Optimization of Purification Technology of Total Lignans from in Conjunction with Macroporous Resin and Aluminum Oxide Column
Objective:To optimize purification technology of total lagnans from Schisandra sphenanthera extract by macroporous resin combined with aluminum oxide. Method: With composite score of impurity removal rate and transfer rate of schisantherin A as index
effect of the amount of resin
ethanol concentration and amount on macroporous resin purification technology of total lignans was investigated by L9(34) orthogonal test;alumina adsorption purifying technology of total lignans from S. sphenanthera was investigated by single factor test.The content of total lignans was determined UV spectrophotometry
HPLC was used to determine the contents of schisantherin A and deoxyschizandrin. Result: Optimum purification conditions of total ligans in S. sphenanthera were as follows:took 2/3-4/3 times the amount of LSA-7 macroporous resin
eluted with 6 BV 85% ethanol
ratio of aluminum oxide and crude drug 1∶1. Conclusion: This optimized purification process was feasible
stable
reasonable and suitable for industrial production.
