Objectif : L’angélique, en tant que plante médicinale alimentaire largement utilisée, est fréquemment contrefaite sur le marché, ce qui nuit gravement à l’efficacité clinique des préparations. Actuellement, les techniques d’identification qualitative des contrefaçons sont relativement matures, mais il n’existe pas encore de méthodes quantitatives pour détecter les contrefaçons dans les produits transformés. Cette étude vise à développer une méthode de détection quantitative moléculaire des herbes (Herb-Q) en prenant pour exemple l’angélique et son proche apparenté facilement confondu « l’angélique du sol » (angélique coréenne), afin d’établir une méthode quantitative moléculaire pour l’angélique et ses produits transformés, fournissant un exemple pour la mise en place de techniques de détection quantitative pour les matières premières d’angélique et les produits de santé connexes. La méthode est basée sur la sélection de sites spécifiques de polymorphisme nucléotidique simple (SNP) dans les chloroplastes des angéliques et de l’angélique coréenne selon le génome chloroplastique complet. Des sites spécifiques à l’angélique ont été choisis pour évaluer la méthodologie quantitative à l’aide de poudres d’herbes mixtes à différents taux de contrefaçon (linéarité, limite quantitative, limite de détection et reproductibilité). Trois proportions différentes de contrefaçon (15%, 25%, 35%) dans une poudre analgésique activant la circulation sanguine ont été utilisées pour valider la précision de la méthode Herb-Q pour la détection quantitative dans la médecine traditionnelle chinoise contenant de l’angélique. Résultats : En comparant 123 séquences de génomes chloroplastiques du genre Angélique, en se basant sur la conservation intra-espèce, la spécificité inter-espèce et les exigences du pyroséquençage à haut débit, les positions 9674 (A/G) dans la séquence NC_042826.1 et 38592 (T/C) dans NC_029393.1 ont été identifiées comme sites moléculaires spécifiques pour l’angélique et l’angélique coréenne respectivement. Le site spécifique 9674 (A/G) de l’angélique a été choisi pour établir la relation linéaire de la méthode Herb-Q avec une valeur R² de 0.9974 (>0.99), indiquant une bonne linéarité. La limite quantitative et la limite de détection étaient de 2.0%, avec une bonne reproductibilité (écart type relatif (RSD) <2.0%). Le système quantitatif construit sur la méthode Herb-Q a détecté la quantité de contrefaçon d’angélique coréenne dans la poudre analgésique activant la circulation sanguine, avec une déviation moyenne des répétitions moléculaires triplicates de 1.3%. Conclusion : Cette étude montre que la méthode de détection quantitative Herb-Q basée sur la position 9674 (A/G) du génome chloroplastique NC_042826.1 de l’angélique est bien applicable, objective et précise pour l’angélique et «l’angélique du sol» ainsi que leurs produits transformés, fournissant un support technique pour la détection quantitative des produits dérivés de l’angélique vendus sur le marché tels que les préparations de médecine traditionnelle chinoise, les compléments alimentaires et les produits de santé.
