Objetivo: La angélica, como materia prima medicinal y alimentaria comúnmente utilizada, presenta frecuentes casos de adulteración en el mercado, lo que afecta gravemente la eficacia clínica de las preparaciones. Actualmente, las técnicas para la identificación cualitativa de adulterantes están bastante maduras, pero los métodos cuantitativos para la detección de adulteración en productos procesados aún están ausentes. Este estudio utiliza la angélica y su principal especie cercana fácilmente confundible “angélica terrestre” (angélica coreana) como ejemplo para llevar a cabo una investigación de detección cuantitativa, con el objetivo de establecer un método de detección molecular cuantitativa de hierbas (Herb-Q) para la angélica y sus productos procesados, proporcionando un modelo para establecer técnicas de detección cuantitativa para materias primas de angélica y productos de salud relacionados. El método se basa en la selección de sitios específicos de polimorfismo de nucleótido único (SNP) en los genomas de los cloroplastos de la angélica y la angélica coreana. Se eligieron sitios específicos de la angélica para estudiar la metodología cuantitativa mediante mezclas de polvos de hierbas con diferentes proporciones de adulteración (linealidad, límite cuantitativo, límite de detección y reproducibilidad). Se validó la precisión del método Herb-Q en una preparación de polvo para aliviar el dolor con efecto activador sanguíneo con tres proporciones diferentes de adulteración (15%, 25%, 35%). Resultados: mediante la comparación de 123 secuencias del genoma del cloroplasto del género angélica, basándose en la conservación intraespecífica, especificidad interespecífica y los requisitos del secuenciamiento por pirosecuenciación de alto rendimiento, se identificaron los sitios 9674 (A/G) en la secuencia NC_042826.1 y 38592 (T/C) en NC_029393.1 como sitios moleculares específicos para la angélica y la angélica coreana, respectivamente. Se seleccionó el sitio específico 9674 (A/G) de la angélica para establecer la relación lineal del método Herb-Q con un valor R² de 0.9974 (>0.99), mostrando buena linealidad. Los límites cuantitativos y de detección fueron del 2.0%, con buena reproducibilidad (desviación estándar relativa (RSD) <2.0%). El sistema cuantitativo basado en el método Herb-Q detectó la cantidad de adulteración de angélica coreana en el polvo para aliviar el dolor activador sanguíneo, con un error promedio de las repeticiones moleculares triples del 1.3%. Conclusión: este estudio demuestra que el método de detección cuantitativa Herb-Q basado en el sitio 9674 (A/G) del genoma del cloroplasto NC_042826.1 de la angélica tiene buena aplicabilidad, objetividad y precisión para la angélica y la “angélica terrestre” y sus productos procesados, proporcionando soporte técnico para la detección cuantitativa de productos derivados de angélica en el mercado como preparaciones de medicina tradicional china, suplementos dietéticos y productos de salud.
