El objetivo de este estudio fue investigar los efectos y el mecanismo de la acción preventiva y terapéutica de Dendrobium officinale en la enfermedad del hígado graso no alcohólico (EHGNA) mediante farmacología de redes y experimentos en animales. Se empleó cromatografía líquida de alta resolución acoplada a un espectrómetro de masas cuadrupolo-orbitrap de alta resolución (UPLC-QE-HF-MS/MS) para analizar e identificar los componentes en sangre de Dendrobium officinale; se utilizaron métodos de farmacología de redes y de acoplamiento molecular para identificar los componentes farmacológicamente activos y los posibles blancos terapéuticos de Dendrobium officinale en el EHGNA; se empleó una dieta alta en grasas para inducir un modelo de rata de EHGNA, y se utilizó un kit de reactivos bioquímicos para medir los niveles de lípidos sanguíneos, lípidos hepáticos y la expresión de la función hepática en las ratas, así como tinción de Hematoxilina y Eosina (HE) y tinción con rojo aceite para observar los cambios patológicos en el hígado de las ratas, y reacción en cadena de la polimerasa cuantitativa en tiempo real (PCR) para validar los posibles blancos identificados por farmacología de redes. Los resultados mostraron que en Dendrobium officinale se identificaron un total de 13 componentes en sangre como la berberina, la berberastina, la prenilina I, etc.; farmacología de redes identificó un total de 14 posibles blancos, entre ellos el factor de transcripción forkhead box protein O1 (FoxO1), el receptor del factor de crecimiento epidérmico (EGFR), el receptor de la insulina tipo 1 (IGF1R), afectando vías tales como la señalización del receptor de productos finales de la glicación avanzada (AGE)/receptor para la AGE (RAGE), metabolismo lipídico y aterosclerosis, resistencia a la insulina y señalización de FoxO. Los resultados de los experimentos en animales mostraron que se logró inducir un modelo de rata de EHGNA, y que el tratamiento preventivo y terapéutico con Dendrobium officinale resultó en la normalización de los niveles de lípidos sanguíneos, lípidos hepáticos, y la función hepática, mejorando significativamente la acumulación de lípidos y las lesiones patológicas en el hígado, así como la disminución significativa de la expresión de ARNm de FoxO1 en el hígado (P<0.05), y el aumento significativo de la expresión de ARNm de PI3 quinasa (PI3K), proteína quinasa B (Akt), EGFR y IGF-1R (P<0.05). En conclusión, Dendrobium officinale tiene efectos preventivos y terapéuticos en ratas con EHGNA, y su mecanismo de acción puede estar relacionado con la regulación de los blancos IGF1R y EGFR, y la activación de la vía de Pi3 quinasa/Akt/FoxO.
