Abstract
Objective: To develop an HPLC method for the determination of ellagic acid tablets in rabbits plasma and to study the pharmacokinetics of ellagic acid tablets in rabbits. Method: An Agilent C18 (4.6 mm×250 mm
5 μm) column was adopted
the mobile phase was a mixture of acetonitrile and 0.1% phosphoric acid solution (23∶77) at the flow rate of 0.8 mL·min -1
the detection wavelength was set at 254 nm and the column temperature was controlled at 30 ℃. Result: The retention time of ellagic acid was observed at 8.5 min. The linear range of calibration curve was within drug concentrations of 0.013-1.635 mg·L-1 (r=0.998 9). The quantitation limit was 0.010 mg·L-1. The recoveries of three levels were more than 95%. The plasma ellagic acid concentration-time curve could be evaluated by the two compartment model. Conclusion: The developed HPLC method for the pharmacokinetic study of ellagic acid tablets is precise.
