L'objectif de l'étude était d'explorer le mécanisme par lequel l'anmidan améliore les fonctions cognitives et régule le processus inflammatoire immunitaire en régulant la voie de signalisation cGAS/STING chez les rats insomniaques. Méthodes : 60 rats de race SD ont été répartis au hasard en un groupe témoin, un groupe modèle, un groupe de sulpiride (30 mg·kg-1), des groupes à faible, moyen et fort dosage d'anmidan (4,55, 9,09, 18,18 g·kg-1), 10 rats par groupe ; le modèle de rat insomniaque a été établi par injection intrapéritonéale d'acide propionique (AP), puis les rats ont reçu une perfusion gastrique de l'infusion aqueuse d'anmidan correspondant à la dose pendant 28 jours et de solution saline physiologique ; les fonctions cognitives ont été évaluées par les tests du labyrinthe d'eau de Morris, de la reconnaissance d'objets nouveaux ; la morphologie des cellules de l'hippocampe a été observée par coloration à l'hématoxyline-éosine (HE) et à la nissel ; la teneur en interleukine-1 (IL-1), interleukine-1β (IL-1β), interleukine-6 (IL-6), interleukine-8 (IL-8), interleukine-12 (IL-12), interleukine-18 (IL-18), facteur de nécrose tumorale-α (TNF-α) a été déterminée par ELISA ; l'expression relative de la protéine et de l'ARNm de cGAS et STING dans l'hippocampe a été mesurée par immunobuvardage et PCR en temps réel. Résultats: Comparé au groupe témoin, le groupe modèle a montré une diminution significative de la teneur en 5-hydroxytryptophane (5-HT) (P<0.01) ; une augmentation significative du temps pour atteindre la plateforme supérieure et de la distance totale parcourue, une diminution significative du temps dans le quadrant cible et du nombre de traversées de la plateforme (P<0,01), une diminution significative de l'indice de reconnaissance d'objet nouveau (P<0,01) ; une dysplasie marquée et une répartition désordonnée des neurones dans la zone étudiée de l'hippocampe ont été observées, le nombre de corps nissels intracellulaires a diminué ; la teneur en IL-1, IL-1β, IL-6, IL-8, IL-12, IL-18, TNF-α, les protéines et l'ARNm de cGAS, STING a considérablement augmenté (P<0,01) ; Comparé au groupe modèle, le groupe à forte dose d'anmidan a montré une diminution significative du temps pour atteindre la plateforme supérieure (P<0,05), une augmentation significative du temps passé dans le quadrant cible, du nombre de traversées de la plateforme dans les groupes à doses moyenne et élevée d'anmidan et dans le groupe utilisant du sulpiride (P<0,01), une diminution significative de la distance totale dans tous les groupes de traitement (P<0,01), une augmentation significative de l'indice de reconnaissance d'objet nouveau dans tous les groupes de traitement (P<0,01), un aspect et un nombre de corps nissels intracellulaires plus normaux dans les groupes à doses moyenne et élevée d'anmidan; dans les groupes à doses moyenne et élevée d'anmidan, les niveaux de IL-1, IL-1β, IL-6, IL-8, IL-12, IL-18, TNF-α, et des protéines et de l'ARNm du cGAS ont considérablement diminué (P< 0,05, P<0,01) ; Conclusions: L'amélioration des fonctions cognitives chez les rats insomniaques par l'anmidan est probablement liée à l'inhibition du processus inflammatoire immunitaire via la voie de signalisation cGAS/STING.
