Objective:To explore the factors influencing the germ-free germination of seeds of Paris polyphylla var.Yunnanensis and the plant morphogenesis. Method: The effects of culture medium
culture temperature and light intensity on the germ-free germination of seeds of P. polyphylla var.Yunnanensis were studied
and the plant morphogenesis during the course of germ-free germination of seeds of P. polyphylla var.Yunnanensis was observed at the same time. Result: The best culture medium for the germ-free germination of seeds of P. polyphylla var.Yunnanensis was Knudson C
and seed protrusion rate and germination rate were 91.0% and 76.7%
respectively. B5 culture medium was suitable for the growth of radicle
and the radicle length is (4.567±0.251 7)cm. The seed protrusion rate was higher than 81.7% when culture temperatures were changed with 15 ℃/25 ℃ and 20 ℃/25 ℃. Culture by changed temperatures with 15/25 ℃ could promote seed germination
and the seed germination rate was as high as 55.3%. Some embryos could break through the seed coat and grow into cotyledon. Culture by changed temperatures with 20 ℃/25 ℃ is suitable for the growth of radicle
and the radicle length is (3.667±0.208 2) cm. Weak light irradiation of 750-1 500 lx is suitable for the seed germination
the seed protrusion rate and germination rate were more than 86.7% and 58.3%
respectively. Under 750 1x weak light
the radicle grow rapidly
and the radicle length could be up to(4.867±0.152 8) cm. 3 000 lx strong light irradiation exhibited some inhibition for the seed germination and radicle growth. Conclusion: This study established the germ-free germination culture system of radicle and embryo for the seeds of P. polyphylla var.Yunnanensis
which could lay the foundation for further study the growth
physiological and biochemical change of radicle and embryo of P. polyphylla var.Yunnanensis under aseptic condition.
