Effect of Sarcandra Glabra on Apoptosis of the Parotid Glands in Miniature Pig Irradiated by Co γ Rays
Objective:To explore the influence and mechanism of Sarcandra Glabra on apoptosis and expression of B-cell lymphoma-2(bcl-2)/bcl-2 associated x protein(bax) of parotid glands in the miniature pig irradiated by 60Co γ rays. Method: Sixty miniature pigs were randomly divided into normal control group
radiation control group and medicine radiation group(0.3 g·kg-1)
then each group was randomly divided into parallel group numbered a

c and d. Sarcandra Glabra Herb was given to the corresponding groups from a week before exposure to the irradiation until the parotid glands were removed. Each pig in the radiation control group and the medicine radiation group was received 60Co gamma-rays with parotid gland(total radiation dose:30 Gy)
which in the normal control group was not received rays.On the 1th day
10th day
40th day and 90th day after irradiation
the parotid glands of miniature pigs were removed for experiment. The expression of bcl-2/bax was detected by Western blotting
and the apoptosis was detected by TUNEL method. Result: The expression of bcl-2 in medicine radiation group was incresed compared with radiation control group on every time point. The expression of bax in three groups decreased. The apoptosis rate in the radiation control group and the medicine radiation group was higher than the normal control group on every time point(P<0.05). In radiation to control group
the minimum of expression of bcl-2 was in 10th day
and the maximumwas in 90th day. The highest expression of apoptosis is in 10th day
then decreased with time(P<0.05). Conclusion: Sarcandra Glabra diminished the expression of bax and apoptosis
while augmented the expression of bcl-2. The mechanism of Sarcandra Glabra reducing apoptosis may be related to augmenting the expression of bcl-2 and diminishing the expression of bax.
