Determination of Content of Chlorogenic Acid and Bacalin in Yinhuang Granules by RP-HPLC
Objective: To establish an RP-HPLC method for determination of chlorogenic acid and bacalin in Yinhuang granules. Method: The analysis was performed on a Kromasil C18 column (4.6 mm×250 mm
5 μm) with acetonitrile and water containing 0.4% phosphoric acid as the mobile phase in gradient elution. The flow rate was set at 1.0 mL·min-1. The UV detector wavelength was at 325 nm
and the column temperature was at 30 ℃. Result: The linear ranges of chlorogenic acid and bacalin were 0.060 0-1.20 μg (r = 0.999 9) and 0.272-5.44 μg (r=0.999 9); the average recoveries of chlorogenic acid and bacalin were 99.5% and 97.9%
and RSD were 0.73% and 1.79% respectively. Conclusion: The method was simple
feasible
and it can be used for the quality control of Yinhuang granule.
