El objetivo de este estudio fue usar cromatografía líquida de ultra alta resolución acoplada a espectrometría de masas en tándem de tiempo de vuelo (UPLC-Q-TOF-MS/MS) combinada con farmacología de red y acoplamiento molecular para investigar el efecto terapéutico de la decocción Erdiao Sitan en ratas eczematosas de tipo calor-humedad y su mecanismo de acción. El método consistió en el establecimiento de un modelo de ratas eczematosas de tipo calor-humedad inducido por el clorobenceno trinitro (DNCB) combinado con la intervención de una cámara climática artificial, las ratas fueron asignadas al azar en un grupo normal, un grupo modelo, un grupo de dosis baja y un grupo de dosis alta de Erdiao Sitan (3,40 y 6,80 g·kg-1), así como un grupo de acetato de prednisolona (2,51 mg·kg-1), con 8 ratas por grupo y un total de 46 ratas, incluyendo 6 ratas en el experimento de suero medicamentoso. Se realizó un análisis químico del suero de las ratas después de la administración de medicamentos mediante UPLC-Q-TOF-MS/MS, junto con la farmacología de red para predecir los componentes clave, los blancos centrales y las vías de señalización, y la experimentación de acoplamiento molecular en el sitio web CB-Dock2. El efecto terapéutico de la decocción Erdiao Sitan sobre el eczema de tipo calor-humedad se estudió combinando indicadores epigenéticos con la tinción de tejidos patológicos; los niveles séricos de gastrina (GAS), interleucina-4 (IL-4) e interleucina-13 (IL-13) fueron evaluados por ELISA; los niveles de expresión proteica de la interleucina-6 (IL-6), la Janus quinasa 1 (JAK1), la JAK1 fosforilada (p-JAK1), el transductor de señales y activador de la transcripción 3 (STAT3) y la STAT3 fosforilada (p-STAT3) fueron detectados por Western blot. Los tejidos de lesiones cutáneas mostraron una queratinización incompleta, acumulación de células inflamatorias, una separación poco clara entre la epidermis y la dermis, una mucosa gástrica adelgazada y perturbada, así como una acumulación evidente de células inflamatorias; los niveles séricos de GAS, de IL-4 y de IL-13 disminuyeron significativamente (P<0,05, P<0,01), mientras que los niveles de expresión proteica de IL-6, de JAK1, de p-JAK1 y de p-STAT3 en los tejidos de lesiones cutáneas aumentaron significativamente (P<0,05, P<0,01); en comparación con el grupo modelo, las ratas de los grupos tratados mostraban un estado mental estable, heces formadas, un área anal limpia, una epidermis esencialmente normal con solo una pequeña cantidad de costras en escamas, aumento de peso, disminución de la temperatura rectal y del consumo de agua, así como una disminución del bazo, del timo y del índice gástrico (P<0,05, P<0,01), una disminución del grosor de la epidermis, una separación clara entre la epidermis y la dermis, aunque la queratinización excesiva y la acumulación de células inflamatorias aún eran visibles, un aumento del grosor de la mucosa gástrica con diferentes grados de recuperación. Los niveles séricos de GAS, de IL-4 y de IL-13 en las ratas de los grupos tratados aumentaron en diferente medida, mientras que los niveles de expresión proteica de IL-6, de JAK1, de p-JAK1 y de p-STAT3 en los tejidos de lesiones cutáneas disminuyeron significativamente (P<0,05, P<0,01). En conclusión, la decocción Erdiao Sitan podría ejercer un efecto terapéutico en las ratas eczematosas de tipo calor-humedad mediante la regulación de la vía de señalización JAK/STAT.
