目的: 优选复方一枝蒿速释微丸的提取工艺。 方法: 采用正交试验设计
以(R
S)-告依春含量和浸膏得率的综合评分为指标
优选板蓝根和大青叶水提取工艺;以一枝蒿酮酸含量和浸膏得率为评价指标
优选一枝蒿醇提取工艺。采用体外抗病毒药效学试验对优选工艺与原工艺进行对比研究。 结果: 板蓝根、大青叶的最佳提取工艺为板蓝根加8倍量水浸泡48 h
加入大青叶
提取3次
每次0.5 h;一枝蒿最佳提取工艺为加8倍量70%乙醇提取2次
每次1.0 h。体外药效学试验表明
优选的提取工艺抗病毒作用效果优于原工艺。 结论: 优选的提取工艺稳定可行
能较好保留药效成分。
Abstract
Objective:To optimize extraction technology of compound Yizhihao immediate-release pellets. Method: With composite score of the content of (R
S)-epigoitrin and extract yield as index
water extraction technology of Radix Isatidis and Folium Isatidis were optimized by orthogonal test;With the content of rupestonic acid and extract yield as indexes
ethanol extraction technology of Artemisia rupetris was optimized by orthogonal test.Comparison of optimum extraction technology and original extraction technology was investigated by in vitro antiviral pharmacodynamic test. Result: Optimum extraction technology was as following:Radix Isatidis was soaked for 48 h with 8 times the amount of water
then added Folium Isatidis
extracted 3 times with 0.5 h per time; A. rupetris was extracted 2 times with 8 times the amount of 70% ethanol
1.0 h per time.It showed that antiviral activity of optimized extraction technology was better than original extraction technology by in vitro pharmacodynamic test. Conclusion: This optimized technology was stable and feasible
