Effect of Dihuang Yinzi on Expression of Bax,Bcl-2 and Caspase-3 Proteins in Rat Model of Cerebral Ischemia Reperfusion
Objective:To explore the effect of Dihuang Yinzi on the expression of Bax
Bcl-2 and Caspase-3 proteins and protective cerebral mechanism after cerebral ischemia reperfusion. Method: Sixty SD rats were selected and randomized into six groups: sham group
model group and high
medium
low dose groups of Dihuang Yinzi(32

8 g·kg-1) and nimodipine positive drug group(1 mg·kg-1)
each involved ten rats. The cerebral ischemia/reperfusion model was established. The expression of Bax
Bcl-2 and Caspase-3 proteins were detected 120 min after cerebral ischemia/reperfusion treated by Dihuang Yinzi. Result: The expression of Bax and Caspase-3 proteins in the models were significantly higher than those in the sham group (P<0.05). The expression of Bcl-2 proteins in the model group were significantly lower than those in the sham group (P<0.05). While the expression of Bax and Caspase-3 proteins in the treatment group were significantly lower than those in the model group (P<0.05)
the expression of Bcl-2 proteins in the treatment group were significantly higher than those in the model group (P<0.05). Conclusion: Dihuang Yinzi can upregulate the expression of Bcl-2 protein and downregulate the expression of Bax and Caspase-3 proteins. Dihuang Yinzi may protect the cerebral injury induced by ischemia/reperfusion through inhibiting neuron apoptosis.
