L’objectif est d’étudier les variations des composants chimiques avant et après la carbonisation du Gardenia afin de fournir un soutien de données pour la recherche sur la base matérielle de la carbonisation du Gardenia. La méthode utilise la chromatographie liquide ultra-performante couplée à la spectrométrie de masse haute résolution quadrupolaire à piégeage orbital électrique (UPLC-Q-Orbitrap MS/MS) pour collecter des données en mode ion positif et négatif, combinée au logiciel Compound Discoverer 3.3 et aux bases de données en ligne pour une analyse complète des composants chimiques dans le Gardenia cru et carbonisé. Le logiciel SIMCA14.1 est utilisé pour l’analyse en composantes principales et l’analyse discriminante par moindres carrés partiels. Les métabolites secondaires et primaires différents avant et après la carbonisation sont sélectionnés selon des valeurs VIP supérieures à 1, et le site MetaboAnalyst est utilisé pour l’enrichissement des voies KEGG afin d’éclaircir le mécanisme de préparation. Au total, 185 composants ont été identifiés, y compris 96 métabolites secondaires et 89 métabolites primaires; classés en neuf catégories dominées par des glycosides terpènes éthers, flavonoïdes et terpènes, dont les schémas de fragmentation ont été analysés. En outre, une analyse statistique multivariée a été effectuée sur les métabolites secondaires et primaires, identifiant respectivement 70 et 59 métabolites différentiels. Les métabolites secondaires sont enrichis dans deux voies métaboliques, y compris le métabolisme des acides branchés C5 et la biosynthèse des flavonoïdes et flavonols, tandis que les métabolites primaires différemment enrichis concernent sept voies métaboliques, incluant le métabolisme de l’acide linoléique et de la tyrosine. La conclusion est que la composition chimique change de manière significative après la carbonisation du Gardenia, avec une réduction notable des teneurs en glycosides terpènes éthers tels que l’hydroxy-gardénoside, la crocine II, l’acide safranal et le jasminoside B, et une augmentation significative des teneurs en 4-hydroxycoumarine, acide genipodique, gentiopicroside et méthyl ester de gardenoside. On suppose que cette variation est liée à l’amélioration des effets de refroidissement du sang et d’arrêt du saignement induite par la carbonisation. Les composants clés identifiés fournissent une base matérielle pour expliquer les variations d’efficacité avant et après la carbonisation du Gardenia.
