L'objectif est d'explorer le mécanisme par lequel la formule Jianpi Xiaoaifang régule l'axe métabolique de la synthase inductible de l'oxyde nitrique - arginase 1 (iNOS-ARG1), induisant une dysfonction structurale et fonctionnelle des mitochondries médiée par les espèces réactives de l'oxygène mitochondriale (mito-ROS), afin d'inhiber la progression du cancer du côlon. Un modèle de désordre du métabolisme de l'arginine dans les cellules humaines de cancer du côlon HCT116 a été établi par une intervention conjointe avec de l'interféron recombiné humain-γ (IFN-γ, 10 μg·L⁻¹) et N-(ω)-hydroxy-L-arginine (Nor-NOHA, 200 μmol·L⁻¹) pendant 24 h, suivi d'interventions avec des sérums médicamenteux contenant 5 %, 10 % et 20 % de la formule Jianpi Xiaoaifang. La prolifération cellulaire a été détectée par CCK-8, coloration EdU, et expérience de formation de colonies ; l'invasion et la migration ont été évaluées par chambre Transwell et test de cicatrisation ; le potentiel membranaire mitochondrial et les niveaux de ROS ont été évalués par colorations JC-1 et MitoSOX ; la structure ultra-mitochondriale a été observée par microscopie électronique à transmission (TEM) ; l'expression des protéines iNOS, ARG1, MFN2 et DRP1 a été analysée par Western blot et immunofluorescence ; les niveaux de L-arginine, citrulline et urée ont été mesurés par dosage colorimétrique/ELISA. Par rapport au groupe témoin, le groupe modèle montrait une expression significativement augmentée de iNOS et diminuée de ARG1, une diminution de ARG1-iNOS, une diminution de la L-arginine et de l'urée, et une augmentation significative de la citrulline (P<0,05) ; parallèlement, l'accumulation de mito-ROS était augmentée, le ratio JC-1 rouge/vert diminuait, les mitochondries présentaient un gonflement et une rupture des crêtes, indiquant que le désordre métabolique peut induire des lésions mitochondriales. Par rapport au groupe modèle, toutes les doses de la formule Jianpi Xiaoaifang pouvaient davantage réduire ARG1/iNOS, augmenter l'accumulation du monoxyde d'azote (NO) et des espèces réactives de l'azote, diminuer davantage les niveaux de L-arginine et d'urée, et augmenter significativement la citrulline (P<0,01) ; les taux positifs EdU, le taux de formation de colonies, le taux de cicatrisation des blessures et le nombre de cellules traversant la membrane Transwell diminuaient significativement avec l'augmentation de la concentration de sérum médicamenteux (P<0,01) ; le niveau de mito-ROS augmentait, le ratio JC-1 rouge/vert diminuait davantage. La TEM révélait un gonflement et une vacuolisation mitochondriale aggravés, une expression diminuée de la protéine MFN2, une expression augmentée de DRP1 (P<0,01), dépendant de la dose. Conclusion : la formule Jianpi Xiaoaifang peut activer davantage le stress oxydatif et nitrosatif dominé par le NO sur la base du déséquilibre du métabolisme de l'arginine, induire une accumulation de ROS mitochondriaux, une dépolarisation membranaire et un déséquilibre dynamique, inhiber la prolifération et la migration des cellules du cancer colorectal, révélant ainsi un mécanisme synergique anticancéreux ciblant le métabolisme et les mitochondries.
