El objetivo del estudio fue estudiar el mecanismo de acción de las gotas de corazón de cerezo silvestre mejora la disfunción de las células musculares lisas vasculares, alivia la transdiferenciación y migración y mejora la señal trastornada inducida por la angiotensina II (Ang II). Los métodos utilizados para establecer un modelo de migración de células musculares lisas vasculares Ang II utilizan un método efectivo para Ang II y están establecidos como grupo de control (Control), grupo de modelo (Modelo) y grupo de intervención (STDP) (5%, 10%, 20%) durante 24 horas. A través de pruebas de inmunofluorescencia para detectar los niveles de proteína alfa del músculo liso vascular (α-SMA) y metaloproteinasa-3 (MMP-2), así como pruebas de proliferación y viabilidad celular (CCK-8) y pruebas de tinción EdU para verificar la migración de células musculares lisas vasculares. En comparación con el grupo modelo, los resultados de las pruebas de inmunofluorescencia mostraron que el STDP puede aumentar significativamente los niveles de α-SMA y suprimir claramente los niveles de MMP-2 (P <0.05) ; Los resultados de las pruebas CCK-8 y EdU mostraron que el STDP puede suprimir significativamente la migración de células musculares lisas vasculares (P <0.05) ; Los resultados de las pruebas de raspadura mostraron que el STDP puede suprimir significativamente la migración de células musculares lisas vasculares (P <0.05) ; Los resultados de Western blot mostraron que el STDP puede aumentar significativamente los niveles de ACE2, AT2, MAS1 y suprimir significativamente los niveles de AT1 (P <0.05), suprimir significativamente los niveles de PCNA, Col I, MMP-9, Rock1, Rock2, SRF (P <0.05) . En comparación con el grupo STDP, el inhibidor ACE2 revirtió el efecto del STDP. Conclusiones: el STDP puede inhibir la transdiferenciación y migración de células musculares lisas vasculares, regular la expresión de proteínas relacionadas con la proliferación y migración celular, mejorar los trastornos de la función de las células musculares lisas vasculares, cuyo mecanismo está relacionado con el aumento de la expresión de proteínas de la vía ACE2-AT2/MAS y la supresión de la expresión de proteínas de la vía de señalización AT1/Rock.
