Objective:To observe digestive system disease of rats after oral administration by Gamboges
compare the toxicity on rat intestinal epithelial cell line IEC-6 of the gamboges and the processed products and discuss the possible mechanism of detoxication. Method: Single administration of the gamboges
the gamboges processed with water and the gambogic acid was carried out
and the heart
liver
kidney
esophagus
stomach
duodenum
jejunum
ileum and colon were collected to observe digestive system change. The method of MTT was adopted to detect the cell proliferation of intestinal epithelial cell line IEC-6; the shapes of cells were observed by inverted microscope. Result: The results of the pathological section showed that there was no visible toxicity in the liver
kidney
heart
stomach and esophagus. But there was toxicity in duodenum and jejunum
showed the intestinal villus dropsical. All the experimental drugs could cause the toxicities in intestinal tract
and the gamboges were harmer than the other two. Cell toxicity results showed that the gamboges could obviously inhibit the cell proliferation when compared with control group (P<0.01)
the cell toxicities of produced products were obviously lower than gamboges (P<0.01). Conclusion: The results of the pathological section and the cell toxicity both showed that the toxicity of gamboges was obviously reduced after processing
which provides a basis for further exploration of detoxication mechanism of gamboges and the processed products.
