肺線維症に対する伝統方剤由来のオリジナル新薬開発戦略

サマリー

「精密選別-知能構築-全連鎖検証」新薬開発体系は、筆者が提唱する肺線維症に対する伝統方剤由来の漢方単体薬開発の新戦略である。本稿では、組織指向抗線維化単体の知能選別、ターゲットおよび生源駆動単体構造最適化、候補薬の跨スケール評価などの観点から本戦略の主要内容を説明し、臨床で確かな効果がある方剤における組織標的新薬発見、抗線維化新薬創製および機序解析、候補薬の臨床転化と二次開発における応用を展開する。本戦略は、伝統漢方方剤に由来し、有効性が顕著で、構造が新規、標的が明確で、機序が明瞭、安全性が高い単体オリジナル新薬を開発し、肺線維症治療の重大かつ緊急な臨床ニーズを満たし、患者の「生存延長、生活の質向上」における飛躍的突破を実現し、自主的知的財産権を持つ中国独自の革新的医薬品開発に新たな戦略と新たなパラダイムを提供することを目的とする。
