Ce mémoire a pour but d'utiliser la chromatographie liquide à haute efficacité couplée à la spectrométrie de masse à tempête quadrupolaire (UPLC-Q-TOF-MS/MS) en combinaison avec la pharmacologie en réseau et le docking moléculaire - pour étudier l'effet pharmacologique et le mécanisme d'action d'un médicament sur l'eczéma chaud humide chez le rat. Méthode établissement du modèle d'eczéma chaud humide chez le rat par le biais d'une intervention de la serre climatique artificielle combinée à une induction de la sensibilisation au dinitrochlorobenzène, répartition aléatoire en groupe normal, groupe modèle, basse et haute dose du médicament UPLC-Q-TOF-MS/MS (3.40, 6.80 g∙kg-1), groupe acétate de prednisone (2.51 mg∙kg-1), 8 rats par groupe, total de 46 rats dont 6 avec sérum médicamenteux; analyse chimique des sérums des rats par UPLC-Q-TOF-MS/MS, prédictions des composés clés, points cibles principaux et voies de signalisation par la pharmacologie en réseau, expérimentations de docking moléculaire sur le site en ligne CB-Dock2;examen de la pharmacologie du complexe pour l'eczéma humide chaud par des indicateurs d'apparence combinés à la coloration des tissus pathologiques; détermination des niveaux sériques de gastrine, d'interleukine-4 (IL-4) et d'interleukine-13 (IL-13) par dosage immuno-enzymatique (ELISA); détection des niveaux de protéines dans le sérum de l'interleukine-6 (IL-6), de la Janus kinase 1 (JAK1), de la JAK1 phosphorée, de la signalisation et de la transcription activée par le facteur 3 (STAT3), et de la STAT3 phosphorée par immunobuvatelaD??resultats de l'expérience sur les animaux a montré la présence de 19 composites actifs dans les échantillons de sérum de rats médicamenteux, qui, selon les prédictions, agissaient sur 198 cibles pour traiter l'eczéma chaud humide, dont les composés clés comprennent la berbérine, la rhénatine, l'harpagide, entre autres, et les principales cibles incluent le STAT3, le facteur de nécrose tumorale (TNF), l'IL-6, entre autres, avec des implications majeures dans les voies de signalisation pour le cancer, la phosphoinositrol-3-kinase (PI3K)/voie de signalisation protéine kinase B (Akt), la voie de différenciation des cellules T auxiliaires 17 (Th17), la voie de signalisation JAK/STAT, entre autres. Les résultats de l'expérience de docking moléculaire ont montré que les composés clés avaient une forte activité de liaison avec les cibles principales de la voie JAK/STAT d'IL-6, de JAK1 et de STAT3. Les résultats de l'expérience sur les animaux ont montré que, comparé au groupe normal, le groupe modèle de rats était maussade, avec des poils clairsemés, de la diarrhée, des épanchements cutanés, des érosions et une érosion épaisse se présentant sous la forme d'une squame, une baisse de la masse corporelle, une poussée de la température anale et de la consommation d'eau, une augmentation de l'indice de la rate, du thymus et de l'estomac (P<0.05, P<0.01), la pathologie cutanée présentant une kératose incomplète, un agrégat de cellules inflammatoires, une séparation indistincte entre l'épiderme et le derme, une paroi gastrique mince dépourvue de structure, et un agrégat évident de cellules inflammatoires, des niveaux sériques significativement inférieurs de gastrine, d'IL-4, d'IL-13 (P<0.05, P<0.01), des niveaux de protéines significativement supérieurs d'IL-6, de JAK1, de JAK1 phosphorée, de STAT3 et de STAT3 phosphorée présents dans la pathologie cutanée (P<0.05, P<0.01);comparé au groupe modèle, le groupe de traitement de rats est devenu plus stable, avec des matières fécales formées, des fesses propres, une peau presque normale, avec seulement une petite quantité d'écailles, une augmentation de la masse corporelle, une diminution de la température anale et de la consommation d'eau, une diminution de l'indice de la rate, du thymus et de l'estomac (P<0.05, P<0.01), une réduction de l'épaisseur de la peau, une séparation nette entre l'épiderme et le derme, mais une kératose excessive et un agrégat de cellules inflammatoires présents, une augmentation de l'épaisseur de la muqueuse gastrique, une récupération de la structure de divers degrés, une augmentation de différents degrés des niveaux sériques de gastrine, d'IL-4, d'IL-13 dans la pathologie cutanée, une diminution significative des niveaux de protéines d'IL-6, de JAK1, de JAK1 phosphorée, de STAT3 et de STAT3 phosphorée présent dans la pathologie cutanée (P<0.05, P<0.01);conclusion l'effet de l'élixir des cinq terres sur l'eczéma chaud humide chez le rat pourrait être exercé par la régulation de la voie de signalisation JAK/STAT.
