Abstract
Objective:To establish a HPLC method for simultaneous determination of rutin
forsythoside A and phillyrin of Forsythiae Fructus and its extract.Method: The procedure of HPLC-PDA was performed on the chromatographic column of Sunfire C18(4.6 mm×150 mm
5 μm)
and the mobile phase in the gradient elution(0 min
14%A;15 min
18%A;40 min
30%A) was acetonitrile(A)-0.2% acetic acid solution(B).The flow rate was 1.0 mL · min-1;the temperature was kept at 40 ℃
and the detection wavelength was set at 354
329 nm and 277 nm respectively.Result: Rutin
forsythoside A and phillyrin were linear in the range of 0.1-2.0
0.12-2.4 μg and 0.05-1.0 μg respectively.The average recoveries were 98.63%
98.97% and 102.18% respectively(RSD 7.94%
0.93% and 1.63%).Conclusion: The method is accurate with higher repeatability
which can be used in the quality control of Forsythiae Fructus and its extract.
