Optimization of Purification Technology for Alcohol Extract of K-CoxB-JN by Macroporous Resin
Objective: To optimize purification technology of total saponins
flavonoids and lignins from K-CoxB-JN by macroporous resin. Method: Taking the contents of total saponins
flavonoids and lignins as indexes
adsorption and desorption properties of 10 kinds macroporous resins were compared
based on these
maximum sample volume
sample flow rate and elution solvent were investigated by single-factor test. Result: D101 macroporous resin gave good static-dynamic and adsorption-desorption characteristics for all components
optimized purification technology was:crude drug concentration of alcohol extraction solution 0.5 g ·mL-1
herbs-wet resin 1: 4
adsorption flow rate 2 BV ·h-1
washed with 3 BV distilled water
4 BV of 70% ethanol as elution solvent.Under these conditions
purity of total saponins
total flavonoids and total lignins were enriched from 5.36%
0.3%
2.26% to 53.82%
3.01%
22.87%. Conclusion: This optimum purification technology of K-CoxB-JN was simple
stable and effective with good purification effect
