum, cinnabar and realgar. Using measurement conversion standards of Qing dynasty, the modern single-batch formulation comprised 37.30 g of mercury, 149.20 g of saltpeter, 37.30 g of alum, 22.38 g of soap alum, 18.65 g of cinnabar, and 18.65 g of realgar. In modern refining of Hongshengdan, most medical practitioners take the core medicines, with dosages approximately 30 g of mercury, 30 g of saltpeter, and 30 g of alum. Refining method involves pretreatment stewing the materials during preparation, and alum, soap alum, and saltpeter are first ground together, then combined with mercury, cinnabar, and realgar for grinding until mercury and other drugs grind to the degree of no star points. The mixture is then placed in a pot or vessel by cold-forming method. After covering, the opening is sealed using either raw gypsum salt mud or honey-dipped cotton paper strips. Sand is packed around the vessel and then pressurized. During the calcination process, begin with a low flame(30 min), then increase to a medium flame(30 min), followed by a high flame(30 min), after removing fire toxins, collect the final product. Hongshengdan has the efficacy of lifting the poison, removing the corrosion, producing muscle and dispersing, and is often used in the treatment of surgical sore and carbuncle type of diseases. Modern research indicates that Hongshengdan is commonly used to treat skin system diseases such as ulcers and herpes. The aforementioned findings provide a reference basis for the subsequent refining method and clinical application of Hongshengdan.

      摘要:ObjectiveTo investigate the alleviating effect of calcium cyanamide (CaCN2) soil fumigation on replanting problems of Rehmannia glutinosa.MethodsNewly soil (NP) was used as the control group, while three treatment groups were established: replanted soil (RP), newly soil treated with CaCN2 (120 g·m², tillage depth 25 cm) (NPCC), and replanted soil treated with CaCN2 (RPCC). R. glutinosa was cultivated in all groups. At harvest, the tuber agronomic traits (number of enlarged roots, maximum root diameter, fresh weight, dry weight) were measured. The content of catalpol and rehmannioside D was quantified by ultra-high-performance liquid chromatography (UPLC) to evaluate medicinal quality. Rhizosphere soil available nutrients and enzyme activities were analyzed by assay kits. The community structure and composition of fungi and bacteria in rhizosphere soil were assessed via internal transcribed spacer 2 (ITS2) sequencing and 16S rDNA sequencing, respectively.ResultsCompared with NP, the RP group showed obviously reduced in tuber agronomic traits and quality indicators (P0.05). However, the RPCC group showed significant improvement in agronomic traits and a notable increase in rehmannioside D content compared to RP (P0.05). The contents of available phosphorus and potassium in RPCC and NP groups were obviously lower than those in RP (P0.05). The polyphenol oxidase soil (S-PPO) activity in RP was obviously lower than in NP (P0.05), while sucrose soil (S-SC), acid phosphatase soil (S-ACP), and S-PPO activities in RPCC were obviously higher than in RP (P0.05). Microbial richness and diversity in RP were obviously higher than in NP (P0.05), whereas no significant differences were observed between the RPCC and NP. The relative abundances of fungal genera Nectria, Myrothecium, Tomentella, and bacterial genus Skermanella were obviousl lower in RPCC and NP than in RP (P0.05). Correlation analysis that S-ACP activity was positively correlated with the content of rehmannioside D (P0.05). Fungal genera Engyodontium and Alternaria, and bacterial genera Pir4 lineage, Pirellula, Methyloversatilis, Brevundimonas, Ralstonia, and Acidibacter were obviously positively correlated with tuber dry weight (P0.05). Conversely, fungal genera Pseudaleuria, Nectria, Haematonectria, Ceratobasidium, and bacterial genera Streptomyces, Skermanella, RB41, Gemmatimonas, and Bacillus were obviously negatively correlated with dry weight (P0.05). The fungal genus Alternaria and bacterial genera Brevundimonas, Ralstonia, Acidibacter, and Dongia showed positive correlations with medicinal quality of R.glutinosa tuber, while fungal genera Pseudaleuria, Nectria, Stachybotrys, Fusarium, Gibberella, Ceratobasidium, and bacterial genera Sphingomonas, Skermanella, RB41, Gemmatimonas, and Bacillus were obviously negatively correlated (P0.05).ConclusionCaCN2 soil fumigation can significantly improve enzyme activities in replanted Rehmannia rhizosphere soil, enhance the utilization of available nutrients, reshape microbial community structure of replanted R.glutinosa at the family and genus level, and notably improve tuber agronomic traits and medicinal quality. This study provides a novel approach to alleviating replanting problems and offers insights for the integrated development of standardized cultivation techniques, including soil disinfection, nutrient-targeted regulation, and microbial inoculant application.

      摘要:Lianggesan was first recorded in Taiping Huimin Heji Jufang, which was composed of Rhei Radix et Rhizoma, Natrii Sulfas, Gardeniae Fructus, Forsythiae Fructus, Scutellariae Radix, Glycyrrhizae Radix et Rhizoma(GRR), Menthae Haplocalycis Herba, Lophatheri Herba and Mel. It was clinically applied to treat fire-heat syndrome in the upper and middle Jiao, and the curative effect was positive. In this study, the bibliometric method was used to conduct a detailed textual research on the formula name, medicinal composition, dosage evolution, origin and processing, functional indications and other aspects of Lianggesan. Research revealed that Lianggesan has six other names, such as Lianqiao Yinzi, Lianqiao Jiedusan, Jufang Lianggesan, Jiegu Lianggesan, Hejian Lianggesan and Qingji Lianggesan. Based on the edition of Taiping Huimin Heji Jufang, an analysis of the evolution of its formula composition revealed that the missing Chinese medicines were predominantly bamboo leaves and honey, while the added Chinese medicines were primarily supplements introduced to address changes in disease manifestations. After textual research, the dosage for one dose of Lianggesan from Taiping Huimin Heji Jufang was as follows:826 g of Rhei Radix et Rhizoma, 826 g of Natrii Sulfas, 826 g of GRR, 413 g of Gardeniae Fructus, 413 g of Menthae Haplocalycis Herba, 413 g of Scutellariae Radix, and 1652 g of Forsythiae Fructus. Decocting method was as following:Grinding the Chinese medicines into coarse powder(2-4 mm), taking 8.16 g per dose, adding 300 mL of water, along with 2 g of Lophatheri Herba and 5 g of Mel, and decocting to 140 mL. The residue was removed and taken warmly 30 min after meals. It was recommended to take it three times daily until improvement was achieved. The origins of the 9 Chinese medicines were consistent with the 2020 edition of Pharmacopoeia of the People's Republic of China. Except for GRR, which required single frying(stir-frying), the remaining medicines were all raw products. The description of the function of this formula in ancient books was summarized as purging fire and promoting bowel movements, clearing heat from the upper body and purging the lower body, and the main syndromes included facial redness, tongue swelling, red eyes, etc. In modern applications, the formula is primarily used for respiratory and digestive system diseases, including acute lung injury, chronic obstructive pulmonary disease, herpetic angina and aphthous stomatitis, covering 142 types of diseases. In summary, this paper can provide a basis for further research and development of Lianggesan through the literature review and key information combing.

      摘要:The rupture of carotid vulnerable plaques is the core pathological basis for major cardiovascular and cerebrovascular events. However, the insufficient alignment between existing animal models and the clinical disease and syndrome characteristics of traditional Chinese and western medicine has limited research progress. In this study, biomedical databases in China and abroad were systematically searched, and the modeling mechanisms and evaluation systems of carotid vulnerable plaque animal models were systematically assessed based on diagnostic criteria of both traditional Chinese and western medicine. Analysis of the clinical correspondence indicated that existing animal models can be categorized into four types: simple high-fat diet, surgical induction combined with high-fat feeding, genetic engineering combined with high-fat feeding, and drug induction combined with high-fat feeding. Among these, the compound strategy of surgical induction combined with high-fat feeding has become the current mainstream approach, showing good concordance with western medicine. The study found that the double balloon injury rabbit model and the ApoE-/- mouse carotid artery tandem constriction combined with high-fat feeding model demonstrated a high degree of clinical correspondence with both traditional Chinese and western medicine in terms of vulnerable plaque imaging and pathological features. Nevertheless, existing models still face significant technical limitations in faithfully simulating plaque pathology and in translating findings to clinical applications. To address these challenges, integrating complex comorbidity mechanism construction, multimodal dynamic mechanism monitoring, and collaborative evaluation systems of traditional Chinese and western medicine could enable the development of highly concordant carotid vulnerable plaque disease-syndrome combination animal models. Such models would provide a reproducible experimental platform for targeted drug development to regulate plaque stability and for individualized precision treatment, as well as a theoretical basis for innovation in clinical diagnostic and therapeutic strategies.

      摘要:By systematically combing ancient and modern literature, this paper examined Tribuli Fructus and Astragali Complanati Semen(ACS) used in the famous classical formulas from the aspects of name, origin, production area, harvesting and processing, clinical efficacy, so as to provide a basis for the development of famous classical formulas containing such medicinal materials. The results showed that the names of Tribuli Fructus in the past dynasties were mostly derived from its morphology, and there were nicknames such as Baijili, Cijili and Dujili. The name of ACS in the past dynasties were mostly originated from its production areas, and there were nicknames such as Baijili, Shayuan Jili and Tongjili. Because both of them had the name of Baijili, confusion began to appear in the Song dynasty. In ancient and modern times, the main origin of Tribuli Fructus were Tribulus terrestris, and ancient literature recorded the genuine producing areas of Tribuli Fructus was Dali in Shaanxi and Tianshui in Gansu, but today it is mainly cultivated in Anhui and Shandong. The fruit is the medicinal part, harvested in autumn throughout history. There is no description of the quality of Tribuli Fructus in ancient times, and the plump, firm texture, grayish-white color is the best in modern times. Traditional processing methods for Tribuli Fructus included stir-frying and wine processing, while modern commonly used is purified, fried and salt-processed. The ancient records of Tribuli Fructus were spicy, bitter, and warm in nature, with modern research adding that it is slightly toxic. The main effects of ancient and modern times include treating wind disorders, improving vision, promoting muscle growth, and treating vitiligo. The mainstream base of ACS used throughout history is Astragalus complanatus. Ancient texts indicated ACS primarily originated from Shaanxi province. Today, the finest varieties come from Tongguan and Dali in Shaanxi. The medicinal part is the seed, traditionally harvested in autumn. Modern harvesting occurs in late autumn or early winter, followed by sun-drying. Ancient texts valued seeds with a fragrant aroma as superior, while modern standards prioritize plump, uniform and free of impurities. Traditional processing methods for ACS included frying until blackened and wine-frying, while modern practice commonly employs purification methods. In terms of medicinal properties, the ancient and modern records are sweet and warm in nature. Due to originally classified under Tribuli Fructus, its effects were thus regarded as equivalent to those of Tribuli Fructus, serving as the medicine for treating wind disorders, additional functions included tonifying the kidneys and treating vitiligo. The present record of its efficacy is to tonify the kidney and promote Yang, solidify sperm and reduce urine, nourish the liver and brighten the eye, etc. Based on the textual research results, it is suggested that when developing the famous classical formulas of Tribuli Fructus medicinal materials, we should pay attention to the specific reference object of Baijili, T. terrestris and A. complanatus should be identified and selected, and the processing method should be in accordance with the requirements of the formulas.

      摘要:Breast cancer, as one of the major cancers threatening women's health globally, is characterized by high aggressiveness, high malignancy, and poor prognosis. In 2022, according to the World Health Organization, breast cancer ranked second in the incidence of female cancers globally, accounting for 11.6% of all new cancer cases. Western medical doctors mainly use surgery, chemotherapy, radiotherapy, endocrine therapy, and molecular targeted therapy to treat breast cancer, which can effectively improve the recurrence rate and death rate of breast cancer patients and prolong the survival period of patients. However, its treatment process is often accompanied by a series of side effects, which bring challenges to patients' quality of life. However, traditional Chinese medicine (TCM) has demonstrated significant therapeutic effects in inhibiting the proliferation and metastasis of breast cancer cells, reducing toxic side effects produced by chemotherapy, and improving patients' survival rate and quality of life. It is therefore particularly necessary to investigate the pharmacological effects and mechanisms of TCM in breast cancer treatment. The authors combed the pharmacological effects and mechanisms of the etiology and pathogenesis of breast cancer, identification and treatment of breast cancer, TCM compound, TCM single medicine, TCM monomer, and external treatment of TCM to prevent and control breast cancer and found that TCM has a therapeutic effect on breast cancer. It can play a role in increasing the effectiveness, reducing the toxicity, and alleviating the adverse reactions. It can inhibit breast cancer cell proliferation, cell cycle, migration, invasion, immune escape, epithelial-mesenchymal transition (EMT), aerobic glycolysis, mitochondrial biosynthesis, aminoacyl-tRNA biosynthesis, reduce drug resistance, promote apoptosis, ferroptosis, cell autophagy, and regulate the tumor immune microenvironment by regulating signaling pathways. This paper aims to provide new ideas and methods for experimental research and clinical treatment of breast cancer.

      摘要:Aromatherapy refers to the method of using the aromatic components of plants in appropriate forms to act on the entire body or a specific area to prevent and treat diseases. Essential oils used in aromatherapy are hydrophobic liquids containing volatile aromatic molecules, such as limonene, linalool, linalool acetate, geraniol, and citronellol. These chemicals have been extensively studied and shown to have a variety of functions, including reducing anxiety, relieving depression, promoting sleep, and providing pain relief. Terpenoids are a class of organic molecules with relatively low lipid solubility. After being inhaled, they can pass through the nasal mucosa for transfer or penetrate the skin and enter the bloodstream upon local application. Some of these substances also have the ability to cross the blood-brain barrier, thereby exerting effects on the central nervous system. Currently, the academic community generally agrees that products such as essential oils and aromatherapy from aromatic plants have certain health benefits. However, the process of extracting a single component from it and successfully developing it into a drug still faces many challenges. Its safety and efficacy still need to be further verified through more rigorous and systematic experiments. This article systematically elaborated on the efficacy of aromatic substances, including plant extracts and natural small molecule compounds, in antibacterial and antiviral fields and the regulation of nervous system activity. As a result, a deeper understanding of aromatherapy was achieved. At the same time, the potential of these aromatic substances for drug development was thoroughly explored, providing important references and insights for possible future drug research and application.

      摘要:The pathogenesis of central nervous system (CNS) diseases is complex, seriously affecting patients' physical and mental health and imposing a heavy economic burden on society. Western medicine shows limited efficacy in treating CNS diseases and is often associated with numerous adverse reactions and contraindications. Chinese medicine Lycii Fructus exhibits multiple pharmacological effects, including immune regulation, enhancement of hematopoietic function, liver protection, anti-tumor, hypoglycemic, antipyretic, anti-aging, and anti-radiation activities, and has gradually been applied in clinical treatment. In recent years, the active components of Lycii Fructus have attracted considerable attention for their potential therapeutic effects on CNS diseases. Studies indicate that these active components may exert neuroprotective effects through anti-inflammatory and antioxidant actions, inhibition of neuronal apoptosis, and repair of neuronal damage, involving multiple targets and pathways. This review summarizes the therapeutic effects of Lycii Fructus active components in CNS diseases over the past decade by searching PubMed, CNKI, Wanfang Data, and other electronic databases, aiming to provide new treatment strategies and insights for future research on Lycii Fructus in CNS disorders.

      摘要:As the core technology of processing, the stir-frying method of traditional Chinese medicine(TCM) realizes the regulation of efficacy of medicinal substances through the coordination of fire gradient and excipients. This study aims to systematically analyze the influence of different stir-frying degrees(stir-frying until yellow, stir-frying to brown, stir-frying to charcoal) and excipient types(solid excipients such as rice, wheat bran, soil, liquid excipients such as rice wine, vinegar, honey) on the physicochemical properties, efficacy and toxicity of TCM, summarizing their common regulatory mechanisms and characteristics. Then, the three-dimensional regulatory common mechanisms and five-dimensional regulatory specificity mechanisms of different stir-frying processes were obtained. The three-dimensional regulatory common mechanisms are as follows:①Thermal effects break down inherent barriers of medicinal materials and improve the dissolution of components. ②Inducing Maillard reaction, carbonization and other reactions of medicinal materials to promote the transformation of components. ③Combining the catalytic and harmonizing effects of excipients to achieve targeted enrichment of efficacious substances and precise control of toxic components. The five-dimensional regulatory specificity mechanisms manifest as:①The efficacy gradient pattern where stir-frying until yellow strengthens the spleen, stir-frying to brown aids digestion, and stir-frying to charcoal stops bleeding. ②According to the polarity difference and intrinsic properties, the liquid excipients form a directional synergistic mechanism of wine processing enhances ascending nature, ginger processing to warm and disperse, salt processing directs effects to the kidneys, vinegar processing targets the liver, honey processing imparts sweetness and moderation. ③Through porous structure and interfacial properties, solid excipients give the synergistic effect of rice processing for diarrhea relief, bran processing for stomach tonification, soil processing for warming the middle warmer, and clam powder/talcum powder/sand processing for texture optimization. ④Thermal effects induce decomposition/oxidation/polymerization and other reactions to reshape the material basis, directing changes in the efficacy. ⑤The targeted distribution of efficacy is regulated by component enrichment and meridian-guiding effect, and the meridian tropism is changed. The system, driven by thermal effect, excipient synergy and physicochemical reconstruction, integrates five-dimensional regulation of fire, solvent, interface, composition and distribution. It systematically explains the inherent law of efficacy-substance-process of traditional stir-frying, and provides theoretical basis and practical guidance for standardizing TCM processing and enhancing the efficacy.

      摘要:Diabetic cardiomyopathy (DCM), a cardiovascular complication caused by diabetes mellitus, is a major cause of heart failure and even sudden cardiac death in diabetic patients. Traditional Chinese medicine (TCM) posits that the core pathogenesis of DCM lies in internal deficiency and superficial excess, characterized by deficiency of both Qi and Yin combined with phlegm and blood stasis. Modern medical treatments for DCM primarily focus on blood glucose control and symptom alleviation yet lack targeted therapeutic strategies. In contrast, TCM offers a wealth of practical experience and a complete theoretical system, demonstrating definite clinical efficacy and high medication safety in DCM management. As a classic formula for tonifying Qi and nourishing Yin, Shengmaisan comprises Ginseng Radix et Rhizoma, Ophiopogonis Radix, and Schisandrae Chinensis Fructus. It contains multiple bioactive components, including ginsenosides, ophiopogonin, schisandrins, and homoisoflavonoids, which exhibit cardioprotective properties. The therapeutic mechanisms of Shengmaisan-like formulae for DCM involve enhancing myocardial contractility, attenuating myocardial fibrosis, modulating mitochondrial quality control, regulating glucose metabolism, mitigating oxidative stress, and suppressing inflammatory responses. Clinically, Shengmaisan-like formulae not only manage hyperglycemic status but also ameliorate cardiac structural and functional impairments and enhance exercise tolerance in DCM patients, playing a vital role in the prevention, treatment, and rehabilitation of DCM. This paper analyzes the feasibility of Shengmaisan-like formulae in DCM management and synthesizes current research achievements regarding their chemical components, mechanisms of action, and clinical applications, aiming to provide a scientific foundation for the use of such formulae in the treatment of DCM.
