以0.4mL/min的流速进行梯度洗脱
检测波长238 nm
柱温30℃
。结果 在21min内得到金银花药材的指纹图谱
对其中5个色谱峰进行了初步归属
并对14批药材样品进行了分析
其相似度为0.915~0.987。结论UHPLC指纹图谱方法较HPLC大大缩短了分析时间
可用于金银花药材的质量评价。
Abstract
Objective To develop an UHPLC method of determining the characteristic chromatographic profile of Lonicera japonica Thunb for controlling the drug quality quickly and accurately. Methods The analysis was performed on an Agilent C18 column(50 mm?2.1 mm
1.8μm) with acetonitrile-0.2%phosphoric acid as mobile phase in gradient elution mode. The flow rate was 0.4 ml.min-1and the injection volume was 2.0μl with the detection wavelength of238 nm.The column temperature was set at 30℃. Results The fingerprint of Lonicera japonica Thunb was obtained within 21min and five peaks in the fingerprint were designed.14 samples were analyzedand the similarities were 0.915-0.987. Conclusion the UHPLC system enjoys the merits of a higher speed
sensitivity
and resolution
It can be used to control the quality of Lonicera japonica Thunb.
