Effect of Chaipu Decoction on Expression of Bcl-2 in Lung Tissue in Asthma Guinea Pigs
Objective: To observe the effect of Chaipu decoction on expression of Bcl-2 in lung tissue in asthma guinea pigs. Method: Forty guinea pigs were randomized into normal control
model
dexamethasone (0.001 g·kg-1) and chaipu decoction (0.35 g·kg-1) groups with 10 guinea pigs each group. Asthma mouse model was prepared by injecting ovalbumin (OVA) into the abdominal cavity and inhalation of aerosol
and normal group
was given saline water. Each group was given relevant medicines by oral administration before 30 mins of suscitation corresponding
once every other day
for 2 weeks. All animals were sacrificed after the last stimulation. The white blood cell(WBC) count
the number of eosinophil(EOS) and the percentage of EOS in bronchoalveolar lavage fluid (BALF) of guinea pigs and the number of EOS in whole blood were examined. Pulmonary pathological changes were observed by HE stain. Bcl-2 protein expression was examined by immunohistochemistry. And Bcl-2 mRNA expression was detected with in situ hybridization. Result: Compared with model group
chaipu decoction group could significantly reduce the EOS count in whole blood
the WBC count
the number of EOS and the percentage of EOS in BALF(P<0.05); and the expression level of Bcl-2 mRNA and protein in the lung tissue were significantly decreased(P<0.05). In Chaipu decoction group
compared with dexamethasone group
the significant difference was not found. Conclusion: Chaipu decoction for asthma was able to reduce the proportion of EOS in BALF
alleviate the inflammation of airway and lung tissue of the asthmatic guinea pigs. Chaipu decoction for asthma could reduce the Bcl-2 mRNA and protein expression level.
