摘要

目的: 探讨抵当汤对高血压脑出血模型大鼠脑组织低氧诱导因子-1α(HIF-1α)表达的影响。方法: 将健康雄性SD大鼠随机分为假手术组、模型组、抵当汤低剂量组、中剂量组、高剂量组

采用腹腔注射垂体后叶注射液复制大鼠高血压模型

脑内注射自体血复制脑出血模型

给予低、中、高(3.78

15.12 g·kg-1)3个剂量的抵当汤灌胃

治疗7 d

分别用免疫组织化学法、蛋白免疫印迹法、聚合酶联反应法检测大鼠脑组织HIF-1α的表达。结果: 抵当汤各治疗组HIF-1α阳性细胞、HIF-1α蛋白、HIF-1α mRNA表达均显著高于模型组(P<0.05)

其中低剂量组尤为明显。结论: 抵当汤药理作用与上调大鼠脑组织HIF-1α表达密切相关。

Abstract

Objective: To investigate the ameliorating effect of Didang decoction on hypoxia-inducible factor-1α(HIF-1α) in hypertensive cerebral hemorrhage model rats. Method: Healthy male Sprague-Dawsley rats were randomly allotted to five groups as following: sham operation group

model group

Didang decoction low-dose group

middle-dose group

high-dose group

respectively. The hypertension was reproduced by intraperitoneal injection posterior pituitary injection

and then intracerebral injection autoblood was used to establish cerebral hemorrhage.Didang decoction was continuously treated for 7 days. The HIF-1α was detected by immunohistochemistry

Western blotting

and polymerase chain reaction(PCR). Result: The HIF-1α positive cells

HIF-1α protein content

HIF-1α mRNA expression significantly enhanced in Didang decoction treatment groups

particularly in the low-dose group (P<0.05). Conclusion: Up-regulated the expression of HIF-1α maybe involved in effects of Didang decoction.
