L'objectif est d'analyser le mécanisme pharmacologique de la lutéoline contre l'ischémie cérébrale à partir de données transcriptomiques, en considérant plusieurs dimensions telles que les tissus, les cellules, les processus pathologiques, les processus biologiques et les voies de signalisation. Méthodes : 30 rats SD ont été répartis aléatoirement en groupe chirurgie simulée, groupe modèle et groupe lutéoline (40 mg/kg). Après 30 minutes d'administration aux deux groupes sauf le groupe chirurgie simulée, un modèle d'occlusion de l'artère cérébrale moyenne (MCAO) a été induit par la méthode de fil. Après 24 heures, la technologie de séquençage transcriptomique a été utilisée pour détecter le profil d'expression génique dans le cortex cérébral et la zone de pénombre des rats, suivie d'une analyse d'enrichissement des gènes différentiellement exprimés via l'ontologie génétique (GO) et la base de données Kyoto Encyclopedia of Genes and Genomes (KEGG). Une plateforme de pharmacologie réseau multi-échelle basée sur le transcriptome (TMNP) a été employée pour analyser le mécanisme d'action de la lutéoline contre l'ischémie cérébrale selon cinq dimensions : tissus, cellules, processus pathologiques, processus biologiques et voies de signalisation. Résultats : Les résultats du séquençage et de la quantification des gènes ont révélé 667 gènes inversés de manière significative par la lutéoline. Les analyses KEGG et GO ont montré que les voies d'action de la lutéoline impliquent la régulation de la réponse au stress, le transport ionique, la prolifération et différenciation cellulaires, la fonction synaptique, entre autres. L'étude TMNP a démontré qu'au niveau tissulaire, la lutéoline peut considérablement améliorer les lésions cérébrales dues à l'ischémie ; aux niveaux cellulaire et pathologique, elle agit contre l'ischémie en améliorant l'état de diverses cellules nerveuses, en mobilisant les cellules immunitaires et en régulant la réponse inflammatoire ; au niveau des processus biologiques et des voies de signalisation, la lutéoline agit principalement sur la revascularisation, les voies de signalisation inflammatoires et la transmission synaptique. Conclusion : Le mécanisme d'action de la lutéoline contre l'ischémie cérébrale pourrait être lié à l'intervention dans la réponse au stress et l'inflammation, l'impact sur la revascularisation et la régulation de la fonction synaptique, fournissant ainsi une base et une référence pour des recherches approfondies sur le mécanisme pharmacologique de la lutéoline.
