摘要

目的: 研究丹皮酚和芍药苷配伍对实验性心肌缺血大鼠心肌损伤的保护作用。方法: 采用在体结扎大鼠心脏冠状动脉前降支法建立大鼠心肌缺血损伤模型。实验分为假手术组、模型组、阳性药对照组(复方丹参片840 mg ·kg-1)、丹芍配伍高剂量组(丹皮酚100 mg ·kg-1+芍药苷25 mg ·kg-1)、丹芍配伍中剂量组(丹皮酚50 mg ·kg-1+芍药苷12.5 mg ·kg-1)、丹芍配伍低剂量组(丹皮酚25 mg ·kg-1+芍药苷6.25 mg ·kg-1)。ig给药容积10 mL ·kg-1

连续给药5 d

1次/d

假手术组和模型组ig等量生理盐水。观测大鼠心电图(ECG)

测定大鼠血清天门冬氨酸氨基转移酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)的活性

硝基四氮唑蓝(N-BT)染色测定心肌梗死面积。结果: 与梗死模型组比较

丹芍配伍高、中剂量组均可明显减轻冠脉结扎形成的大鼠急性心肌缺血性心电图的(ST段抬高及T波高耸)变化程度(P<0.05)

缩小心肌梗死面积(P<0.05或P<0.01)

同时能够降低血清中AST、CK、LDH的升高程度(P<0.05或P<0.01)。结论: 丹芍配伍对大鼠急性心肌缺血损伤具有明显保护作用。

Abstract

Objective: To investigate protective effects of the compatibility of paeonol and paeoniflorin on experimental myocardial ischemia in rats. Method: Using the method of ligation of left descending coronary artery the rat model of myocardial ischemia was established. Experiment group:sham group

paeonol 100 mg · kg-1+paeoniflorin 25 mg · kg-1

paeonol 50 mg · kg-1 +paeoniflorin 12.5 mg · kg-1

paeonol 25 mg · kg-1 +paeoniflorin 6.25 mg · kg-1. ig 10 mL · kg-1 for 5 d. Sham group and model group was given ig physiological saline. electrocardiogram(ECG)

transaminase(AST)

crealine kinase(CK)

lactate dehydrogenase(LDH) and the myocardial infarct size were measured. Result: Compared with the model group

the S-T segment and T waves elevation levels in myocardial ischemia rats in the medium dose and high dose groups of the compatibility of paeonol and paeoniflorin were obviously reduced(P<0.05 or P<0.01).The myocardial infarct sizes were decreased(P<0.05 or P<0.01). The content of AST

CK and LDH in rat serum were reduced. Conclusion: The compatibility of paeonol and paeoniflorin can effectively protect myocardial ischemic injury of rats.
