Este estudio tiene como objetivo investigar el mecanismo de acción de los antidepresivos en la activación de células gliales en el cerebro de ratones después de la separación de la madre y la descendencia y el doble impacto. En comparación con el grupo NC, los ratones del grupo MRS mostraron comportamiento depresivo, los niveles de neurotransmisores en el hipocampo disminuyeron significativamente (P<0.05, P<0.01), la proteína Iba-1 aumentó (P<0.01), los niveles de proteínas JAK2 y STAT3 aumentaron (P<0.05), los CD68, CD11b, IL-1β, JAK2, STAT3 mRNA aumentaron significativamente (P<0.01), los niveles de IL-10 mRNA disminuyeron significativamente (P<0.01). En comparación con el grupo MRS, los grupos XSF y minociclina mostraron una mejora en el comportamiento depresivo, el contenido de neurotransmisores en el hipocampo de los ratones de los grupos XSF y minociclina aumentó significativamente (P<0.05, P<0.01), el nivel de Iba-1 disminuyó significativamente (P<0.01), los niveles de proteínas JAK2 y STAT3 disminuyeron (P<0.05), los niveles de mRNA de CD68, CD11b, JAK2, STAT3 y IL-1β disminuyeron significativamente (P<0.05, P<0.01), los niveles de mRNA de IL-10 aumentaron significativamente (P<0.05, P<0.01). Conclusión: Los antidepresivos pueden aliviar el comportamiento depresivo de los ratones expuestos a la separación de la madre y la descendencia después de un doble impacto, y es posible que el mecanismo de alivio de la activación de las células gliales del hipocampo por los antidepresivos esté relacionado con la vía JAK2/STAT3.
