Simultaneous Determination of Quercetin and Kaempferol in Leaf of by HPLC
Objective:To establish an analytical method on simultaneous determination of quercetin and kaempferol in leaf of Ligustrum sinense. Method: The analysis was carried out on a column eluted with a mobile phase of methanol-0.4% phosphoric acid solution (55∶45). The flow rate was 1.0 mL·min-1; the detection wavelength was set at 360 nm. Result: The calibration curves of quercetin and kaempferol were linear in the ranges of 0.058-0.525 μg and 0.203-0.712 μg
and the average recoveries were 103.0% and 107.6% (n=6)
respectively. Conclusion: The method is accurate
reliable and repeatable with stable result. It can be used as a method of quantitative determination of quercetin and kaempferol in the leaf of L. sinense
