El objetivo es investigar el efecto del alcaloide de raíz medicinal sobre los metabolitos endógenos y las vías metabólicas en ratones con colitis ulcerosa. Método: utilizando un modelo de enfermedad de colitis ulcerosa en ratones inducido por 3 % de dextrano sulfato de sodio (DSS), se administraron diferentes dosis orales del alcaloide de raíz medicinal a los ratones. Se observó el efecto terapéutico mediante el peso corporal, la longitud del colon, el índice de actividad de la enfermedad (DAI) y la histopatología del tejido del colon; mediante cromatografía líquida ultraeficiente acoplada a espectrometría de masas de tiempo de vuelo con cuadrupolo (UPLC-Q-TOF/MS) se midieron los niveles de metabolitos en suero y heces de los ratones, combinando métodos metabolómicos para seleccionar metabolitos diferenciales y examinar el posible mecanismo terapéutico del alcaloide en el tratamiento de la colitis ulcerosa inducida por DSS en ratones. Resultados: tras la intervención con el alcaloide, comparado con el grupo modelo, el DAI de los ratones disminuyó significativamente, la longitud del colon se restauró y la histopatología del colon mejoró de manera notable. El estudio metabolómico mostró 13 metabolitos diferenciales en el suero y 8 en las heces, y el análisis de enriquecimiento de vías identificó 3 rutas metabólicas principales potenciales: biosíntesis de ácidos grasos insaturados, biosíntesis de fenilalanina, tirosina y triptófano, así como metabolismo de fenilalanina. Conclusión: el alcaloide de raíz medicinal posiblemente ejerce su acción terapéutica en la colitis ulcerosa mediante su participación en rutas metabólicas como la síntesis y metabolismo de aminoácidos y biosíntesis de ácidos grasos insaturados.
