Objective: To establish a HPLC assay method for trollioside and determine its content in the flowers of Trollius chinensis commercially available in China. Method: High performance liquid chromatography was employed using kromasil C18 ODS column (4.6 mm×250 mm
5 μm)
and mobile phase comprising acetonitrile-0.5% acetic acid solution (gradient:0-30 min
2%-100% A) at a flow rate of 1.0 mL·min-1 and detecting wavelength of 258 nm. Result: A good linear relationship was demonstrated for trollioside within 0.115 8-1.042 2 μg and the content of trollioside in nine batches of the flowers of T. chinensis commercially available in China was within 0-0.058%. Conclusion: This assay method is good in terms of precision
accuracy and repeatability and can be used for determination of the content of trollioside.
