El objetivo de este estudio fue investigar el efecto del Sanjoin negro en la función cognitiva en la enfermedad de Alzheimer (EA) al discutir el mecanismo de apoptosis celular mediado por la vía de señalización fosfoinositol 3-quinasa (PI3K) / proteína quinasa B (Akt) / factor de transcripción nuclear-κB (NF-κB). Métodos: El experimento in vivo (animales) dividió aleatoriamente ratas macho SD de 4 meses en grupo control, grupo de cirugía ficticia, grupo modelo, grupo de clorhidrato de donepezilo (0,45 mg · kg-1) y grupos de dosis alta, media y baja de Sanjoin negro (15,30, 7,65, 3,82 g · kg-1), cada uno compuesto por 10 ratas. En los grupos de cirugía ficticia, modelo, clorhidrato de donepezilo y en los grupos de dosis alta, media y baja de Sanjoin negro, se estableció la enfermedad de Alzheimer (EA) inyectando bilateralmente una solución de péptido β-amiloide 1-42 (Aβ1-42) en el hipocampo. Una vez al día durante 42 días, las ratas recibieron tratamiento por gavage. Las ratas fueron sometidas a una prueba de laberinto de agua de Morris para evaluar sus habilidades de aprendizaje y memoria, una tinción de hematoxilina-eosina (HE) para observar cambios patológicos en la región del hipocampo, un ensayo inmunoenzimático (ELISA) para medir los niveles de caspasa-3, linfoma B de células-B-2 (Bcl-2) y proteína X asociada a Bcl-2 (Bax), y un Western blot para determinar los niveles de expresión de las proteínas PI3K, Akt y NF-κB. Para el experimento in vitro (células), un modelo de enfermedad de Alzheimer se estableció cultivando células PC12 con péptido β-amiloide 1-42 (Aβ1-42), y la viabilidad celular se evaluó utilizando el kit de ensayo de actividad de proliferación celular (CCK-8) y la apoptosis celular se midió mediante citometría de flujo (CF). Conclusiones: Los resultados de los experimentos in vivo y in vitro mostraron que el Sanjoin negro podría mejorar las habilidades de aprendizaje y memoria de las ratas con enfermedad de Alzheimer (EA) a través del mecanismo de apoptosis celular, al tiempo que aumenta la vitalidad celular de los modelos de enfermedad de Alzheimer (EA) y reduce la tasa de apoptosis celular, probablemente regulando la vía de señalización de PI3K/Akt/NF-κB.
