L’objectif est d’étudier le rôle de la voie de signalisation Toll-like receptor 4 (TLR4) / facteur de différenciation myéloïde 88 (MyD88) / facteur nucléaire de transcription-κB (NF-κB) dans les lésions de la barrière muqueuse intestinale dans la colite ulcéreuse et le mécanisme d’intervention de la décoction de Paeonia. Méthodes : 60 rats SD ont été répartis en groupe contrôle, groupe modèle, groupe mésalazine (0,42 g·kg⁻¹) et groupes dose faible, moyenne et élevée de décoction de Paeonia (11,1, 22,2, 44,4 g·kg⁻¹). Le modèle de colite ulcéreuse a été induit par le TNBS (2,4,6-trinitrobenzènesulfonique). Après confirmation du modèle, les groupes ont reçu respectivement du sérum physiologique, de la mésalazine ou la décoction de Paeonia par gavage pendant 7 jours. La longueur et la masse du côlon ont été mesurées, la morphologie pathologique du tissu colique observée par coloration hématoxyline-éosine (HE), l’immunohistochimie a détecté l’expression des facteurs inflammatoires IL-8, COX-2, JAM-1, Claudin-1 dans les tissus, et le Western blot a analysé l’expression des protéines TLR4, MyD88, NF-κB. Résultats : comparé au groupe normal, le score d’indice d’activité de la maladie (DAI) a significativement augmenté (P<0,01) dans le groupe modèle, la longueur et la masse du côlon ont significativement diminué (P<0,01), l’expression des protéines JAM-1 et Claudin-1 a significativement diminué (P<0,01), tandis que les expressions d’IL-8, COX-2, TLR4, MyD88 et NF-κB p65 ont significativement augmenté (P<0,01). Par rapport au groupe modèle, les groupes traités ont montré une diminution significative du score DAI (P<0,05, P<0,01), une augmentation significative de la longueur et de la masse colique (P<0,05, P<0,01), une augmentation significative des protéines JAM-1 et Claudin-1 (P<0,01), et une diminution des niveaux d’IL-8, COX-2, TLR4, MyD88 et NF-κB p65 (P<0,01). Conclusion : la décoction de Paeonia peut réduire l’inflammation intestinale, améliorer les lésions muqueuses et protéger l’intégrité de la barrière intestinale, probablement en régulant la voie de signalisation TLR4/MyD88/NF-κB.
