Optimization of Extraction and Precipitation Technology for Liuyueqing Polysaccharide Capsule by Orthogonal Design
Objective: To optimize extraction and precipitation technology conditions of Liuyueqing polysaccharide capsule. Method: With the content of polysaccharide as index
orthogonal test was adopted to investigate influence of solid-liquid ratio
decoction times and decoction time on extraction technology of Liuyueqing polysaccharide capsule;With the content and yield of polysaccharide as comprehensive evaluation index
alcohol precipitation time and times
the amount of ethanol and alcohol precipitation concentration were chosen as factors
alcohol precipitation technology was optimized by orthogonal test.The content of total polysaccharides was determined by phenol-concentrated sulfuric acid method. Result: Optimal extraction technology of Liuyueqing polysaccharide capsule was as follows:decocted three times with 10-fold water
2 hours each time.Optimum alcohol precipitation conditions were:filtered extraction liquid
mixed and concentrated to 0.5 g ·mL-1
and then precipitated one time with 7-fold 95% ethanol for 12 hours.Under these conditions
purity of polysaccharide was up to 71.05%
yield of it was 6.83%. Conclusion: This optimized extraction and precipitation procedure was proved to be stable and reliable
it could provide experimental basis for Liuyueqing polysaccharide capsule.
