Effect and Mechanism of Wogonin on NF-B Nuclear Translocation and Expression in NR8383 Cells Infected by Influenza Virus
Objective: To investigate the effect of wogonin on the nuclear translocation and expression of nuclear factor-kappa B(NF-κB) in NR8383 cells infected by influenza virus and the relationship with Toll like receptor 7(TLR7) mediated myeloid differentiation factor 88(MyD88)-dependent signaling pathway. Method: After 1 h adsorption of influenza virus
the medium containing 0.016 g · L-1 wogonin was added to NR8383 cells in wells. At 24 h after drug application
reverse transcription polymerase chain reaction(RT-PCR) was used to detect the mRNA level of MyD88
NF-κB p65 and TLR7. At 4

immunocytochemistry was used to detect nuclear translocation of NF-κB p65 in the cells and perform semi-quantity analysis. At 24 h after wogonin application
Western blot was used to detect the expression of NF-κB p65 protein. Result: Wogonin reduced the mRNA level of TLR7

and NF-κB p65(P<0.05

P<0.05)and inhibited the nuclear translocation and expression of NF-κB p65(P<0.01). Conclusion: Wogonin inhibited the nuclear translocation and expression of NF-κB by repressing TLR7 mediated MyD88-dependent signaling pathway
and alleviated the inflammatory reaction in influenza virus infection.
